isi High-resolution double vision of the allosteric phosphatase PTP1B By journals.iucr.org Published On :: 2024-01-01 Protein tyrosine phosphatase 1B (PTP1B) plays important roles in cellular homeostasis and is a highly validated therapeutic target for multiple human ailments, including diabetes, obesity and breast cancer. However, much remains to be learned about how conformational changes may convey information through the structure of PTP1B to enable allosteric regulation by ligands or functional responses to mutations. High-resolution X-ray crystallography can offer unique windows into protein conformational ensembles, but comparison of even high-resolution structures is often complicated by differences between data sets, including non-isomorphism. Here, the highest resolution crystal structure of apo wild-type (WT) PTP1B to date is presented out of a total of ∼350 PTP1B structures in the PDB. This structure is in a crystal form that is rare for PTP1B, with two unique copies of the protein that exhibit distinct patterns of conformational heterogeneity, allowing a controlled comparison of local disorder across the two chains within the same asymmetric unit. The conformational differences between these chains are interrogated in the apo structure and between several recently reported high-resolution ligand-bound structures. Electron-density maps in a high-resolution structure of a recently reported activating double mutant are also examined, and unmodeled alternate conformations in the mutant structure are discovered that coincide with regions of enhanced conformational heterogeneity in the new WT structure. These results validate the notion that these mutations operate by enhancing local dynamics, and suggest a latent susceptibility to such changes in the WT enzyme. Together, these new data and analysis provide a detailed view of the conformational ensemble of PTP1B and highlight the utility of high-resolution crystallography for elucidating conformational heterogeneity with potential relevance for function. isi Scattered high-energy synchrotron radiation at the KARA visible-light diagnostic beamline By journals.iucr.org Published On :: 2024-03-26 To characterize an electron beam, visible synchrotron light is often used and dedicated beamlines at synchrotron sources are becoming a more common feature as instruments and methods for the diagnostics are, along with the accelerators, further developed. At KARA (Karlsruhe Research Accelerator), such a beamline exists and is based on a typical infrared/visible-light configuration. From experience at such beamlines no significant radiation was expected (dose rates larger than 0.5 µSv h−1). This was found not to be the case and a higher dose was measured which fortunately could be shielded to an acceptable level with 0.3 mm of aluminium foil or 2.0 mm of Pyrex glass. The presence of this radiation led to further investigation by both experiment and calculation. A custom setup using a silicon drift detector for energy-dispersive spectroscopy (Ketek GmbH) and attenuation experiments showed the radiation to be predominantly copper K-shell fluorescence and is confirmed by calculation. The measurement of secondary radiation from scattering of synchrotron and other radiation, and its calculation, is important for radiation protection, and, although a lot of experience exists and methods for radiation protection are well established, changes in machine, beamlines and experiments mean a constant appraisal is needed. isi The diamond–silicon carbide composite Skeleton® as a promising material for substrates of intense X-ray beam optics By journals.iucr.org Published On :: 2024-08-06 The paper considers the possibility of using the diamond-silicon carbide composite Skeleton® with a technological coating of polycrystalline silicon as a substrate for X-ray mirrors used with powerful synchrotron radiation sources (third+ and fourth generation). Samples were studied after polishing to provide the following surface parameters: root-mean-square flatness ≃ 50 nm, micro-roughness on the frame 2 µm × 2 µm σ ≃ 0.15 nm. The heat capacity, thermal conductivity and coefficient of linear thermal expansion were investigated. For comparison, a monocrystalline silicon sample was studied under the same conditions using the same methods. The value of the coefficient of linear thermal expansion turned out to be higher than that of monocrystalline silicon and amounted to 4.3 × 10−6 K−1, and the values of thermal conductivity (5.0 W cm−1 K−1) and heat capacity (1.2 J K−1 g−1) also exceeded the values for Si. Thermally induced deformations of both Skeleton® and monocrystalline silicon samples under irradiation with a CO2 laser beam have also been experimentally studied. Taking into account the obtained thermophysical constants, the calculation of thermally induced deformation under irradiation with hard (20 keV) X-rays showed almost three times less deformation of the Skeleton® sample than of the monocrystalline silicon sample. This study introduces a method that employs the Noise2Noise approach combined with neural networks to effectively mitigate these ambiguities. We applied this methodology to hundreds of ambiguous reconstructed images retrieved from a single diffraction pattern using a conventional retrieval algorithm. Our results demonstrate that ambiguous features in these reconstructions are effectively treated as inter-reconstruction noise and are significantly reduced. The post-Noise2Noise treated images