MicroED with the Falcon III direct electron detector

Microcrystal electron diffraction (MicroED) combines crystallography and electron cryo-microscopy (cryo-EM) into a method that is applicable to high-resolution structure determination. In MicroED, nanosized crystals, which are often intractable using other techniques, are probed by high-energy electrons in a transmission electron microscope. Diffraction data are recorded by a camera in movie mode: the nanocrystal is continuously rotated in the beam, thus creating a sequence of frames that constitute a movie with respect to the rotation angle. Until now, diffraction-optimized cameras have mostly been used for MicroED. Here, the use of a direct electron detector that was designed for imaging is reported. It is demonstrated that data can be collected more rapidly using the Falcon III for MicroED and with markedly lower exposure than has previously been reported. The Falcon III was operated at 40 frames per second and complete data sets reaching atomic resolution were recorded in minutes. The resulting density maps to 2.1 Å resolution of the serine protease proteinase K showed no visible signs of radiation damage. It is thus demonstrated that dedicated diffraction-optimized detectors are not required for MicroED, as shown by the fact that the very same cameras that are used for imaging applications in electron microscopy, such as single-particle cryo-EM, can also be used effectively for diffraction measurements.

R3c-type LnNiO3 (Ln = La, Ce, Nd, Pm, Gd, Tb, Dy, Ho, Er, Lu) half-metals with multiple Dirac cones: a potential class of advanced spintronic materials

In the past three years, Dirac half-metals (DHMs) have attracted considerable attention and become a high-profile topic in spintronics becuase of their excellent physical properties such as 100% spin polarization and massless Dirac fermions. Two-dimensional DHMs proposed recently have not yet been experimentally synthesized and thus remain theoretical. As a result, their characteristics cannot be experimentally confirmed. In addition, many theoretically predicted Dirac materials have only a single cone, resulting in a nonlinear electromagnetic response with insufficient intensity and inadequate transport carrier efficiency near the Fermi level. Therefore, after several attempts, we have focused on a novel class of DHMs with multiple Dirac crossings to address the above limitations. In particular, we direct our attention to three-dimensional bulk materials. In this study, the discovery via first principles of an experimentally synthesized DHM LaNiO3 with many Dirac cones and complete spin polarization near the Fermi level is reported. It is also shown that the crystal structures of these materials are strongly correlated with their physical properties. The results indicate that many rhombohedral materials with the general formula LnNiO3 (Ln = La, Ce, Nd, Pm, Gd, Tb, Dy, Ho, Er, Lu) in the space group R3c are potential DHMs with multiple Dirac cones.

Throughput and resolution with a next-generation direct electron detector

Direct electron detectors (DEDs) have revolutionized cryo-electron microscopy (cryo-EM) by facilitating the correction of beam-induced motion and radiation damage, and also by providing high-resolution image capture. A new-generation DED, the DE64, has been developed by Direct Electron that has good performance in both integrating and counting modes. The camera has been characterized in both modes in terms of image quality, throughput and resolution of cryo-EM reconstructions. The modulation transfer function, noise power spectrum and detective quantum efficiency (DQE) were determined for both modes, as well as the number of images per unit time. Although the DQE for counting mode was superior to that for integrating mode, the data-collection throughput for this mode was more than ten times slower. Since throughput and resolution are related in single-particle cryo-EM, data for apoferritin were collected and reconstructed using integrating mode, integrating mode in conjunction with a Volta phase plate (VPP) and counting mode. Only the counting-mode data resulted in a better than 3 Å resolution reconstruction with similar numbers of particles, and this increased performance could not be compensated for by the increased throughput of integrating mode or by the increased low-frequency contrast of integrating mode with the VPP. These data show that the superior image quality provided by counting mode is more important for high-resolution cryo-EM reconstructions than the superior throughput of integrating mode.

