They taught "that sinners had the natural ability to believe, and that evangelistic methods could overcome their 'moral' inability through the persuasive power of the Gospel." "Finney and Mahan applied this same understanding to the Christian's growth toward spiritual maturity. To be sanctified, they insisted, required only the same kind of simple, instantaneous faith one exercised to be converted." In 1836 both men experienced what they called "baptisms of the Holy Ghost" which they believed not only freed them from committing sin but also removed their tendency toward sin. Contributing to the spread of this "Holiness" doctrine were the popular camp meeting revivals of the first half of the 1800s, the ministry of Phoebe Palmer (1807-1874) (who taught that sanctification could be reached instantaneously by an act of faith) and the "Prayer Revival" of 1857-1858 (sometimes called the Third Great Awakening). There was also much unrest in Methodist circles as many felt the denomination had lost its fervor. The Wesleyan Methodist (in 1843) and the Free Methodist (in 1860) left the denomination to form the first Holiness denominations. Until the 1890s the Holiness Movement was largely a Methodist phenomenon, but as the Methodists settled more into mainstream Christianity tensions escalated into a schism which resulted in new, non-Methodist, Holiness denominations. These included the Church of God, Anderson, Indiana (1880), Church of the Nazarene (1908) and Pilgrim Holiness Church (1897). The Holiness adherents saw themselves as the true descendents of the Wesleys and practiced strict moral ethics, abstinence from worldly pleasures and amusements and a strong belief in entire sanctification (also known as the "second blessing" and the baptism of the Holy Spirit). More importantly "Holiness teaching offered 19th-century evangelicals a means of overcoming their sectarian conflicts. Doctrine might divide, but the experience of a pure heart would unite all true believers against the threats posed by religious formalism, atheism, and Roman Catholicism." This Holiness emphasis would continue to be spread throughout the 19th century by individuals and groups as diverse as the Salvation Army, Quakers, D. L. Moody, Hannah Whitall Smith, the Y.M.C.A., the Keswick Movement and Oswald Chambers. A brief explanation concerning some of those might prove helpful. ... But that holiness is not found in either short-cuts or self effort. **It is found as we pursue righteousness (2 Timothy 2:22) laying aside the deeds of the flesh (Colossians 3:5-10) through the power of the Holy Spirit (Galatians 5:16) and as we behold the glory of the Lord (2 Corinthians 3:18). The Holiness Movement was in many ways a reaction to the dead orthodoxy and lifeless spirituality that infiltrated so much of Christianity during the nineteenth-century. initial Modern Transdisciplinarity: Results of the Development of the Prime Cause and Initial Ideas By Published On :: 2022-05-11 Aim/Purpose This paper focuses on systematizing and rethinking the conformity of modern transdisciplinarity with its prime cause and initial ideas. Background The difficulties of implementing transdisciplinarity into science and education are connected with the fact that its generally accepted definition, identification characteristics, and methodological features are still missing. In order to eliminate these disadvantages of transdisciplinarity, its prime cause and initial ideas had to be detected. It is also important to analyze the correspondence of the existing opinions about transdisciplinarity with the content of these cause and ideas. Methodology The qualitative analysis of the literature reviews on the subject of transdisciplinary was used in order to determine the correspondence of the opinions about the transdisciplinarity with the meaning of its prime cause and initial ideas. These opinions had to be generalized as well. Through this method, it was possible to detect and classify opinions into 11 groups including 39 stereotypes of transdisciplinarity. For substantiation of transdisciplinary approaches that are consistent with the approaches of contemporary science, C.F. Gauss random variables normal distribution was used. The “Gauss curve” helped to show the place of transdisciplinary and systems transdisciplinary approaches in the structure of academic and systems approaches. The “Gauss curve” also demonstrated the step-by-step “broadening of the scientific worldview horizon due to sequential intensification of synthesis, integration, unification, and generalization of the disciplinary knowledge.” Contribution After reconsideration of the results on qualitative analysis of the literature reviews, the generalized definition of transdisciplinarity could be formulated, including the definition for transdisciplinary and systems transdisciplinary approaches. It was proven that transdisciplinarity is a natural stage for the development of contemporary science and education, and the transdisciplinary approaches were able to suggest the methods and tools to solve the complex and poorly structured problems of science and the society. Findings Many existing stereotypes of transdisciplinarity do not meet its prime cause and initial ideas. Such stereotypes do not have deep philosophic and theoretical substantiation. They also do not suggest the transdisciplinary methods and tools. Thus, the authors of such stereotypes often claim them to be transdisciplinary or suggest perceiving them as transdisciplinarity. This circumstance is the reason why many disciplinary scientists, practitioners, and initiators of higher