closely approximate the average and singular value decomposition analysis of various reconstructions, providing consistent and reliable reconstructions. isi Pillar data-acquisition strategies for cryo-electron tomography of beam-sensitive biological samples By journals.iucr.org Published On :: 2024-06-03 For cryo-electron tomography (cryo-ET) of beam-sensitive biological specimens, a planar sample geometry is typically used. As the sample is tilted, the effective thickness of the sample along the direction of the electron beam increases and the signal-to-noise ratio concomitantly decreases, limiting the transfer of information at high tilt angles. In addition, the tilt range where data can be collected is limited by a combination of various sample-environment constraints, including the limited space in the objective lens pole piece and the possible use of fixed conductive braids to cool the specimen. Consequently, most tilt series are limited to a maximum of ±70°, leading to the presence of a missing wedge in Fourier space. The acquisition of cryo-ET data without a missing wedge, for example using a cylindrical sample geometry, is hence attractive for volumetric analysis of low-symmetry structures such as organelles or vesicles, lysis events, pore formation or filaments for which the missing information cannot be compensated by averaging techniques. Irrespective of the geometry, electron-beam damage to the specimen is an issue and the first images acquired will transfer more high-resolution information than those acquired last. There is also an inherent trade-off between higher sampling in Fourier space and avoiding beam damage to the sample. Finally, the necessity of using a sufficient electron fluence to align the tilt images means that this fluence needs to be fractionated across a small number of images; therefore, the order of data acquisition is also a factor to consider. Here, an n-helix tilt scheme is described and simulated which uses overlapping and interleaved tilt series to maximize the use of a pillar geometry, allowing the entire pillar volume to be reconstructed as a single unit. Three related tilt schemes are also evaluated that extend the continuous and classic dose-symmetric tilt schemes for cryo-ET to pillar samples to enable the collection of isotropic information across all spatial frequencies. A fourfold dose-symmetric scheme is proposed which provides a practical compromise between uniform information transfer and complexity of data acquisition. Full Article text

However, despite extensive processing of large image sets collected in cryo-EM experiments to amplify the signal-to-noise ratio, the reconstructed 3D protein-density maps are often limited in quality due to residual noise, which in turn affects the accuracy of the macromolecular representation. Here, crefDenoiser is introduced, a denoising neural network model designed to enhance the signal in 3D cryo-EM maps produced with standard processing pipelines. The crefDenoiser model is trained without the need for `clean' ground-truth target maps. Instead, a custom dataset is employed, composed of real noisy protein half-maps sourced from the Electron Microscopy Data Bank repository. Competing with the current state-of-the-art, crefDenoiser is designed to optimize for the theoretical noise-free map during self-supervised training. We demonstrate that our model successfully amplifies the signal across a wide variety of protein maps, outperforming a classic map denoiser and following a network-based sharpening model. Without biasing the map, the proposed denoising method leads to improved visibility of protein structural features, including protein domains, secondary structure elements and modest high-resolution feature restoration. isi Crystal structure of a three-coordinate lithium complex with monodentate phenyloxazoline and hexamethyldisilylamide ligands By journals.iucr.org Published On :: 2024-05-17 The reaction of lithium hexamethyldisilylamide, [Li{N(Si(CH3)3)2}] (LiHMDS), with 4,4-dimethyl-2-phenyl-2-oxazoline (Phox, C11H13NO) in hexane produced colourless crystals of bis(4,4-dimethyl-2-phenyl-2-oxazoline-κN)(hexamethyldisilylamido-κN)lithium, [Li(C6H18NSi2)(C11H13NO)2] or [Li{N(Si(CH3)3)2}(Phox)2] in high yield (89%). Despite the 1:1 proportion of the starting materials in the reaction mixture, the product formed with a 1:2 amide:oxazoline ratio. In the unit cell of the C2/c space group, the neutral molecules lie on twofold rotation axes coinciding with the Li—N(amide) bonds. The lithium(I) centre adopts a trigonal–planar coordination geometry with three nitrogen donor atoms, one from the HMDS anion and two from the oxazolines. All ligands are monodentate. In the phenyloxazoline units, the dihedral angle defined by the five-membered heterocyclic rings is 35.81 (5)°, while the phenyl substituents are approximately face-to-face, separated by 3.908 (5) Å. In the amide, the methyl groups assume a nearly eclipsed arrangement to minimize steric repulsion with the analogous substituents on the oxazoline rings. The non-covalent interactions in the solid-state structure of [Li{N(Si(CH3)3)2}(Phox)2] were assessed by Hirshfeld surface analysis and fingerprint plots. isi Revisiting the hydrogenation behavior of NdGa and its hydride phases By journals.iucr.org Published On :: 2024-02-16 NdGa hydride and deuteride phases were prepared from high-quality NdGa samples and their structures characterized by powder and single-crystal X-ray diffraction and neutron