X-ray structure of the direct electron transfer-type FAD glucose dehydrogenase catalytic subunit complexed with a hitchhiker protein

The bacterial flavin adenine dinucleotide (FAD)-dependent glucose dehydrogenase complex derived from Burkholderia cepacia (BcGDH) is a representative molecule of direct electron transfer-type FAD-dependent dehydrogenase complexes. In this study, the X-ray structure of BcGDHγα, the catalytic subunit (α-subunit) of BcGDH complexed with a hitchhiker protein (γ-subunit), was determined. The most prominent feature of this enzyme is the presence of the 3Fe–4S cluster, which is located at the surface of the catalytic subunit and functions in intramolecular and intermolecular electron transfer from FAD to the electron-transfer subunit. The structure of the complex revealed that these two molecules are connected through disulfide bonds and hydrophobic interactions, and that the formation of disulfide bonds is required to stabilize the catalytic subunit. The structure of the complex revealed the putative position of the electron-transfer subunit. A comparison of the structures of BcGDHγα and membrane-bound fumarate reductases suggested that the whole BcGDH complex, which also includes the membrane-bound β-subunit containing three heme c moieties, may form a similar overall structure to fumarate reductases, thus accomplishing effective electron transfer.

Noncrystallographic symmetry-constrained map obtained by direct density optimization

Noncrystallographic symmetry (NCS) averaging following molecular-replacement phasing is generally the major technique used to solve a structure with several molecules in one asymmetric unit, such as a spherical icosahedral viral particle. As an alternative method to NCS averaging, a new approach to optimize or to refine the electron density directly under NCS constraints is proposed. This method has the same effect as the conventional NCS-averaging method but does not include the process of Fourier synthesis to generate the electron density from amplitudes and the corresponding phases. It has great merit for the solution of structures with limited data that are either twinned or incomplete at low resolution. This method was applied to the case of the T = 1 shell-domain subviral particle of Penaeus vannamei nodavirus with data affected by twinning using the REFMAC5 refinement software.

Updating direct methods

The standard method of joint probability distribution functions, so crucial for the development of direct methods, has been revisited and updated. It consists of three steps: identification of the reflections which may contribute to the estimation of a given structure invariant or seminvariant, calculation of the corresponding joint probability distribution, and derivation of the conditional distribution of the invariant or seminvariant phase given the values of some diffracted amplitudes. In this article the conditional distributions are derived directly without passing through the second step. A good feature of direct methods is that they may work in the absence of any prior information: that is also their weakness. Different types of prior information have been taken into consideration: interatomic distances, interatomic vectors, Patterson peaks, structural model. The method of directly deriving the conditional distributions has been applied to those cases. Some new formulas have been obtained estimating two-, three- and four-phase invariants. Special attention has been dedicated to the practical aspects of the new formulas, in order to simplify their possible use in direct phasing procedures.

Monday's fat stack of news also includes some views about what to do about drought and Western water supplies.

The New York Times has published six answers to the questions "What are the best ways to share the water? And how can we ensure it lasts for the foreseeable future?" Pat Mulroy, former general manager of the Southern Nevada Water Authority, weighs in, as do several other think tankers:

[N]ew energy and fuel production options have become more water intensive. Unconventional oil and gas production methods such as hydraulic fracturing have significant implications for local and regional water quality and quantity. Bioenergy consumes water at various stages of production (including irrigation for crops) and also has impacts on water quality and quantity...We should be pursuing cleaner energy and streamlined approaches to conserving water in order to truly safeguard our water supply. Dirty for the drought: LA Waterkeeper wants you to take the no-car wash pledge

Washing your car with a garden hose can use up to 120 gallons of water. Most corner conveyor belt operations use less, but all that water can add up, as we told you in August.

That’s why the group LA Waterkeeper is challenging motorists to drive dirty and pledge to skip car washes for 60 days. 

"Water conservation is the easiest and most affordable way to quickly reduce water demand and also extend supplies into next year," says Liz Crosson, the group's executive director. 

