Nano-resolution X-ray microscopy has been demonstrated as a powerful technique for characterizing the evolution of battery electrodes under operating conditions with sensitivity to their morphology, compositional distribution and redox heterogeneity. In real-world batteries, the electrode could deform upon battery operation, causing challenges for the image registration which is necessary for several experimental modalities, e.g. XANES imaging. To address this challenge, this work develops a deep-learning-based method for automatic particle identification and tracking. This approach was not only able to facilitate image registration with good robustness but also allowed quantification of the degree of sample deformation. The effectiveness of the method was first demonstrated using synthetic datasets with known ground truth. The method was then applied to an experimental dataset collected on an operating lithium battery cell, revealing a high degree of intra- and interparticle chemical complexity in operating batteries. It was observed that the temperature-induced vibrational dynamics involves switching occupancy between antisymmetric and symmetric configurations of the carboxylate oxygen lone pair orbitals, through which electron density around carboxylate oxygen sites is redistributed and metal–ligand interactions are tuned. It will have an effective area above 1.1 m2 at 1 keV, which corresponds to a polished mirror surface of about 300 m2 due to the grazing incidence. As such a mirror area is not achievable with an acceptable mass even with nested shells, silicon pore optics (SPO) technology will be utilized. In the PTB laboratory at BESSY II, two dedicated beamlines are in use for their characterization with monochromatic radiation at 1 keV and a low divergence well below 2 arcsec: the X-ray Pencil Beam Facility (XPBF 1) and the X-ray Parallel Beam Facility (XPBF 2.0), where beam sizes up to 8 mm × 8 mm are available while maintaining low beam divergence. This beamline is used for characterizing mirror stacks and controlling the focusing properties of mirror modules (MMs) – consisting of four mirror stacks – during their assembly at the beamline. A movable CCD based camera system 12 m from the MM registers the direct and the reflected beams. The positioning of the detector is verified by a laser tracker. The energy-dependent reflectance in double reflection through the pores of an MM with an Ir coating was measured at the PTB four-crystal monochromator beamline in the photon energy range 1.75 keV to 10 keV, revealing the effects of the Ir M edges. The confiscated powder material was highly crystalline and was found to consist of two very different species, accidentally of sizes convenient for X-ray diffraction. Thus, one each was selected and redundant complete sets of data were collected at 100 K using Cu Kα radiation. The selected crystals contained: (a) 1,2-diphenyl-2-(pyrrolidin-1-yl)ethanone hydrochloride hemihydrate or 1-(2-oxo-1,2-diphenylethyl)pyrrolidin-1-ium chloride hemihydrate, C18H20NO+·Cl−·0.5H2O, (I), a synthetic cathinone called `α-D2PV', and (b) the sugar myo-inositol, C6H12O6, (II), probably the only instance in which the drug and its diluent have been fully characterized from a single confiscated sample. Moreover, the structural details of both are rather attractive showing: (i) interesting hydrogen bonding observed in pairwise interactions by the drug molecules, mediated by the chloride counter-anions and the waters of crystallization, and (ii) π–π interactions in the case of the phenyl rings of the drug which are of two different types, namely, π–π stacking and edge-to-π. (2023), C79, 445–455] stated that the product of the reaction of [(Cp*Rh)2(μ-OH)3]+ (Cp* is 1,2,3,4,5-pentamethylcyclopentadiene) with 1-methylthymine (1-MT) at pH 10 and 60 °C, to synthesize the anionic component [RhI(η1-N3-1-MT)2]−, was not an RhI complex, but rather an AgI complex, due to the use of silver triflate (AgOTf) to remove Cl− from [Cp*RhCl2]2 to synthesize [Cp*Rh(H2O)3](OTf)2, a water-soluble crystalline complex. We will clearly show that this premise, as stated, is invalid, while the authors have simply avoided several important facts, including that Cp*OH, a reductive elimination product, at pH 10 and 60 °C, was unequivocally identified, thus leading to the RhI anionic component [RhI(η1-N3-1-MT)2]−. More importantly, AgOH, from the reaction of NaOH at pH 10 with any potentially remaining AgOTf, after the AgCl was filtered off, would be insoluble in water. Furthermore, a control experiment with the inorganic complex Rh(OH)3, reacting with 1-methylthymine at pH 10, provided no product, and this bodes well for a similar fate with AgOTf and 1-methylthymine, i.e. at pH 10, AgOTf would again be converted to the water-insoluble AgOH; therefore, no reaction would occur! Finally, a 1H NMR spectroscopy experiment was carried out with synthesized