powder diffraction. NdGa with the orthorhombic CrB-type structure absorbs hydrogen at hydrogen pressures ≤ 1 bar until reaching the composition NdGaH(D)1.1, which maintains a CrB-type structure. At elevated hydrogen pressure additional hydrogen is absorbed and the maximum composition recovered under standard temperature and pressure conditions is NdGaH(D)1.6 with the Cmcm LaGaH1.66-type structure. This structure is a threefold superstructure with respect to the CrB-type structure. The hydrogen atoms are ordered and distributed on three fully occupied Wyckoff positions corresponding to tetrahedral (4c, 8g) and trigonal–bipyramidal (8g) voids in the parent structure. The threefold superstructure is maintained in the H-deficient phases NaGaH(D)x until 1.6 ≥ x ≥ 1.2. At lower H concentrations, coinciding with the composition of the hydride obtained from hydrogenation at atmospheric pressure, the unit cell of the CrB-type structure is resumed. This phase can also display H deficiency, NdGaH(D)y (1.1 ≥ y ≥ 0.9), with H(D) exclusively situated in partially empty tetrahedral voids. The phase boundary between the threefold superstructure (LaGaH1.66 type) and the onefold structure (NdGaH1.1 type) is estimated on the basis of phase–composition isotherms and neutron powder diffraction to be x = 1.15. However, for measurements of samples in the sub-millimetre range, many experimental setups show limitations. Position drifts on the order of hundreds of micrometres during thermal stabilization of the system can last for hours of expensive beam time. To compensate for drifts, sample tracking and feedback systems must be used. However, in complex sample environments where sample access is very limited, many existing solutions cannot be applied. In this work, we apply a robust computer vision algorithm to automatically track and readjust the sample position in the dozens of micrometres range. Our approach is applied in a complex sample environment, where the sample is in an ultra-high vacuum chamber, surrounded by cooled thermal shields to reach sample temperatures down to 2.5 K and in the center of a superconducting split coil. Our implementation allows sample-position tracking and adjustment in the vertical direction since this is the dimension where drifts occur during sample temperature change in our setup. The approach can be easily extended to 2D. The algorithm enables a factor of ten improvement in the overlap of a series of X-ray absorption spectra in a sample with a vertical size down to 70 µm. This solution can be used in a variety of experimental stations, where optical access is available and sample access by other means is reduced. Full Article text

The more than 3,600-hectare project covers 64 claim blocks contiguous to projects owned by Sayona Mining Ltd. and Refined Metals Corp.'s Lac Simard property that sampled 2.1% lithium (4.52% lithium oxide or Li2O) and 5.88% tantalum oxide (Ta2O5), the company said. "This is the company's entry point into Quebec with the intention of building a strategic portfolio of hard rock lithium projects to complement our lithium brine assets," Director and Chief Executive Officer R. Nick Horsley said. "The company's long-term belief in a lithium price rebound is steadfast and now is the time to build a multi-jurisdictional lithium company." The Lac Simard South project is about 80 kilometers southwest of Sayona’s Authier lithium project and spans the townships of Beauneville, Clérion, Delbreuil, and ChabertIt, and is accessible by gravel road off Route 117 near the municipality of Cadillac, American Salars said. "The claims in the eastern sector are accessible by a network of logging roads; the southern and western sectors are accessible by boat or all-terrain vehicle and has very little overburden," the company said in a release. "It was on the outside of this piece of core," Callaghan said. "And then the core had split … and there was more gold on the inside of it, as well." Drill hole QGQ24-17 was terminated at a depth of 477.32 meters, and the "only thing that stopped us from drilling further was the claim boundary with Osisko," Callaghan said at the time. On Tuesday, the company announced rock sample results from its 2024 field campaign, which found up to 8.47 grams per tonne gold (g/t Au) in one outcrop in the Halo zone and 1.13 g/t Au in another outcrop near the Pioneer showing. "Although there is a lot of glacial cover on this project, our geologists still managed to find new gold-bearing outcrops in areas of great significance," Callaghan said in a release announcing the results. Drilling 'Nonstop' and 'Underbudget' Golden Cariboo, a Canadian explorer-developer, is targeting a potential multimillion-ounce gold resource at the 3,814-hectare Quesnelle project, where gold, silver, lead and zinc were produced historically, according to its Investor Presentation. The company's neighbors in the mining district include Osisko's Cariboo Gold Project, Spanish Mountain Gold Ltd. (SPA:TSX.V) (Spanish Mountain deposit), Omineca Mining and Metals Ltd. Levey and Phil Galewitz | NPRThe Biden administration is quietly engineering a series of expansions to Medicaid that may bolster protections for millions of low-income Americans and bring more people into the program. Biden's efforts — which have been largely overshadowed by other economic and health initiatives — represent an abrupt reversal of the Trump administration's moves to scale back the safety-net program. The changes could further boost Medicaid