Crystal structure, Hirshfeld surface analysis and HOMO–LUMO analysis of (E)-N'-(3-hy­droxy-4-meth­oxy­benzyl­idene)nicotinohydrazide monohydrate

The mol­ecule of the title Schiff base compound, C14H13N3O3·H2O, displays a trans configuration with respect to the C=N bond. The dihedral angle between the benzene and pyridine rings is 29.63 (7)°. The crystal structure features inter­molecular N—H⋯O, C—H⋯O, O—H⋯O and O—H⋯N hydrogen-bonding inter­actions, leading to the formation of a supramolecular framework. A Hirshfeld surface analysis indicates that the most important contributions to the crystal packing are from H⋯H (37.0%), O⋯H/H⋯O (23.7%)), C⋯H/H⋯C (17.6%) and N⋯H/H⋯N (11.9%) inter­actions. The title compound has also been characterized by frontier mol­ecular orbital analysis.

Engineering a surrogate human heteromeric α/β glycine receptor orthosteric site exploiting the structural homology and stability of acetylcholine-binding protein

Protein-engineering methods have been exploited to produce a surrogate system for the extracellular neurotransmitter-binding site of a heteromeric human ligand-gated ion channel, the glycine receptor. This approach circumvents two major issues: the inherent experimental difficulties in working with a membrane-bound ion channel and the complication that a heteromeric assembly is necessary to create a key, physiologically relevant binding site. Residues that form the orthosteric site in a highly stable ortholog, acetylcholine-binding protein, were selected for substitution. Recombinant proteins were prepared and characterized in stepwise fashion exploiting a range of biophysical techniques, including X-ray crystallography, married to the use of selected chemical probes. The decision making and development of the surrogate, which is termed a glycine-binding protein, are described, and comparisons are provided with wild-type and homomeric systems that establish features of molecular recognition in the binding site and the confidence that the system is suited for use in early-stage drug discovery targeting a heteromeric α/β glycine receptor.

Structural and kinetic insights into flavin-containing monooxygenase and calponin-homology domains in human MICAL3

MICAL is an oxidoreductase that participates in cytoskeleton reorganization via actin disassembly in the presence of NADPH. Although three MICALs (MICAL1, MICAL2 and MICAL3) have been identified in mammals, only the structure of mouse MICAL1 has been reported. Here, the first crystal structure of human MICAL3, which contains the flavin-containing monooxygenase (FMO) and calponin-homology (CH) domains, is reported. MICAL3 has an FAD/NADP-binding Rossmann-fold domain for mono­oxygenase activity like MICAL1. The FMO and CH domains of both MICAL3 and MICAL1 are highly similar in structure, but superimposition of the two structures shows a different relative position of the CH domain in the asymmetric unit. Based on kinetic analyses, the catalytic efficiency of MICAL3 dramatically increased on adding F-actin only when the CH domain was available. However, this did not occur when two residues, Glu213 and Arg530, were mutated in the FMO and CH domains, respectively. Overall, MICAL3 is structurally highly similar to MICAL1, which suggests that they may adopt the same catalytic mechanism, but the difference in the relative position of the CH domain produces a difference in F-actin substrate specificity.

The use of local structural similarity of distant homologues for crystallographic model building from a molecular-replacement solution

The performance of automated protein model building usually decreases with resolution, mainly owing to the lower information content of the experimental data. This calls for a more elaborate use of the available structural information about macromolecules. Here, a new method is presented that uses structural homologues to improve the quality of protein models automatically constructed using ARP/wARP. The method uses local structural similarity between deposited models and the model being built, and results in longer main-chain fragments that in turn can be more reliably docked to the protein sequence. The application of the homology-based model extension method to the example of a CFA synthase at 2.7 Å resolution resulted in a more complete model with almost all of the residues correctly built and docked to the sequence. The method was also evaluated on 1493 molecular-replacement solutions at a resolution of 4.0 Å and better that were submitted to the ARP/wARP web service for model building. A significant improvement in the completeness and sequence coverage of the built models has been observed.

This work has been motivated by recent papers that quantify the density of values of generic quadratic forms and other polynomials at integer points, in particular ones that use Rogers' second moment estimates. In this paper we establish such results in a very general framework. Namely, given any subhomogeneous function (a notion to be defined) $f: mathbb{R}^n o mathbb{R}$, we derive a necessary and sufficient condition on the approximating function $psi$ for guaranteeing that a generic element $fcirc g$ in the $G$-orbit of $f$ is $psi$-approximable; that is, $|fcirc g(mathbf{v})| le psi(|mathbf{v}|)$ for infinitely many $mathbf{v} in mathbb{Z}^n$. We also deduce a sufficient condition in the case of uniform approximation. Here, $G$ can be any closed subgroup of $operatorname{ASL}_n(mathbb{R})$ satisfying certain axioms that allow for the use of Rogers-type estimates.