education view transdisciplinarity as a marginal direction of contemporary science. Based on the generalized definition of transdisciplinarity, as well as its prime cause and initial ideas, it was shown that transdisciplinarity is presented in contemporary science in the form of two different approaches, i.e., the transdisciplinary approach and systems transdisciplinary approach. The objective of the transdisciplinary approach is to ensure science development at the stage of synthesis and integration of disciplinary knowledge, while the objective of the systems transdisciplinary approach is to ensure that the problems of modern society are solved through unification and generalization of the disciplinary knowledge. Recommendation for Researchers The researchers should consider that within the limits of the transdisciplinary approach, the disciplinary specialists are managed. Within the limits of the systems transdisciplinary approach, the disciplinary knowledge is managed. Thus, the transdisciplinary approach is efficient for organization and research with participation of the scientists of the complementary disciplines. An example of such research can be a team of researchers of medical disciplines and complementary disciplines from chemistry, physics, and engineering. The systems transdisciplinary approach is efficient for organization and performance of research with participation of the scientists of non-complementary disciplines such as economics, physics, meteorology, chemistry, ecology, geology, and sociology. Future Research In terms of the main initial idea, transdisciplinarity is formed as a global approach. The global approach should have a traditional institutional form: it should be a science discipline (meta-discipline) and have carriers with the transdisciplinary worldview. Training for such carriers can be organized by the universities within the limits of the systems transdisciplinarity departments and Centers of Systems Transdisciplinary Retraining for Disciplinary Specialists. Thus, it is reasonable to initiate discussion for the idea to reform the disciplinary structure of the universities considering creation of such departments and centers. Full Article

The main idea is to combine all the different values of every single criterion or attribute, to form the first proposal of the so-called multiclusters, obtaining in this way the maximum number of clusters for the whole dataset. The multiclusters thus obtained, are themselves clustered in a second step, according to the desired final number of clusters. Background: Popular cluster methods for categorical data, such as the well-known K-Modes, usually select the initial sets by means of some random process. This fact introduces some randomness in the final results of the algorithms. We explore a different application of the clustering methodology for categorical data that overcomes the instability problems and ultimately provides a greater clustering efficiency. Methodology: For assessing the performance of the proposed algorithm and its comparison with K-Modes, we apply both of them to categorical databases where the response variable is known but not used in the analysis. In our examples, that response variable can be identified to the real clusters or classes to which the observations belong. With every data set, we perform a two-step analysis. In the first step we perform the clustering analysis on data where the response variable (the real clusters) has been omitted, and in the second step we use that omitted information to check the efficiency of the clustering algorithm (by comparing the real clusters to those given by the algorithm). Contribution: Simplicity, efficiency and stability are the main advantages of the multicluster method. Findings: The experimental results attained with real databases show that the multicluster algorithm has greater precision and a better grouping effect than the classical K-modes algorithm. Recommendations for Practitioners: The method can be useful for those researchers working with small and medium size datasets, allowing them to detect the underlying structure of the data in an intuitive and reasonable way. Recommendation for Researchers: The proposed algorithm is slower than K-Modes, since it devotes a lot of time to the calculation of the initial combinations of attributes. The reduction of the computing time is therefore an important research topic. Future Research: We are concerned with the scalability of the algorithm to large and complex data sets, as well as the application to mixed data sets with both quantitative and qualitative attributes. To secure investment, entrepreneurs can signal about their venture's latent potential by aligning themselves with reliable third parties. Such affiliations affirm the new venture's legitimacy and provide substantive benefits in the form of mentoring, access to resources, and ongoing monitoring. However, early stage financing is an especially "noisy" signaling environment owing to the large number of startups seeking funding, many of which will not survive. The real value of third party affiliations in this context resides in their ability to unlock the potential of other more pedestrian signals, such as the entrepreneur's characteristics and actions that might otherwise go unnoticed. We borrow from the sensemaking literature to explain how third party affiliation signals disambiguate signals with multiple possible interpretations so that potential investors interpret them positively. Findings support our theory that a startup's characteristics and actions are signals that remain