enrollment — which the pandemic has already pushed to a record 80.5 million. Some of the expansion is funded by the COVID-19 relief bill that passed in March, including coverage for new mothers. Others who could also gain coverage under Biden are inmates and undocumented immigrants. At the same time, the administration is opening the door to new Medicaid-funded services such as food and housing that the government insurance plan hasn't traditionally offered. "There is a paradigm change underway," said Jennifer Langer Jacobs, Medicaid director in New Jersey, one of a growing number of states trying to expand home-based Medicaid services to keep enrollees out of nursing homes and other institutions. "We've had discussions at the federal level in the last 90 days that are completely different from where we've ever been before," Langer Jacobs said. Taken together, the Medicaid moves represent some of the most substantive shifts in federal health policy undertaken by the new administration. "They are taking very bold action," said Rutgers University political scientist Frank Thompson, an expert on Medicaid history, noting in particular the administration's swift reversal of Trump policies. "There really isn't a precedent." The Biden administration seems unlikely to achieve what remains the holy grail for Medicaid advocates: getting 12 holdout states, including Texas and Florida, to expand Medicaid coverage to low-income working-age adults through the Affordable Care Act. And while some of the recent expansions – including for new mothers -- were funded by close to $20 billion in new Medicaid funding in the COVID relief bill Biden signed in March, much of that new money will stop in a few years unless Congress appropriates additional money. The White House strategy has risks. Medicaid, which swelled after enactment of the 2010 health law, has expanded further during the economic downturn caused by the pandemic, pushing enrollment to a record 80.5 million, including those served by the related Children's Health Insurance Program. That's up from 70 million before the COVID crisis began. The programs now cost taxpayers more than $600 billion a year. And although the federal government will cover most of the cost of the Biden-backed expansions, surging Medicaid spending is a growing burden on state budgets. The costs of expansion are a frequent target of conservative critics, including Trump officials like Seema Verma, the former administrator of the Centers for Medicare & Medicaid Services, who frequently argued for enrollment restrictions and derided Medicaid as low-quality coverage. But even less partisan experts warn that Medicaid, which was created to provide medical care to low-income Americans, can't make up for all the inadequacies in government housing, food and education programs. "Focusing on the social drivers of health ... is critically important in improving the health and well-being of Medicaid beneficiaries. But that doesn't mean that Medicaid can or should be responsible for paying for all of those services," said Matt Salo, head of the National Association of Medicaid Directors, noting that the program's financing "is simply not capable of sustaining those investments." Restoring federal support However, after four years of Trump administration efforts to scale back coverage, Biden and his appointees appear intent on not only restoring federal support for Medicaid, but also boosting the program's reach. "I think what we learned during the repeal-and-replace debate is just how much people in this country care about the Medicaid program and how it's a lifeline to millions," Biden's new Medicare and Medicaid administrator, Chiquita Brooks-LaSure, told KHN, calling the program a "backbone to our country." The Biden administration has already withdrawn permission the Trump administration had granted Arkansas and New Hampshire to place work requirements on some Medicaid enrollees. In April, Biden blocked a multibillion-dollar Trump administration initiative to prop up Texas hospitals that care for uninsured patients, a policy that many critics said effectively discouraged Texas from expanding Medicaid coverage through the Affordable Care Act, often called Obamacare. Texas has the highest uninsured rate in the nation. The moves have drawn criticism from Republicans, some of whom accuse the new administration of trampling states' rights to run their Medicaid programs as they choose. "Biden is reasserting a larger federal role and not deferring to states," said Josh Archambault, a senior fellow at the conservative Foundation for Government Accountability. But Biden's early initiatives have been widely hailed by patient advocates, public health experts and state officials in many blue states. "It's a breath of fresh air," said Kim Bimestefer, head of Colorado's Department of Health Care Policy and Financing. Chuck Ingoglia, head of the National Council for Mental Wellbeing, said: "To be in an environment where people are talking about expanding health care access has made an enormous difference." Mounting evidence shows that expanded Medicaid coverage improves enrollees' health, as surveys and mortality data in recent years have identified greater health improvements in states that expanded Medicaid through the 2010 health law versus states that did not. Broadening eligibility In addition to removing Medicaid restrictions imposed by Trump administration officials, the Biden administration has backed a series of expansions to broaden