Cdc24 interacts with the septins to create a positive feedback during bud site assembly in yeast [RESEARCH ARTICLE]

Julian Chollet, Alexander Dünkler, Anne Bäuerle, Laura Vivero-Pol, Medhanie A. Mulaw, Thomas Gronemeyer, and Nils Johnsson

Yeast cells select the position of their new bud at the beginning of each cell cycle. The recruitment of the septins to this prospective bud site is one of the critical events in a complex assembly pathway that culminates in the outgrowth of a new daughter cell. Hereby, the septin-rods follow the high concentration of Cdc42GTP that is generated by the focused localization of its GEF Cdc24. We show that shortly before budding Cdc24 not only activates Cdc42 but also transiently interacts with Cdc11, the septin subunit that caps both ends of the septin rods. Mutations in Cdc24 reducing the affinity to Cdc11 impair septin recruitment and decrease the stability of the polarity patch. The interaction between septins and Cdc24 thus reinforces bud assembly at sites where septin structures are formed. Once the septins polymerize into the ring, Cdc24 is found at the cortex of the bud and directs its further outgrowth from this position.

DC think tank: California online schools group should be investigated

A Washington, D.C., think tank issued a report that says California Virtual Acadmies, a major online school network, has had more dropouts than graduates in most years.; Credit: Jeff J Mitchell/Getty Images

Adolfo Guzman-Lopez

A report released Thursday by a labor group-affiliated Washington think tank is questioning the education provided by an online public school program that says it is in a union fight.

The report by In the Public Interest, a group funded by unions, says the thousands of students enrolled in the California Virtual Academies online public school known as CAVA are receiving a  substandard education by most measures.

"So in every year since CAVA began graduating students, with the exception of 2013, it has produced more dropouts than graduates,” said Shahrzad Habibi, who authored the report.

She said state test score data show that 71 percent of California public schools performed better than the virtual academies.

The report calls on California officials to investigate the online schools’ administration and finances.

California Virtual Academies enrolls about 14,000 kindergarten to 12th grade students through 11 sites, including those in Los Angeles, San Diego, and Fresno. It is run by a national for-profit company called K12 Inc.

In a written statement, California Virtual Academies did not dispute the reported low student performance numbers, but denied other allegations in the study, which it called “inaccurate and deeply flawed.”

“The report relies primarily on misinformation from the California Teachers Association — the union currently engaged in a coordinated and well-funded distortion campaign to unionize the eleven independent California Virtual Academies charter schools.”

In the Public Interest, which supports the work of labor unions, partnered with the American Federation of Teachers last year on a website to track for-profit charter school companies.

White House Rejected 'Overly Prescriptive' CDC Guidance For Reopening Communities

President Trump has said he wants to see the country begin to reopen. The pandemic crashed the economy by keeping people at home, leading to millions of job losses.; Credit: Pool/Getty Images

Franco Ordoñez and Alana Wise | NPR

The White House coronavirus task force rejected detailed guidance drafted by the Centers for Disease Control and Prevention on how workplaces ranging from schools to bars to churches should resume operations to prevent the spread of the virus because it was viewed as "overly prescriptive."

President Trump has said he wants to see the country begin to reopen. The pandemic crashed the economy by keeping people at home, leading to millions of job losses. The White House task force issued guidelines on how to gradually and safely reopen but left decisions up to governors based on conditions in their states.

Experts have warned that a rush to reopen could have disastrous implications for containing COVID-19. Many businesses have said they want more details about how to do things safely.

The draft detailed guidance was provided to the task force in late April, a couple of weeks after it released its April 16 guidance to states for reopening.

The task force sought "certain revisions" to the CDC's detailed guidance, two administration officials told NPR. But revised recommendations were never returned to the task force.

The Associated Press first reported on the task force decision to shelve the detailed guidance. Copies obtained and published by the AP, The New York Times and The Washington Post revealed detailed, staged directions for child care centers, schools, camps, restaurants and bars, churches and mass transit providers about how to safely resume operations.

"I think many people would argue that it is not the role of the federal government to tell specific entities — whether they be schools or churches or businesses — how they should go about doing things because the nation is so diverse," one of the administration officials said.

The task force said that some of the points may be helpful, but they needed to "zoom out a little bit and not be so prescriptive," according to the official. The task force said they would welcome a new set of recommendations, but that never happened, the official said.