relatively unnoticed unless a startup combines them with a third party affiliation that enhances the signal's value, thus increasing the likelihood of receiving external capital. Under uncertainty, acolytes (i.e., subordinates with work connections to high-reputation industry leaders) are likely to benefit in terms of signaling fitness for promotion in the external job market. Analysis of promotion outcomes of coaches in the NFL over 31 years showed that the acolyte effect was reduced for individuals for whom uncertainty was the least (acolytes with considerable industry experience or high centrality in the co-worker industry network). There was no support for either a knowledge-transfer or an intrinsic quality explanation for why acolytes initially gained advantage. Rather, the evidence supported the idea that ties to high-reputation leaders were somewhat randomly distributed so that acolytes faced ex post settling up consequences after their promotions: fewer further promotions or lateral moves, more demotions. Thus, acolytes initially benefited from a loose-linkage between their unobservable quality and signals offered by their industry-leader ties, but they also suffered as the unreliability of social network signals became evident. The results suggest that a competitive job market may exhibit self-correction over time. We offer countervailing theory and evidence to the prevailing view that high-reputation third-party endorsements perpetuate a rich-get-richer social structure resistant to performance outcomes. The aims were to highlight the link to infrastructures and processes like GEOSS or DataONE and to discuss the data standards and informatics architecture that will be followed by the EU BON project. During the three-days of the workshop, the participants of the meeting discussed the important aspects regarding the informatics architecture and decided on the next steps to develop a new open-access platform for sharing biodiversity data and tools in order to advance the European biodiversity knowledge. On the first day, the aim was to highlight the link of EU BON with GEOSS, GEO BON and other processes like DataONE to find synergies and to build on work that was conducted in these processes. On the second day, detailed discussion on the specific tasks of the workgroup took place. The afternoon session was split into 3 different tracks where issues like architectural design, review and guidelines for using data standards, the design of monitoring sites and the gap analysis of existing biodiversity data were analyzed and discussed. It was agreed that a new platform is needed which should be built on existing solutions. Thus, the platform will use the technical solutions of the DataONE network that will be adjusted to the specific needs of the EU BON project. EU BON Partners will implement DataONE Member Nodes to start the process and a DataOne coordinating node may be established towards 2015. The microtubule-associated protein tripartite motif containing 46 (TRIM46) was reported to regulate axon specification, AIS assembly, and neuronal polarity through the bundling, or fasciculation, of microtubules in the proximal axon. However, these claims are based on TRIM46 knockdown in cultured neurons. To investigate TRIM46 function in vivo, we examined male and female TRIM46 knock-out mice. Contrary to previous reports, we find that TRIM46 is dispensable for axon specification and AIS formation. TRIM46 knock-out mice are viable, have normal behavior, and have normal brain structure. Thus, TRIM46 is not required for AIS formation, axon specification, or nervous system function. However, we confirm that TRIM46 is required for microtubule fasciculation. We also show TRIM46 enrichment in the first ~100 μm of axon occurs independently of ankyrinG (AnkG) in vivo, although AnkG is required to restrict TRIM46 only to the AIS. Our results highlight the need for further investigation of the mechanisms by which the AIS and microtubules interact to shape neuronal structure and function. Full Article

initial Tempus ECO initial setup summary not matching timing report results By community.cadence.com Published On :: Sat, 29 Jun 2024 01:51:01 GMT We are currently setting up the Tempus flow and have ran into some mismatched data regarding ECO and timing reports. I generated a timing report before running ECO and saw six total setup violations. When running opt_signoff -setup, the initial setup summary that was printed in the shell only showed one violation. I can see that violation from the initial setup summary in my pre-ECO timing report and it is not the worst path. Upon further investigation, I forced the tool to try to fix setup on one of the other five violations from the timing report using the opt_signoff_select_setup_endpoints attribute and the tool said that the endpoint had positive slack and would be ignored. Since the time of regulatory approval, however, real-world data have been lacking. This study investigated the efficacy, safety, and outcome predictors of [177Lu]Lu-PSMA-617 at a major U.S. academic center. Methods: Patients with mCRPC who received [177Lu]Lu-PSMA-617 at the Johns Hopkins Hospital outside clinical trials were screened for inclusion. Patients who underwent [177Lu]Lu-PSMA-617 and had available outcome data were included in this study. Outcome data included prostate-specific antigen (PSA) response (≥50% decline), PSA progression-free survival (PFS), and overall survival (OS). Toxicity data were evaluated according to the Common Terminology Criteria for Adverse Events version 5.03. The study tested the association of baseline