and crystallized [Cp*Rh(H2O)3](OTf)2 in D2O at various pD values; at pD 8.65 no reaction took place, while at pD 13.6, and at 60 °C for 2 h, a reductive elimination reaction caused the precipitation of Cp*OH. The subsequent 1H NMR spectrum clearly demonstrated, in the absence of any AgI complexes, that the solution structure and the X-ray crystals in D2O were similar. A postulated mechanism for this novel anionic component structure, as published previously [Smith et al. (2014). Organometallics, 33, 2389–2404], will be presented, along with the experimental data, to insure the credibility of our results. In recent years, high-resolution cryo-EM studies of complex I from various species have greatly enhanced the understanding of the structure and function of this important membrane-protein complex. Less well studied is the structural basis of complex I biogenesis. The assembly of this complex of more than 40 subunits, encoded by nuclear or mitochondrial DNA, is an intricate process that requires at least 20 different assembly factors in humans. These are proteins that are transiently associated with building blocks of the complex and are involved in the assembly process, but are not part of mature complex I. Although the assembly pathways have been studied extensively, there is limited information on the structure and molecular function of the assembly factors. ath Crystallographic fragment-binding studies of the Mycobacterium tuberculosis trifunctional enzyme suggest binding pockets for the tails of the acyl-CoA substrates at its active sites and a potential substrate-channeling path between them By journals.iucr.org Published On :: 2024-07-16 The Mycobacterium tuberculosis trifunctional enzyme (MtTFE) is an α2β2 tetrameric enzyme in which the α-chain harbors the 2E-enoyl-CoA hydratase (ECH) and 3S-hydroxyacyl-CoA dehydrogenase (HAD) active sites, and the β-chain provides the 3-ketoacyl-CoA thiolase (KAT) active site. Linear, medium-chain and long-chain 2E-enoyl-CoA molecules are the preferred substrates of MtTFE. Previous crystallographic binding and modeling studies identified binding sites for the acyl-CoA substrates at the three active sites, as well as the NAD binding pocket at the HAD active site. These studies also identified three additional CoA binding sites on the surface of MtTFE that are different from the active sites. It has been proposed that one of these additional sites could be of functional relevance for the substrate channeling (by surface crawling) of reaction intermediates between the three active sites. Here, 226 fragments were screened in a crystallographic fragment-binding study of MtTFE crystals, resulting in the structures of 16 MtTFE–fragment complexes. Analysis of the 121 fragment-binding events shows that the ECH active site is the `binding hotspot' for the tested fragments, with 41 binding events. The mode of binding of the fragments bound at the active sites provides additional insight into how the long-chain acyl moiety of the substrates can be accommodated at their proposed binding pockets. In addition, the 20 fragment-binding events between the active sites identify potential transient binding sites of reaction intermediates relevant to the possible channeling of substrates between these active sites. These results provide a basis for further studies to understand the functional relevance of the latter binding sites and to identify substrates for which channeling is crucial. Full Article text

Herein, the synthesis and crystal structure of diammonium {μ-1,3,4,7,8,10,12,13,16,17,19,22-dodecaazatetracyclo[8.8.4.13,17.18,12]tetracosane-5,6,14,15,20,21-hexaonato}ferrate(IV) acetic acid trisolvate, (NH4)2[FeIV(C12H12N12O6)]·3CH3COOH or (NH4)2[FeIV(L–6H)]·3CH3COOH is reported. The FeIV ion is encapsulated by the macropolycyclic ligand, which can be described as a dodeca-aza-quadricyclic cage with two capping triazacyclohexane fragments making three five- and six six-membered alternating chelate rings with the central FeIV ion. The local coordination environment of FeIV is formed by six deprotonated hydrazide nitrogen atoms, which stabilize the unusual oxidation state. The FeIV ion lies on a twofold rotation axis (multiplicity 4, Wyckoff letter e) of the space group C2/c. Its coordination geometry is intermediate between a trigonal prism (distortion angle φ = 0°) and an antiprism (φ = 60°) with φ = 31.1°. The Fe—N bond lengths lie in the range 1.9376 (13)–1.9617 (13) Å, as expected for tetravalent iron. Structure analysis revealed that three acetic acid molecules additionally co-crystallize per one iron(IV) complex, and one of them is positionally disordered over four positions. In the crystal structure, the ammonium cations, complex dianions and acetic acid molecules are interconnected by an intricate system of hydrogen bonds, mainly via the oxamide oxygen atoms acting as acceptors. ath Time-resolved high-energy