Apolipoprotein A-I (ApoA-I) of high-density lipoprotein (HDL) is essential for the transportation of cholesterol between peripheral tissues and the liver. However, specific mutations in Apolipoprotein A-I (ApoA-I) of high-density lipoprotein (HDL) are responsible for a late-onset systemic amyloidosis, the pathological accumulation of protein fibrils in tissues and organs. Carriers of these mutations do not exhibit increased cardiovascular disease risk despite displaying reduced levels of ApoA-I/ HDL-cholesterol. To explain this paradox, we show that the HDL particle profile of patients carrying either L75P or L174S ApoA-I amyloidogenic variants a higher relative abundance of the 8.4 nm vs 9.6 nm particles, and that serum from patients, as well as reconstituted 8.4 and 9.6 nm HDL particles (rHDL), possess increased capacity to catalyze cholesterol efflux from macrophages. Synchrotron radiation circular dichroism and hydrogen-deuterium exchange revealed that the variants in 8.4 nm rHDL have altered secondary structure composition and display a more flexible binding to lipids compared to their native counterpart. The reduced HDL-cholesterol levels of patients carrying ApoA-I amyloidogenic variants are thus balanced by higher proportion of small, dense HDL particles and better cholesterol efflux due to altered, region-specific protein structure dynamics.

The backbone of all sphingolipids (SLs) is a sphingoid long-chain base (LCB) to which a fatty acid is N-acylated. Considerable variability exists in the chain length and degree of saturation of both of these hydrophobic chains, and recent work has implicated ceramides with different LCBs and N-acyl chains in distinct biological processes; moreover, they may play different roles in disease states and possibly even act as prognostic markers. We now demonstrate that the half-life, or turnover rate, of ceramides containing diverse N-acyl chains is different. By means of a pulse-labeling protocol using stable-isotope, deuterated free fatty acids, and following their incorporation into ceramide and downstream SLs, we show that very-long-chain (VLC) ceramides containing C24:0 or C24:1 fatty acids turn over much more rapidly than long-chain (LC) ceramides containing C16:0 or C18:0 fatty acids due to the more rapid metabolism of the former into VLC sphingomyelin and VLC hexosylceramide. In contrast, d16:1 and d18:1 ceramides show similar rates of turnover, indicating that the length of the sphingoid LCB does not influence the flux of ceramides through the biosynthetic pathway. Together, these data demonstrate that the N-acyl chain length of SLs may not only affect membrane biophysical properties but also influence the rate of metabolism of SLs so as to regulate their levels and perhaps their biological functions.

Bacterial regulatory RNAs (sRNAs) are important players to control gene expression. In Staphylococcus aureus, SprC is an antivirulent trans-acting sRNA known to base-pair with the major autolysin atl mRNA, preventing its translation. Using MS2-affinity purification coupled with RNA sequencing, we looked for its sRNA-RNA interactome and identified 14 novel mRNA targets. In vitro biochemical investigations revealed that SprC binds two of them, czrB and deoD, and uses a single accessible region to regulate its targets, including Atl translation. Unlike Atl regulation, the characterization of the SprC-czrB interaction pinpointed a destabilization of the czrAB cotranscript, leading to a decrease of the mRNA level that impaired CzrB zinc efflux pump expression. On a physiological standpoint, we showed that SprC expression is detrimental to combat against zinc toxicity. In addition, phagocyctosis assays revealed a significant, but moderate, increase of czrB mRNA levels in a sprC-deleted mutant, indicating a functional link between SprC and czrB upon internalization in macrophages, and suggesting a role in resistance to both oxidative and zinc bursts. Altogether, our data uncover a novel pathway in which SprC is implicated, highlighting the multiple strategies used by S. aureus to balance virulence using an RNA regulator.

JUDY WOODRUFF: But first: Sunday’s elections Austria were the latest ample of a shift to the right Europe’s politics, as 31-year-old Christian Kurz was elected chancellor on an anti-immigration platform.

He may now form a government with a far-right party founded in the 1950s by former Nazis.

That follows recent elections in Germany, where a far-right party roiled the race and dealt a blow to returning leader Angela Merkel.

In Sweden, too, there is a strong challenge from the right and a neo-Nazi group that looks stand in elections next year.

