Nevertheless, laboratory rodent studies examining the neural bases of stress responses have generally attributed increments in these systems to be monolithic, regardless of whether an active or passive coping strategy is employed. Using the shock probe defensive burying test (SPDB) to measure stress-coping features naturalistically in male and female rats, we identify a neural pathway whereby activity changes may promote distinctive response patterns of hemodynamic and HPA indices typifying active and passive coping phenotypes. Optogenetic excitation of the rostral medial prefrontal cortex (mPFC) input to the ventrolateral periaqueductal gray (vlPAG) decreased passive behavior (immobility), attenuated the glucocorticoid hormone response, but did not prevent arterial pressure and heart rate increases associated with rats’ active behavioral (defensive burying) engagement during the SPDB. In contrast, inhibition of the same pathway increased behavioral immobility and attenuated hemodynamic output but did not affect glucocorticoid increases. Further analyses confirmed that hemodynamic increments occurred preferentially during active behaviors and decrements during immobility epochs, whereas pathway manipulations, regardless of the directionality of effect, weakened these correlational relationships. Finally, neuroanatomical evidence indicated that the influence of the rostral mPFC->vlPAG pathway on coping response patterns is mediated predominantly through GABAergic neurons within vlPAG. These data highlight the importance of this prefrontal->midbrain connection in organizing stress-coping responses and in coordinating bodily systems with behavioral output for adaptation to aversive experiences. In this study, we used male and female mice to relate taste bud innervation patterns to the function of a subset of proenkephalin-expressing (Penk+) taste neurons. We found that taste arbors (the portion of the axon within the taste bud) stemming from Penk+ neurons displayed diverse branching patterns and lacked stereotypical endings. The range in complexity observed for individual taste arbors from Penk+ neurons mirrored the entire population, suggesting that taste arbor morphologies are not primarily regulated by the neuron type. Notably, the distinguishing feature of arbors from Penk+ neurons was their propensity to come within 110 nm (in apposition with) different types of taste-transducing cells within the taste bud. This finding is contrary to the expectation of genetically defined taste neuron types that functionally represent a single stimulus. Consistently, further investigation of Penk+ neuron function revealed that they are more likely to respond to innately aversive stimuli—sour, bitter, and high salt concentrations—as compared with the full taste population. Penk+ neurons are less likely to respond to nonaversive stimuli—sucrose, umami, and low salt—compared with the full population. Our data support the presence of a genetically defined neuron type in the geniculate ganglion that is responsive to innately aversive stimuli. This implies that genetic expression might categorize peripheral taste neurons into hedonic groups, rather than simply identifying neurons that respond to a single stimulus. Full Article

His mother died of tuberculosis in July, 1732, about two weeks before his seventh birthday. Two years later, he went to live in Aveley, the home of his father's new wife. Newton spent two years at boarding school. At age eleven he went to sea with his father. Newton sailed six voyages before his father retired in 1742. Newton's father made plans for him to work at a sugar plantation in Jamaica. Instead, Newton signed on with a merchant ship sailing to the Mediterranean Sea. In 1743, while on the way to visit some friends, Newton was captured and pressed into the naval service by the Royal Navy. He became a midshipman aboard HMS Harwich. At one point, Newton attempted to desert and was punished in front of the crew of 350. Stripped to the waist, tied to the grating, he received a flogging of one dozen lashes, and was reduced to the rank of a common seaman. Following that disgrace and humiliation, Newton initially contemplated suicide. He recovered, both physically and mentally. Later, while Harwich was on route to India, he transferred to Pegasus, a slave ship bound for West Africa. They left him in West Africa with Amos Clowe, a slave dealer. Clowe took Newton to the coast, and gave him to his wife Princess Peye, an African duchess. Newton was abused and mistreated along with her other slaves. It was this period that Newton later remembered as the time he was "once an infidel and libertine, a servant of slaves in West Africa." Early in 1748 he was rescued by a sea captain who had been asked by Newton's