axis Synaxis of the Icon of our Most Holy Lady the Theotokos “Of the Three Hands” By www.ancientfaith.com Published On :: 2022-06-01T02:21:00+00:00 Saint John of Damascus (December 4), the great defender of Orthodoxy against the iconoclasts, was falsely accused of plotting against the Caliph of Damascus through the intrigues of the iconoclast Emperor Leo the Isaurian (reigned 717-741). The Caliph ordered St John's hand to be cut off for his suspected treachery. The saint asked for the severed hand, and passed the night praying fervently for the aid before an icon of the most holy Theotokos. Waking in the morning, he found his hand miraculously restored, with only a scar around the wrist where it had been completely severed. In thanksgiving, St John had a silver hand mounted on the icon. When he became a monk in the monastery of St Sabbas in the Holy Land, he took the icon with him. It remained there until it was given to St Sabbas (Sava) of Serbia (January 14), who brought it to Serbia. Later it was miraculously taken to the Hilandar Monastery on the Holy Mountain (carried, according to legend, from Serbia to Mt Athos by an unguided donkey), where it may now be found. Full Article

The Archangels Michael and Gabriel, whom we especially commemorate today along with all the other bodiless powers, have served as messengers to man. "Michael" means "Who is like God?";"Gabriel" means "God is mighty." Gabriel especially was appointed to announce the coming of Christ in the flesh. There is no reckoning the number of the Heavenly Host, though we know that they are a great multitude. They are ranked in nine orders, called Thrones, Cherubim, Seraphim, Dominions, Powers, Authorities, Principalities, Archangels and Angels. "Angel" means "herald" or "messenger" and is properly applied only to those who serve as messengers from God to man; but the name is often applied to the entire host of bodiless powers. axis Observations of specimen morphology effects on near-zone-axis convergent-beam electron diffraction patterns By journals.iucr.org Published On :: 2024-03-21 This work presents observations of symmetry breakages in the intensity distributions of near-zone-axis convergent-beam electron diffraction (CBED) patterns that can only be explained by the symmetry of the specimen and not the symmetry of the unit cell describing the atomic structure of the material. The specimen is an aluminium–copper–tin alloy containing voids many tens of nanometres in size within continuous single crystals of the aluminium host matrix. Several CBED patterns where the incident beam enters and exits parallel void facets without the incident beam being perpendicular to these facets are examined. The symmetries in their intensity distributions are explained by the specimen morphology alone using a geometric argument based on the multislice theory. This work shows that it is possible to deduce nanoscale morphological information about the specimen in the direction of the electron beam – the elusive third dimension in transmission electron microscopy – from the inspection of CBED patterns. The method makes use of the lengths of the three shortest reciprocal-lattice vectors in each pattern and the angles between them. No prior pattern calibration is required for the method to work, as the Rn ratio method is based entirely on geometric relationships. In the first step the pattern is assigned to one of three possible pattern types according to the angles that are measured between the three reciprocal-lattice vectors. The lattice direction [uvw] and possible Bravais type(s) and Laue indices of the corresponding reflections can then be determined by using lookup tables. In addition to determining the lattice direction, this simple geometric analysis allows one to distinguish between the P, I and F Bravais lattices for spot patterns aligned along [013], [112], [114] and [233]. Key to this goal is the elimination of cancer stem cells (CSCs) endowed with tumor-initiating capacity and drug resistance. However, current therapeutic strategies capable of accomplishing this are insufficient. Using in vitro models of CSCs and in vivo models of tumor initiation in which CSCs give rise to xenograft tumors, we show that dexamethasone induces expression of MKP-1, a MAPK phosphatase, via glucocorticoid receptor activation, thereby inactivating JNK, which is required for self-renewal and tumor initiation by pancreatic CSCs as well as for their expression of survivin, an anti-apoptotic protein implicated in multidrug resistance. We also demonstrate that systemic administration of clinically relevant doses of dexamethasone together with gemcitabine prevents tumor formation by CSCs in a pancreatic cancer xenograft model. Our study thus provides preclinical evidence for the efficacy of dexamethasone as an adjuvant therapy to prevent postoperative recurrence in patients with pancreatic cancer. Full Article

Statins, which inhibit both cholesterol biosynthesis and protein prenylation branches of the mevalonate pathway, increase anti-tubercular antibiotic efficacy in animal models. However, the underlying molecular mechanisms are unknown. In this study, we used an in vitro macrophage infection model to investigate simvastatin’s anti-tubercular activity by systematically inhibiting each branch of the mevalonate pathway and evaluating the effects of the branch-specific inhibitors on mycobacterial growth. The anti-tubercular activity of simvastatin used at clinically relevant doses specifically targeted the cholesterol biosynthetic branch rather than the prenylation branches of the mevalonate pathway. Using Western blot analysis and AMP/ATP measurements, we found that simvastatin treatment blocked activation of mechanistic target of rapamycin complex 1 (mTORC1), activated AMP-activated protein kinase (AMPK) through increased intracellular AMP:ATP ratios, and favored nuclear translocation of transcription factor EB (TFEB). These mechanisms all induce autophagy, which is anti-mycobacterial. The biological effects of simvastatin on the AMPK-mTORC1-TFEB-autophagy axis were reversed by adding exogenous cholesterol to the cells. Our data demonstrate that the anti-tubercular activity of simvastatin requires inhibiting cholesterol biosynthesis, reveal novel links between cholesterol homeostasis, the AMPK-mTORC1-TFEB axis, and Mycobacterium tuberculosis infection control, and uncover new anti-tubercular therapy targets. For the layout, I'd prefer a brighter color. Is there a way to realize this?
