Glial TIM-3 Modulates Immune Responses in the Brain Tumor Microenvironment

T-cell immunoglobulin and mucin domain–containing molecule 3 (TIM-3), a potential immunotherapeutic target for cancer, has been shown to display diverse characteristics in a context-dependent manner. Thus, it would be useful to delineate the precise functional features of TIM-3 in a given situation. Here, we report that glial TIM-3 shows distinctive properties in the brain tumor microenvironment. TIM-3 was expressed on both growing tumor cells and their surrounding cells including glia and T cells in an orthotopic mouse glioma model. The expression pattern of TIM-3 was distinct from those of other immune checkpoint molecules in tumor-exposed and tumor-infiltrating glia. Comparison of cells from tumor-bearing and contralateral hemispheres of a glioma model showed that TIM-3 expression was lower in tumor-infiltrating CD11b+CD45mid glial cells but higher in tumor-infiltrating CD8+ T cells. In TIM-3 mutant mice with intracellular signaling defects and Cre-inducible TIM-3 mice, TIM-3 affected the expression of several immune-associated molecules including iNOS and PD-L1 in primary glia-exposed conditioned media (CM) from brain tumors. Further, TIM-3 was cross-regulated by TLR2, but not by TLR4, in brain tumor CM- or Pam3CSK4-exposed glia. In addition, following exposure to tumor CM, IFNγ production was lower in T cells cocultured with TIM-3–defective glia than with normal glia. Collectively, these findings suggest that glial TIM-3 actively and distinctively responds to brain tumor, and plays specific intracellular and intercellular immunoregulatory roles that might be different from TIM-3 on T cells in the brain tumor microenvironment.Significance:TIM-3 is typically thought of as a T-cell checkpoint receptor. This study demonstrates a role for TIM-3 in mediating myeloid cell responses in glioblastoma.

[PERSPECTIVES] Brain Metastasis Organotropism

Brain metastases are associated with poor prognosis irrespective of the primary tumor they originate from. Current treatments for brain metastases are palliative, and patients with symptomatic brain metastasis have a one-year survival of <20%. Lung cancer, breast cancer, and melanoma have higher incidences of brain metastases compared with other types of cancers. However, it is not very clear why some cancers metastasize to the brain more frequently than others. Studies thus far suggest that brain-specific tropism of certain types of cancers is defined by a winning combination of the following factors: unique genetic subtypes of primary tumors or its subclones enabling detachment, dissemination, blood–brain barrier penetration, plus proliferation and survival in hypoxic low-glucose microenvironment; specific transcriptomic and epigenetic changes of colony-forming metastatic cells, allowing their outgrowth; favorable metastasis-permissive microenvironment of the brain created by interactions of cancer cells and cells in the brain through triggering inflammation, recruiting myeloid-derived suppressor cells, and promoting metabolic adaptation; immunosuppression resulting in the failure of adaptive immune response to recognize or kill cancer cells in the brain. Here, we briefly review recent advances in understanding brain metastasis organotropism and outline directions for future research.

Thalamic Massa Intermedia in Children with and without Midline Brain Malformations [PEDIATRICS]

BACKGROUND AND PURPOSE:

The massa intermedia is a normal midline transventricular thalamic connection. Massa intermedia aberrations are common in schizophrenia, Chiari II malformation, X-linked hydrocephalus, Cornelia de Lange syndrome, and diencephalic-mesencephalic junction dysplasia, among others. We have noticed that massa intermedia abnormalities often accompany other midline malformations. The massa intermedia has never been formally evaluated in a group of exclusively pediatric patients, to our knowledge. We sought to compare and contrast the prevalence, size, and location of the massa intermedia in pediatric patients with and without congenital midline brain abnormalities.

