Informing at the Crossroads of Design Science Research, Academic Entrepreneurship, and Digital Transformation: A Platform Ecosystem Roadmap

Aim/Purpose: Developing Digital Platform Ecosystems (DPE) to transform conventional Knowledge Management Systems (KM/KMS) scenarios promises significant benefits for individuals, institutions, as well as emerging knowledge economies. Background: The academic entrepreneurship project presented is aiming for such a KMS-DPE configuration. Having consolidated this author’s own and external re-search findings, realization is currently commencing with a start-up in a business incubator. Methodology: Design science research applying mixed one-sample case study and illustrative scenario approach focusing on conceptual analysis and entrepreneurship. Contribution: Although (academic) entrepreneurship is a young research area with recently growing interest, publications focusing on this transitional stage between maturing research and projected commercial viability of digital technologies are rare. Findings: A roadmap looking beyond the immediate early-start-up perspective is out-lined by integrating recent development-stage-related DPE-research and by addressing stakeholders diverse informing needs essential for system realization. Recommendations for Practitioners and Researchers: As this transdisciplinary perspective combines KM, informing, design science, and entrepreneurial research spaces, it may assist other researchers and practitioners facing similar circumstances and/or start-up opportunities. Impact on Society: The article advances the understanding of how DPE communities may serve members with highly diverse skills and ambitions better to gainfully utilize the platform’s resources and generative potential in their personal and local settings. Future Research: As the entrepreneurial agenda will complement (not substitute) the academic research, research priorities have been highlighted aligned to three future stages.

Environment: Commission launches new platform to help resolve social conflicts over large carnivores

Europe's brown bear, wolf, wolverine, lynx – at least one of these species can now be found in 21 EU Member States. After a lengthy period of decline their numbers are growing once more, but coexistence with man can be problematic. In an effort to solve the social and economic problems that sometimes result from this new expansion, the European Commission has launched a platform where farmers, conservationists, hunters, landowners and scientists can exchange ideas and best practices on sharing the same land with large carnivores.

The EU Platform on Coexistence between People and Large Carnivores will support constructive dialogue between key stakeholder organisations at the European level. Launching the platform, EU Environment Commissioner Janez Potočnik said: "We need to treat our natural neighbours with respect – but we also need to heed the concerns of those whose lives are genuinely affected by their close proximity. My warm congratulations to the organisations that have worked together to set up this important platform, which represents a major step forward in efforts to address the issue of peaceful coexistence."

The European Union is home to five species of large carnivores. All suffered dramatic declines in numbers and distribution as a consequence of human activity, but increasing protection and public awareness about their vital role in healthy ecosystems have caused many populations to stabilize or increase, and to return to areas from which they had been absent for decades or even centuries.

While this recovery is seen by some as a great conservation success, it has not been without its opponents. The issue involves a diversity of stakeholders such as hunters, foresters, livestock producers, reindeer herders, landowners, rural communities, conservation organizations and the wider public. These groups are influenced by and perceive large carnivores in different ways, and in some cases these differences can be a source of conflict. The platform will facilitate exchanges of knowledge and promote ways and means to minimize, and wherever possible, find equitable solutions to these conflicts.

The platform launched today follows a number of efforts to understand the conflicts between stakeholders over large carnivores, the results of which were set out in workshops conclusions and in a report.

Next steps

The Platform will hold its first working session immediately following the official launch today, on 10 June. It will adopt terms of reference and a work plan. The Platform will hold one annual meeting and organize additional workshops on selected topics. It will be supported by a web-based resource centre that will serve as the main tool to disseminate information on the activities of the platform, identify good practices in the form of documents or a manual, act as a gateway to the portals of the member organisations, and host media resources such as press kits for journalists.

HEIDI: an experiment-management platform enabling high-throughput fragment and compound screening

The Swiss Light Source facilitates fragment-based drug-discovery campaigns for academic and industrial users through the Fast Fragment and Compound Screening (FFCS) software suite. This framework is further enriched by the option to utilize the Smart Digital User (SDU) software for automated data collection across the PXI, PXII and PXIII beamlines. In this work, the newly developed HEIDI webpage (https://heidi.psi.ch) is introduced: a platform crafted using state-of-the-art software architecture and web technologies for sample management of rotational data experiments. The HEIDI webpage features a data-review tab for enhanced result visualization and provides programmatic access through a representational state transfer application programming interface (REST API). The migration of the local FFCS MongoDB instance to the cloud is highlighted and detailed. Surface-mutagenesis strategies to enable structural biology crystallization platforms

A key prerequisite for the successful application of protein crystallography in drug discovery is to establish a robust crystallization system for a new drug-target protein fast enough to deliver crystal structures when the first inhibitors have been identified in the hit-finding campaign or, at the latest, in the subsequent hit-to-lead process. The first crucial step towards generating well folded proteins with a high likelihood of crystallizing is the identification of suitable truncation variants of the target protein. In some cases an optimal length variant alone is not sufficient to support crystallization and additional surface mutations need to be introduced to obtain suitable crystals. In this contribution, four case studies are presented in which rationally designed surface modifications were key to establishing crystallization conditions for the target proteins (the protein kinases Aurora-C, IRAK4 and BUB1, and the KRAS–SOS1 complex). The design process which led to well diffracting crystals is described and the crystal packing is analysed to understand retrospectively how the specific surface mutations promoted successful crystallization. The presented design approaches are routinely used in our team to support the establishment of robust crystallization systems which enable structure-guided inhibitor optimization for hit-to-lead and lead-optimization projects in pharmaceutical research.

EMhub: a web platform for data management and on-the-fly processing in scientific facilities

Most scientific facilities produce large amounts of heterogeneous data at a rapid pace. Managing users, instruments, reports and invoices presents additional challenges. To address these challenges, EMhub, a web platform designed to support the daily operations and record-keeping of a scientific facility, has been introduced. EMhub enables the easy management of user information, instruments, bookings and projects. The application was initially developed to meet the needs of a cryoEM facility, but its functionality and adaptability have proven to be broad enough to be extended to other data-generating centers. The expansion of EMHub is enabled by the modular nature of its core functionalities. The application allows external processes to be connected via a REST API, automating tasks such as folder creation, user and password generation, and the execution of real-time data-processing pipelines. EMhub has been used for several years at the Swedish National CryoEM Facility and has been installed in the CryoEM center at the Structural Biology Department at St. Jude Children's Research Hospital. A fully automated single-particle pipeline has been implemented for on-the-fly data processing and analysis. At St. Jude, the X-Ray Crystallography Center and the Single-Molecule Imaging Center have already expanded the platform to support their operational and data-management workflows.
