The series is set in a completely furry world, with a complete lack of humans – something I can always get behind. And yet, I can't ever quite get behind them. I think if I had to put my finger on what's wrong, it's that the Sing movies feel like the Illumination version of Oscar bait, being behind the scenes musicals that ostensibly celebrate the performing arts, something Academy Awards voters should, in theory, love; and yet, they can't even get the easy lay up of Best Original Song, which is straightforwardly embarrassing for movies called, well, Sing. It's not that they're failed Oscar Bait, it's that they're not going for Best Picture, or even Best Animated Feature, but simply seem to be aiming to be nominated in that category. They're not aiming for the top, and they're still missing! Or maybe I'm just being too hard on them, and displacing my own Oscar obsessions on this otherwise innocuous series of jukebox musicals with no higher goal than to be entertaining bits of fluff. The newest entry in the franchise, if it can be called that, is just that. Sing: Thriller is a short available on Netflix, and it features a simple take on a nightmare zombie apocalypse, but furry and kid-friendly; an obvious homage to Michael Jackson's Thriller. In a normal year I’d wait for the four and a half hour Wiseman documentary to arrive on television rather than taking up two time slots to watch it from the confines of a cinema seat at TIFF. But this is not such a year and with a digital screening you get a pause button when you need it. This is bound for PBS and due to the breadth of its subject matter will serve as an excellent introduction to those unfamiliar with this pillar of the documentary form. Usually when I fail to post on time and then illegally backdate the post, yielding a penalty of -1,000 points, it's shortly after midnight. Like, as I'm trying to fall asleep (which of course involves and involuntary inventory of everything I may have failed to do), I'm struck with a panic and then get back out of bed to write some dumb pro-forma apology post. This time I just went to bed and actually fell asleep and now here I am noticing that it is May 1. Still, the whole point of doing this every month is to make the grid of months line up nicely, so the post is backdated by 9 hours and nets -1,000 points. Speaking of lining up nicely: I did get my SIGBOVIK papers in on time and gave a lightning talk at the conference. SIGBOVIK was very popular this year, with our longest-ever proceedings (see SIGBOVIK 2024 PDF or bound volume). This year my project is a paper about a new typesetting system that I wrote to produce the paper (and the talk's slides). That system is called BoVeX and the paper is called Badness 0, which you can read as Badness 0 (Knuth's version) and/or Badness 0 (Epsom's version). You can also maybe find a recorded livestream of the breakneck 5 min presentation, but I would wait for the proper video (in progress now!), which is the same content with much better pacing and details. Speaking of details: I also presented at An Evening of Unnecessary Detail, which is one of Matt Parker ("Standup Maths")'s live shows. Other than the part where I tried to pack a dense months-long technical project about details into 12 minutes, this was a blast! Lots of cool, interesting people. This took place in a proper comedy club in Brooklyn, like with posters of people that I watch on TV (e.g. Taskmaster legend Fern Brady is performing there in a few weeks, so it seems I'm a mere 5 or 6 steps away from my dream of being a contestant on Taskmaster now), and was sold out (due exclusively to the eminence of others, since it was sold out before I even joined the bill). I finally hung out with Grant Sanderson ("3blue1brown") and told him about math. The audience was amazingly attentive and wholesome, and quite a few of them recognized me and wanted to talk after the show, which is fun. (I do not envy the queue that Matt and Grant endured, though!) After a complex week-long courtship ritual with the FedEx guy, that finally arrived last night, at which point I immediately realized that I need further accessories. But I'm excited to shoot on this thing and to make my computer suffer with 8k video. It seems to have gotten too complacent with "Full HD." Finally, I think the main reason I failed to post on time last night was that I was up late playing Balatro. This game is all over the place so you probably don't need me to tell you about it, but it is indeed a good (and addictive) deck-building game that I am enjoying instead of sleep. I am not interested in 100%ing this one, but there are still lots of appealing challenges left for me to do. I'd recommend it if you have the self control to avoid firing it up "for a quick game" when you should be working on your projects or sleeping. Full Article