This summer Ventura County’s water agency asked people to skip washing their rides for a month; some people got detailing and car washes as a reward. Crosson’s group is hoping bragging rights will be enough of an incentive. (Though there are some as-yet-undisclosed prizes.)

People who live in LA are really excited and interested by the cars that they drive," Crosson says. "And frankly, you can have a dirty car and you can have a sticker and have an excuse for having a dirty car so it saves your image too."

The campaign’s just kicking off this weekend (at the Abbott-Kinney festival), but Crosson’s already optimistic for success. Early adopters of the pledge are hundreds of people from the online site Reddit and the general manager for the Los Angeles Department of Water and Power.

How Director Eliza Hittman’s Journey To Pregnancy Centers In Rural America Inspired Her New Film ‘Never Rarely Sometimes Always’

The film “Never Rarely Sometimes Always” was slated for a theatrical release in March, but due to COVID-19 screenings were postponed. Instead, the film is out on digital this week, currently sporting a 98 percent rating on Rotten Tomatoes and receiving critical acclaim both here on FilmWeek and nationwide as one of the best films of 2020 so far.

Writer-director Eliza Hittman’s third feature-length film is about two teenage girls Skylar (Talia Ryder) and Autumn (Sidney Flanagan) from rural Pennsylvania who travel to New York City for medical help after an unplanned pregnancy. Hittman says the idea for the film came to her when she read in a book about how some women in Ireland, which up until recently had very strict laws against abortions, would travel from Ireland to London in 24 hours just to get a procedure. It struck her as worthy of a screenplay, and the idea was born. As part of her research for the film Hittman went to a small coal-mining community in rural Pennsylvania and, even though she wasn’t pregnant, visited pregnancy centers, got tested, and talked with women getting treatment and counseling so she could, as she says, “write the scenes with credibility.”

Animator, Director Brad Bird Teams Up With Turner Classic Movies To Curate ‘The Essentials’

Brad Bird has made a name for himself as the director of new age, animated classics, like “The Iron Giant” and “The Incredibles”. Now he’s teaming up with Turner Classic Movies to share his picks for classic movie watching that will keep you busy and entertained while you’re stuck at home.

Bird has curated a list of his favorite films which will air on this season’s “The Essentials” with TCM Primetime host Ben Mankiewicz. Every Saturday from May up to January of next year, TCM will air one of twenty movies chosen by Bird. His list includes big-name classics that any movie buff will recognize, like “Casablanca” and “Dr. Strangelove”. But Bird throws in a few surprises, one being that there are no animated films on his list. Another surprising thing viewers will notice is that about a third of the films included are musicals, like “Singin’ In The Rain” and “Guys And Dolls”. With TCM’s seemingly infinite library of movie classics to choose from, Bird says narrowing it down to only twenty favorites was a challenge. According to Bird, his list comprises movies that are not only entertaining but “transcend time” and “speak in a language that is still in many ways current”.

“The Essentials” was filmed in early December, but Bird’s list is coming out during a time when movie-going is looking very different these days. The virus has put the entire film industry on pause, with studios halting production and theaters closing their doors indefinitely. In lieu of new movie releases and regular theater-going, many are taking the opportunity to revisit the classics and Bird’s list provides a starting guide for which ones should be on your watch-list.

Today on FilmWeek, Brad Bird joins us to talk about his favorite movies, what projects he has lined up, and his thoughts on how the film industry is adjusting to and getting through the pandemic.

Director Of New Documentary ‘Spaceship Earth’ Explores Quarantining In The Name Of Science

Two months is a long time to be quarantined in one place. Just ask, well, pretty much anyone in the era of COVID-19. But imagine if you were quarantined for two years instead of two months, all in the name of science, and it was by choice!

