warsaw, poland — Polish President Andrzej Duda marked Independence Day on Monday with a call for sustained U.S. commitment to Europe's security in view of Russian aggression in the region and argued that Ukraine's pre-2014 borders should be restored. Weighing on the minds of many is the war across Poland's border in Ukraine, and an expectation that Donald Trump's return to the White House will bring a change in the security situation in the region. Some fear Trump could end the U.S. commitment to NATO or make a deal with Russian President Vladimir Putin that could result in a permanent loss of territory for Ukraine and encourage Russia to attack other nations. Others believe Trump could persuade Putin to stop the fighting. Duda, who has had friendly relations with Trump, said in a speech in Warsaw that Europe will continue to need U.S. protection. “It is a pipe dream — as some people think — that Europe can ensure its own security today,” Duda said. He emphasized that the security guarantees of successive U.S. presidents are extremely important in times of resurgent Russian imperialism. “Today we have no doubts that for the security of Europe and the world, it is necessary to strengthen Euro-Atlantic ties,” Duda said. He said the territorial integrity of all countries, especially Ukraine, should be respected and it “must return to its borders from before the Russian attack, not only the one in 2022 but also the first one, in 2014." Russia annexed Ukraine's Crimean Peninsula in 2014 and launched its full-scale invasion in 2022. Later in a news conference, Duda said he had spoken to Trump and that they would meet before the inauguration in January. Duda spoke as Poland marked the 106th anniversary of its restored independence at the end of World War I after more than a century of being partitioned and ruled by Russia, Germany and Austro-Hungary. The date of Nov. 11 carries powerful weight for a nation where the trauma of losing national sovereignty endures. In Warsaw, tens of thousands of people took part in a march organized by nationalist groups that has sometimes seen violent clashes in past years but passed without major incident Monday. Organizers estimated turnout at 250,000 while city hall put it at 90,000. Police said they detained 75 people and seized banned items from participants, including pyrotechnic materials, knives, telescopic batons and brass knuckles. Jaroslaw Kaczynski, leader of the conservative Law and Justice party, which governed Poland from 2015-2023 and is seeking a comeback, joined the march with other party members.

The transcription factor (TF) cone-rod homeobox (CRX) is essential for the differentiation and maintenance of photoreceptor cell identity. Several human CRX variants cause degenerative retinopathies, but most are variants of uncertain significance. We performed a deep mutational scan (DMS) of nearly all possible single amino acid substitutions in CRX using a cell-based transcriptional reporter assay, curating a high-confidence list of nearly 2000 variants with altered transcriptional activity. In the structured homeodomain, activity scores closely aligned to a predicted structure and demonstrated position-specific constraints on amino acid substitution. In contrast, the intrinsically disordered transcriptional effector domain displayed a qualitatively different pattern of substitution effects, following compositional constraints without specific residue position requirements in the peptide chain. These compositional constraints were consistent with the acidic exposure model of transcriptional activation. We evaluated the performance of the DMS assay as a clinical variant classification tool using gold-standard classified human variants from ClinVar, identifying pathogenic variants with high specificity and moderate sensitivity. That this performance could be achieved using a synthetic reporter assay in a foreign cell type, even for a highly cell type-specific TF like CRX, suggests that this approach shows promise for DMS of other TFs that function in cell types that are not easily accessible. Together, the results of the CRX DMS identify molecular features of the CRX effector domain and demonstrate utility for integration into the clinical variant classification pipeline.

It is of utmost importance that medicines are available at all times for our patients. Historically, medication unavailability has typically, if not exclusively, affected low- and middle-income countries [1]. More recently however, drug shortages have also been reported in high-income European countries [2]. Drug shortages have negative health consequences for patients [3], and a profound economic impact, with the need to resort to more expensive alternatives and demands on healthcare professionals’ time to find, prescribe and dispense alternatives [4].

Targeting the endocannabinoid (eCB) signaling system for pain relief is an important treatment option that is only now beginning to be mechanistically explored. In this review, we focus on two recently appreciated cannabinoid-based targeting strategies, treatments with cannabidiol (CBD) and α/β-hydrolase domain containing 6 (ABHD6) inhibitors, which have the exciting potential to produce pain relief through distinct mechanisms of action and without intoxication. We review evidence on plant-derived cannabinoids for pain, with an emphasis on CBD and its multiple molecular targets expressed in pain pathways. We also discuss the function of eCB signaling in regulating pain responses and the therapeutic promises of inhibitors targeting ABHD6, a 2-arachidonoylglycerol (2-AG)-hydrolyzing enzyme. Finally, we discuss how the novel cannabinoid biosensor GRAB_eCB2.0 may be leveraged to enable the discovery of targets modulated by cannabinoids at a circuit-specific level.

Ethnic cleansing is not only a modern phenomenon. The medieval Catholic Church saw non-Christians as a threat and facilitated the ethnoreligious cleansing of Muslim and Jewish communities across Western Europe. Three conditions made this possible: The rising power of the papacy as a supranational religious authority; its dehumanization of non-Christians; and competition among Catholic Western European monarchs that left them vulnerable to papal-clerical demands to eradicate non-Christians. These findings revise our understanding twentieth- and twenty-first-century ethnic cleansing in places like Cambodia, Iraq, Myanmar, the Soviet Union, and Syria.