eligibility and add services enrollees can receive. Biden supported a provision in the COVID relief bill that gives states the option to extend Medicaid to new mothers for up to a year after they give birth. Many experts say such coverage could help reduce the U.S. maternal mortality rate, which is far higher than rates in other wealthy nations. Several states, including Illinois and New Jersey, had sought permission from the Trump administration for such expanded coverage, but their requests languished. The COVID relief bill — which passed without Republican support — also provides additional Medicaid money to states to set up mobile crisis services for people facing mental health or substance use emergencies, further broadening Medicaid's reach. And states will get billions more to expand so-called home and community-based services such as help with cooking, bathing and other basic activities that can prevent Medicaid enrollees from having to be admitted to expensive nursing homes or other institutions. Perhaps the most far-reaching Medicaid expansions being considered by the Biden administration would push the government health plan into covering services not traditionally considered health care, such as housing. This reflects an emerging consensus among health policy experts that investments in some non-medical services can ultimately save Medicaid money by keeping patients out of the hospital. In recent years, Medicaid officials in red and blue states — including Arizona, California, Illinois, Maryland and Washington — have begun exploring ways to provide rental assistance to select Medicaid enrollees to prevent medical complications linked to homelessness. The Trump administration took steps to support similar efforts, clearing Medicare Advantage health plans to offer some enrollees non-medical benefits such as food, housing aid and assistance with utilities. But state officials across the country said the new administration has signaled more support for both expanding current home-based services and adding new ones. That has made a big difference, said Kate McEvoy, who directs Connecticut's Medicaid program. "There was a lot of discussion in the Trump administration," she said, "but not the capital to do it." Other states are looking to the new administration to back efforts to expand Medicaid to inmates with mental health conditions and drug addiction so they can connect more easily to treatment once released. Kentucky health secretary Eric Friedlander said he is hopeful federal officials will sign off on his state's initiative. Still other states, such as California, say they are getting a more receptive audience in Washington for proposals to expand coverage to immigrants who are in the country without authorization, a step public health experts say can help improve community health and slow the spread of communicable diseases. "Covering all Californians is critical to our mission," said Jacey Cooper, director of California's Medicaid program, known as Medi-Cal. "We really feel like the new administration is helping us ensure that everyone has access." The Trump administration moved to restrict even authorized immigrants' access to the health care safety net, including the "public charge" rule that allowed immigration authorities to deny green cards to applicants if they used public programs such as Medicaid. The White House issued a formal announcement of the trip shortly afterward. The official death toll in the collapse has risen to 11, with some 150 people unaccounted for. The Biden administration has responded to the disaster, dispatching FEMA administrator Deanne Criswell to the scene earlier this week. "[The agency] has deployed an Incident Management Assistance Team, as well as building science experts, structural engineers and geotechnical experts to support search-and-rescue operations, and a mobile command center," White House press secretary Jen Psaki said Monday. Psaki said the U.S. Army Corps of Engineers is also providing technical assistance for debris removal. Two FEMA-supported search-and-rescue teams are also involved in the response to the collapse. Florida Gov. Ron DeSantis has praised FEMA and the Biden administration for "stepping up to the plate" in providing assistance in the search and recovery effort. Miami-Dade Mayor Daniella Levine Cava said Biden's upcoming trip would be "an important reminder that our county, our state and our nation are giving everything we have to search for the victims of this tragedy and support the families in this incredibly devastating time." Last week, the USC Annenberg Inclusion Initiative released a report documenting the prevalence of Asian and Pacific Islanders both on-and off-screen across the top-grossing films each year from 2007 to 2019. Of the 1,300 films examined, only 44 featured API actors playing lead roles, nearly a third of which were played by Johnson. The report offers more staggering statistics: In 2019, over a quarter of API characters in the top-grossing films died. Most died by drowning, explosions, stabbing or suicides Of the over 51,000 speaking characters in the 1,300 films examined, only 6% were Asian, Asian American or Native Hawaiian/Pacific Islanders Only 50 of the 1,447 directors in the 1,300 films examined were of API heritage. It was the first cosmic event detected in both gravitational waves and the entire spectrum of light, from gamma rays to radio emissions. The impact also created a kilonova -- a turbocharged explosion that