X-ray diffraction studies of ultrathin Ni ferrite films on MgO(001) By journals.iucr.org Published On :: 2023-11-29 Time-resolved high-energy X-ray diffraction was used during growth of ultrathin NixFe3−xO4 films with varying Ni content (0 ≤ x ≤ 1.5) deposited on MgO(001) substrates by reactive molecular beam epitaxy, providing an insight into the growth dynamics of these films. In order to obtain structural information, reciprocal-space maps were recorded and the temporal evolution of the Bragg peaks specific to the octahedral and tetrahedral lattice sites of the inverse spinel structure of NixFe3−xO4 was observed during growth of the films. A time delay, corresponding to a coverage of 1.2–1.8 nm, between the appearance of the Bragg reflections originating from octahedral sites and reflections originating exclusively from tetrahedral sites indicates that the ferrite films grow in two stages. In the initial growth phase, a rock salt interface layer is formed. Afterwards, a structural transition occurs and the films grow in an inverse spinel structure. The thickness of the initial rock salt phase was found to increase with Ni content and to be responsible for atypical strain in the thin films. Films with Ni contents x > 1 do not show a structural transition. These films remain in a (deficient) rock salt structure consisting of a mixed Ni–Fe oxide and do not form a spinel structure at all. They show an increased number of NiO clusters as detected by X-ray photoelectron spectroscopy of the valence band, accompanied by a significant roughening of the films. Full Article text
ath Energy-dispersive Laue diffraction analysis of the influence of statherin and histatin on the crystallographic texture during human dental enamel demineralization By journals.iucr.org Published On :: 2024-09-25 Energy-dispersive Laue diffraction (EDLD) is a powerful method to obtain position-resolved texture information in inhomogeneous biological samples without the need for sample rotation. This study employs EDLD texture scanning to investigate the impact of two salivary peptides, statherin (STN) and histatin-1 (HTN) 21 N-terminal peptides (STN21 and HTN21), on the crystallographic structure of dental enamel. These proteins are known to play crucial roles in dental caries progression. Three healthy incisors were randomly assigned to three groups: artificially demineralized, demineralized after HTN21 peptide pre-treatment and demineralized after STN21 peptide pre-treatment. To understand the micro-scale structure of the enamel, each specimen was scanned from the enamel surface to a depth of 250 µm using microbeam EDLD. Via the use of a white beam and a pixelated detector, where each pixel functions as a spectrometer, pole figures were obtained in a single exposure at each measurement point. The results revealed distinct orientations of hydroxyapatite crystallites and notable texture variation in the peptide-treated demineralized samples compared with the demineralized control. Specifically, the peptide-treated demineralized samples exhibited up to three orientation populations, in contrast to the demineralized control which displayed only a single orientation population. The texture index of the demineralized control (2.00 ± 0.21) was found to be lower than that of either the STN21 (2.32 ± 0.20) or the HTN21 (2.90 ± 0.46) treated samples. Hence, texture scanning with EDLD gives new insights into dental enamel crystallite orientation and links the present understanding of enamel demineralization to the underlying crystalline texture. For the first time, the feasibility of EDLD texture measurements for quantitative texture evaluation in demineralized dental enamel samples is demonstrated. Full Article text

(SYH:TSX.V; SYHBF:OTCQX; SC1P:FSE) announced that it has completed its earn-in requirements for a 51% interest at its co-flagship Russell Lake Uranium Project in the central core of Canada's Eastern Athabasca Basin in Saskatchewan. The company and Rio Tinto have formed a joint venture (JV) to further explore the property, with Skyharbour holding 51% ownership interest and Rio Tinto holding 49%. This summer, Skyharbour announced that in the first phase of drilling it had found what was historically the best uranium intercept mineralization at the project when hole RSL24-02 at the recently identified Fork Target returned a 2.5-meter-wide intercept of 0.721% U3O8 at a relatively shallow depth of 338.1 meters, including 2.99% U3O8 over 0.5 meters at 339.6 meters. The second phase of drilling included three holes totaling 1,649 meters, with emphasis "at the MZE (M-Zone Extension) target, approximately 10 km northeast of the Fork target, identified prospective faulted graphitic gneiss accompanied by anomalous sandstone and basement geochemistry," Skyharbour said. "The discovery of multi-percent, high-grade, sandstone-hosted uranium mineralization at a new target is a