Leveraging symmetric Bragg reflections known as Friedel pairs, our method enables diffraction events to be precisely located within the sample volume. This method allows for fitting the phase, crystal structure and unit-cell parameters at the intra-grain scale on a voxel grid. The processing workflow incorporates several new modules, designed to (i) efficiently match Friedel pairs in large s3DXRD datasets containing up to 108 diffraction peaks; (ii) assign phases to each pixel or voxel, resolving potential ambiguities arising from overlap in scattering angles between different crystallographic phases; and (iii) fit the crystal orientation and unit cell locally on a point-by-point basis. We demonstrate the effectiveness of our technique on fractured granite samples, highlighting the ability of the method to characterize complex geological materials and show their internal structure and mineral composition. Additionally, we include the characterization of a metal gasket made of a commercial aluminium alloy, which surrounded the granite sample during experiments. The results show the effectiveness of the technique in recovering information about the internal texture and residual strain of materials that have undergone high levels of plastic deformation. The Conceptual Design Report for the AfLS is in its final editing stages. This document specifies the socio-economic and scientific rationales and the technical aspects amongst others. The AfLS is supported by many national and Pan-African scientific professional bodies and voluntary associates across many scientific disciplines, and there are stakeholders throughout the continent and beyond. The current roadmap phases have expanded to include national and Pan-African level conversations with policy makers through new Strategic Task Force groups. While it is crucial to obtain overlapping scans in small increments over the region of interest for achieving high-resolution sample reconstruction, a significant number of these scans often redundantly measure the empty background within the wide field of view. To address this inefficiency, an innovative algorithm is proposed that introduces automatic guidance for data acquisition. The algorithm first directs the scan point to actively search for the object of interest within the field of view. Subsequently, it intelligently scans along the perimeter of the sample, strategically acquiring measurements exclusively within the boundary of the region of interest. By employing this approach, a reduction in the number of measurements required to obtain high-resolution reconstruction images is demonstrated, as compared with conventional raster scanning methods. Furthermore, the automatic guidance provided by the algorithm offers the added advantage of saving valuable time during the reconstruction process. Through practical implementation on real experiments, these findings showcase the efficacy of the proposed algorithm in enhancing the efficiency and accuracy of X-ray ptychography experiments. A vector tomography algorithm has been developed to reconstruct the full 3D magnetic vector field without any prior noise assumptions or knowledge about the sample. Two tomographic scans around the vertical axis are acquired on single-crystalline Nd2Fe14B pillars tilted at two different angles, with 2D STXM projections recorded using a focused 120 nm X-ray beam with left and right circular polarization. Image alignment and iterative registration have been implemented based on the 2D STXM projections for the two tilts. Due to averaging over the illuminated sample volume, these measurements traditionally overlooked the compositional and morphological heterogeneity within the sample. Here, a scanning tomographic imaging method is described, using contrast derived from the X-ray scattering intensity, for virtually sectioning the sample to reveal its internal structure at a resolution of a few micrometres. This method provides a means for retrieving the local scattering signal that corresponds to any voxel within the virtual section, enabling characterization of the local structure using traditional data-analysis methods. This is accomplished through tomographic reconstruction of the spatial distribution of a handful of mathematical components identified by non-negative matrix factorization from the large dataset of X-ray scattering intensity. Joint analysis of multiple datasets, to find similarity between voxels by clustering of the decomposed data, could help elucidate systematic differences between samples, such as those expected from genetic modifications, chemical treatments or fungal decay. The spatial distribution of the microfibril angle can also be analyzed, based on the tomographically reconstructed scattering intensity as a function of the azimuthal angle. Full Article text

