This modification sterically blocks binding of several classes of antibiotics to 23S rRNA, resulting in a multidrug-resistant phenotype in bacteria expressing the enzyme. ErmC is an erythromycin resistance methyltransferase found in many Gram-positive pathogens, whereas ErmE is found in the soil bacterium that biosynthesizes erythromycin. Whether ErmC and ErmE, which possess only 24% sequence identity, use similar structural elements for rRNA substrate recognition and positioning is not known. To investigate this question, we used structural data from related proteins to guide site-saturation mutagenesis of key residues and characterized selected variants by antibiotic susceptibility testing, single turnover kinetics, and RNA affinity–binding assays. We demonstrate that residues in α4, α5, and the α5-α6 linker are essential for methyltransferase function, including an aromatic residue on α4 that likely forms stacking interactions with the substrate adenosine and basic residues in α5 and the α5-α6 linker that likely mediate conformational rearrangements in the protein and cognate rRNA upon interaction. The functional studies led us to a new structural model for the ErmC or ErmE-rRNA complex. Full Article

Even before the COVID-19 pandemic, it was clear that the world is undergoing a transition away from fossil fuels and carbon-intensive sectors, towards renewable energy and clean growth. The collapse of oil demand and prices have simply compounded the challenges that oil and gas producers already faced.What happens next will have significant implications for Norway, as one of the world’s largest exporters of both energy and capital, and for the UK, as it plans its recovery and looks ahead to its hosting of the next major climate change summit in 2021 - COP26.While the speed and scale of the transition has always been uncertain and contested, an accelerated transition with deep implications for future oil and gas demand looks plausible.There has long been a debate over when global demand will peak, but what happens after demand has peaked is perhaps the more critical question. Now there is the additional uncertainty of how this post-peak demand might be affected by an oncoming global recession and potentially by the greening of recovery measures implemented in response to it. Will there be an extended plateau, a gentle decline or a sudden collapse?The post-peak trend will impact oil producers and exporters to varying degrees, in terms of their vulnerability to reduced volumes and lower prices, and their ability to compete in a shrinking market. There is also growing scepticism over whether natural gas can act as a bridge between coal-fired power and renewables, as increasingly, renewables directly replace coal. There is also significant uncertainty over extent to which hydrogen, either produced from fossil fuels or renewable energy, will play a significant role in a decarbonizing energy sector.Even before the pandemic, there was growing public and political pressure in most EU member states for more ambitious action on climate change. More challenging climate targets now look certain as a growing number of governments and companies commit to becoming carbon-neutral by ever-earlier dates.While market developments, such as the rate of change and the costs of technologies such as renewable energy and electric vehicles will heavily influence their deployment rates, policy interventions and large-scale investment in core infrastructure are still crucial to their scaling up. We are now seeing the EU refocus its Green Deal in support of post-COVID recovery, and scale its support for transition in coal-dependent and carbon-intensive regions with its €100bn Just Transition Mechanism. These developments have significant implications for fossil fuel producers and energy consumers both inside and outside the EU. It will particularly affect Norway, not only as a significant supplier of energy to the EU, but as a member of the European Economic Area, with likely pressure to adopt similarly binding domestic carbon reduction legislation. Similarly, as the UK forges new post-Brexit trading and regulatory relationships, it will need to align with European policies for efficiency.As the host of the critical COP26 UN Climate Change Summit in Glasgow next year, the UK will also need to at least match the EU in terms of its ambition on national emissions reductions, and in placing decarbonization and sustainability at the heart of COVID-19 recovery measures. However, unfortunately, the early indications are that 'Project Speed' will focus on traditional infrastructure projects are less than promising. The UK and Norway face similar challenges, as oil and gas producers that recognize the importance of climate change, and will rightly face scrutiny where they reinvest in their oil and gas sectors. They are both outside, yet highly dependent on developments within the EU. However, they are also both, somewhat surprisingly, world leaders in different aspects of decarbonization, such as off-shore wind or electric vehicle deployment, in part due their offshore capabilities and advanced manufacturing capabilities. This presents an opportunity for both countries and their industries to place an accelerated energy transition at the heart of their economic recovery and their relationship with the EU.There will of course be different opinions on how to do this. A new Chatham House paper – Expert Perspectives on Norway’s Energy Future – explores these issues in the Norwegian context, and draws upon the views of 15 international experts on energy transition and climate change, each interviewed in depth. While unsurprisingly there is little consensus, these views provide valuable background from which to consider the future of future of energy for Norway, and for its partners including the UK and the EU. We found that in Drosophila melanogaster, a set of neurons, which we call CL062, previously shown to mediate male aggression also mediate female aggression. These neurons elicit aggression acutely and without the presence of a target. Although the same set of actions is elicited in males and females, the overall behavior is sexually dimorphic. The CL062 neurons do not express fruitless, a gene required for sexual dimorphism in flies, and expressed by most other neurons important for controlling fly aggression. Connectomic analysis in a female electron microscopy dataset suggests that these neurons have limited connections with fruitless expressing neurons that have been shown to be important for aggression and signal to different descending neurons. Thus, CL062 is part of a monomorphic circuit for aggression that functions parallel to the known dimorphic circuits. Full Article