circulating tumor DNA mutational status in homologous recombination repair, PI3K alteration pathway, and aggressive-variant prostate cancer–associated genes with treatment outcome. Baseline PSMA PET/CT images were analyzed using SelectPSMA, an artificial intelligence algorithm, to predict treatment outcome. Associations with the observed treatment outcome were evaluated. Results: All 76 patients with PSMA-positive mCRPC who received [177Lu]Lu-PSMA-617 met the inclusion criteria. A PSA response was achieved in 30 of 74 (41%) patients. The median PSA PFS was 4.1 mo (95% CI, 2.0–6.2 mo), and the median OS was 13.7 mo (95% CI, 11.3–16.1 mo). Anemia of grade 3 or greater, thrombocytopenia, and neutropenia were observed in 9 (12%), 3 (4%), and 1 (1%), respectively, of 76 patients. Transient xerostomia was observed in 23 (28%) patients. The presence of aggressive-variant prostate cancer–associated genes was associated with a shorter PSA PFS (median, 1.3 vs. 6.3 mo; P = 0.040). No other associations were observed between circulating tumor DNA mutational status and treatment outcomes. Eighteen of 71 (25%) patients classified by SelectPSMA as nonresponders had significantly lower rates of PSA response than patients classified as likely responders (6% vs. 51%; P < 0.001), a shorter PSA PFS (median, 1.3 vs. 6.3 mo; P < 0.001), and a shorter OS (median, 6.3 vs. 14.5 mo; P = 0.046). Conclusion: [177Lu]Lu-PSMA-617 offered in a real-world setting after regulatory approval in the United States demonstrated antitumor activity and a favorable toxicity profile. Artificial-intelligence–based analysis of baseline PSMA PET/CT images may improve patient selection. Validation of these findings on larger cohorts is warranted. Full Article

initial Inherit, initial, unset, revert By www.quirksmode.org Published On :: Wed, 02 Jun 2021 11:55:44 +0100 Today we’re going to take a quick look at a few special CSS keywords you can use on any CSS property: inherit, initial, revert, and unset. Also, we will ask where and when to use them to the greatest effect, and if we need more of those keywords. The first three were defined in the Cascading Level 3 spec, while revert was added in Cascading Level 4. Despite 4 still being in draft revert is already supported. See also the MDN revert page, Chris Coyier’s page, and my test page inherit The inherit keyword explicitly tells an element that it inherits the value for this declaration from its parent. Let’s take this example: .my-div { margin: inherit; } .my-div a { color: inherit; } The second declaration is easiest to explain, and sometimes actually useful. It says that the link colour in the div should be the same as the text colour. The div has a text colour. It’s not specified here, but because color is inherited by default the div gets the text color of its parent. Let’s say it’s black. Links usually have a different colour. As a CSS programmer you frequently set it, and even if you don’t browsers automatically make it blue for you. Here, however, we explicitly tell the browsers that the link colour should be inherited from its parent, the div. In our example links become black. (Is this a good idea? Occasionally. But if you remove the colour difference between links and main text, make sure your links are underlined. Your users will thank you.) Now let’s look at the margin: inherit. Normally margins don’t inherit, and for good reason. The fact that an element has margin-left: 10% does not mean all of its descendents should also get that margin. In fact, you most likely don’t want that. Margins are set on a per-case basis. This declaration tells the div to use the margin specified on its parent, however. This is an odd thing to specify, and I never saw a practical use case in the wild. Still CSS, being ridiculously powerful, allows it. In any case, that’s how the inherit keyword works. Using it for font sizes or colours may occasionally be a good idea. In other contexts - rarely. And keep the difference between inheriting and non-inheriting properties in mind. It’s going to be important later on. initial The initial keywords sets the property back to its initial value. This is the value specified in the W3C specification for that property. Initial values from the spec are a bit of a mixed bag. Some make sense, others don’t, really. float: none and background-color: transparent are examples of the first category. Of course an element does not have a background colour without you specifying one, nor does it float automatically. Others are historically determined, such as background-repeat: repeat. Back in the Stone Age before CSS all background images repeated, and the CSS1 specification naturally copied this behaviour. Still others are essentially arbitrary, such as display: inline. Why did W3C opt for inline instead of block? I don’t know, and it doesn’t really matter any more. They had to decide on an initial value, and while inline is somewhat strange, block would be equally strange. In any case, the initial keyword makes the property revert to this initial value from the specification, whether that makes sense or not. unset When we get to the unset value the distinction between inheriting and non-inheriting properties becomes important. unset has a different effect on them. If a property is normally inherited, unset means inherit. If a property is normally not inherited, unset means initial. revert revert, the newest of these keywords, also distinguishes between inheriting and non-inheriting properties. If a property is normally inherited, revert means inherit. If a property is normally not inherited, revert reverts to the