Similar adaptive mirrors are used on X-ray beamlines to compensate optical aberrations, control wavefronts and tune mirror focal distances at will. Controlled by 1D arrays of piezoceramic actuators, these glancing-incidence mirrors can provide nanometre-scale surface shape adjustment capabilities. However, significant engineering challenges remain for mounting them with low distortion and low environmental sensitivity. Finite-element analysis is used to predict the micron-scale full actuation surface shape from each channel and then linear modeling is applied to investigate the mirrors' ability to reach target profiles. Using either uniform or arbitrary spatial weighting, actuator voltages are optimized using a Moore–Penrose matrix inverse, or pseudoinverse, revealing a spatial dependence on the shape fitting with increasing fidelity farther from the mount. There is an urgent need for techniques of absolute crystal diffraction wavefront measurement. At the Beijing Synchrotron Radiation Facility (BSRF), a novel edge scan wavefront metrology technique has been developed. This technique employs a double-edge tracking method, making diffraction-limited level absolute crystal diffraction wavefront measurement a reality. The results demonstrate an equivalent diffraction surface slope error below 70 nrad (corresponding to a wavefront phase error of 4.57% λ) r.m.s. within a nearly 6 mm range for a flat crystal in the crystal surface coordinate. The double-edge structure contributes to exceptional measurement precision for slope error reproducibility, achieving levels below 15 nrad (phase error reproducibility < λ/100) even at a first-generation synchrotron radiation source. They are not found as natural building blocks of canonical DNA. Reports on their conformational properties are limited. Herein, the single-crystal X-ray structure of α-d-2'-deoxyadenosine (α-dA), C10H13N5O3, and its conformational parameters were determined. In the crystalline state, α-dA forms two conformers in the asymmetric unit which are connected by hydrogen bonds. The sugar moiety of each conformer is arranged in a `clamp'-like fashion with respect to the other conformer, forming hydrogen bonds to its nucleobase and sugar residue. For both conformers, a syn conformation of the nucleobase with respect to the sugar moiety was found. This is contrary to the anti conformation usually preferred by α-nucleosides. The sugar conformation of both conformers is C2'-endo, and the 5'-hydroxyl groups are in a +sc orientation, probably due to the hydrogen bonds formed by the conformers. The formation of the supramolecular assembly of α-dA is controlled by hydrogen bonding and stacking interactions, which was verified by a Hirshfeld and curvedness surface analysis. Chains of hydrogen-bonded nucleobases extend parallel to the b direction and are linked to equivalent chains by hydrogen bonds involving the sugar moieties to form a sheet. A comparison of the solid-state structures of the anomeric 2'-deoxyadenosines revealed significant differences of their conformational parameters. Full Article text

The synthesis process, involved the reaction between N-cycloamino-2-sulfanilamide and various substituted o-salicylaldehydes, resulted in a set of compounds that were subjected to rigorous characterization using advanced spectral techniques, including 1H NMR, 13C NMR and FT–IR spectroscopy, and single-crystal X-ray diffraction. Furthermore, an in-depth assessment of the synthesized compounds was conducted through Absorption, Distribution, Metabolism, Excretion and Toxicity (ADMET) analysis, in conjunction with docking studies, to elucidate their pharmacokinetic profiles and potential. Impressively, the ADMET analysis showcased encouraging drug-likeness properties of the newly synthesized Schiff bases. These computational findings were substantiated by molecular properties derived from density functional theory (DFT) calculations using the B3LYP/6-31G* method within the Jaguar Module of Schrödinger 2023-2 from Maestro (Schrodinger LLC, New York, USA). The exploration of frontier molecular orbitals (HOMO and LUMO) enabled the computation of global reactivity descriptors (GRDs), encompassing charge separation (Egap) and global softness (S). Notably, within this analysis, one Schiff base, namely, 4-bromo-2-{N-[2-(pyrrolidine-1-sulfonyl)phenyl]carboximidoyl}phenol, 20, emerged with the smallest charge separation (ΔEgap = 3.5780 eV), signifying heightened potential for biological properties. Conversely, 4-bromo-2-{N-[2-(piperidine-1-sulfonyl)phenyl]carboximidoyl}phenol, 17, exhibited the largest charge separation (ΔEgap = 4.9242 eV), implying a relatively lower propensity for biological activity. Moreover, the synthesized Schiff bases displayed remarkeable inhibition of tankyrase poly(ADP-ribose) polymerase enzymes, integral in colon cancer, surpassing the efficacy of a standard drug used for the same purpose. Additionally, their bioavailability scores aligned closely with established medications such as trifluridine and 5-fluorouracil. The exploration of molecular electrostatic potential through colour mapping delved into the electronic behaviour and reactivity tendencies intrinsic to this diverse range of molecules. The TcA protein forms a pentameric assembly that attaches to the surface of target cells and penetrates the cell membrane. The TcB and TcC proteins assemble as a heterodimeric TcB–TcC subcomplex that makes