Special correspondent Malcolm Brabant ha been surveying the political landscape in Germany and Sweden, and he begins his report in Scandinavia.

MALCOLM BRABANT, Special Correspondent: In a Gothenburg parking lot, supporters of the Nordic Resistance Movement form up for what they hope will be their biggest-ever march, to propagate an ideology espoused by mother of eight Paulina Forslund.

PAULINA FORSLUND, Nordic Resistance Movement: When white becomes the minority, they will be destroyed. I want my children to have a secure future. I want them not only for them to have a secure Sweden. I want them to have a secure world. And I want other people to fight for the same thing.

MALCOLM BRABANT: When addressing her fellow neo-Nazis, Forslund’s rhetoric sharpens.

PAULINA FORSLUND (through interpreter): I’m the welder’s daughter, the forester’s grandchild. My line consists of hardworking men and women. It’s people like them we can thank for the welfare system that our lying politicians are now giving away to imported scum.

MALCOLM BRABANT: Clearly expecting trouble, the movement’s leaders have a muscular protection detail, marching past a silent protest. The sign reads “No Nazis on our streets.”

This protester would only give her name as Johanna.

JOHANNA, Anti-Nazi Protester: They are racist people. They are people who think that certain people are better than others, and I will not stand for that. It’s not something I think has a place in a modern society.

MALCOLM BRABANT: Experts say the resistance movement is recruiting aggressively, and believe this demonstration is emblematic of the rise of the far right.

It took place on Yom Kippur, the Jewish day of atonement.

Allan Stutzinky is leader of Gothenburg’s Jewish community.

ALLAN STUTZINKY, Jewish Community Leader (through interpreter): Nazism has returned. The descendants of the murderers are organizing the same marches today, waving the same flags, shouting the same slogans, and have the same racist agenda.

MALCOLM BRABANT: Anna Johansson is a member of the governing Social Democrat Party. It’s considering outlawing the Nordic Resistance Movement.

ANNA JOHANSSON, Swedish Social Democratic Party: In Sweden and in Denmark, and in other countries, extreme parties are growing, and the hatred is spreading around.

MALCOLM BRABANT: “Go home to mama,” he shouts. “Nazi pigs,” chant the anti- fascist protesters, as a bottle flies through the air.

DAMON, Nordic Resistance Movement: If someone calls themselves a Nazi, most of us would dissociate with that person. That’s nothing we stand for ourselves. I never call myself a Nazi. I’m a national socialist.

MALCOLM BRABANT: Hitler’s party was also called National Socialist, but Damon, a 40-year-old welder, insists he’s a nonviolent family man.

DAMON: The demographic landscape of our — of the whole of Europe is changing, so, basically, it’s a concern on preserving my heritage for my family and our kin.

MALCOLM BRABANT: This demonstration has been stopped short of its destination. The Nordic Resistance Movement is currently trapped between a line of police and anti-fascist protesters. And it looks as though this demonstration isn’t going any further.

Violence briefly erupts as the resistance movement tries to break through police lines, and several marchers are arrested.

PAULINA FORSLUND: We are not your enemy. We are the government’s enemy.

They say we live in a democracy, but we have never had an election about if we want to take all these people in.

MALCOLM BRABANT: When Europe’s refugee crisis began in 2015, Sweden copied Germany’s open-door policy, and 160,000 migrants entered the country. Two years on, Sweden has tighter borders and has begun deporting some of the newcomers.

The new atmosphere alarms Floid Gumbo, entertaining an anti-Nazi rally.

FLOID GUMBO, Singer Originally from Zimbabwe: I came to Sweden over 20 years ago. The climate in Sweden, the people were so friendly, and things were completely different, more welcoming. And I feel like things have sort of gradually changed.

I’m very concerned, because I have children, because I’m thinking what I experienced here is not the same kind of climate, atmosphere that they are going to experience here.

ANNA JOHANSSON: It’s not so long ago that the Nazis ruined Europe. And that makes me very worried. The German elections were terrifying, I think.

MALCOLM BRABANT: Johansson is referring to last month’s success of the right-wing Alternative For Germany Party, or AFD, when it entered Parliament for the first time with 13 percent of the vote.