major breakthrough in the discovery process at Russell — something that hasn't been seen before at the project with the potential to quickly grow with more drilling," President and Chief Executive Officer Jordan Trimble said at the time. ANT Survey, Upcoming Drilling Program The company also announced on Thursday that it had completed an Ambient Noise Tomography (ANT) survey in preparation for further drilling at the Russell Lake Project, set to commence in the fall. The survey used Fleet Space Technologies' Exosphere technology to acquire 3D passive seismic velocity data over the highly prospective Grayling and Fork target areas, where previous drilling has intersected high-grade uranium mineralization. "The ANT technology has been successfully employed in mapping significant sandstone and basement structures and associated alteration zones related to hydrothermal fluids pathways in the Athabasca Basin," the company said. Results from the survey will be used to further refine drill targets for the upcoming drilling program. Skyharbour is fully funded and permitted for the follow-up fall drill campaign consisting of approximately 7,000 metres of drilling at its main Russell and Moore Projects, with 2,500 meters of drilling at Moore and 4,500 meters of drilling at Russell. A Great Neighborhood Russell Lake is a large, advanced-stage uranium exploration property totaling 73,294 hectares strategically located between Cameco's Key Lake and McArthur River projects and Denison's Wheeler River Project to the west, and Skyharbour's Moore project to the east. "Skyharbour's acquisition of a majority interest in Russell Lake creates a large, nearly contiguous block of highly prospective uranium claims totaling 108,999 hectares between the Russell Lake and the Moore uranium projects," the company said. Most of the historical exploration at Russell Lake was conducted before 2010, prior to the discovery of several major deposits in/around the Athabasca Basin, Skyharbour said. Notable exploration targets on the property include the Grayling Zone, the M-Zone Extension target, the Little Man Lake target, the Christie Lake target, the Fox Lake Trail target and the newly identified Fork Zone target. "More than 35 kilometers of largely untested prospective conductors in areas of low magnetic intensity also exist on the property," the company noted. In an updated research note in July, Analyst Sid Rajeev of Fundamental Research Corp. wrote that Skyharbour "owns one of the largest portfolios among uranium juniors in the Athabasca Basin." "Given the highly vulnerable uranium supply chain, we anticipate continued consolidation within the sector," wrote Rajeev, who rated the stock a Buy with a fair value estimate of CA$1.21 per share. "Additionally, the rapidly growing demand for energy from the AI (artificial intelligence) industry is likely to accelerate the adoption of nuclear power, which should, in turn, spotlight uranium juniors in the coming months." The Catalyst: Uranium is 'BACK!' The growth of AI, new data centers, electric vehicle (EV) adoption, and the need for more net-zero power means more nuclear energy and the uranium needed to fuel it. Uranium prices are expected to move higher by the end of this quarter, when Trading Economics' global macro models and analyses forecast uranium to trade at US$84.15 per pound, Nuclear Newswire reported on Oct. 3. In another year, the site estimates that the metal will trade at US$91.80 per pound. Just last month, Microsoft Corp. (MSFT:NASDAQ) announced a deal with Constellation Energy Group (CEG:NYSE) to restart and buy all of the power from one of the shut-down reactors at its infamous Three Mile Island plant in Pennsylvania and the Biden administration also announced a plan to restart the Palisades plant in Michigan. Judge Brenda Penny denied the request, which was first filed by Spears' attorney, Samuel D. Ingham III, last November. The judge's decision comes after the singer appeared in court last Wednesday to make a direct appeal to the court. In that emotional statement, Spears said that she was being exploited and "bullied" by the conservatorship — and specifically, by her father. Until recently, both the financial and personal arms of the conservatorship were controlled by Spears' father, Jamie Spears. Last year, Ingham stated in a filing that Spears "strongly opposed" her father as conservator, and that she refused to perform if he remained in charge of her career. In February, Judge Penny allowed a wealth-management company, Bessemer Trust, to come in as a co-conservator for the financial arm of Spears' arrangement. Jamie Spears remains the main conservator for all other aspects of Spears' conservatorship. The next hearing in the case is currently scheduled for July 14. It is possible that Spears will submit a petition for the conservatorship to be terminated. In her comments to Judge Penny last week, Spears