ski Striatal Kir2 K+ channel inhibition mediates the antidyskinetic effects of amantadine By www.jci.org Published On :: Levodopa-induced dyskinesia (LID) poses a significant health care challenge for Parkinson’s disease (PD) patients. Amantadine is currently the only drug proven to alleviate LID. Although its efficacy in treating LID is widely assumed to be mediated by blockade of N-methyl-D-aspartate (NMDA) glutamate receptors, our experiments demonstrate that at therapeutically relevant concentrations, amantadine preferentially blocks inward-rectifying K+ channel type 2 (Kir2) channels in striatal spiny projection neurons (SPNs) — not NMDA receptors. In so doing, amantadine enhances dendritic integration of excitatory synaptic potentials in SPNs and enhances — not antagonizes — the induction of long-term potentiation (LTP) at excitatory, axospinous synapses. Taken together, our studies suggest that the alleviation of LID in PD patients is mediated by diminishing the disparity in the excitability of direct- and indirect-pathway SPNs in the on state, rather than by disrupting LTP induction. This insight points to a pharmacological approach that could be used to effectively ameliorate LID and improve the quality of life for PD patients. Full Article

Other than being in a sandwich-generation holding pattern (when will the next phase of my life begin? no one knows), things are pretty good. I'm making more money than I ever have, I love my work, I get lots of downtime to read and hang out with James and mom, and my health is better than it's been in years. These armoured orcs caused quite a problem as to who was going to lead them. Originally, it was going to be the guy with the fur cloak, but he looked a little underwhelming. So a quick look into the spares box produced a much bigger, better suited leader for the task. This crazed, motley lot are in fact a skirmish line of small snaga (slave orcs). They are being whipped and yelled at by larger orcs. They don’t look like they will able to do much with all their squabbling going on, but then again skirmishers in Dragon Rampant are rather limited. I was lucky enough to find a few goblins in chains which worked well whip armed orc from Northstar. I think the goblins came from Alternative Armies. The serious orc leader with his impressive uniformed band, he obviously has higher standards than most.The unruly mob scuffle across the battlefield, making a dreadful din as they go. Using 2011-2016 American Community Survey data (ACS), we show that workers employed in low-wage occupations are older and more likely to have a high school diploma following recent statutory minimum wage increases. To better understand the role of firms, we examine the Burning Glass vacancy data. We find increases in a high school diploma requirement following minimum wage hikes, consistent with our ACS evidence on stocks of employed workers. In this paper we design and test a new method for identifying individual contributions to group performance. We randomly assign people to multiple teams and predict team performance based on previously assessed individual skills. Some people consistently cause their group to exceed its predicted performance. We call these individuals “team players”. Team players score significantly higher on a well-established measure of social intelligence, but do not differ across a variety of other dimensions, including IQ, personality, education and gender. Social skills – defined as a single latent factor that combines social intelligence scores with the team player effect – improve group performance about as much as IQ.