MATERIALS AND METHODS:

Successive 3T brain MR imaging examinations from pediatric patients with and without midline malformations were procured from the imaging data base at a pediatric hospital. Massa intermedia presence, size, morphology, and position were determined using 3D-TIWI with 1-mm isotropic resolution. The brain commissures, septum pellucidum, hypothalamus, hippocampus, vermis, and brain stem were evaluated to determine whether alterations were related to or predictive of massa intermedia abnormalities.

RESULTS:

The massa intermedia was more frequently absent, dysmorphic, and/or displaced in patients with additional midline abnormalities than in those without. The massa intermedia was absent in 40% of patients with midline malformations versus 12% of patients with normal findings (P < .001). Massa intermedia absence, surface area, and morphology were predictable by various attributes and alterations of the commissures, hippocampus, hypothalamus, vermis, brain stem, and third ventricle.

CONCLUSIONS:

Most pediatric patients have a thalamic massa intermedia centered in the anterior/superior third ventricle. Massa intermedia abnormalities are commonly associated with other midline malformations. Normal-variant massa intermedia absence is a diagnosis of exclusion.

Advanced ADC Histogram, Perfusion, and Permeability Metrics Show an Association with Survival and Pseudoprogression in Newly Diagnosed Diffuse Intrinsic Pontine Glioma: A Report from the Pediatric Brain Tumor Consortium [FUNCTIONAL]

BACKGROUND AND PURPOSE:

Diffuse intrinsic pontine glioma is a lethal childhood brain cancer with dismal prognosis and MR imaging is the primary methodology used for diagnosis and monitoring. Our aim was to determine whether advanced diffusion, perfusion, and permeability MR imaging metrics predict survival and pseudoprogression in children with newly diagnosed diffuse intrinsic pontine glioma.

MATERIALS AND METHODS:

A clinical trial using the poly (adenosine diphosphate ribose) polymerase (PARP) inhibitor veliparib concurrently with radiation therapy, followed by maintenance therapy with veliparib + temozolomide, in children with diffuse intrinsic pontine glioma was conducted by the Pediatric Brain Tumor Consortium. Standard MR imaging, DWI, dynamic contrast-enhanced perfusion, and DSC perfusion were performed at baseline and approximately every 2 months throughout treatment. ADC histogram metrics of T2-weighted FLAIR and enhancing tumor volume, dynamic contrast-enhanced permeability metrics for enhancing tumors, and tumor relative CBV from DSC perfusion MR imaging were calculated. Baseline values, post-radiation therapy changes, and longitudinal trends for all metrics were evaluated for associations with survival and pseudoprogression.

RESULTS:

Fifty children were evaluable for survival analyses. Higher baseline relative CBV was associated with shorter progression-free survival (P = .02, Q = 0.089) and overall survival (P = .006, Q = 0.055). Associations of higher baseline mean transfer constant from the blood plasma into the extravascular extracellular space with shorter progression-free survival (P = .03, Q = 0.105) and overall survival (P = .03, Q = 0.102) trended toward significance. An increase in relative CBV with time was associated with shorter progression-free survival (P < .001, Q < 0.001) and overall survival (P = .004, Q = 0.043). Associations of longitudinal mean extravascular extracellular volume fraction with progression-free survival (P = .03, Q = 0.104) and overall survival (P = .03, Q = 0.105) and maximum transfer constant from the blood plasma into the extravascular extracellular space with progression-free survival (P = .03, Q = 0.102) trended toward significance. Greater increases with time were associated with worse outcomes. True radiologic progression showed greater post-radiation therapy decreases in mode_ADC_FLAIR compared with pseudoprogression (means, –268.15 versus –26.11, P = .01.)

CONCLUSIONS:

ADC histogram, perfusion, and permeability MR imaging metrics in diffuse intrinsic pontine glioma are useful in predicting survival and pseudoprogression.