Cardiac involvement is a major risk factor for mortality. Using MAS solid-state NMR, we studied the fibril structure of a recombinant light chain fragment corresponding to the fibril protein from patient FOR005, together with fibrils formed by protein sequence variants that are derived from the closest germline (GL) sequence. Both analyzed fibril structures were seeded with ex-vivo amyloid fibrils purified from the explanted heart of this patient. We find that residues 11-42 and 69-102 adopt β-sheet conformation in patient protein fibrils. We identify arginine-49 as a key residue that forms a salt bridge to aspartate-25 in the patient protein fibril structure. In the germline sequence, this residue is replaced by a glycine. Fibrils from the GL protein and from the patient protein harboring the single point mutation R49G can be both heterologously seeded using patient ex-vivo fibrils. Seeded R49G fibrils show an increased heterogeneity in the C-terminal residues 80-102, which is reflected by the disappearance of all resonances of these residues. By contrast, residues 11-42 and 69-77, which are visible in the MAS solid-state NMR spectra, show 13Cα chemical shifts that are highly like patient fibrils. The mutation R49G thus induces a conformational heterogeneity at the C terminus in the fibril state, whereas the overall fibril topology is retained. ril Fibrillar {alpha}-synuclein toxicity depends on functional lysosomes [Cell Biology] By www.jbc.org Published On :: 2020-12-18T00:06:18-08:00 Neurodegeneration in Parkinson's disease (PD) can be recapitulated in animals by administration of α-synuclein preformed fibrils (PFFs) into the brain. However, the mechanism by which these PFFs induce toxicity is unknown. Iron is implicated in PD pathophysiology, so we investigated whether α-synuclein PFFs induce ferroptosis, an iron-dependent cell death pathway. A range of ferroptosis inhibitors were added to a striatal neuron-derived cell line (STHdhQ7/7 cells), a dopaminergic neuron–derived cell line (SN4741 cells), and WT primary cortical neurons, all of which had been intoxicated with α-synuclein PFFs. Viability was not recovered by these inhibitors except for liproxstatin-1, a best-in-class ferroptosis inhibitor, when used at high doses. High-dose liproxstatin-1 visibly enlarged the area of a cell that contained acidic vesicles and elevated the expression of several proteins associated with the autophagy-lysosomal pathway similarly to the known lysosomal inhibitors, chloroquine and bafilomycin A1. Consistent with high-dose liproxstatin-1 protecting via a lysosomal mechanism, we further de-monstrated that loss of viability induced by α-synuclein PFFs was attenuated by chloroquine and bafilomycin A1 as well as the lysosomal cysteine protease inhibitors, leupeptin, E-64D, and Ca-074-Me, but not other autophagy or lysosomal enzyme inhibitors. We confirmed using immunofluorescence microscopy that heparin prevented uptake of α-synuclein PFFs into cells but that chloroquine did not stop α-synuclein uptake into lysosomes despite impairing lysosomal function and inhibiting α-synuclein toxicity. Together, these data suggested that α-synuclein PFFs are toxic in functional lysosomes in vitro. Therapeutic strategies that prevent α-synuclein fibril uptake into lysosomes may be of benefit in PD. ril Perilipin 5 S155 phosphorylation by PKA is required for the control of hepatic lipid metabolism and glycemic control [Research Articles] By www.jlr.org Published On :: 2020-12-17T12:30:20-08:00 Perilipin (PLIN) 5 is a lipid droplet-associated protein that coordinates intracellular lipolysis in highly oxidative tissues and is thought to regulate lipid metabolism in response to phosphorylation by protein kinase A (PKA). We sought to identify PKA phosphorylation sites in PLIN5 and assess their functional relevance in cultured cells and the livers of mice. We detected phosphorylation on S155, S161 and S163 of recombinant PLIN5 by PKA in vitro and identified S155 as a functionally important site for lipid metabolism. Expression of phosphorylation-defective PLIN5 S155A in Plin5 null cells resulted in decreased rates of lipolysis and triglyceride-derived fatty acid oxidation compared with cells expressing wildtype PLIN5. These differences in lipid metabolism were not associated with differences in the cellular distribution of PLIN5. Rather, FLIM-FRET analysis of protein-protein interactions showed that PLIN5 S155 phosphorylation regulates PLIN5 interaction with adipose