Sleep restless, anxiety dreams, and if there wasn’t enough to be concerned about I am worrying about our baby beans and peas.

It is often like this in spring. The responsibility, it comes with the shorter nights and longer light, maybe I have more time on my hands. I have saved two hours a day on travelling and I only work a few miles’ walk from home. This extra time has now become a trip to the plot, or perhaps pottering on the terrace. A more intimate gardening relationship cemented in the spring mornings. Deepened, more dependent.

Crystallographic snapshots of the EF-hand protein MCFD2 complexed with the intracellular lectin ERGIC-53 involved in glycoprotein transport

The transmembrane intracellular lectin ER–Golgi intermediate compartment protein 53 (ERGIC-53) and the soluble EF-hand multiple coagulation factor deficiency protein 2 (MCFD2) form a complex that functions as a cargo receptor, trafficking various glycoproteins between the endoplasmic reticulum (ER) and the Golgi apparatus. It has been demonstrated that the carbohydrate-recognition domain (CRD) of ERGIC-53 (ERGIC-53CRD) interacts with N-linked glycans on cargo glycoproteins, whereas MCFD2 recognizes polypeptide segments of cargo glycoproteins. Crystal structures of ERGIC-53CRD complexed with MCFD2 and mannosyl oligosaccharides have revealed protein–protein and protein–sugar binding modes. In contrast, the polypeptide-recognition mechanism of MCFD2 remains largely unknown. Here, a 1.60 Å resolution crystal structure of the ERGIC-53CRD–MCFD2 complex is reported, along with three other crystal forms. Comparison of these structures with those previously reported reveal that MCFD2, but not ERGIC-53–CRD, exhibits significant conformational plasticity that may be relevant to its accommodation of various polypeptide ligands.

Crystal structure and Hirshfeld surface analysis of new polymorph of racemic 2-phenyl­butyramide

A new polymorph of the title compound, C10H13NO, was obtained by recrystallization of the commercial product from a water/ethanol mixture (1:1 v/v). Crystals of the previously reported racemic and homochiral forms of 2-phenyl­butyramide were grown from water–aceto­nitrile solution in 1:1 volume ratio [Khrustalev et al. (2014). Cryst. Growth Des. 14, 3360–3369]. While the previously reported racemic and enanti­opure forms of the title compound adopt very similar supra­molecular structures (hydrogen-bonded ribbons), the new racemic polymorph is stabilized by a single N—H⋯O hydrogen bond that links mol­ecules into chains along the c-axis direction with an anti­parallel (centrosymmetric) packing in the crystal. Hirshfeld mol­ecular surface analysis was employed to compare the inter­molecular inter­actions in the polymorphs of the title compound.

Reaction of 2-(anthracen-9-yl)phenol (HOPhAn, 1) with divalent Yb[N(SiMe3)2]2·2THF in THF–toluene mixtures affords the mixed-valence YbII–YbIII dimer {[2-(anthracen-9-yl)phenolato-κO]bis­(tetra­hydro­furan)­ytterbium(III)}-tris­[μ-2-(anthracen-9-yl)phenolato]-κ4O:O;κO:1,2-η,κO-{[2-(anthracen-9-yl)phenolato-κO]ytterbium(II)} toluene tris­olvate, [Yb2(C20H13O)5(C4H8O)2]·3C7H7 or [YbIII(THF)2(OPhAn)](μ-OPhAn)3[YbII(OPhAn)]·3C7H7 (2), as the major product. It crystallized as a toluene tris­olvate. The Yb—O bond lengths in the crystal structure of this dimer clearly identify the YbII and YbIII centres. Inter­estingly, the formally four-coordinate YbII centre shows a close contact with one anthracene C—C bond of a bridging OPhAn ligand, bringing the formal coordination number to five.

In the mol­ecule of the title anthracene derivative, C22H17NO2, the benzene ring is inclined to the mean plane of the anthracene ring system (r.m.s. deviation = 0.024 Å) by 75.21 (9)°. In the crystal, mol­ecules are linked by pairs of O—H⋯O hydrogen bonds, forming classical carb­oxy­lic acid inversion dimers with an R22(8) ring motif. The dimers are linked by C—H⋯π inter­actions, forming a supra­molecular framework.

The title compound, C22H17NO2·C3H7NO, was synthesized by condensation of an aromatic aldehyde with a secondary amine and subsequent reduction. It was crystallized from a di­methyl­formamide solution as a monosolvate, C22H17NO2·C3H7NO. The aromatic mol­ecule is non-planar with a dihedral angle between the mean planes of the aniline moiety and the methyl anthracene moiety of 81.36 (8)°. The torsion angle of the Car­yl—CH2—NH—Car­yl backbone is 175.9 (2)°. The crystal structure exhibits a three-dimensional supra­molecular network, resulting from hydrogen-bonding inter­actions between the carb­oxy­lic OH group and the solvent O atom as well as between the amine functionality and the O atom of the carb­oxy­lic group and additional C—H⋯π inter­actions. Hirshfeld surface analysis was performed to qu­antify the inter­molecular inter­actions.