a hollow shell. This TcB–TcC subcomplex self-cleaves and encapsulates within the shell a cytotoxic `cargo' encoded by the C-terminal region of the TcC protein. Here, we describe the structure of a previously uncharacterized TcC protein from Yersinia entomophaga, encoded by a gene at a distant genomic location from the genes encoding the rest of the toxin complex, in complex with the TcB protein. When encapsulated within the TcB–TcC shell, the C-terminal toxin adopts an unfolded and disordered state, with limited areas of local order stabilized by the chaperone-like inner surface of the shell. We also determined the structure of the toxin cargo alone and show that when not encapsulated within the shell, it adopts an ADP-ribosyltransferase fold most similar to the catalytic domain of the SpvB toxin from Salmonella typhimurium. Our structural analysis points to a likely mechanism whereby the toxin acts directly on actin, modifying it in a way that prevents normal polymerization. Full Article text

The current crystallographic phase identification, however, is very time consuming. To address this challenge, we have developed a real-time crystallographic phase identifier based on a convolutional self-attention neural network (CPICANN). Trained on 692 190 simulated powder X-ray diffraction (XRD) patterns from 23 073 distinct inorganic crystallographic information files, CPICANN demonstrates superior phase-identification power. Single-phase identification on simulated XRD patterns yields 98.5 and 87.5% accuracies with and without elemental information, respectively, outperforming JADE software (68.2 and 38.7%, respectively). Bi-phase identification on simulated XRD patterns achieves 84.2 and 51.5% accuracies, respectively. In experimental settings, CPICANN achieves an 80% identification accuracy, surpassing JADE software (61%). Integration of CPICANN into XRD refinement software will significantly advance the cutting-edge technology in XRD materials characterization. Full Article text

Focusing on research-active universities in the East African countries of Kenya, Ethiopia, Tanzania and Uganda, the effort has run a series of schools focused on materials for sustainable energy and materials for sustainable development. These bring together early-career researchers from Africa, the US, and beyond, for two weeks in a close-knit environment. The program includes lectures on cutting-edge research from internationally renowned speakers, highly interactive tutorial lectures on the science behind the research, also from internationally known researchers, and hands-on practicals and team-building exercises that culminate in group proposals from self-formed student teams. The schools have benefited more than 300 early-career students and led to proposals that have received funding and have led to research collaborations and educational non-profits. JUAMI continues and has an ongoing community of alumni who share resources and expertise, and is open to like-minded people who want to join and develop contacts and collaborations internationally. The results showed that the deprotonated 8-hydroxyquinoline (C9H6NO) coordinates with the PtII atom via the N and O atoms while the ethylene coordinates in the η2 manner and in the trans position compared to the coordinating N atom. The crystal packing is characterized by C—H⋯O, C—H⋯π, Cl⋯π and Pt⋯π interactions. Complex (I) showed high selective activity against Lu-1 and Hep-G2 cell lines with IC50 values of 0.8 and 0.4 µM, respectively, 54 and 33-fold more active than cisplatin. In particular, complex (I) is about 10 times less toxic to normal cells (HEK-293) than cancer cells Lu-1 and Hep-G2. Furthermore, the reaction of complex (I) with guanine at the N7 position was proposed and investigated using the DFT method. The results indicated that replacement of the ethylene ligand with guanine is thermodynamically more favorable than the Cl ligand and that the reaction occurs via two consecutive steps, namely the replacement of ethylene with H2O and the water with the guanine molecule. can Characterization of sub-micrometre-sized voids in fixed human brain tissue using scanning X-ray microdiffraction By journals.iucr.org Published On :: 2024-10-01 Using a 5 µm-diameter X-ray beam, we collected scanning X-ray microdiffraction in both the small-angle (SAXS) and the wide-angle (WAXS) regimes from thin sections of fixed human brain tissue from Alzheimer's subjects. The intensity of scattering in the SAXS regime of these patterns exhibits essentially no correlation with the observed intensity in the WAXS regime, indicating that the structures responsible for these two portions of the diffraction patterns, which reflect different length scales, are distinct. SAXS scattering exhibits a power-law behavior in which the log of intensity decreases linearly with the log of the scattering angle. The slope of the log–log curve is roughly proportional to the intensity in the SAXS regime and, surprisingly, inversely proportional to the intensity in the WAXS regime. We interpret these observations as being due to the presence of sub-micrometre-sized voids