Treatment Outcomes of Endovascular Embolization Only in Patients with Unruptured Brain Arteriovenous Malformations: A Subgroup Analysis of ARUBA (A Randomized Trial of Unruptured Brain Arteriovenous Malformations) [INTERVENTIONAL]

BACKGROUND AND PURPOSE:

Endovascular embolization only has been advocated for treatment of brain arteriovenous malformations in recent trials. Our aim was to evaluate the results of embolization only in a cohort of patients who were enrolled in the A Randomized Trial of Unruptured Brain Arteriovenous Malformations (ARUBA) study at 39 clinical sites in 9 countries.

MATERIALS AND METHODS:

We analyzed the rates and severity of stroke and death in patients who underwent embolization only. Events were identified through in-person neurologic follow-up visits performed at 6-month intervals during the first 2 years and annually, with telephone contact every 6 months thereafter. All event-related data were reviewed by independent adjudicators.

RESULTS:

Among 30 patients who had embolization planned, 26 underwent embolization only. A total of 13 stroke events were reported in the follow-up period among 26 subjects (ischemic, hemorrhagic, or both in 4, 7, and 2 subjects, respectively). The adverse event occurred after the first embolization in 11 of 13 patients. One patient had a major motor deficit, and 2 patients developed major visual field deficits. One event was fatal. The modified Rankin Scale score was 0–2 at last follow-up in 11 of the 12 stroke survivors. Estimated stroke-free survival was 46% at 12 months.

CONCLUSIONS:

Although the rates of stroke and/or death were high in patients treated with embolization only in ARUBA, the rates of favorable outcomes following stroke were high during follow-up.

Complications of Endovascular Treatments for Brain Arteriovenous Malformations: A Nationwide Surveillance [INTERVENTIONAL]

BACKGROUND AND PURPOSE:

Embolization is widely performed to treat brain arteriovenous malformations, but little has been reported on factors contributing to complications. We retrospectively reviewed a nationwide surveillance to identify risk factors contributing to complications and short-term clinical outcomes in the endovascular treatment of brain arteriovenous malformations.

MATERIALS AND METHODS:

Data for endovascular treatment of brain arteriovenous malformations were extracted from the Japanese nationwide surveillance. Patient characteristics, brain arteriovenous malformation features, procedures, angiographic results, complications, and clinical outcomes at 30 days postprocedure were analyzed.

RESULTS:

A total of 1042 endovascular procedures (788 patients; mean, 1.43 ± 0.85 procedures per patient) performed in 111 institutions from 2010 to 2014 were reviewed. Liquid materials were used in 976 procedures (93.7%): to perform presurgical embolization in 638 procedures (61.2%), preradiosurgical embolization in 160 (15.4%), and as sole endovascular treatment in 231 (22.2%). Complete or near-complete obliteration of brain arteriovenous malformations was obtained in 386 procedures (37.0%). Procedure-related complications occurred in 136 procedures (13.1%), including hemorrhagic complications in 59 (5.7%) and ischemic complications in 57 (5.5%). Univariate analysis identified deep venous drainage, associated aneurysms, infratentorial location, and preradiosurgical embolization as statistically significant risk factors for complications. Multivariate analysis showed that embolization of brain arteriovenous malformations in the infratentorial location was significantly associated with complications. Patients with complications due to endovascular procedures had worse clinical outcomes 30 days after the procedures than those without complications.

CONCLUSIONS:

Complications arising after endovascular treatment of brain arteriovenous malformations are not negligible even though they may play a role in adjunctive therapy, especially in the management of infratentorial brain arteriovenous malformations.

Anoxic Brain Injury Detection with the Normalized Diffusion to ASL Perfusion Ratio: Implications for Blood-Brain Barrier Injury and Permeability [FUNCTIONAL]

BACKGROUND AND PURPOSE:

Anoxic brain injury is a result of prolonged hypoxia. We sought to describe the nonquantitative arterial spin-labeling perfusion imaging patterns of anoxic brain injury, characterize the relationship of arterial spin-labeling and DWI, and evaluate the normalized diffusion-to-perfusion ratio to differentiate patients with anoxic brain injury from healthy controls.