HUGH BRONSON, Alternative For Germany Party: The AFD only came into existence because Merkel deserted the traditional conservative Christian voters. They were looking for a home, and the AFD has offered them a safe place.

MALCOLM BRABANT: Hugh Bronson is deputy leader of the AFD in Berlin.

Now his party, the third largest in Parliament, is demanding that Angela Merkel imposes tougher immigration rules.

Your opponents claim that you are a party of hate. What’s your response to that?

HUGH BRONSON: We embrace foreigners who respect our laws, pay their taxes, send their children to school, and go about their normal life. The problem is with people who abuse the system to have a better life, or let others pay for their better lives, or who are criminals.

MALCOLM BRABANT: Outside the opera house in Dresden, former East Germany, singer Luca Bergelt is dismayed by the political landscape shifting to the right.

LUCA BERGELT, Singer: My fear is that they will tear Europe apart. They are going to raise up the walls again. They’re going to build new walls between the countries, and that Europe will get more close into itself.

MALCOLM BRABANT: Anti-immigrant sentiment is strong in Dresden. The city was the birthplace of a pan-European anti-Islamic movement, and it delivered the largest number of votes for the right-wing party.

On a holiday to celebrate German unification after the fall of communism, retired engineer Wilfried Schmidt explained why he sent a message to Angela Merkel.

WILFRIED SCHMIDT, Retired Engineer (through interpretor): Let’s put it this way. We all need to recognize that Germany is undergoing social changes that are becoming harder to control. For one, there is mass immigration from difficult regions that is increasingly uncontrollable, of people with entirely different concepts of life, from fundamental differently structured societies that are problematic.

MALCOLM BRABANT: About one million migrants poured into Germany in 2015. Chancellor Merkel consistently defended her pro-refugee policies, but now she has been punished by voters who believe she ignored their concerns.

Chancellor Merkel has promised to listen to the people who voted for the AFD, and she says she’s going to try to win them over with what she calls good politics. But she will not countenance having the party in her coalition.

But the chancellor needs to find new partners who are prepared to be tough on immigration.

As she tries to forge a coalition, the chancellor has agreed to put an annual cap of 200,000 on the number of immigrants, something she previously refused to do. But will it be enough to woo back people who deserted her at the election?

A question for Werner Patzelt, a political scientist at Dresden University.

WERNER PATZELT, Dresden University: Since Chancellor Merkel has made so many U-turns in German domestic politics, it wouldn’t be a surprise if she would try to do a U-turn, also winning back AFD voters.

But this is a really hard political task, because so many of them are so much disappointed by the Christian Democratic Union in general, and by Chancellor Merkel in particular, that they will do anything to avoid going back.

MALCOLM BRABANT: Back in Sweden, the governing party is horrified at the concept of conceding ground to right-wingers, and is trying to isolate them.

ANNA JOHANSSON: Experience shows that, when you adopt the ideas from these right-wing parties, they spread. These parties have their agenda implemented by other parties. And I wouldn’t want to see that happen in Sweden.

FLOID GUMBO: We’re all human beings. We share this world. We’re all here. There’s enough space for us all.

MALCOLM BRABANT: But that’s an appeal that an increasing number of Swedes are rejecting, as the country and much of Europe go through a crisis of identity.

Estimates of forest carbon stores and flux for California circa 1990 were modeled from forest inventory data in support of California's legislatively mandated greenhouse gas inventory. Reliable estimates of live-tree carbon stores and flux on timberlands outside of national forest could be calculated from periodic inventory data collected in the 1980s and 1990s; however, estimation of circa 1990 flux on national forests and forests other than timberland was problematic owing to a combination of changing inventory protocols and definitions and the lack of remeasurement data on those land categories. We estimate annual carbon flux on the 7.97 million acres of timberlands outside of national forests (which account for 24 percent of California's forest area and 28 percent of its live tree aboveground biomass) at 2.9 terragrams per year.