formed during dehydration of the fixed tissue. The SAXS intensity is due largely to scattering from these voids, while the WAXS intensity derives from the secondary structures of macromolecular material surrounding the voids. The ability to detect and map the presence of voids within thin sections of fixed tissue has the potential to provide novel information on the degradation of human brain tissue in neurodegenerative diseases. Understanding their bulk physical and chemical properties requires characterization down to the nano-scale. A powerful technique to study the three-dimensional microstructure is X-ray tomography, but it lacks information about the chemistry of samples. To develop a methodology for measuring the multi-scale 3D microstructure of geological samples, correlative X-ray micro- and nanotomography were performed on two rocks followed by scanning electron microscopy with energy-dispersive spectroscopy (SEM-EDS) analysis. The study was performed in five steps: (i) micro X-ray tomography was performed on rock sample cores, (ii) samples for nanotomography were prepared using laser milling, (iii) nanotomography was performed on the milled sub-samples, (iv) samples were mounted and polished for SEM analysis and (v) SEM imaging and compositional mapping was performed on micro and nanotomography samples for complimentary information. Correlative study performed on samples of serpentine and basalt revealed multiscale 3D structures involving both solid mineral phases and pore networks. Significant differences in the volume fraction of pores and mineral phases were also observed dependent on the imaging spatial resolution employed. This highlights the necessity for the application of such a multiscale approach for the characterization of complex aggregates such as rocks. Information acquired from the chemical mapping of different phases was also helpful in segmentation of phases that did not exhibit significant contrast in X-ray imaging. Adoption of the protocol used in this study can be broadly applied to 3D imaging studies being performed at the Advanced Light Source and other user facilities. Full Article text

Though surprisingly ignored by Grammy Awards voters, Isbell's album of the year winner "Southeastern" reverberated through the Americana community and made many of 2013's best-of lists. He performed song of the year "Cover Me Up" with Shires, a significant figure on the album as muse and collaborator. "I wrote this song for my wife," Isbell said. "I've had a lot of people ask me to dedicate it to their wives, girlfriends or cousin's wife or something strange like that. This was probably the hardest song I ever had to write because I wrote it for her and then I played it for her. It was very difficult. Do the things that scare you. That's the good stuff." Isbell was one of this year's top nominees along with Rosanne Cash and Robert Ellis. Each had three nominations and all were up for artist, album and song of the year. Many of the top nominees and honors recipients performed, including all five emerging artist nominees. Former couple Patty Griffin and Robert Plant made a surprise appearance and sang their collaboration "Ohio." Sturgill Simpson, something of a modern cosmic cowboy, earned emerging artist of the year and the Milk Carton Kids took group/duo of the year. And Buddy Miller, now executive music producer for the television show "Nashville" and theAmericana's winningest performer, won his fifth instrumentalist of the year award. The Americana Music Association also honored several pioneering musicians. Loretta Lynn received the lifetime achievement award for songwriting from Kacey Musgraves and Angaleena Presley. "The truth is we both might cry giving out this award," Musgraves said. Lynn, writer of some of country music's most important female empowerment songs, accepted the award in a sparkly lavender dress and her usual humble manner. "When they told me I was going to get this award," she told the crowd, "I said, 'Naw, you got the wrong one.'" Jackson Browne received the Spirit of Americana-Free Speech in Music award, Flaco Jimenez received the lifetime achievement award for instrumentalist and Taj Mahal earned the lifetime achievement award for performance. "I was affected deeply by American music, near and far — my mother's interest in Southern music and my dad's interest in jazz and bebop and classical, all that kind of stuff," Mahal said in an interview. The first project under review is about 150 kilometers north of Matagami, where there is a small town with a rail link to much of James Bay, and has the Billy Diamond Highway running through it. American Salars said it's close to Q2 Metals Corp. (QTWOTSX-V; OTCQB:QUEXF) Cisco Lithium Project, which reported drill intercepts of 215.6 meters at 1.69% Li2O, including 64.6 meters at 2.29%. Also nearby are multiple projects owned by Sayona Mining Ltd (SYA:ASX), which is currently Canada's only lithium producer. "Our primary objective remains the acquisition of low-cost lithium brine assets in Argentina while expanding our existing NI 43-101 lithium brine resources," said Chief