OPTN recruitment to a Golgi-proximal compartment regulates immune signalling and cytokine secretion [RESEARCH ARTICLE]

Optineurin (OPTN) is a multifunctional protein involved in autophagy, secretion as well as NF-B and IRF3 signalling and OPTN mutations are associated with several human diseases. Here we show that, in response to viral RNA, OPTN translocates to foci in the perinuclear region, where it negatively regulates NF-B and IRF3 signalling pathways and downstream pro-inflammatory cytokine secretion. These OPTN foci consist of a tight cluster of small membrane vesicles, which are positive for ATG9A. Disease mutations linked to POAG cause aberrant foci formation in the absence of stimuli, which correlates with the ability of OPTN to inhibit signalling. Using proximity labelling proteomics, we identify the LUBAC complex, CYLD and TBK1 as part of the OPTN interactome and show that these proteins are recruited to this OPTN-positive perinuclear compartment. Our work uncovers a crucial role for OPTN in dampening NF-B and IRF3 signalling through the sequestration of LUBAC and other positive regulators in this viral RNA-induced compartment leading to altered pro-inflammatory cytokine secretion.

XGANDALF – extended gradient descent algorithm for lattice finding

Serial crystallography records still diffraction patterns from single, randomly oriented crystals, then merges data from hundreds or thousands of them to form a complete data set. To process the data, the diffraction patterns must first be indexed, equivalent to determining the orientation of each crystal. A novel automatic indexing algorithm is presented, which in tests usually gives significantly higher indexing rates than alternative programs currently available for this task. The algorithm does not require prior knowledge of the lattice parameters but can make use of that information if provided, and also allows indexing of diffraction patterns generated by several crystals in the beam. Cases with a small number of Bragg spots per pattern appear to particularly benefit from the new approach. The algorithm has been implemented and optimized for fast execution, making it suitable for real-time feedback during serial crystallography experiments. It is implemented in an open-source C++ library and distributed under the LGPLv3 licence. An interface to it has been added to the CrystFEL software suite.

Rachel Mason had, to a certain extent, the normal upbringing you’d imagine a family of five with small business owner parents would have. But in her documentary, ‘Circus of Books’, she pulls the curtain on the double-life her parents led as modest business owners and pillars of the LGBTQ community.

Karen and Barry Mason established West Hollywood’s Circus of Books on Santa Monica Boulevard in the 1980s. What seemed like an unassuming bookstore was actually a gay porn shop that became an institution in the LGBTQ community during a time when homosexuality was still largely unaccepted. The store was far from being a “bookstore with a circus theme”. The Los Angeles-based shop was the central hub for gay pornography around the country, once one of the main distributors for adult films. 

While the store was becoming a home for gay culture and pride, the Masons largely kept their business a secret from colleagues, friends, family, even their own children. It’s a central conflict that Rachel Mason explores throughout the film as the daughter of two shop owners caught between the pressures of maintaining a traditional family image and making a living as gay pornography distributors.

Today on FilmWeek, we’re joined by ‘Circus of Books’ director Rachel Mason for a conversation on her documentary and the experience of creating a film with her parents and their secret as the subject.

Microalgae could be used to effectively bio-refine brewery wastewater

A circular economy, in which waste is minimised and resources are kept within the system, relies upon inventive ways of turning waste into a resource. A new study explored the possibility of using the microalga Scenedesmus obliquus to refine and process brewery wastewater. The alga efficiently removed pollutants from the effluent, produced biomass and biofuels in a range of different forms — and with different bioactive compounds — and encouraged waste barley and wheat to germinate at increased rates. This is especially important for breweries, as barley seeds are one of the main feedstocks for the industry — and thus are key to increasing its sustainability and circularity.

DISCUSSION AND OUTLOOK
----------------------
At 1000 PM CDT (0300 UTC), the center of Post-Tropical Cyclone Olga
was located near latitude 27.8 North, longitude 92.2 West. The
post-tropical cyclone is moving toward the northeast near 17 mph (28
km/h). Olga is forecast to move quickly northward to
north-northeastward on Saturday and then turn northeastward late
Saturday or Sunday. On the forecast track, the center of the
post-tropical cyclone will move up the Mississippi Valley tomorrow
and toward the Great Lakes later this weekend.

Maximum sustained winds are near 50 mph (85 km/h) with higher gusts.
Weakening is expected after the cyclone moves over land Saturday
morning.

Tropical-storm-force winds extend outward up to 140 miles (220 km)
from the center.

The estimated minimum central pressure based on earlier data from
an Air Force Reserve hurricane hunter aircraft and surface
observations over the northern Gulf of Mexico is 999 mb (29.50
inches).


HAZARDS AFFECTING LAND
----------------------
WIND: Gale-force winds associated with Olga and its remnants should
spread over portions of the northern Gulf coast tonight and Saturday
morning.

RAINFALL: The post-tropical cyclone, along with rainfall ahead of
the system along and north of the frontal boundary across the
Central Gulf coast, is expected to produce total rainfall
accumulations of 3 to 6 inches with maximum totals of 8 inches
across the Central Gulf coast into portions of the Lower Mississippi
Valley and western Tennessee Valley through Saturday morning. These
rains may produce flash flooding across the Central Gulf coast into
the Lower Mississippi Valley and western Tennessee Valley.

COASTAL FLOODING: Above-normal tides and associated coastal
flooding are possible across portions of the northern Gulf coast.

TORNADOES: Isolated tornadoes are possible tonight into Saturday

Earlier this evening, the last 2 passes through Olga made by an Air
Force Reserve Hurricane Hunter aircraft showed that the cyclone
becoming embedded within a cold front. Strong northwesterly flow was
observed within 10 n mi northwest of Olga's center and a sharp
temperature and dew point gradient was measured across the cyclone.
It does not appear that Olga has separated from the front in any
significant way since the plane left. In fact, recent surface
observations suggest that either the front passes through the center
of the cyclone or its circulation has become poorly defined. Based
on all these data, Olga is now classified as post-tropical and this
is the last NHC advisory.

Since Olga is entangled with the front, the winds and seas
associated the front are now considered to be representative of the
overall system. The intensity of the post-tropical cyclone is set at
45 kt based on SFMR winds of 48 kt and flight-level winds of 54 kt
found by the plane behind the front. No substantial change in Olga's
strength is anticipated before it reaches the coast Saturday
morning, and the winds should decrease quickly thereafter. The
post-tropical cyclone is forecast to move quickly up the Mississippi
Valley on Saturday, before turning northeastward toward the Great
Lakes late Saturday or early Sunday. Dissipation is anticipated
before the end of the weekend.

Severe weather, including heavy rain, gusty winds, and isolated
tornadoes will be the primary hazard associated with post-tropical
Olga as it races northward. Coastal flooding is also possible
tonight along portions of the Louisiana coast. For more information