On the mechanism of solid-state phase transitions in molecular crystals – the role of cooperative motion in (quasi)racemic linear amino acids

During single-crystal-to-single-crystal (SCSC) phase transitions, a polymorph of a compound can transform to a more stable form while remaining in the solid state. By understanding the mechanism of these transitions, strategies can be developed to control this phenomenon. This is particularly important in the pharmaceutical industry, but also relevant for other industries such as the food and agrochemical industries. Although extensive literature exists on SCSC phase transitions in inorganic crystals, it is unclear whether their classications and mechanisms translate to molecular crystals, with weaker interactions and more steric hindrance. A comparitive study of SCSC phase transitions in aliphatic linear-chain amino acid crystals, both racemates and quasi-racemates, is presented. A total of 34 transitions are considered and most are classified according to their structural change during the transition. Transitions without torsional changes show very different characteristics, such as transition temperature, enthalpy and free energy, compared with transitions that involve torsional changes. These differences can be rationalized using classical nucleation theory and in terms of a difference in mechanism; torsional changes occur in a molecule-by-molecule fashion, whereas transitions without torsional changes involve cooperative motion with multiple molecules at the same time.

An extracellular domain of the EsaA membrane component of the type VIIb secretion system: expression, purification and crystallization

The membrane protein EsaA is a conserved component of the type VIIb secretion system. Limited proteolysis of purified EsaA from Staphylococcus aureus USA300 identified a stable 48 kDa fragment, which was mapped by fingerprint mass spectrometry to an uncharacterized extracellular segment of EsaA. Analysis by circular dichroism spectroscopy showed that this fragment folds into a single stable domain made of mostly α-helices with a melting point of 34.5°C. Size-exclusion chromatography combined with multi-angle light scattering indicated the formation of a dimer of the purified extracellular domain. Octahedral crystals were grown in 0.2 M ammonium citrate tribasic pH 7.0, 16% PEG 3350 using the hanging-drop vapor-diffusion method. Diffraction data were analyzed to 4.0 Å resolution, showing that the crystals belonged to the enantiomorphic tetragonal space groups P41212 or P43212, with unit-cell parameters a = 197.5, b = 197.5, c = 368.3 Å, α = β = γ = 90°.

Election 2015: iPad controversy looms large in LAUSD District 3 board race

As the city's March 3 primary election draws near, Los Angeles Unified school board candidates are blasting incumbents for the controversial iPad program.

Opponents sharply criticized the $1.3 billion bond-funded program at a debate Tuesday in West San Fernando Valley, where District 3 school board member Tamar Galatzan was elected in 2007.

"Galatzan said the district is going in the right direction," declared candidate Carl Petersen, a parent and businessman. "I don’t know how anyone can look at the events of the past year and come to that conclusion."

The program attracted national attention last December when the FBI raided district offices and carted off 20 boxes of bids, evaluations and correspondences with executives at Apple and its subcontractor Pearson, the manufacturer of the learning software loaded on to each device. The investigation is ongoing.

At the debate, teachers dressed in dark windbreakers with FBI plastered on the back in protest to Galatzan's support of the program. (They have not held similar demonstrations at election events in East Los Angeles' District 5, where Bennett Kayser, a teacher union ally, is running for re-election.)

Tom Richards, a Granada Hills parent, said he considers the iPad program a central issue as he weighs candidates.

"I think it's absolutely ridiculous," Richards said. "I don't believe that's a good way to spend the money that they have. Looking at some really fundamental needs — we don't have a librarian, but we want to give iPads?" 

Galatzan was an early advocate for more technology in the classroom; it was her goal even before the iPad was on the market.

"There is a whole world out there that can be accessed through technology, and we need to take advantage of that," Galatzan told KPCC.

Her advocacy of technology hasn't always been controversial. Galatzan points to her 2010 initiative to fund school computer labs with a settlement from Microsoft.

The school board's support of the iPad program varied the first year, but waned in August after KPCC published a series of emails showing district administrators had close ties with Pearson, calling into question whether the bidding process was fair. Problems with the rollout of the devices and the effectiveness of the software they contained also eroded support for the program.

Still, school board members unanimously approved more iPad purchases after the FBI investigation came to light. Superintendent Ramon Cortines said the tablets were necessary for new digital state tests scheduled this spring and offered to purchase them under a different contract with Apple to avoid complications involving the federal probe.

If the candidates' positions are a measure of support for the program, it's unpopular at best.  All of Galatzan's opponents are against it. 

When asked in a KPCC election survey conducted if he supported the iPad program, Scott Schmerelson, a retired administrator and District 3 contender, responded: "Not when you are paying for them from LAUSD Bond Money! The taxpayers generously supported the bond issue with the belief that the money would be used to repair and modernize our schools." 

Candidate Ankur Patel said in his answer to the survey, "I oppose the LAUSD’s iPad program. Throughout the program, important questions were not asked enough, and when they were, they were not answered properly."

Filiberto Gonzalez, another Galatzan challenger, said of the iPad project: "It was a mistake and ill-conceived from the very beginning. As was noted in the report by the U.S. Department of Education last month, the Common Core Technology Project (iPad program) lacked 'established metrics of success' and 'was difficult to show the impact of the investment.'

Elizabeth Badger Bartels is also running for the District 3 seat, but did not respond to the survey by deadline.