said that she had been unaware that she could take such an action. "I didn't know I could petition the conservatorship to end it," she said. Prior to his current role as a professor of oceanography at Oregon State University, Spinrad served as NOAA's top scientist under President Obama and the U.S. representative to the United Nations' Intergovernmental Oceanographic Commission. The nomination comes at a difficult moment in NOAA's history. The agency has been without an official, Senate-confirmed leader since former President Donald Trump took office in January 2017, after his two nominees to lead the agency failed to garner enough support to win a full vote before the Senate. If Spinrad manages to win over the Senate, he will have to contend with a challenge beyond the agency's already-rigorous scientific mandate: restoring public confidence in a traditionally apolitical agency marred by political scandal. In September 2019, then-President Trump wrongly said Alabama was in the projected path of Hurricane Dorian. He continued to reassert the claim for several days, including during an Oval Office briefing on the storm — in which he displayed what appeared to be an official National Weather Service map in which the storm's projected path was extended to Alabama by someone using a black marker. After a National Weather Service office in Birmingham put out a tweet correctly stating that Alabama would not feel the effects of the storm, NOAA published an unsigned defense of the president's claims and rebuking its professional staff who posted the message. Dan Sobien, then-president of the National Weather Service Employees Organization, said at the time that "the hard working employees of the NWS had nothing to do with the utterly disgusting and disingenuous tweet sent out by NOAA management." If confirmed, Spinrad will lead a 12,000-person agency charged with a diverse portfolio that spans daily weather forecasts, climate monitoring, fisheries management and coastal restoration. In a statement, the Environmental Defense Fund's Eric Schwaab applauded Spinrad's nomination, saying that NOAA's workers "couldn't ask for a better leader to restore scientific integrity and honor the agency's mission." Biden, whose administration has made climate action a central focus, has proposed the largest budget in NOAA's history — $6.9 billion, a $1.5 billion increase over the 2021 budget allocated by Congress. Authorities have identified 26 of the bodies. Another 113 people were unaccounted for, though local officials said they had only been able to confirm that about 70 of those people were in the building at the time of the collapse nearly two weeks ago. Detectives are continuing to follow up on reports submitted about possible victims in the partial collapse, but in some cases they have been unable to reach those who submitted the tips in the first place. "People call anonymously. People call and don't leave return phone numbers. People call with partial information, not enough to really secure whether that person may or may not have been in the building," Miami-Dade County Mayor Daniella Levine Cava told reporters on Tuesday. Some of the reports of possible victims are also incomplete, she said, including a name but no apartment number or no date of birth. Levine Cava urged people who are missing loved ones to communicate with local authorities. She said there may also be victims of the collapse who have not been reported missing. The rescue effort stopped briefly overnight due to lightning from a passing storm. Surfside Mayor Charles Burkett said high winds were hampering the cranes moving heavy debris from the collapse site. Lt. Gov. Jeanette Nuñez said Florida has declared a state of emergency ahead of Tropical Storm Elsa, which is expected to reach hurricane strength before making landfall Wednesday morning on the state's west coast. ath Prosecutors Get Their 1st Guilty Plea In The Jan. 6 Oath Keepers Conspiracy Case By www.scpr.org Published On :: Wed, 23 Jun 2021 16:20:11 -0700 Ryan Lucas | NPR Updated June 23, 2021 at 6:56 PM ET Federal prosecutors secured their first guilty plea Wednesday in the Justice Department's sprawling conspiracy case involving the Oath Keepers extremist group in connection with the Jan. 6 attack on the U.S. Capitol. At a hearing in federal court in Washington, D.C., Graydon Young pleaded guilty to one count of conspiracy and one count of obstruction of an official proceeding. The 55-year-old Florida resident agreed to cooperate with investigators, which could prove critical as the government pursues the remaining defendants in the high-profile case. Young is one of 16 people associated with the Oath Keepers to be charged with conspiracy, obstruction and other offenses over the Capitol riot. Prosecutors say the defendants coordinated their efforts and actions to try to disrupt Congress' certification of the Electoral College