Executive Officer and Director R. Nick Horsley. "We believe that Quebec-based hard rock lithium assets can now be acquired at deeply discounted prices and advanced with critical mineral flow through financing incentives in anticipation of the next lithium rally." American Salars said it is reviewing additional projects close to Sayona, which will be subject to disclosure "once due diligence is completed and a deal is completed." The company stressed that its "intent to acquire property and current strategic review does not necessarily mean that a transaction will occur." Current Assets Also in Argentina, Nevada The company's existing portfolio of lithium deposits includes two NI 43-101-compliant Inferred Mineral Resource Estimates (MREs) consisting of 457,000 tonnes of lithium carbonate equivalent (LCE) at the Candela 2 Lithium Brine Project and a shared MRE at the Pocitos 1 Lithium Brine Project consisting of 760,000 tonnes LCE. The Pocitos MRE is shared with the neighboring Pocitos 2 property, which is not under contract or owned by American Salars, but the company noted that none of the drilling that makes up a partial basis for the MRE took place on the Pocitos 2 block. Both brine projects are located in Salta Province, Argentina. Major mining company Rio Tino recently invested in Argentina by acquiring Argentina lithium producer Arcadium Lithium for US$6.7 billion, making the company the world's third-largest lithium producer. American Salars recently released assay results from soil samples collected during its Phase 1 exploration program at its 100%-owned Black Rock South lithium project close to Tesla's Gigafactory in Nevada. Technical Analyst Clive Maund wrote that the entire "San Emidio Desert basin is a highly prospective lithium exploration zone and is about 38 kilometers long and up to 11 kilometers wide at the widest point, with the central playa measuring about 8.5 kilometers north-south and 4.5 kilometers east-west." Out of 38 samples, 33 recorded lithium concentration of more than 100 parts per million (ppm) or higher, the company said. The highest grade was 180.5 ppm with an average grade of 131 ppm across the 33 samples of the surface of the property. Technical Analyst Clive Maund wrote that the entire "San Emidio Desert basin is a highly prospective lithium exploration zone and is about 38 kilometers long and up to 11 kilometers wide at the widest point, with the central playa measuring about 8.5 kilometers north-south and 4.5 kilometers east-west."* "After a massive speculative runup in 2020 and especially in 2021, the lithium price fell victim to a severe bear market that ran from mid-2022 through the end of 2023," Maund continued. "By the end of last year, this bear market had exhausted itself, and a basing process began that has continued up to the present." In addition to its location near the Gigafactory, Black Rock South is 93 miles southwest of Thacker and 215 miles northwest of the United States' only producing lithium mine, the Silver Peak lithium brine mine owned by Albermarle. The Catalyst: More Growth Coming Lithium is critical in the energy transition for its use in batteries for EVs and other applications. It also is used in electronics, medicine and other industries. According to a report by Grand View Research, market size for the metal was estimated at US$31.75 billion last year and is projected to grow at a compound annual growth rate (CAGR) of 17.7% from this year through 2030. "The automotive application segment is expected to witness substantial growth, driven by stringent regulations imposed by government bodies on ICE automakers to reduce carbon dioxide emissions from vehicles," researchers at Grand View said. "This has shifted the interest of automakers toward producing EVs, which is anticipated to benefit the demand for lithium and related products." EVs and battery storage primarily will fuel future growth of the lithium market, Marin Katusa of Katusa Research wrote recently. He pointed out that all major electric vehicle batteries require lithium, about 1.55 pounds per kilowatt hour of battery capacity, on average. "I think the data speaks for itself that there's more growth and opportunity on the horizon," Katusa wrote. According to FastMarkets, prices for the metal have fallen over the past 18 months as weaker demand improved availability. However, this "has done little to deter the appetite for expansion," raw battery materials analyst Jordan Roberts told the publication.