father to search for him. And he made it to freedom. In 1750 he married his childhood sweetheart in St. Margaret's Church, Rochester. -- Spiritual conversion: He sailed back to England in 1748 aboard the merchant ship Greyhound, which was carrying beeswax and dyer's wood, now referred to as camwood. During this voyage, he experienced a spiritual conversion. The ship encountered a severe storm off the coast of Donegal and almost sank. Newton awoke in the middle of the night and finally called out to God as the ship filled with water. After he called out, the cargo came out and stopped up the hole, and the ship was able to drift to safety. It was this experience which he later marked as the beginnings of his conversion to evangelical Christianity. As the ship sailed home, Newton began to read the Bible and other religious literature. By the time he reached Britain, he had accepted the doctrines of evangelical Christianity. The date was March 10, 1748, an anniversary he marked for the rest of his life. From that point on, he avoided profanity, gambling, and drinking. Although he continued to work in the slave trade, he had gained a considerable amount of sympathy for the slaves. He later said that his true conversion did not happen until some time later: "I cannot consider myself to have been a believer in the full sense of the word, until a considerable time afterwards." Newton returned to Liverpool, England and, partly due to the influence of his father's friend Joseph Manesty, obtained a position as first mate aboard the slave ship Brownlow, bound for the West Indies via the coast of Guinea. During the first leg of this voyage, while in west Africa (1748-1749), Newton acknowledged the inadequacy of his spiritual life. While he was sick with a fever, he professed his full belief in Christ and asked God to take control of his destiny. He later said that this experience was his true conversion and the turning point in his spiritual life. He claimed it was the first time he felt totally at peace with God. Still, he did not renounce the slave trade until later in his life. Although associated closely with Rotterdam, he lived there for only four years, never to return. Information on his family and early life comes mainly from vague references in his writings. As far as the years of 1000 are concerned, it is assumed that the Ions came to this region, lead by Androckles. Ephesus was captured by the Kimmers (Cimmerians) in the 7th century B.C., by the Lydians in 560, and later in 546 B.C. by the Persians; and was rescued from the Persian domination when Alexander the Great defeated the Persians in 334 B.C. -- Lysimachos, a commander of Alexander's, had the settlement removed from the whereabouts of the Temple of Artemis to the location between the Mount of Panayir and the Mount of Bülbül, and had a wall built around the city. The city was taken by the Kingdom of Pergamon after 190 B.C., by Rome in 133 B.C., and later by Byzantium. Ephesus maintained its importance during the period of Christianity; the apostle St. Paul arrived there during the years of 50 A.D., and St. John was buried on the hill of Ayasuluk (Selcuk, near Izmir) at the beginning of the 2nd century. Ephesus lived through its third glorious period during the reign of Justinian in the middle of the 6th century A.D. At this time, the Church of St. John was built by the Byzantine emperor. -- The ruins of Ephesus, situated near Selçuk town at 70 kilometers (44 miles) south of Izmir, is a main center of archaeological interest owing to the ancient remains that still exist. When you enter through the Magnesia Gate (south gate or upper gate), you can see the State Agora (or Upper Agora). The Temple of Isis is situated at the center of the Agora, and Stoa is placed on the North side of it. The Odeion (Bouletarion or Parliament) with a capacity of 1,400 persons is placed behind it and the Prytaneion (Town Hall) where the sacred fire used to burn, is on its flank. The Baths of Varius are placed on the east side of Odeion. On the west of the Agora, the Monument of Memmius built in the 1st century BC., the fountain of Sextilius Pollio built in the year 93 A.D., and the Temple of Domitian (81-93 A.D) are placed. On the south of the Agora, the fountain of Laecanius Bassus is situated. The Curetes street starts downwards from the Temple of Memmius. The Gate of Heracles (Hercules) is placed on this avenue. After passing through this part, the fountain of Trajan built in the years 102-114 is seen on the right hand side and after this, the Temple of Hadrian appears in front of us, in all its splendid beauty (117-138 A.D). The Scholastica Baths, built in the 4th century A.D., are situated behind the Temple of Hadrian. The houses of the rich people of Ephesus