instantly forged several hundred planets’ worth of gold and platinum. The observations provided the first compelling evidence that kilonovae produce large quantities of heavy metals, a finding long predicted by theory. Astronomers suspect that all of the gold and platinum on Earth formed as a result of ancient kilonovae created during neutron star collisions. Based on data from the 2017 event, first spotted by the Laser Interferometer Gravitational-wave Observatory (LIGO), astronomers began to adjust their assumptions of how a kilonova should appear to Earth-bound observers. The agreement was reached by U.S. special envoy for climate John Kerry and his Chinese counterpart Xie Zhenhua during two days of talks in Shanghai last week, according to a joint statement. The two countries "are committed to cooperating with each other and with other countries to tackle the climate crisis, which must be addressed with the seriousness and urgency that it demands," the statement said. China is the world's biggest carbon emitter, followed by the United States. The two countries pump out nearly half of the fossil fuel fumes that are warming the planet's atmosphere. Their cooperation is key to a success of global efforts to curb climate change, but frayed ties over human rights, trade and China's territorial claims to Taiwan and the South China Sea have been threatening to undermine such efforts. Meeting with reporters in Seoul on Sunday, Kerry said the language in the statement is "strong" and that the two countries agreed on "critical elements on where we have to go." But the former secretary of state said, "I learned in diplomacy that you don't put your back on the words, you put on actions. We all need to see what happens." Noting that China is the world's biggest coal user, Kerry said he and Chinese officials had a lot of discussions on how to accelerate a global energy transition. "I have never shied away from expressing our views shared by many, many people that it is imperative to reduce coal, everywhere," he said. Biden has invited 40 world leaders, including Chinese President Xi Jinping, to the April 22-23 summit. The U.S. and other countries are expected to announce more ambitious national targets for cutting carbon emissions ahead of or at the meeting, along with pledging financial help for climate efforts by less wealthy nations. It's unclear how much Kerry's China visit would promote U.S.-China cooperation on climate issues. While Kerry was still in Shanghai, Chinese Vice Foreign Minister Le Yucheng signaled Friday that China is unlikely to make any new pledges at next week's summit. "For a big country with 1.4 billion people, these goals are not easily delivered," Le said during an interview with The Associated Press in Beijing. "Some countries are asking China to achieve the goals earlier. I am afraid this is not very realistic." During a video meeting with German and French leaders Friday, Xi said that climate change "should not become a geopolitical chip, a target for attacking other countries or an excuse for trade barriers," the official Xinhua News Agency reported. On whether Xi would join the summit, Le said "the Chinese side is actively studying the matter." The joint statement said the two countries "look forward to" next week's summit. Major emitters of greenhouse gases are preparing for the next U.N. climate summit taking place in Glasgow, U.K., in November. The summit aims to relaunch global efforts to keep rising global temperatures to below 1.5 degrees Celsius (2.7 degrees Fahrenheit) as agreed in the Paris accord. According to the U.S.-China statement, the two countries would enhance "their respective actions and cooperating in multilateral processes, including the United Nations Framework Convention on Climate Change and the Paris Agreement." It said both countries also intend to develop their respective long-term strategies before the Glasgow conference and take "appropriate actions to maximize international investment and finance in support of" the energy transition in developing countries. It's an ambitious goal that requires transforming much of the economy. Renewable energy would need to make up half of the U.S. power supply from roughly 21% currently. Electric cars make up about 2% of sales now — by 2030, at least half, potentially all, new car sales would need to be electric, according to estimates. Many industrial manufacturing facilities would need to use technologies that haven't been developed. It's part of Biden's effort to get the U.S. on track to reach the goals of the 2015 Paris Climate Agreement to keep global temperatures from rising more than 1.5 degrees Celsius. Former President Donald Trump withdrew the U.S. from the agreement but Biden has formally rejoined. John Kerry is Biden's special envoy for climate, a position that involves meeting with countries around the world about efforts to stem emissions. He calls the threat of climate change "existential." "That means life and death. And the question is, are we behaving as if it is? And the answer is no," Kerry said in an interview on NPR's All Things Considered. This interview has been edited for length and clarity and includes extended Web-only answers. Interview Highlights Is this more a matter of shoot for the moon and if you miss, at least you'll land among the stars? No, I think it's achievable. And I think that people who've really studied this, analyzed it and thought about it for a long period of time believe it is achievable. Already the [car] marketplace is moving towards electric. I mean, you know, Joe Biden didn't create the value of Tesla as the most valuable automobile company in the world. The market did that. And the market did it because that's where people are moving. The scale of change that you're talking about in the timeframe that is required is something we've never seen in human history. Let me put it to you this way. How many politicians, how many scientists, how many people have stood up and said, "This is existential for us on this planet"? Existential. That means life and death. And the question is, are we behaving as if it is? And the answer is no. So why are younger generation folks so angry? Why are they standing up and demonstrating and asking adults to accept adult responsibility to move our nations in the right direction? Because the scientists are telling them that. They learn about this in high school and college. They read. They know what's happening. They know we're experiencing the hottest day in human history, the hottest week, the hottest month, the hottest year. And we see the results. Fires, floods, mudslides, drought, crop disruption, ice melting in the Arctic, run the list. Climate change is still seen as a partisan issue in the U.S., and Republicans could take over Congress next year. A Republican could win the White House in three years. So why should global leaders view this as a reliable commitment from the United States when GOP leaders have not bought in? For two reasons. No. 1, when Donald Trump was president of the United States and he pulled out of the agreement, 37 governors in the United States, Republican and Democrats alike, stood up and said, "We're still in." And states, those 37 states, have passed renewable portfolio laws. So at the state level, people are moving because they know it's better for their state. It's a safer, better delivery of power to their state, and it's the way it's going to move. The second part of the answer: Masses of capital, trillions of dollars, are going to move into the energy market, which is the largest market the world has ever seen and going to grow now. Multiple double-digit trillions of dollars of market. And no politician can come along and tell those banks, or those asset managers or those investors or those venture capitalists or the companies, the corporations that are doing this, they know this is where the market's going to be in the future. If the $2 trillion infrastructure and jobs plan that the president has put forth does not pass the Senate, does this goal to cut emissions in half by 2030 effectively die with the bill? Well, it doesn't die, but it certainly takes a blow, a serious one. But the companies I've talked about are going to move in this direction no matter what. I mean, if you look at the biggest companies in America, these folks are all pushing to get this done because they know that the world is going to be better off and that their businesses are going to be better off if we do that. This is a real challenge for all of us, and I think people are waking up to it all around the world. Let me ask you a question. Why do you think 40 heads of state, including President Xi of China, President Putin of Russia, Prime Minister Modi of India, huge populations come together and say, "We have to do this"? Do they know something that some of these opponents of it don't know or aren't willing to admit? I mean, the only leader in the entire world that saw fit to pull out of the Paris agreement was Donald Trump. But it's so easy to make commitments and we haven't seen countries follow through on those commitments. This is accurate. They're doing things; they're not doing enough. There are very few countries that are doing enough. Most countries are not. And of the 20 countries that equal 81% of all the emissions, they are the critical ones that have to do more. isi How Do You Help Girls Thrive In School? But a new study, published in May in the journal World Bank Economic Review, begs to differ. Researchers David Evans and Fei Yuan reviewed 267 studies of education programs from 54 low- and middle-income countries to find the most effective ways to get more girls in school and improve their learning. Globally, more than 130 million girls remain out of school, according to the World Bank, due to poverty, child marriage and violence. Instead of only examining girls' education programs, they looked at all kinds of programs. To measure access, they analyzed enrollment rates, attendance, drop-out, graduation and completion rates, and to measure performance, they looked at test scores. Their biggest finding is that gender-neutral programs — such as handing out cash aid to families of school-aged children — can be just as effective at improving girls' education as programs designed just for girls. The study is among the first to look both at ways to boost girls' access to school as well as their classroom performance, says Markus Goldstein, lead economist at the World Bank's Africa Gender Innovation Lab, who did not work on the report. We spoke with Evans, a senior fellow at the Center for Global Development, and Yuan, a doctoral candidate in education policy and program evaluation at Harvard University, to discuss the best ways to boost education for girls in low- and middle-income countries. This interview has been edited for length and clarity. What inspired you to conduct this study? Evans: A lot of the previous work that examined this issue have focused on programs