Dubbed by one user as the “nerve center” of the Silicon Beach tech scene, Cross Campus opened its membership-based workspace facility in Santa Monica in 2012. But co-founder Ronen Olshansky said the shared workspace phenomenon isn't limited to coders. "Fewer and fewer people are making the traditional drive into the corporate office," Olshansky said. "They're working remotely as professionals, going off on their own as freelancers, or they're starting their own companies as entrepreneurs." A forecast from Forrester Research says that 43 percent of workers will telecommute by 2016, compared to estimates of about a quarter of the workforce telecommuting last year. Olshansky said that, for many people, working from home or in a coffee shop isn't productive. That's led shared workspaces to pop up in Los Angeles, Culver City and Santa Monica. Among them: Maker City L.A., WeWork, NextSpace, Coloft and Hub LA. Los Angeles-based tech investor David Waxman said these kind of shared spaces are crucial for the early stages of tech ventures. "When you’re just starting out, and capital is very scarce, having not to commit to an entire office but having part of an office is very important," Waxman said. “There comes a collective energy when a bunch of entrepreneurs get together in the same space, even if they’re not working on the same project." And he said Pasadena is a good choice for a shared workspace. "It is the home of Caltech, the Arts Center, and IdeaLab — probably the world’s first tech incubator — started there," he said. But he said the need isn't limited to Pasadena. "In Silver Lake, in South Pasadena, in Glendale, you see a lot of little pockets of people getting together, and as soon as there’s a critical mass, we’ll see co-working spaces like Cross Campus come into being," said Waxman, who named his investment firm TenOneTen after the two freeways that connect Santa Monica and the Westside to Pasadena. (arXiv:2005.03652v1 [cs.RO]) By arxiv.org Published On :: Assistive shared-control robots have the potential to transform the lives of millions of people afflicted with severe motor impairments. The usefulness of shared-control robots typically relies on the underlying autonomy's ability to infer the user's needs and intentions, and the ability to do so unambiguously is often a limiting factor for providing appropriate assistance confidently and accurately. The contributions of this paper are four-fold. First, we introduce the idea of intent disambiguation via control mode selection, and present a mathematical formalism for the same. Second, we develop a control mode selection algorithm which selects the control mode in which the user-initiated motion helps the autonomy to maximally disambiguate user intent. Third, we present a pilot study with eight subjects to evaluate the efficacy of the disambiguation algorithm. Our results suggest that the disambiguation system (a) helps to significantly reduce task effort, as measured by number of button presses, and (b) is of greater utility for more limited control interfaces and more complex tasks. We also observe that (c) subjects demonstrated a wide range of disambiguation request behaviors, with the common thread of concentrating requests early in the execution. As our last contribution, we introduce a novel field-theoretic approach to intent inference inspired by dynamic field theory that works in tandem with the disambiguation scheme. Full Article

However, these tasks can be complex, containing both coarse reaching motions and fine-grained manipulation. For example, when eating, not only does one need to move to the correct food item, but they must also precisely manipulate the food in different ways (e.g., cutting, stabbing, scooping). Shared autonomy methods make robot teleoperation safer and more precise by arbitrating user inputs with robot controls. However, these works have focused mainly on the high-level task of reaching a goal from a discrete set, while largely ignoring manipulation of objects at that goal. Meanwhile, dimensionality reduction techniques for teleoperation map useful high-dimensional robot actions into an intuitive low-dimensional controller, but it is unclear if these methods can achieve the requisite precision for tasks like eating. Our insight is that---by combining intuitive embeddings from learned latent actions with robotic assistance from shared autonomy---we can enable precise assistive manipulation. In this work, we adopt learned latent actions for shared autonomy by proposing a new model structure that changes the meaning of the human's input based on the robot's confidence of the goal. We show convergence bounds on the robot's distance to the most likely goal, and develop a training procedure to learn a controller that is able to move between goals even in the presence of shared autonomy.