which were in front of it, have been restored and opened for visits at present with special permits. -- At the corner formed by the Curetes street and the Marble Road, the House of Love (Pornaion or Brothel) is placed and the Library of Celsus, restored and reestablished in recent years, stands right in front of this. The library which had been built in the name of proconsul Gaius Celsus completed in the year 135 A.D. by his son Tiberius Giulius Aquila, is entered by way of a stairway, 21 meters (69 feet) in width and having 9 steps. The southeastern gate of the Trade Agora opens to the Library of Celsus. Emperor Augustus' slaves, Mazaeus and Mithridates, liberated by him had this gate built in the year 1st century A.D.; it comprises three sections and has been restored today. The Corinthian columns of the Stoa encircling the Trade Agora with the dimensions 110 x 110 meters (361 x 361 feet), are standing erect today. The Temple of Serapes built in the period of Antony (138-192 A.D.) is placed behind the Trade Agora. -- One of the magnificent buildings of Ephesus is the Great Theater, largest in Asia Minor, which had a capacity of more than 24.000 people and is in a rather well preserved condition. The construction had started during the Hellenistic period but it could only be completed during the time of Trajan (98-117 A.D.). St. Paul was dragged into this theater to face the crowed because of his famous letter to Ephesians, but rescued by the security corps of the city. Festivals are celebrated in this theater today. -- All the streets of Ephesus were illuminated at night with oil lamps, this shows us the richness of the city. The Port Avenue extends in front of the theater. The avenue is 11 meters (36 feet) wide and 600 meters (1970 feet) long, and it has been called Arcadian Street because it was renewed during the time of Arcadius. On the whole north side of the avenue, there are the Harbor Gymnasium, baths and the Theater Gymnasium. The avenue that passes along the front of the theater, extends towards the Stadium built during the Nero period (54-68 A.D.) and towards the Vedius Gymnasium. The Church of the Virgin Mary built at the beginning of the 4th century A.D. is situated behind the Port Gymnasium just before the exit from the lower gate (north gate). **This was also the meeting place of the 3rd Ecumenical Council [The Council of Ephesus may refer to: The First Council of Ephesus of 431 AD. The House of Virgin Mary: On Bulbul Dag (Nightingale mountain) there is the House of the Virgin where it's believed that she passed last years of her life and passed away. She came to Ephesus together with St. John and taken up to Panaghia Kapulu mountain to survive the Roman persecutions. The House was destroyed by many earthquakes and not discovered until 1951 thanks to a German nun, Catherine Emmerich, who saw its location in her visions. The site is recognized as a shrine by Vatican and visited by the Popes. Today, the House of Virgin Mary is renovated by George Quatman Foundation from Ohio and serves as a small church which attracts many Christians as well as Muslims coming to pray for Her. The Mass is held here every Sunday. On the 15th August 2000 there was a great ceremony for the Assumption of the Virgin, the year which marked the two thousand years of the birth of Jesus. peri Customer Experience and Satisfaction Key for Marketers in 2009 By bryanong.blogspot.com Published On :: Sat, 31 Jan 2009 18:20:00 +1030 Companies Missing Big Opportunity to Turn Customer Pain Into Competitive Gain, Says CMO Council ReportDespite overwhelming agreement on the importance of customer experience and word-of-mouth, senior marketers admit their companies are failing to take decisive, company-wide action to integrate customer voice and experience into key business and marketing processes, according to a new study by the Chief Marketing Officer (CMO) Council.Sponsored by Satmetrix, the Net Promoter Company, the study, titled "Giving Customer Voice More Volume," revealed that a surprising 58 percent of the 480 executives surveyed said their companies do not compensate any employees or executives based on customer loyalty, satisfaction improvements or analytics. Some 38 percent said their companies have no programs in place to track or propagate positive word of mouth among customers. In addition, only 29 percent said their companies rate highly in their ability handle and resolve customer problems or complaints.The CMO Council study underscores critical deficiencies in the way companies measure, optimize and leverage customer experience to drive loyalty, improve brand value and increase business performance and growth, including:-- Insufficient availability and aggregation of real-time customer experience data across touch