MATERIALS AND METHODS:

We identified all patients diagnosed with anoxic brain injuries from 2002 to 2019. Twelve ROIs were drawn on arterial spin-labeling with coordinate-matched ROIs identified on DWI. Linear regression analysis was performed to examine the relationship between arterial spin-labeling perfusion and diffusion signal. Normalized diffusion-to-perfusion maps were generated using a custom-built algorithm.

RESULTS:

Thirty-five patients with anoxic brain injuries and 34 healthy controls were identified. Linear regression analysis demonstrated a significant positive correlation between arterial spin-labeling and DWI signal. By means of a combinatory cutoff of slope of >0 and R2 of > 0.78, linear regression using arterial spin-labeling and DWI showed a sensitivity of 0.86 (95% CI, 0.71–0.94) and specificity of 0.82 (95% CI, 0.66–0.92) for anoxic brain injuries. A normalized diffusion-to-perfusion color map demonstrated heterogeneous ratios throughout the brain in healthy controls and homogeneous ratios in patients with anoxic brain injuries.

CONCLUSIONS:

In anoxic brain injuries, a homogeneously positive correlation between qualitative perfusion and DWI signal was identified so that areas of increased diffusion signal showed increased ASL signal. By exploiting this relationship, the normalized diffusion-to-perfusion ratio color map may be a valuable imaging biomarker for diagnosing anoxic brain injury and potentially assessing BBB integrity.

Discrimination between Glioblastoma and Solitary Brain Metastasis: Comparison of Inflow-Based Vascular-Space-Occupancy and Dynamic Susceptibility Contrast MR Imaging [FUNCTIONAL]

BACKGROUND AND PURPOSE:

Accurate differentiation between glioblastoma and solitary brain metastasis is of vital importance clinically. This study aimed to investigate the potential value of the inflow-based vascular-space-occupancy MR imaging technique, which has no need for an exogenous contrast agent, in differentiating glioblastoma and solitary brain metastasis and to compare it with DSC MR imaging.

MATERIALS AND METHODS:

Twenty patients with glioblastoma and 22 patients with solitary brain metastasis underwent inflow-based vascular-space-occupancy and DSC MR imaging with a 3T clinical scanner. Two neuroradiologists independently measured the maximum inflow-based vascular-space-occupancy–derived arteriolar CBV and DSC-derived CBV values in intratumoral regions and peritumoral T2-hyperintense regions, which were normalized to the contralateral white matter (relative arteriolar CBV and relative CBV, inflow-based vascular-space-occupancy relative arteriolar CBV, and DSC-relative CBV). The intraclass correlation coefficient, Student t test, or Mann-Whitney U test and receiver operating characteristic analysis were performed.

RESULTS:

All parameters of both regions had good or excellent interobserver reliability (0.74~0.89). In peritumoral T2-hyperintese regions, DSC-relative CBV (P < .001), inflow-based vascular-space-occupancy arteriolar CBV (P = .001), and relative arteriolar CBV (P = .005) were significantly higher in glioblastoma than in solitary brain metastasis, with areas under the curve of 0.94, 0.83, and 0.72 for discrimination, respectively. In the intratumoral region, both inflow-based vascular-space-occupancy arteriolar CBV and relative arteriolar CBV were significantly higher in glioblastoma than in solitary brain metastasis (both P < .001), with areas under the curve of 0.91 and 0.90, respectively. Intratumoral DSC-relative CBV showed no significant difference (P = .616) between the 2 groups.

CONCLUSIONS:

Inflow-based vascular-space-occupancy has the potential to discriminate glioblastoma from solitary brain metastasis, especially in the intratumoral region.

Brain Metastases: Insights from Statistical Modeling of Size Distribution [ADULT BRAIN]

BACKGROUND AND PURPOSE:

Brain metastases are a common finding on brain MRI. However, the factors that dictate their size and distribution are incompletely understood. Our aim was to discover a statistical model that can account for the size distribution of parenchymal metastases in the brain as measured on contrast-enhanced MR imaging.