targeted to needs that are unique to girls, such as menstrual health. Those are worthy interventions, but if we only focus on programs that target girls, we might miss programs that benefit girls a lot but happen to help boys as well. That's why we decided to look at all of the interventions we know of to identify the ones that are most effective at improving outcomes for girls, regardless of whether they're specifically for girls or not. You found that the most effective programs for getting more girls into school cut the cost of education for students, regardless of gender, and their families. What are some examples of programs that worked well? Evans: A lot of the most effective programs are ones that either eliminate school fees, provide scholarships or provide families a cash transfer to cover the other costs of having their daughter in school. For example, in Ghana, lots of girls and boys pass their secondary school entrance exam, but they don't have the money to pay school fees. So, a program there provided scholarships to students who had already passed the entrance exam. It dramatically increased the high-school graduation rate of girls by 66%. But the most effective interventions are those that address costs related to specific obstacles that girls face in a particular setting. In Afghanistan, for example, a [non-gendered] program built schools in rural communities. It decreased [the cost of] travel to school for both girls and boys and led to a more than 50% increase in girls' participation in primary school. That's dramatic. Which programs were the most helpful for improving a girl's school performance, as opposed to just getting them into the classroom? Evans: The most effective interventions to increase learning were programs that improved the quality of teaching. But it's not just throwing teachers into a conference room and giving them some lecture. It's also not about throwing fancy technology, like laptops or tablets, at classrooms. Hardware doesn't work. It's distracting for teachers and students. Instead, a literacy program – which included coaching teachers, providing them with detailed teachers' guides and providing students with books – had a big impact on girls' education [in terms of test scores] in Kenya. So did another program in Kenya that helped teachers to teach children in a language they spoke at home (rather than English). Were there any other types of programs that helped girls learn better in the classroom? Yuan: Another intervention worth mentioning is called Teaching at the Right Level, based in India. The idea is that students in the same classroom may have many different reading levels. But because of constraints like large class sizes, teachers may not be able to tailor their teaching to the right level for every student. This leaves some students behind. Teaching at the Right Level facilitated summer camps in which children were grouped by reading level, instead of age or grade. This allowed teachers to target their teaching to the specific levels of these students. In one region, after 50 days of focused teaching in these camps, children at the lowest achievement levels in India were able to catch up to the learning level of the third-highest achieving state in the country. Many of the high-impact interventions you're referencing don't target girls specifically. Are you saying that girls' programs aren't necessary? Evans: Not at all! We particularly focused on how to increase access to education and improve quality of learning. Some [girl-focused] programs have other goals – such as reducing violence against girls, improving girls' psychological and emotional wellbeing, reducing adolescent pregnancy or helping girls to transition from school to the workforce. But when teaching is of bad quality, we just need to help schools improve the teaching. That's not necessarily a gender-specific problem. Wouldn't it be more cost-effective to just offer scholarships or cash transfers to girls only instead of both genders, especially if far fewer girls are attending school than boys? Evans: Sure, if you don't have the budget to waive school fees for everyone, eliminating school fees for girls is an effective way to do a girl-targeted program. That's what The Gambia did. But sometimes general, non-targeted interventions are more politically palatable for governments, since constituents have both daughters and sons. Were you concerned that some of the gender-neutral programs might benefit boys more than girls? Evans: That was something we were worried about – increasing inequality. But we found that overall, the impact of gender-neutral programs tends to be slightly larger on girls than boys both in terms of access and learning. These differences, for the most part, were not statistically significant. They were small. But it does mean that these general, non-targeted interventions are not increasing inequality between boys and girls. If anything, they're likely to decrease it. What changes do you hope to see in how we work on girls' education around the world? Evans: We want to make sure that people who care about girls' education draw on the full toolbox of programs that can improve girls' education. That includes girl-targeted programs. It also includes general programs. We don't anyone to walk away from this and say, 'Oh, we don't need to worry about girls.' In the Court's view, 270 months, which amounts to an additional ten years over the presumptive 150-month sentence, is the appropriate sentence."
