A flexible network of vimentin intermediate filaments promotes migration of amoeboid cancer cells through confined environments [Cell Biology]

Tumor cells can spread to distant sites through their ability to switch between mesenchymal and amoeboid (bleb-based) migration. Because of this difference, inhibitors of metastasis must account for each migration mode. However, the role of vimentin in amoeboid migration has not been determined. Because amoeboid leader bleb–based migration (LBBM) occurs in confined spaces and vimentin is known to strongly influence cell-mechanical properties, we hypothesized that a flexible vimentin network is required for fast amoeboid migration. To this end, here we determined the precise role of the vimentin intermediate filament system in regulating the migration of amoeboid human cancer cells. Vimentin is a classic marker of epithelial-to-mesenchymal transition and is therefore an ideal target for a metastasis inhibitor. Using a previously developed polydimethylsiloxane slab–based approach to confine cells, RNAi-based vimentin silencing, vimentin overexpression, pharmacological treatments, and measurements of cell stiffness, we found that RNAi-mediated depletion of vimentin increases LBBM by ∼50% compared with control cells and that vimentin overexpression and simvastatin-induced vimentin bundling inhibit fast amoeboid migration and proliferation. Importantly, these effects were independent of changes in actomyosin contractility. Our results indicate that a flexible vimentin intermediate filament network promotes LBBM of amoeboid cancer cells in confined environments and that vimentin bundling perturbs cell-mechanical properties and inhibits the invasive properties of cancer cells.

The WTO Ministerial Conference in June 2020 presents a critical opportunity to move ahead on better alignment of trade and environmental sustainability objectives, policymaking and governance. In light of the challenges facing the WTO, meaningful efforts to address environmental sustainability would also help to reinvigorate the organization and strengthen its relevance. 

In this context, the meeting aims to advance discussion on two questions: How can the multilateral trade system better contribute to meeting the UN Sustainable Development Goals and the Paris climate goals? What priorities and tangible outcomes on trade and environmental sustainability should be advanced at the WTO Ministerial Conference in Nur Sultan in June and beyond?

The event will be hosted by the US and the Americas Programme and the Hoffmann Centre for Sustainable Resource Economy at Chatham House in partnership with both the Global Governance Centre and the Centre for Trade and Economic Integration at the Graduate Institute, Geneva.

"Researchers of DETCRC 3.1 project acquired substantial experience in seismic exploration of Nickel deposits across Yilgarn craton, Western Australia. There are many lessons learnt and various approaches trialled. Research quality was scrutinised by publishing in high level journals. In general high quality images are achieved over nickel rich Kambalda province that allowed for an accurate structural interpretation. Several case histories are documented and briefly analysed. Abstract:
To predict time series of counts with small values and remarkable fluctuations, an available model is the $r$ states random environment process based on the negative binomial thinning operator and the geometric marginal. However, we argue that the aforementioned model may suffer from the following two drawbacks. First, under the condition of no prior information, the overdispersed property of the geometric distribution may cause the predictions fluctuate greatly. Second, because of the constraints on the model parameters, some estimated parameters are close to zero in real-data examples, which may not objectively reveal the correlation relationship. For the first drawback, an $r$ states random environment process based on the binomial thinning operator and the Poisson marginal is introduced. For the second drawback, we propose a generalized $r$ states random environment integer-valued autoregressive model based on the binomial thinning operator to model fluctuations of data. Yule–Walker and conditional maximum likelihood estimates are considered and their performances are assessed via simulation studies. Two real-data sets are conducted to illustrate the better performances of the proposed models compared with some existing models.

Abstract:
Present day bio-medical research is pointing towards the fact that cognizance of gene–environment interactions along with genetic interactions may help prevent or detain the onset of many complex diseases like cardiovascular disease, cancer, type2 diabetes, autism or asthma by adjustments to lifestyle. In this regard, we propose a Bayesian semiparametric model to detect not only the roles of genes and their interactions, but also the possible influence of environmental variables on the genes in case-control studies. Our model also accounts for the unknown number of genetic sub-populations via finite mixtures composed of Dirichlet processes. An effective parallel computing methodology, developed by us harnesses the power of parallel processing technology to increase the efficiencies of our conditionally independent Gibbs sampling and Transformation based MCMC (TMCMC) methods. Applications of our model and methods to simulation studies with biologically realistic genotype datasets and a real, case-control based genotype dataset on early onset of myocardial infarction (MI) have yielded quite interesting results beside providing some insights into the differential effect of gender on MI.