"Issuing overly specific instructions — that CDC leadership never cleared — for how various types of businesses open up would be overly prescriptive and broad for the various circumstances states are experiencing throughout the country," the second administration official said. "Guidance in rural Tennessee shouldn't be the same guidance for urban New York City."

The United States last month reached 1 million known coronavirus cases, representing one-third of all coronavirus cases worldwide. Nearly 74,000 Americans have been felled by the disease as of Thursday, according to data compiled by the Johns Hopkins Coronavirus Resource Center.

CDC Guidance For Reopening Schools, Child Care And Summer Camps Is Leaked

Anya Kamenetz | NPR

No field trips. No game rooms. No teddy bears. These are some of the CDC's guidelines for reopening schools, childcare centers and day camps safely in places where coronavirus cases are on the decline.

The guidance, which also covers restaurants, churches and other public places, was obtained by The Associated Press, which reports that the White House tried to keep it from coming to light. The New York Times quoted Mark Meadows, the chief of staff, as being concerned that the guidelines were "overly prescriptive."

The CDC does not have authority to enforce its guidance, which is intended for public information only; the actual policy decisions are up to state and local governments. Schools are closed through the end of the school year throughout much of the country, with the exception of Montana, which welcomed a handful of students back this week. Child care protocols are different in different states.

But millions of parents need child care so they can work, and socialization and stimulation for children who have been confined to home by lockdowns for weeks on end. This is the guidance that summer camps and day cares have been waiting for to make decisions about reopening safely.

The guidance says that where coronavirus is spreading rapidly, child care should only serve the children of essential workers. This is the case today in much of the country, which the guidelines refer to as "Phase 1".

In Phase 2, programs can expand to serve all children with enhanced social distancing measures, and in Phase 3, with a lower risk, social distancing will continue.

Recommended measures include:

Handwashing;

Cloth masks for staff;

Regular disinfection of all surfaces;

Six-foot distance "if possible," head-to-toe positioning with bedding;

As much outdoor air as possible — open windows, fans;

Restricting mixing of groups;

Restricting visitors, and staggering dropoffs and pickups to reduce contact among parents;

Limiting sharing of materials like art supplies or toys. Disinfecting them in between use.;

Avoiding soft toys that can't be easily disinfected;

Not using common areas like dining halls or playgrounds if possible. If it is necessary, stagger visits and disinfect in between;

Adjust operations based on local health data;

Monitor absenteeism.

The guidelines also emphasize keeping attendance at such programs local, to limit children bringing the disease from high to low transmission areas.

From Sriracha sauce to jet engine parts, LAEDC tries to keep jobs in LA

The LAEDC helped Huy Fong Foods reach a compromise to keep operating its Sriracha factory in Irwindale ; Credit: Maya Sugarman/KPCC

Brian Watt

Even as California loses manufacturing jobs, a program run by the Los Angeles County Economic Development Corporation has fought to save some.

When a company is considering relocating to take advantage of lower costs or an easier business climate, the LAEDC’s business assistance program steps in.

It did so in the well-publicized case of Huy Fung Foods last year.

When the city of Irwindale filed a lawsuit against the Sriracha sauce-maker because of bad smells, politicians from other states - most notably Texas - began to circle, offering the company a new home.

Fighting against those suitors is a  familiar dance for the nonprofit Los Angeles County Economic Development Corporation. Many states and municipalities have similar agencies, whose job it is to try to attract and keep employers.

In the Sriracha case, the LAEDC prepared an economic impact analysis, met with the company and the South Coast Air Quality Management District and negotiated a compromise that kept the hot sauce manufacturer here, according to Carrie Rogers, Vice President of Business Assistance and Development with LAEDC.

"We all love Sriracha," she said, adding that she was happy to keep the "180 jobs and really to thwart the efforts of Governor Perry from Texas to try to lure our company away to their state."

The LAEDC estimates its business assistance program has played a role in keeping or luring 200,000 jobs since 1996, when it was formed. It's being recognized by the County Board of Supervisors for those efforts today.

But plenty of jobs still leave.

In a study published in July, the LAEDC said between 1990 and 2012, California lost about 40 percent of its manufacturing jobs – 842,180.

"We compete internationally so a lot of our competitors have gone to Mexico," said Jeff Hynes, CEO of Covina-based Composites Horizons Incorporated, which makes ceramic structures for jet engines. "A week doesn’t go by that I don’t get a call from an economic development corp out of Texas or the South."

He scored a big contract recently and needed to expand fast to begin fulfilling orders.