MATERIALS AND METHODS:

Tumor volumes were calculated on the basis of measured tumor diameters from contrast-enhanced T1-weighted spoiled gradient-echo images in 68 patients with untreated parenchymal metastatic disease. Tumor volumes were then placed in rank-order distributions and compared with 11 different statistical curve types. The resultant R2 values to assess goodness of fit were calculated. The top 2 distributions were then compared using the likelihood ratio test, with resultant R values demonstrating the relative likelihood of these distributions accounting for the observed data.

RESULTS:

Thirty-nine of 68 cases best fit a power distribution (mean R2 = 0.938 ± 0.050), 20 cases best fit an exponential distribution (mean R2 = 0.957 ± 0.050), and the remaining cases were scattered among the remaining distributions. Likelihood ratio analysis revealed that 66 of 68 cases had a positive mean R value (1.596 ± 1.316), skewing toward a power law distribution.

CONCLUSIONS:

The size distributions of untreated brain metastases favor a power law distribution. This finding suggests that metastases do not exist in isolation, but rather as part of a complex system. Furthermore, these results suggest that there may be a relatively small number of underlying variables that substantially influence the behavior of these systems. The identification of these variables could have a profound effect on our understanding of these lesions and our ability to treat them.

MR Thermometry in Cerebrovascular Disease: Physiologic Basis, Hemodynamic Dependence, and a New Frontier in Stroke Imaging [ADULT BRAIN]

SUMMARY:

The remarkable temperature sensitivity of the brain is widely recognized and has been studied for its role in the potentiation of ischemic and other neurologic injuries. Pyrexia frequently complicates large-vessel acute ischemic stroke and develops commonly in critically ill neurologic patients; the profound sensitivity of the brain even to minor intraischemic temperature changes, together with the discovery of brain-to-systemic as well as intracerebral temperature gradients, has thus compelled the exploration of cerebral thermoregulation and uncovered its immutable dependence on cerebral blood flow. A lack of pragmatic and noninvasive tools for spatially and temporally resolved brain thermometry has historically restricted empiric study of cerebral temperature homeostasis; however, MR thermometry (MRT) leveraging temperature-sensitive nuclear magnetic resonance phenomena is well-suited to bridging this long-standing gap. This review aims to introduce the reader to the following: 1) fundamental aspects of cerebral thermoregulation, 2) the physical basis of noninvasive MRT, and 3) the physiologic interdependence of cerebral temperature, perfusion, metabolism, and viability.

PlayStation Now Adds Rainbow Six Siege, The Evil Within II And Get Even

Sony Interactive Entertainment announced Rainbow Six SiegeThe Evil Within 2 and Get Even are now available for PlayStation Now subscribers. 

Rainbow Six Siege will only be available to download and stream until Monday, November 2. Downloads for the game are limited to the PlayStation 4 only. 

PlayStation Now features over 800 games on the PlayStation 4, PlayStation 3 and PlayStation 2 available to stream and play on the PlayStation 4 and Windows PC. PS4 subscribers can download and install over 300 games to play locally.

The Circle Of Life: Praying Mantis Eating The Brain (And Rest) Of A 'Murder Hornet'

Because we don't have enough on our plates as it is, now we've got 'murder hornets' flying around the United States. Murder hornets (actually Asian Giant Hornets, but that name didn't strike enough fear into the masses) have a sting powerful enough that China recommends medical treatment if stunk more than ten times, and emergency treatment if stung more than 30. *pours out a little liquor for Macaulay Culkin in My Girl*. And where is his glasses?! He can't see without his glasses! Put his glasses on! This is a video of a praying mantis eating a Giant Asian hornet until it's just a pile of limbs. So yeah, praying mantises -- start breeding them. And make sure to selectively breed them too so we can produce them bigger and bigger until they're preying on us because the way things are going how is that not how this ends? For a Georgia Police Force, a Bungled Shooting Case Follows a Trail of Woes