points that should be shared across the organization-- Poor use of customer interactions to collect insights and intelligence or maximize up-sell and advocacy opportunities-- Lack of Internet processes and systems to track online word of mouth and drive customer advocacy-- Intermittent or deficient monitoring of customer experience that fails to provide true and timely insights into problems and opportunities-- Too few compensation programs tied to customer experience, loyalty and satisfaction gains"Customer experience is one of the most critical determinants of brand strength and business growth. Yet, most organizations and senior marketers suffer from major blind spots and gaps in the way they interact, handle and respond to customer issues or problems," said CMO Council executive director Donovan Neale-May. "CMOs must assume ownership for the customer experience and establish enterprise-wide measures and disciplines to ensure continuous improvement. We are missing a major opportunity to turn customer pain into competitive gain at every touch point through better use of web and contact center technologies and processes."Customer listening, learning and leveling are critical qualities that need to be part of an institutionalized corporate culture, notes the CMO Council. Yet, survey data demonstrates that most companies are not taking advantage of these opportunities to drive company-wide performance improvement and business growth. Instead, most companies treat customer interactions around service situations and incidents only as a problem that needs quick resolution:-- Only 38 percent of companies gather customer insight from customer engagement situations.-- Just 32 percent look for ways to turn problems into new sales opportunities, and only 15 percent introduce new products or services to further monetize the relationship.-- Merely 17 percent use the opportunity to identify and cultivate potential customer champions and advocates. While companies have a long way to go in turning detractors into brand advocates, senior marketers are clearly aware of the importance of customer experience. In fact, 83 percent of respondents said it is either "essential" or "increasingly important" in driving brand advocacy and business performance. In addition, 84 percent said positive customer experiences and word of mouth have helped their brands and businesses grow. There were 44 percent of respondents who admitted that high-profile negative customer experiences had at some time compromised their brands.Only 31 percent rate their company's commitment to customer listening highly, but another 35 percent say it is "getting better." Although 34 percent of respondents said their companies have made no changes to the way they track and analyze customer experience in recent years, it can be seen as a positive development that 45 percent of respondents say their companies have taken steps to better integrate and analyze customer data. Another 39 percent said they have increased personalization and intimacy in their customer communications, 20 percent say they have embraced intelligent Internet analytics and 18 percent are capturing real-time information at the "point of pain.""Companies must become more sophisticated and committed to both leveraging customer experience as a key business metric and instituting company-wide processes that drive improvement," said Laura Brooks, Ph.D. and vice president of research for Satmetrix. "The Net Promoter Score has been proven to be the most reliable customer metric for business growth, but measurement is not an end in itself. Companies need to commit themselves to understanding the key determinants of their score and continuously strive to improve their customer experience competitiveness." Brooks is co-author, with Satmetrix CEO Richard Owen, of a new book, titled "Answering the Ultimate Question," that offers a new operating model for NetPromoter success.Other key findings of the study include:-- Nearly two-thirds of companies do not have a formal Voice of Customer program in place.-- Only 13 percent of companies have deployed real-time systems to collect, analyze and distribute customer feedback.-- While 74 percent say they receive customer feedback via e-mail, only 23 percent say they track and measure the volume and nature of these messages.-- Customer voice has gone online, but only 14.5 percent track word of mouth on the Internet-- Only 12 percent are using a word-of-mouth marketing platform to drive online customer advocacy. I got a simple "map" drawing colored squares that can be dragged around. I found a perlin noise library, and made the map look like wavy colors. It was pretty slow at that point, so I rewrote the drawing code so it writes directly into the image data.