[OWNERSHIP_CHART-11095] The consensus among market analysts points to a recovery in lithium prices in the fourth quarter of 2024, Fastmarkets reported. Franklin/AP Ben Giles | NPRArizona Republicans have stripped the secretary of state's office — currently held by a Democrat — of the right to defend the state's election laws in court, or choose not to, a change enacted as part of Arizona's newly signed budget. The spending blueprint that Gov. Doug Ducey signed into law Wednesday declares that the attorney general — currently a position occupied by Republican Mark Brnovich — has sole authority over election-related litigation. If the secretary of state and attorney general were to disagree over a legal strategy when Arizona election laws are challenged, the new law states that "the authority of the attorney general to defend the law is paramount." Republicans also adopted language stating it's their intent for the law to apply through Jan. 2, 2023, coinciding with the end of Democrat Katie Hobbs' term as secretary of state. Hobbs, the top election official in Arizona who's now running for governor, says her lawyers are looking at whether this change violates the Arizona Constitution. "This is a petty, partisan power grab that is absolutely retaliation towards my office," Hobbs said. "It's clear by the fact that it ends when my term ends. ... It is at best legally questionable, but at worst, likely unconstitutional." Republicans have generally cast the law as a cost-saving measure, citing Hobbs and Brnovich's frequent disagreements over how to defend state election laws that have been challenged in court. In 2020, Hobbs filed complaints with the state bar against Brnovich and other lawyers in his office. Other election provisions in the budget The budget includes a number of other election provisions, and it comes weeks after Republicans enacted new restrictions on early voting in the state, and as a controversial review of 2020 election results in Maricopa County continues. Here are some of the other election-related measures in the budget: New laws could soon require watermarks, QR codes and other security measures to be printed on ballots. There's a new mandate to inspect state and county voter registration databases and create a report on voters who cast federal-only ballots — an option available to Arizonans who don't show proof of citizenship to register to vote in the state, but are still allowed to register under federal law. And a new task force would investigate alleged social media bias as an unreported in-kind political contribution. The ballot security measures, though not mandated by law in the budget, have the potential to be the most cumbersome and costly requirement for county election officials to implement. The budget amendment provides a list of 10 "ballot fraud countermeasures" for counties to choose from — features like holographic foil, background designs similar to those found on banknotes and ultraviolet or infrared ink. If mandated, counties would have to implement any combination of at least three features from the list on their ballots. The budget provides $12 million to pay for those features, to be split among Arizona's 15 counties. "By everyone's admission, there is only one company that can do any of this," said Jennifer Marson, executive director of the Arizona Association of Counties. "And so now, we can't have a competitive bid process or a traditional procurement process at the county or state level to use these countermeasures because we're locked into one company." That company is Authentix, a Texas-based firm that provided Republican Rep. Mark Finchem with a sample ballot that included watermarks, QR codes and other security measures. Finchem had the sample ballot on display at the Capitol in March. According to the Yellow Sheet Report, it could be five times more expensive to print ballots with those security measures as it is to print paper ballots currently in use. Marson said Finchem has acknowledged the security levels required of companies in the budget amendment could only be met by Authentix, and has vowed to mandate the ballot security measures in the "very near future." Finchem defended the company in a brief email. He wrote that Authentix "offers these countermeasures to governments around the world for document and tax stamp security." As the budget was being considered, Democrats like Sen. Tony Navarrete said the amendment is part of a broad effort to solidify conspiracy theories of election fraud. "It's important for us to make sure we vote down conspiracy-laced amendments that are going to hurt the integrity of our election system in the state of Arizona and encourage other states to have these bad copycat laws spread like wildfire," he said. "For Democrats, Jan. 6 undoubtedly looms large," said Lee Miringoff, director of the Marist Institute for Public Opinion, referring to the violence and insurrection at the Capitol, "while, for Republicans, it's more likely about Trump and his claims of a rigged election." Voting access vs. fraud By a 56%-41% margin, survey respondents said making sure that everyone who wants to vote can do so is a bigger concern than making sure that no one who is ineligible votes. But there were wide differences by political party and by race. Among Democrats, almost 9 in 10 said access was more important, but almost three-quarters of Republicans said it was making sure no one votes who isn't eligible. By race, a slim majority of whites said ensuring everyone who wants to vote can was most important, but almost two-thirds of nonwhites said so. Photo ID is popular Nearly 8 in 10 Americans said they believe voters should be required to show government-issued photo identification whenever they vote. Majorities of