triglyceride lipase (ATGL) at the lipid droplet, but not with the co-activator of ATGL, α-β hydrolase domain-containing 5 (ABHD5). Re-expression of PLIN5 S155A in the liver of Plin5 liver-specific null mice reduced lipolysis when compared to mice with wildtype PLIN5 re-expression, but was not associated with other changes in hepatic lipid metabolism, such as fatty acid oxidation, de novo lipogenesis and triglyceride secretion. Furthermore, glycemic control was impaired in mice with expression of PLIN5 S155A compared with mice expressing PLIN5. Chatham House surveyed a total of 937 respondents between 20 and 30 April 2021. Our survey sample corresponds to the general structure of Belarus’s urban population and is corrected and weighted by gender, age, size of respondents’ town of residence and education level. Summary After Lukashenka, the most known political figures in Belarus are pro-protest politicians: Tsikhanouskaya, Babaryka, Tsikhanouski, Kalesnikava, Tsapkala and Latushka. Viktar Babaryka is the most popular presidential candidate among Belarusian urban citizens, and Lukashenka comes second to him. State organizations and bodies are still mistrusted by more than half the population, while the army is slightly more popular than other bodies. Independent media, labour unions and human rights organizations are trusted far more than state ones. Russia leads when it comes to positive attitudes to foreign states: one in three Belarusians feel very positive about their neighbour and 79 per cent feel positive in general. 71 per cent Belarusians feel positive about China, which is the next highest indicator. Despite propaganda efforts, Ukraine, Lithuania, Poland and EU states in general are still perceived positively by more than 60 per cent of Belarusians, with fewer than 20 per cent feeling negative about them. The position on Russian interference in the Belarusian political crisis is consolidated enough: 58 per cent think that Russia should remain neutral. The rest are almost equally divided between those who think Russia should support the protest movement (19 per cent) and those who think Russia should support Lukashenka (23 per cent). 40 per cent have changed their attitude to the Russian government since it supported Lukashenka in the crisis, and 73 per cent are sure that Russia is his sole support. While 32 per cent of Belarusians see union with Russia as the most appropriate foreign policy, 46 per cent would like to be in a union with both Russia and the EU simultaneously. Almost half the population thinks that Belarusian foreign policy in the event of a pro-protest government coming to power would be oriented toward friendship and cooperation with both Russia and the West, which correlates with how the majority thinks it should be. Although 42 per cent expect foreign policy would lean toward the EU in that situation, which is less acceptable: only 25 per cent think it ‘should’. The proportion of respondents who think Belarus should remain in the Collective Security Treaty Organization is 58 per cent, while 35 per cent would like to see Belarus not belonging to any military bloc and a small share (seven per cent) would prefer Belarus to join NATO. Almost half of the population considers Lukashenka to be the major obstacle for political stability and economic development and is sure he does not care about, and in fact is a threat to, independence. In general, most Belarusians have a consolidated opinion that the authorities should start negotiating with opponents and free all political prisoners. More than half of Belarusians think that Lukashenka must leave immediately or before the end of 2021, while 27 per cent think he should leave when his term ends in 2025. The opinion that Lukashenka should rule for another term beginning in 2025 is marginal – only ten per cent support this. Expectations are not so optimistic though: only 26 per cent believe he will leave office this year. One in three thinks he will leave in 2025; a further 19 per cent say that he will stay on for further terms in office. If Lukashenka doesn’t leave, people expect an increase in emigration and unemployment, the continuation of repression in all areas of life, falling exchange rates and the tightening of dependency on Russia. Opinion on abortions is still consolidated, with only 19 per cent thinking they should be banned in most or all cases. The same applies to Lukashenka’s infamous statement that society in Belarus is not ready to have a female president: only 20 per cent agree with the opinion. 52 per cent don’t think there is a problem of wage inequality between men and women in Belarus, while 37 per cent do see an issue.
