Multi-species cryoEM calibration and workflow verification standard
Cryogenic electron microscopy (cryoEM) is a rapidly growing structural biology modality that has been successful in revealing molecular details of biological systems. However, unlike established biophysical and analytical techniques with calibration standards, cryoEM has lacked comprehensive biological test samples. Here, a cryoEM calibration sample consisting of a mixture of compatible macromolecules is introduced that can not only be used for resolution optimization, but also provides multiple reference points for evaluating instrument performance, data quality and image-processing workflows in a single experiment. This combined test specimen provides researchers with a reference point for validating their cryoEM pipeline, benchmarking their methodologies and testing new algorithms.

Crab from the Chinese pet market turns out to be a new species of a new genus

Genetic analysis uncovers 4 species of giraffe, not just 1
Up until now, scientists had only recognized a single species of giraffe made up of several subspecies. But, according to the most inclusive genetic analysis of giraffe relationships to date, giraffes actually aren't one species, but four. For comparison, the genetic differences among giraffe species are at least as great as those between polar and brown bears.

“Killer sperm” prevents mating between worm species
TORONTO, ON — The classic definition of a biological species is the ability to breed within its group, and the inability to breed outside it. A study published today in the journal PLOS Biology offers some important clues about the evolution of barriers to breeding. The vast majority of the time, mating across species is […]

Red-cockaded woodpeckers are no longer an endangered species
The red-cockaded woodpecker, an iconic bird in southeastern forests, has grown more numerous. So the Fish and Wildlife Service no longer lists it as an endangered species, instead classifying it as a threatened species. The Associated Press reports: in the 1970s, the red-cockaded woodpecker population had dipped as low as 1,470 clusters — or groups […]

Interspecies differences in proteome turnover kinetics are correlated with lifespans and energetic demands
Interspecies differences in proteome turnover kinetics are correlated with lifespans and energetic demands [Research]
Cells continually degrade and replace damaged proteins. However, the high energetic demand of protein turnover generates reactive oxygen species (ROS) that compromise the long-term health of the proteome. Thus, the relationship between aging, protein turnover and energetic demand remains unclear. Here, we used a proteomic approach to measure rates of protein turnover within primary fibroblasts isolated from a number of species with diverse lifespans including the longest-lived mammal, the bowhead whale. We show that organismal lifespan is negatively correlated with turnover rates of highly abundant proteins. In comparison to mice, cells from long-lived naked mole rats have slower rates of protein turnover, lower levels of ATP production and reduced ROS levels. Despite having slower rates of protein turnover, naked mole rat cells tolerate protein misfolding stress more effectively than mouse cells. We suggest that in lieu of rapid constitutive turnover, long-lived species may have evolved more energetically efficient mechanisms for selective detection and clearance of damaged proteins.
Transcriptomic Correlates of State Modulation in GABAergic Interneurons: A Cross-Species Analysis
GABAergic inhibitory interneurons comprise many subtypes that differ in their molecular, anatomical, and functional properties. In mouse visual cortex, they also differ in their modulation with an animal’s behavioral state, and this state modulation can be predicted from the first principal component (PC) of the gene expression matrix. Here, we ask whether this link between transcriptome and state-dependent processing generalizes across species. To this end, we analysed seven single-cell and single-nucleus RNA sequencing datasets from mouse, human, songbird, and turtle forebrains. Despite homology at the level of cell types, we found clear differences between transcriptomic PCs, with greater dissimilarities between evolutionarily distant species. These dissimilarities arise from two factors: divergence in gene expression within homologous cell types and divergence in cell-type abundance. We also compare the expression of cholinergic receptors, which are thought to causally link transcriptome and state modulation. Several cholinergic receptors predictive of state modulation in mouse interneurons are differentially expressed between species. Circuit modelling and mathematical analyses suggest conditions under which these expression differences could translate into functional differences.