A survey of Hardy type inequalities on homogeneous groups. (arXiv:2005.03614v1 [math.FA])

In this review paper, we survey Hardy type inequalities from the point of view of Folland and Stein's homogeneous groups. Particular attention is paid to Hardy type inequalities on stratified groups which give a special class of homogeneous groups. In this environment, the theory of Hardy type inequalities becomes intricately intertwined with the properties of sub-Laplacians and more general subelliptic partial differential equations. Particularly, we discuss the Badiale-Tarantello conjecture and a conjecture on the geometric Hardy inequality in a half-space of the Heisenberg group with a sharp constant.

Strong maximum principle and boundary estimates for nonhomogeneous elliptic equations. (arXiv:2005.03338v1 [math.AP])

We give a simple proof of the strong maximum principle for viscosity subsolutions of fully nonlinear elliptic PDEs on the form $$ F(x,u,Du,D^2u) = 0 $$ under suitable structure conditions on the equation allowing for non-Lipschitz growth in the gradient terms. In case of smooth boundaries, we also prove the Hopf lemma, the boundary Harnack inequality and that positive viscosity solutions vanishing on a portion of the boundary are comparable with the distance function near the boundary. Our results apply to weak solutions of an eigenvalue problem for the variable exponent $p$-Laplacian.

NH-HAZE: An Image Dehazing Benchmark with Non-Homogeneous Hazy and Haze-Free Images. (arXiv:2005.03560v1 [cs.CV])

Image dehazing is an ill-posed problem that has been extensively studied in the recent years. The objective performance evaluation of the dehazing methods is one of the major obstacles due to the lacking of a reference dataset. While the synthetic datasets have shown important limitations, the few realistic datasets introduced recently assume homogeneous haze over the entire scene. Since in many real cases haze is not uniformly distributed we introduce NH-HAZE, a non-homogeneous realistic dataset with pairs of real hazy and corresponding haze-free images. This is the first non-homogeneous image dehazing dataset and contains 55 outdoor scenes. The non-homogeneous haze has been introduced in the scene using a professional haze generator that imitates the real conditions of hazy scenes. Additionally, this work presents an objective assessment of several state-of-the-art single image dehazing methods that were evaluated using NH-HAZE dataset.

NTIRE 2020 Challenge on NonHomogeneous Dehazing. (arXiv:2005.03457v1 [cs.CV])

This paper reviews the NTIRE 2020 Challenge on NonHomogeneous Dehazing of images (restoration of rich details in hazy image). We focus on the proposed solutions and their results evaluated on NH-Haze, a novel dataset consisting of 55 pairs of real haze free and nonhomogeneous hazy images recorded outdoor. NH-Haze is the first realistic nonhomogeneous haze dataset that provides ground truth images. The nonhomogeneous haze has been produced using a professional haze generator that imitates the real conditions of haze scenes. 168 participants registered in the challenge and 27 teams competed in the final testing phase. The proposed solutions gauge the state-of-the-art in image dehazing.

Computing-in-Memory for Performance and Energy Efficient Homomorphic Encryption. (arXiv:2005.03002v1 [cs.CR])

Homomorphic encryption (HE) allows direct computations on encrypted data. Despite numerous research efforts, the practicality of HE schemes remains to be demonstrated. In this regard, the enormous size of ciphertexts involved in HE computations degrades computational efficiency. Near-memory Processing (NMP) and Computing-in-memory (CiM) - paradigms where computation is done within the memory boundaries - represent architectural solutions for reducing latency and energy associated with data transfers in data-intensive applications such as HE. This paper introduces CiM-HE, a Computing-in-memory (CiM) architecture that can support operations for the B/FV scheme, a somewhat homomorphic encryption scheme for general computation. CiM-HE hardware consists of customized peripherals such as sense amplifiers, adders, bit-shifters, and sequencing circuits. The peripherals are based on CMOS technology, and could support computations with memory cells of different technologies. Circuit-level simulations are used to evaluate our CiM-HE framework assuming a 6T-SRAM memory. We compare our CiM-HE implementation against (i) two optimized CPU HE implementations, and (ii) an FPGA-based HE accelerator implementation. When compared to a CPU solution, CiM-HE obtains speedups between 4.6x and 9.1x, and energy savings between 266.4x and 532.8x for homomorphic multiplications (the most expensive HE operation). Also, a set of four end-to-end tasks, i.e., mean, variance, linear regression, and inference are up to 1.1x, 7.7x, 7.1x, and 7.5x faster (and 301.1x, 404.6x, 532.3x, and 532.8x more energy efficient). Compared to CPU-based HE in a previous work, CiM-HE obtain 14.3x speed-up and >2600x energy savings. Finally, our design offers 2.2x speed-up with 88.1x energy savings compared to a state-of-the-art FPGA-based accelerator.

The propylene homopolymer composition for a capacitor film contains 50 to 99% by mass of a propylene homopolymer (A1) having predetermined characteristics and 1 to 50% by mass of a propylene homopolymer (B1) having predetermined characteristics, and has (i) a melt flow rate (MFR) of 1.0 to 10.0 g/10 min and (ii) a chlorine content of 2 ppm by mass or less.
