Many rich nation politicians and their media often point to their rising emissions as proof that they urgently need to be bound to emission reduction targets in the same way rich nations are. But what is often easily forgotten or omitted is that greenhouse gases can stay in the atmosphere for a very long time. In other words, historic emissions matter. Historical data show that the majority of greenhouse emissions have been by rich nations, known as "Annex I" countries in climate negotiation speak: Apart from China and India, the remainder of the top 10 historical greenhouse gas emitters have been from Annex I countries. This is why 2 decades ago the climate negotiations started by understanding there were "common but differentiated principles" and why "Annex I" countries were initially given target emissions while the rest were to be given space to grow given the urgent need for poverty alleviation and development. Furthermore, climate negotiations frameworks have always said developing nations need to avoid a polluting path to industrialization, so they can’t just use historic emissions injustice as an excuse not to do anything. At the same time, the dirty path to development was also the cheap and easy path which developing countries need to avoid, so it was also agreed that the Annex I countries should help developing countries in various ways. Needless to say much of this has not really happened. Unlike other autoimmune rheumatic diseases, such as rheumatoid arthritis or systemic lupus erythematosus, autoantibodies have not yet been reported to be a feature of AS. We therefore wished to determine if plasma from patients with AS contained autoantibodies and if so, characterize and quantify this response in comparison to patients with Rheumatoid Arthritis (RA) and healthy controls. Two high-density nucleic acid programmable protein arrays expressing a total of 3498 proteins were screened with plasma from 25 patients with AS, 17 with RA and 25 healthy controls. Autoantigens identified were subjected to Ingenuity Pathway Analysis in order to determine patterns of signalling cascades or tissue origin. 44% of patients with Ankylosing Spondylitis demonstrated a broad autoantibody response, as compared to 33% of patients with RA and only 8% of healthy controls. Individuals with AS demonstrated autoantibody responses to shared autoantigens, and 60% of autoantigens identified in the AS cohort were restricted to that group. The AS patients autoantibody responses were targeted towards connective, skeletal and muscular tissue, unlike those of RA patients or healthy controls. Thus, patients with AS show evidence of systemic humoral autoimmunity and multispecific autoantibody production. Nucleic Acid Programmable Protein Arrays constitute a powerful tool to study autoimmune diseases. Full Article

oric The Maximum Separation Subspace in Sufficient Dimension Reduction with Categorical Response By Published On :: 2020 Sufficient dimension reduction (SDR) is a very useful concept for exploratory analysis and data visualization in regression, especially when the number of covariates is large. Many SDR methods have been proposed for regression with a continuous response, where the central subspace (CS) is the target of estimation. Various conditions, such as the linearity condition and the constant covariance condition, are imposed so that these methods can estimate at least a portion of the CS. In this paper we study SDR for regression and discriminant analysis with categorical response. Motivated by the exploratory analysis and data visualization aspects of SDR, we propose a new geometric framework to reformulate the SDR problem in terms of manifold optimization and introduce a new concept called Maximum Separation Subspace (MASES). The MASES naturally preserves the “sufficiency” in SDR without imposing additional conditions on the predictor distribution, and directly inspires a semi-parametric estimator. Numerical studies show MASES exhibits superior performance as compared with competing SDR methods in specific settings. Full Article

Besides being an exploratory tool used to reduce the dimensionality of multivariate data, it allows estimation of common factors that often have an interesting theoretical interpretation in real problems. However, standard factor analysis is only applicable when the variables are scaled, which is often inappropriate, for example, in data obtained from questionnaires in the field of psychology,where the variables are often categorical. In this framework, we propose a factor model for the analysis of multivariate ordered and non-ordered polychotomous data. The inference procedure is done under the Bayesian approach via Markov chain Monte Carlo methods. Two Monte-Carlo simulation studies are presented to investigate the performance of this approach in terms of estimation bias, precision and assessment of the number of factors. We also illustrate the proposed method to analyze participants' responses to the Motivational State Questionnaire dataset, developed to study emotions in laboratory and field settings. Full Article

Here, we develop and fit a latent discrete Markov random field model for the purpose of identifying and classifying historical forest communities based on spatially referenced multivariate tree species counts across Wisconsin. We show empirically for the actual dataset and through simulation that our latent Markov random field modeling approach improves prediction and parameter estimation performance. For model fitting we introduce a new stochastic approximation algorithm which enables computationally efficient estimation and classification of large amounts of spatial multivariate count data. We show how restricted fusion can be implemented and develop an efficient MCMC (Markov chain Monte Carlo) method for posterior computation. The performance of the proposed method is investigated on simulated data and we illustrate its application on real data from EU-SILC (European Union Statistics on Income and Living Conditions).