Democrats, Republicans, independents, whites and nonwhites all said so. Democrats were far lower, though, with 57% believing photo ID should be required. Biden holding steady President Biden gets a 50% job approval rating, largely unchanged from last month. There is a sharp partisan divide with 9 in 10 Democrats approving, and more than 8 in 10 Republicans disapproving. Biden continues to get his highest ratings when it comes to his handling of the coronavirus pandemic, and his economic approval is holding steady. But Americans have less confidence in his handling of foreign policy, especially immigration. His approval on immigration ticked up slightly from March when it was last measured in the poll. By a 50%-43% margin, respondents said Biden had strengthened America's role on the world stage. Americans are split about whether the country is headed in the right direction or not — 49% said it wasn't, 47% said it was. It's an improvement, however, from right after the Jan. 6 insurrection when three-quarters said the country was on the wrong track. The tone has gotten worse in Washington since Biden was elected, 41% said, but that's better than the two-thirds who said so consistently during the Trump years. Methodology: The poll of 1,115 U.S. adults was conducted using live telephone interviewers from June 22-29. Survey questions were available in English or Spanish. (TCRX:NASDAQ) Phase 1 ALLOHA study, evaluating its lead therapeutic candidates TSC-100 and TSC-101 in hematologic malignancies, showed patients continuing to have a positive response after one year, reported Wedbush analyst Dr. David Nierengarten in a Nov. 5 research note. TSC-100 and TSC-101 are T-cell receptor-engineered T-cell therapies (TCR-Ts). "We see a catalyst-rich next few months ahead with data building in prominence on stock impact," Nierengarten wrote. 87% Return Potential Wedbush has a US$10 per share target price on the Massachusetts-based biotech, trading at the time of the report at about US$5.36 per share, noted the analyst. The difference between these figures implies an 87% return potential for investors. TScan Therapeutics remains rated Outperform. Durability of Response Data Nierengarten presented the clinical trial's latest results. As of the July 8, 2024 data cutoff date, in Phase 1 of ALLOHA, 16 patients with hematologic tumors had been administered TSC-100 or TSC-101, and 11 patients had been given a placebo. Median follow-ups had occurred at 5.8 months and 5.3 months, respectively. At the time, none of the patients in the treatment arm had had a relapse. In the control arm, however, three, or 27% of, the 11 patients had, and the median time to relapse was 159 days. The analyst explained that this is typical for patients receiving a hematopoietic stem cell transplant after reduced-intensity conditioning. One year out from treatment, five patients were evaluable, and all remained relapse free and minimal residual disease negative at the time. These data underscore the durability of response to this TCR-T treatment, Nierengarten commented. Its safety profile was shown to be favorable still, with no patients experiencing dose-limiting toxicities or adverse events associated with allogeneic hematopoietic cell transplantation. "Enrollment continues in dose expansion cohorts, and results could support a registrational trial as early as 2025, pending regulatory feedback," Nierengarten wrote. On the Horizon TScan Therapeutics has several catalysts related to its clinical programs on the horizon, which Nierengarten listed. On Nov. 8 and 9, the company will present preclinical data in the poster sessions at the annual Society for Immunotherapy of Cancer meeting. One poster will show in vitro combinatorial data for T-Plex, TScan's cellular therapy for treating solid tumors. It is comprised of two to three different TCR-Ts that target different tumor antigens on different human leukocyte antigen (HLA) types. A second poster will detail the expansion of ImmunoBank, the biotech's diverse bank of therapeutic T-cell receptors (TCRs) that recognize diverse targets and are associated with multiple HLA types. The third will depict development of a target agnostic platform to evaluate how TCR-Ts affect primary human tissues. On Dec. 9, TScan Therapeutics will present updated one-year data from ALLOHA, at the American Society for Hematology Annual Meeting in December. By year-end, the biotech will announce initial data from administering singleplex therapy, cell therapy engineered using a single TCR, to patients with solid tumors. It's the second year in a row, the popular arts festival won't be held in Nevada's Black Rock Desert due to the pandemic. "We have decided to set our sights on Black Rock City 2022," event officials announced in a blog post on Tuesday. In a frequently-asked-questions section, organizers added: "We've heard from many who don't feel ready to come to Black Rock City. While we're confident in our ability to get a permit and to safeguard public health, we know that co-creating Black Rock City in 2021 would put tremendous strain on our community while we are still ironing out uncertainty." Many