"Los Angeles  - in our particular industry - has a very good supplier base with materials and equipment," he said "but certainly facility costs are lower in other areas of the state and country."

He said the LAEDC helped him get the permits quickly to buy and modify another building on its street and they decided to stay put.

Composites Horizons currently employs 200 people but plans to add 50 employees this year and another 50 next year, he said.

Rogers, of the LAEDC, said that may not seem like much, but it's important to support businesses like this one.

"When you take a step back and think about it, here’s a company that’s growing when many businesses aren’t," she said. "We know there are suppliers that feed into Composites Horizons. So when they get millions of dollars worth of contracts, we know that many more companies and employees around the county will be employed doing work directly for this company."

Seattle Genetics Shares Trade Higher on Q1/20 Earnings and 22% Growth in ADCETRIS Sales

Source: Streetwise Reports   05/01/2020

Seattle Genetics shares traded 8% higher, reaching a new 52-week high, after the company reported Q1/20 financial results which included a 10% y-o-y increase in net revenues fueled by a 22% increase in sales of ADCETRIS® and a strong debut for PADCEV™ in its first full quarter of sales.

Seattle Genetics Inc. (SGEN:NASDAQ) yesterday announced financial results for the first quarter ended March 31, 2020.

The company also provided an update on commercial results achieved in the quarter for its lead medicines including ADCETRIS® (brentuximab vedotin) and PADCEV™ (enfortumab vedotin-ejfv) and the U.S. Food and Drug Administration's (FDA) approval and launch of TUKYSA™ (tucatinib).

The company's President and CEO Clay Siegall, Ph.D., commented, "We have had a remarkable start to 2020, delivering record product sales in the first quarter that are now coming from both ADCETRIS and PADCEV. Notably, strong PADCEV sales in the first full quarter of launch reflect the unmet need among patients with metastatic bladder cancer...With the recent approval of TUKYSA for patients with metastatic HER2-positive breast cancer, we have now launched our third product just four months after our second...We are also preparing for European commercial operations and have hired general managers in major European markets ahead of potential ex-U.S. approvals of TUKYSA. With two new products, growing revenues, and an advancing pipeline of novel cancer programs, we have exciting prospects for future growth."

The company highlighted that ADCETRIS net sales in the U.S. and Canada increased by 22% to $164.1 million in Q1/20, compared to $135 million in Q1/19. The firm indicated that PADCEV net sales in the U.S. reached $34.5 million in Q1/20, which was its first full quarter of commercialization. The company added that royalty revenues in Q1/20 were $20.4 million and collaboration and license agreement revenues in Q1/20 totaled $15.6 million.

The firm reported a net loss for Q1/20 of $168.4 million, or $0.98 per diluted share, compared to net loss of $13.3 million, or $0.08 per diluted share for Q1/19. The company explained that "the net loss in Q1/20 included a net investment loss of $59.1 million primarily associated with its common stock holdings in Immunomedics, which are marked-to-market, compared to a net investment gain of $38.1 million in Q1/19."

The company advised that its TUKYSA was approved by the FDA for patients with HER2-positive metastatic breast cancer who have received one or more prior anti-HER2 regimens in the metastatic setting. The firm mentioned that it also expects to be able to report topline data in late Q2/20 or Q3/20 for the innovaTV 204 pivotal trial of tisotumab vedotin in patients with recurrent and/or metastatic cervical cancer who have relapsed or progressed after standard of care treatment.

The company noted that it is regularly monitoring the effects of the COVID-19 situation and is maintaining its business outlook estimates for FY/20 that it provided previously on February 6, 2020. For FY/20 it expects ADCETRIS net product sales of $675–700 million, royalty revenues of $105–115 million and collaboration and license agreement revenues of $30–50 million. The firm advised that for FY/20 it expects that R&D expenses will range from $860–950 million with SG&A expenses of $475–525 million.

Seattle Genetics is headquartered in Bothell, Wash., and is a global biotechnology company focused on discovering and commercializing cancer medicines.

Seattle Genetics has a market capitalization of around $23.7 billion with approximately 172.5 million shares outstanding. SGEN shares opened 2.75% higher today at $141.00 (+$3.77, +2.75%) over yesterday's $137.23 closing price and reached a new 52-week high price this morning of $157.00. The stock has traded today between $140.05 and $157.00 per share and is currently trading at $148.51 (+$11.28, +8.22%).