Deep Learning Framework for Detecting Ground Deformation in the Built Environment using Satellite InSAR data. (arXiv:2005.03221v1 [cs.CV])

The large volumes of Sentinel-1 data produced over Europe are being used to develop pan-national ground motion services. However, simple analysis techniques like thresholding cannot detect and classify complex deformation signals reliably making providing usable information to a broad range of non-expert stakeholders a challenge. Here we explore the applicability of deep learning approaches by adapting a pre-trained convolutional neural network (CNN) to detect deformation in a national-scale velocity field. For our proof-of-concept, we focus on the UK where previously identified deformation is associated with coal-mining, ground water withdrawal, landslides and tunnelling. The sparsity of measurement points and the presence of spike noise make this a challenging application for deep learning networks, which involve calculations of the spatial convolution between images. Moreover, insufficient ground truth data exists to construct a balanced training data set, and the deformation signals are slower and more localised than in previous applications. We propose three enhancement methods to tackle these problems: i) spatial interpolation with modified matrix completion, ii) a synthetic training dataset based on the characteristics of real UK velocity map, and iii) enhanced over-wrapping techniques. Using velocity maps spanning 2015-2019, our framework detects several areas of coal mining subsidence, uplift due to dewatering, slate quarries, landslides and tunnel engineering works. The results demonstrate the potential applicability of the proposed framework to the development of automated ground motion analysis systems.

Abstract:
Humans are routinely exposed to mixtures of chemical and other environmental factors, making the quantification of health effects associated with environmental mixtures a critical goal for establishing environmental policy sufficiently protective of human health. The quantification of the effects of exposure to an environmental mixture poses several statistical challenges. It is often the case that exposure to multiple pollutants interact with each other to affect an outcome. Further, the exposure-response relationship between an outcome and some exposures, such as some metals, can exhibit complex, nonlinear forms, since some exposures can be beneficial and detrimental at different ranges of exposure. To estimate the health effects of complex mixtures, we propose a flexible Bayesian approach that allows exposures to interact with each other and have nonlinear relationships with the outcome. We induce sparsity using multivariate spike and slab priors to determine which exposures are associated with the outcome and which exposures interact with each other. The proposed approach is interpretable, as we can use the posterior probabilities of inclusion into the model to identify pollutants that interact with each other. We utilize our approach to study the impact of exposure to metals on child neurodevelopment in Bangladesh and find a nonlinear, interactive relationship between arsenic and manganese.

Abstract:
This paper presents a general model framework for detecting the preferential sampling of environmental monitors recording an environmental process across space and/or time. This is achieved by considering the joint distribution of an environmental process with a site-selection process that considers where and when sites are placed to measure the process. The environmental process may be spatial, temporal or spatio-temporal in nature. By sharing random effects between the two processes, the joint model is able to establish whether site placement was stochastically dependent of the environmental process under study. Furthermore, if stochastic dependence is identified between the two processes, then inferences about the probability distribution of the spatio-temporal process will change, as will predictions made of the process across space and time. The embedding into a spatio-temporal framework also allows for the modelling of the dynamic site-selection process itself. Real-world factors affecting both the size and location of the network can be easily modelled and quantified. Depending upon the choice of the population of locations considered for selection across space and time under the site-selection process, different insights about the precise nature of preferential sampling can be obtained. The general framework developed in the paper is designed to be easily and quickly fit using the R-INLA package. We apply this framework to a case study involving particulate air pollution over the UK where a major reduction in the size of a monitoring network through time occurred. It is demonstrated that a significant response-biased reduction in the air quality monitoring network occurred, namely the relocation of monitoring sites to locations with the highest pollution levels, and the routine removal of sites at locations with the lowest. We also show that the network was consistently unrepresenting levels of particulate matter seen across much of GB throughout the operating life of the network. Finally we show that this may have led to a severe overreporting of the population-average exposure levels experienced across GB. This could have great impacts on estimates of the health effects of black smoke levels.