count on Jan. 6. More than 500 people have been charged so far in connection with the Capitol breach, but the Oath Keepers conspiracy case is one of the most closely watched because of the allegations and the link to an extremist organization. Young is the second defendant linked to the Oath Keepers to plead guilty. Jon Schaffer pleaded guilty to obstructing an official proceeding and entering restricted grounds with a dangerous weapon in April. According to Young's statement of offense, he coordinated with his co-conspirators ahead of Jan. 6 and used encrypted messaging apps to maintain "operational security." On the day itself, the document says, Young and some of his co-conspirators pushed through U.S. Capitol Police lines guarding the Capitol and into the building. "Mr. Young believed that he and the co-conspirators were trying to obstruct, influence, and impede an official proceeding, that is, a proceeding before Congress, specifically, Congress's certification of the Electoral College vote," the document says. At Wednesday's hearing, Judge Amit Mehta read that passage to Young to ensure that it was accurate. "Yes, sir," Young replied, "that is correct." According to the plea deal, Young has agreed to cooperate fully with prosecutors, including sitting for interviews with investigators and testifying before the grand jury and at trial. The government, meanwhile, has agreed to dismiss the remaining charges against him. Even so, Mehta said Young is facing a possible prison sentence of 5 to 6 1/2 years under the sentencing guidelines. Wednesday brought another significant development in the Capitol investigation. Anna Morgan-Lloyd, a 49-year-old from Indiana who described Jan. 6 as the "best day ever," became the first Capitol riot defendant to be sentenced. Morgan-Lloyd was not accused of taking part in any of the violence at the Capitol. She pleaded guilty to a single misdemeanor count of "parading, demonstrating, or picketing in a Capitol building." Mark Grods entered a plea of guilty to one count of conspiracy and one count of obstruction of an official proceeding. According to the statement of offense, the conspiracy's aim was to stop Congress' certification of the Electoral College count. The plea marks another step forward for prosecutors pursuing a broader conspiracy case against 16 alleged members or associates of the Oath Keepers, a far-right, anti-government group. Last week, one of the defendants in that case pleaded guilty to conspiracy and obstruction, and agreed to cooperate with investigators. Grods, who was charged separately but admitted to having coordinated with members of the Oath Keepers, has also agreed to cooperate with the government, including testifying before a grand jury or at trial. In a court filing, prosecutors said Grods' case "is part of an ongoing grand jury investigation and plea negotiation related to United States v. Thomas Caldwell, et al.," which is the government's Oath Keepers conspiracy case. At a court hearing Wednesday in Washington, D.C., just blocks from the Capitol, U.S. District Court Judge Amit Mehta went over the charges and the terms of Grods' plea deal, and told him his estimated sentencing guidelines range was 51 to 63 months. "How do you plead on count one, the charge of conspiracy, sir?" Mehta asked. "Guilty," Grods said. "Count two, obstruction of an official proceeding, how do you plead, sir?" Mehta asked. "Guilty," Grods replied again. In his statement of offense, Grods admits to bringing firearms to Washington, D.C., and then stashing them across the Potomac River at a Virginia hotel — a detail the government says buttresses its argument that the Oath Keepers prepared for violence on Jan. 6. The government alleges the group planned to store weapons in Virginia and ferry them into Washington, D.C., on Jan. 6 if the situation in the city got messy. Grods' statement of offense says on Jan. 6, he rode in a golf cart with others through the city before parking a few blocks away from the Capitol and walking the rest of the way. He then linked up with other alleged Oath Keepers, who forged their way through the crowd, up the steps of the Capitol in a military-style "stack" formation and into the building itself. Other members of the "stack" have been charged in the Oath Keepers conspiracy case. Four minutes after entering the Capitol, the statement of offense says, Grods left the building as police shot pepper balls at a wall near him. Two days after the assault on the Capitol, an unnamed individual told Grods to "make sure that all signal comms about the op has been deleted and burned," according to the statement of offense, which Grods confirmed he had done. It is unclear how much additional information Grods will be able to provide investigators, but his plea agreement — the second in the span of a week — may prompt other defendants in the case to cut deals with prosecutors as well.