would-be attendees praised the decision in comments on the Burning Man website and on social media as a safe one; others are anxiously anticipating a bigger and better 2022 Burn. But the cancellation has put many people in the event's host community at ease. Wary of a trend of rising coronavirus cases in some parts of the region, Washoe County's district health officer Kevin Dick said "the right call was made," in order to lower the risk of spreading infection. "The event draws thousands of people from all over the world," Dick said in an email. "We are seeing large outbreaks of COVID-19 occurring in a number of countries, areas where very contagious COVID-19 variants of concern are prevalent and where low rates of vaccination are occurring." The head of a local Paiute tribe is also feeling less burdened knowing there won't be the annual pilgrimage. The main highway to get to the Black Rock Desert playa, which normally draws tens of thousands of people to the summer event, cuts through tribal lands. "For us it is a sigh of relief," said Janet Davis, chairwoman of the Pyramid Lake Paiute Tribe. Although the event — which brings in about $63 million to the state annually — gives the tribal community a welcome financial boost, Davis said. "We don't know who's vaccinated and who's not," she said. "We've been trying to keep our reservation safe and that happening was too soon for us to open." As with last year, the organization will offer virtual programming during Burn Week, from Aug. 29 through Sept. 7, an experience they say drew 165,000 participants in 2020. In response to a request for more details on the reasons for the cancellation, Burning Man organizers declined to comment further. Earlier this month, though, CEO Marian Goodell said the organization was "weighing the gravity" of implementing a vaccination requirement that she said challenged "radical inclusion," one of the group's 10 principles. Still, for many burners, the news won't extinguish their plans to trek to the desert in droves. Just like last year, revelers are preparing to hold unofficial gatherings on public land in place of the annual event. Last summer, those events — the so-called "rogue" and "free" burns or, unmistakably, "Not Burning Man" — drew an estimated 3,000 people to Black Rock Desert during the time Burning Man is normally held, according to the Bureau of Land Management, the federal agency that approves the organization's permits each year. Kevin Jervis, one such attendee who now lives in Gerlach — a tiny desert town near the event site — welcomed this year's cancellation. He called it "more of a relief than anything. ... A lot of us liked it better the way it happened last year." During the informal festivities, Jervis spent a few days between the playa and its outskirts. He said he and his fellow burners felt like it represented the festival's freewheeling roots. "I've had friends that have been going since '94 and they said it was a lot more like it used to be. We didn't have to go by regulations," he said. "We could have guns, dogs ... it was a lot freer." Even before the pandemic, burners increasingly saw an annual gathering under siege. Event-goers who adhere to Burning Man's counterculture beginnings say the festival's explosion in popularity in the past decade has welcomed a host of bad actors who trash the desert and surrounding communities and disregard the event's founding principles, including "decommodification" and the eco-friendly philosophy of "leave no trace." Some of those perceived threats come from festival officials themselves, he said. A ticket to the main event alone cost over $400 in 2019 — a financial hurdle critics say goes against another tenet long espoused, that "everyone is invited." "People that have never been before came out last year because they either couldn't get a ticket other years or they were just kind of curious. Or they didn't have the money to go to the actual Burn," said Jervis. As for the Pyramid Lake Paiute community, with the reservation largely closed during that period last year, Davis said, "we really didn't see the impact" from a public health standpoint. "You're not talking about 65 — 75,000 people." While there was more traffic, she said, "they moseyed on through and moseyed on out." In the years leading up to the pandemic, BLM had been cracking down on the event's growth. Were the festival to return this year, Burning Man organizers said they would have had to meet a population cap of 69,000, down from its 80,000 limit for previous events. Jervis says he won't miss what he describes as organizers' leniency toward "elites" who set up VIP areas at their camps and hire out to construct their art creations instead of making their own. "A lot of people have gotten sick of what Burning Man's kind of become," he said. Even if this year was a go, he said, burners would still be setting up their own Burning Man-adjacent happenings. Following the announcement of the event's cancellation, people are taking to Facebook groups to reminisce about last year's unsanctioned burns and discuss preparations for their own this summer.