BRUNSWICK, Ga. -- When the Glynn County Police Department arrived at the scene of a fatal shooting in February in southeastern Georgia, officers encountered a former colleague with the victim's blood on his hands.They took down his version of events and let him and his adult son, who had fired the shots, go home.Later that day, Wanda Cooper, the mother of the 25-year-old victim, Ahmaud Arbery, received a call from a police investigator. She recounted later that the investigator said her son had been involved in a burglary and was killed by "the homeowner," an inaccurate version of what had happened.More than two months after that fatal confrontation, the Georgia Bureau of Investigation, which took over the case this week, arrested the former officer, Gregory McMichael, and his son, Travis McMichael, on charges of murder and aggravated assault.The charges -- which came after the release of a graphic video showing the killing as the two white men confront Arbery, who was African American -- made clear the depths of the local department's bungling of the case, which was just the latest in a series of troubling episodes involving its officers.And it was one element of the broader potential breakdown of the justice system in South Georgia. Attorney General Chris Carr, through a spokeswoman, said Friday that he planned to start a review of all of the relevant players in that system.Carr's office has already determined that George E. Barnhill, a district attorney who was assigned the case in February but recused himself late last month, should have never taken it on. Among his many conflicts: His son once worked alongside one of the suspects at the local prosecutor's office.S. Lee Merritt, a lawyer representing Arbery's family, has called for a federal civil rights investigation focused not only on the men who pursued Arbery but also the broader justice system."It's small-town America," Merritt said in an interview Thursday. "Those counties, the law enforcement community there, they know each other well; they recycle officers in between themselves -- it's a very tight-knit community."Over the years, Glynn County police officers have been accused of covering up allegations of misconduct, tampering with a crime scene, interfering in an investigation of a police shooting and retaliating against fellow officers who cooperated with outside investigators.The police chief was indicted days after Arbery's killing on charges related to an alleged cover-up of an officer's sexual relationship with an informant. The chief, John Powell, had been hired to clean up the department, which the Glynn County manager described last fall as suffering from poor training, outdated policies and "a culture of cronyism."The Glynn County force was the sort of department where disciplinary records went missing and where evidence room standards were not maintained, leading the state to strip it of its accreditation.Arbery was killed after the McMichaels confronted him while he was running in the Satilla Shores neighborhood just outside of Brunswick, the Glynn County seat. But neither of the McMichaels was arrested immediately after the slaying, which occurred Feb. 23 about 1 p.m.According to a police report, Gregory McMichael said that he saw Arbery running through his neighborhood and thought that he looked like the suspect in a rash of recent break-ins. McMichael, 64, told authorities that he and his son, Travis McMichael, 34, armed themselves and began chasing him in a truck.Gregory McMichael had been a Glynn County police officer from 1982 to 1989 and later worked as an investigator in the local prosecutor's office, before retiring last year.Darren W. Penn, a lawyer and a department critic, said the Ahmaud Arbery case was "another symptom or sign of a police department that appears willing to protect those that they know."Penn is representing a woman who is suing the department over claims that it failed to intervene with her estranged son-in-law, a Glynn County officer, who killed her daughter, a friend and himself in 2018.County officials and a police spokesman could not be reached Friday for comment.From the start, McMichael's connections to the police department and the prosecutor's office presented other challenges.The first district attorney assigned to the case, Jackie Johnson, recused herself because she had worked with McMichael. The second prosecutor, Barnhill, advised Glynn County police that there was "insufficient probable cause" to issue arrest warrants, according to an internal document.Finally, the case moved to Tom Durden, the district attorney in Georgia's Atlantic Judicial Circuit in Hinesville, who this week formally asked the state bureau of investigation to get involved, according to a GBI statement. A Justice Department spokesperson said this week that the FBI was assisting in the investigation.Bob Coleman, a county commissioner at large, was critical of Johnson, saying she should have