value specified in the browser style sheet. all Finally, we should treat all. It is not a value but a property, or rather, the collection of all CSS properties on an element. It only takes one of the keywords we discussed, and allows you to apply that keyword to all CSS properties. For instance: .my-div { all: initial; } Now all CSS properties on the div are set to initial. Examples The reaction of my test page to setting the display of all elements to the four keywords is instructive. My test script sets the following style: body * { display: [inherit | initial | unset | revert] !important; } The elements react as follows: display: inherit: all elements now inherit their display value from the body. Since the body has display: block all elements get that value, whether that makes sense or not. display: initial: the initial value of display is inline. Therefore all elements get that value, whether that makes sense or not. display: unset: display does not inherit. Therefore this behaves as initial and all elements get display: inline. display: revert: display does not inherit. Therefore the defaults of the browser style sheet are restored, and each element gets its proper display — except for the dl, which I had given a display: grid. This value is now supplanted by the browser-provided block. Unfortunately the same test page also contains a riddle I don’t understand the behaviour of <button>s when I set color to the four keywords: color: inherit: all elements, including <button>s, now inherit their colour from the body, which is blue. So all text becomes blue. color: initial: since the initial value of color is black, all elements, including <button>s, become black. color: unset: color inherits. Therefore this behaves as inherit and all elements, including <button>s, become blue. color: revert: This is the weird one. All elements become blue, except for <button>s, which become black. I don’t understand why. Since colors inherit, I expected revert to work as inherit and the buttons to also become blue. But apparently the browser style sheet of button {color: black} (more complicated in practice) is given precedence. Yes, revert should remove author styles (the ones we write), and that would cause the black from the browser style sheet to be applied, but only if a property does not inherit — and color does. I don’t know why the browser style sheet is given precedence in this case. So I’m going to cop out and say form elements are weird. Practical use: almost none The purpose of both unset and revert is to wipe the slate clean and return to the initial and the browser styles, respectively — except when the property inherits; in that case, inheritance is still respected. initial, meanwhile, wipes the slate even cleaner by also reverting inheriting properties to their initial values. This would be useful when you create components that should not be influenced by styles defined elsewhere on the page. Wipe the slate clean, start styling from zero. That would help modularisation. But that’s not how these keywords work. We don’t want to revert to the initial styles (which are sometimes plain weird) but to the browser style sheet. unset comes closest, but it doesn’t touch inherited styles, so it only does half of what we want. So right now these keywords are useless — except for inherit in a few specific situations usually having to do with font sizes and colours. New keyword: default Chris Coyier argues we need a new value which he calls default. It reverts to the browser style sheet in all cases, even for inherited properties. Thus it is a stronger version of revert. I agree. This keyword would be actually useful. For instance: .my-component,.my-component * { all: default; font-size: inherit; font-family: inherit; color: inherit; } Now we have a component that’s wiped clean, except that we decide to keep the fonts and colour of the main page. The rest is a blank slate that we can style as we like. That would be a massive boon to modularisation. New keyword: cascade For years now I have had the feeling that we need yet another keyword, which I’ll call cascade for now. It would mean “take the previous value in the cascade and apply it here.” For instance: .my-component h2 { font-size: 24px; } .my-other-component h2 { font-size: 3em; } h2#specialCase { font-size: clamp(1vw,cascade,3vw) } In this (slightly contrived) example I want to clamp the font-size of a special h2 between 1vw and 3vw, with the preferred value being the one defined for the component I’m working in. Here, cascade would mean: take the value the cascade would deliver if this rule didn’t exist. This would make the clamped font size use either 24px or 3em as its preferred value, depending on which component we’re in. The problem with this example is that it could also use custom properties. Just set --h2size to either 24px or 3em, use it in the clamp, and you’re done. .my-component h2 { --h2size: 24px; font-size: var(--h2size); } .my-other-component h2 { --h2size: 3em; font-size: var(--h2size); } h2#specialCase { font-size: clamp(1vw,var(--h2size),3vw) } Still, this is but the latest example I created. I have had this thought many, many times, but because I didn’t keep track of my use cases I’m not sure if all of them could be served by custom properties. Also, suppose you inherit a very messy CSS code base with dozens of components written at various skill levels. In that case adding custom properties to all components might be impractical, and the cascade keyword might help. Anyway, I barely managed to convince myself, so professional standard writers will certainly not be impressed.