In 1991, eight researchers did exactly that in Oracle, Arizona as part of a first-of-its-kind mission called BIOSPHERE 2. No, there was no failed BIOSPHERE 1 mission -- BIOSPHERE 1 is planet Earth. The mission’s goal was to create a living ecosystem inside a massive glass and steel facility to show that human life could be sustained in outer space. The idea was that whenever humanity finally did gain the ability to travel deeper into space and colonize another planet, a biosphere would need to be built first so that life could be sustained. But what started as a science experiment quickly evolved into a cultural phenomenon, and while some watched with bated breath to see whether the researchers could really create a living ecosystem in a controlled environment, others saw the project and those who were involved as a cult of sorts. Director Matt Wolf explores BIOSPHERE 2  the researchers (“biospherians”) who carried the mission out, what ultimately happened and the good and bad ways in which it became a cultural phenomenon.

Today on FilmWeek, “The Frame” host John Horn talks with Wolf about the making of the film and what can be learned from the biospherians about our current situation staying at home because of COVID-19.

SmileDirectClub Awarded US Patent for SmileShop Concept and Plans to Reopen Stores in May

SmileDirectClub shares traded 20% higher after the company reported it has been awarded a U.S. patent for its SmileShop retail concept and treatment process and that it plans to reopen stores on a rolling basis starting in May.

SmileDirectClub Inc. (SDC:NASDAQ) today announced "it has been issued a patent for its SmileShop intellectual property from the U.S. Patent & Trademark Office which further strengthens the telehealth dentistry pioneer's efforts to bring affordable, accessible oral care to more people through its unique and innovative teledentistry platform and direct-to-consumer business model." The firm claimed that the patent will prevent other clear aligner competitors from duplicating its business model for the next 18 years.

The company's CEO David Katzman commented, "This patent designation is a significant validation of our unique customer-oriented care model, and expands our category ownership, including the manufacturing and retail experience...As the industry pioneer and inventor of the SmileShop concept, this patent is recognition that SmileDirectClub offers an innovative way for consumers to access oral care that is safe, doctor-directed and convenient. We look forward to welcoming customers back to our SmileShops at the earliest and safest possible time."

"We are focused on the entire teeth straightening and care process, and we now own the manufacturing process of our clear aligner products, the customer experience via our teledentistry platform, as well as the retail experience for clear aligner therapy. This patent is another step in our process as we continue to grow and protect our business," Katzman added.

The firm advised that "the patent encompasses the unique SmileShop concept and process" which includes appointment scheduling, conducting an intraoral scan, generating an approved treatment plan by a licensed dentist or orthodontist and then creating and shipping the aligners to the customer. The company noted that so far more than one million customers have used its clear aligner therapy platform.

The company stated that it is planning to slowly reopen its SmileShops in the U.S. and other markets starting in May as local governments begin to lift business restrictions. The firm indicated that it will be supplying all of its SmileShop team members with face shields and other PPE and will institute staggered appointment times, temperature scans and other social distancing and sanitary measures to provide a safe experience for all staff and customers.

SmileDirectClub is an oral care company headquartered in Nashville, Tenn. The firm stated that it is the creator of the first direct-to-consumer medtech platform for teeth straightening. The company has since expanded its business and now offers its products directly through dentist and orthodontists' offices. Some of the products offered by the company include aligners, impression kits, retainers and whitening gel. In addition to the U.S., the company also operates in Australia, Canada, Germany, Hong Kong, Ireland, New Zealand and the U.K.

SmileDirectClub began the day with a market capitalization of around $2.1 billion with approximately 385 million shares outstanding and a short interest of about 10.4%. SDC shares opened more than 26% higher today at $6.76 (+$1.37, +26.42%) over yesterday's $5.39 closing price. The stock has traded today between $6.13 to $6.80 per share and is currently trading at $6.64 (+$1.25, +23.14%).

No males hatched in laboratory colonies that were fed on recommended levels of the pesticide and, even at concentrations 50 times lower, the males that did hatch were deformed, and there were significantly fewer compared to an untreated colony.