given the case to the state attorney general, not Barnhill. After the Georgia Bureau of Investigation made arrests this week, Coleman said, "That's what should have happened a long time ago before the sun went down. They killed a person in the bright sunlight."Glynn County is a marshy coastal corner of Georgia about 300 miles southeast of Atlanta with about 85,000 residents, and is known mostly for its mellow barrier islands and its rich African American coastal culture.Like many Southern communities, its history is studded with racial violence, including three late 19th-century lynchings. Today, the county is about 70% white and 27% black, according to census figures.On Friday, hundreds gathered under the moss-draped trees outside the Glynn County courthouse to protest, arguing that the handling of the case had been botched as months went by without charges."I will never call the Glynn County police to my house!" one of Arbery's aunts said.Mario Baggs, a lifelong resident of Brunswick, said he believed that race was a factor in Arbery's killing, given the unfair treatment black men have long received."The black man is an endangered species," Baggs, 46, said. "We need justice; we need relief; we need the world to pay attention."Yet he also believed that Arbery's case fit into a larger pattern of dysfunction.Over the last decade, the Glynn County Police Department, which has 122 officers, has faced at least 17 lawsuits, including allegations of illegal search and seizure.One suit accused the department of wrongfully killing an unarmed white woman after officers fired through her car windshield. An investigation into that shooting found that Glynn County officers had tried to interfere with the inquiry to protect the officers involved.One of the officers in that shooting later killed his estranged wife and a friend. The wife's mother accused police of ignoring several alarming encounters in the months before the killings.Powell, the police chief, was arrested this year along with three other department officials after an investigation into a disbanded narcotics task force. The inquiry found that Powell had actively tried to shield wrongdoing by the task force. That led to his indictment on charges including violating the oath of office, criminal attempt to commit a felony and influencing a witness.As details of Arbery's death slowly emerged and were reported in The Brunswick News, Arbery's mother, increasingly distraught, called the department. She said that she had been told one thing but that the newspaper had reported something else entirely.Cooper's faith was shaken. "It's hard when you can't really believe what authority tells you, you know?" she said. "When you just cannot believe the people that's supposed to look out for all people. And when you question that, it's not a good feeling."Attempts to reach Gregory McMichael late last month were unsuccessful. In a brief phone conversation late last month, Travis McMichael, who runs a company that gives custom boat tours, declined to comment, citing the continuing investigation.The two men made a brief appearance in Glynn County Magistrate Court on Friday afternoon, but court officials said they did not enter a plea. No information about their lawyers was immediately available.Questions about the handling of Arbery's case extend beyond the police department and to Barnhill, the prosecutor who told police that there was insufficient probable cause to arrest the McMichaels.In an email Barnhill wrote to the state attorney general's office April 7, he asked to be taken off the case, stating that his son, an assistant district attorney in the Brunswick prosecutor's office, had handled a felony probation revocation case involving Arbery. He also said Gregory McMichael had helped with "a previous prosecution of Arbery."Court records show that Arbery was convicted of shoplifting and of violating probation in 2018; according to local news reports, he was indicted five years earlier for taking a handgun to a basketball game.Barnhill's office most recently drew attention beyond south Georgia for its prosecution of a black woman in rural Coffee County who had helped a first-time voter use a voting machine in the 2012 election. In 2018, a jury found the woman not guilty of multiple felonies. Her lawyers called the case "a racially motivated targeted prosecution."J. Peter Murphy, a Glynn County commissioner, on Friday defended the Police Department's decision to make no arrests in the shooting of Arbery. Murphy said the agency had been advised not to make arrests by both Barnhill and officials at the office of Johnson, the district attorney in Brunswick who formally asked to be taken off the case four days after the shooting. Neither prosecutor could be reached for comment."Tell me what the agency did wrong when its men and women were told several times not to arrest anyone?" Murphy said, referring to police. "What were they supposed to do?