Carbon storage and flux estimates for the two national forests in Alaska are provided using inventory data from permanent plots established in 1995–2003 and remeasured in 2004–2010. Estimates of change are reported separately for growth, sapling recruitment, harvest, mortality, snag recruitment, salvage, snag falldown, and decay. Although overall aboveground carbon mass in live trees did not change in the Tongass National Forest, the Chugach National Forest showed a 4.5 percent increase. For the Tongass National Forest, results differed substantially for managed and unmanaged forest: managed lands had higher per-acre rates of sequestration through growth and recruitment, and carbon stores per acre that were higher for decomposing downed wood, and lower for live trees and snags. The species composition of carbon stores is changing on managed lands, with a carbon mass loss for yellow-cedar but increases for red alder and Sitka spruce. On unmanaged lands, the Chugach National forest had carbon mass increases in Sitka spruce and white spruce, and the Tongass National Forest had increases in western redcedar and red alder.

We study the time-averaged flux in a model of particles that randomly hop on a finite directed graph. In the limit as the number of particles and the time window go to infinity but the graph remains finite, the large-deviation rate functional of the average flux is given by a variational formulation involving paths of the density and flux. We give sufficient conditions under which the large deviations of a given time averaged flux is determined by paths that are constant in time. We then consider a class of models on a discrete ring for which it is possible to show that a better strategy is obtained producing a time-dependent path. This phenomenon, called a dynamical phase transition, is known to occur for some particle systems in the hydrodynamic scaling limit, which is thus extended to the setting of a finite graph.

The methods described herein enable the evaluation of compounds on subjects to assess their therapeutic efficacy or toxic effects. The target of analysis is the underlying biochemical process or processes (i.e., metabolic process) thought to be involved in disease pathogenesis. Molecular flux rates within the one or more biochemical processes serve as biomarkers and are quantitated and compared with the molecular flux rates (i.e., biomarker) from control subjects (i.e., subjects not exposed to the compounds). Any change in the biomarker in the subject relative to the biomarker in the control subject provides information to evaluate therapeutic efficacy of an administered drug or a toxic effect and to develop the compound further if desired. In one aspect of the invention, stable isotope-labeled substrate molecules are administered to a subject and the label is incorporated into targeted molecules in a manner that reveals molecular flux rates through metabolic pathways of interest.

Methods are provided herein for: calculating cell growth rates in various environments and genetic backgrounds; calculating the order of substrate utilization from a defined growth medium; calculating metabolic flux reorganization in various environments and at various growth rates; and calculating the maximum metabolic rate and optimal metabolite concentrations and enzyme activities by applying a computational optimization method to a kinetic model of a metabolic pathway. The optimization methods use intracellular molecular crowding parameters and/or well as kinetic rates to assist in modeling metabolic activity.

Embodiments provide a magnetic flux conversion device (MFCD) that may produce a regulated output signal with a target value (e.g., target voltage and/or target current) from a source signal on a power line. The MFCD may include a secondary stage configured to be inductively coupled with the power line. The source signal may cause a secondary electrical signal to flow in the secondary stage. A regulator module may be coupled to the secondary stage and configured to produce the output signal with the target value across output nodes by sensing the output signal and shunting the secondary stage if a value of the output signal is above the target value.

A method for measuring the neutron flux in the core of a nuclear reactor, the method including several steps recurrently performed at instants separated by a period, the method comprising at each given instant the following steps: acquiring a total signal by a cobalt neutron detector placed inside the core of the reactor; assessing a calibration factor representative of the delayed component of the total signal due to the presence of cobalt 60 in the neutron detector; assessing a corrected signal representative of the neutron flux at the detector from the total signal and from the calibration factor; assessing a slope representative of the time-dependent change of the calibration factor between the preceding instant and the given instant; the calibration factor at the given instant being assessed as a function of the calibration factor assessed at the preceding instant, of the slope, and of the time period separating the given instant from the preceding instant.

Disclosed herein are a pre-fill system to improve brake feel and a method of increasing an initial flux using the same which may reduce an invalid travel distance of a brake pedal. The pre-fill system includes a first housing connected with the master cylinder and provided with a first bore having a smaller-diameter portion and a larger-diameter portion, a second housing provided with a stepped second bore communicating with the first bore to be coupled with the first housing to define a hydraulic pressure chamber, a piston arranged in the first bore and provided with an oil channel unit, and a valve assembly adapted to open and close the oil channel unit of the piston according to the hydraulic brake pressure, wherein the piston applies pressure to the hydraulic pressure chamber through the piston to supply hydraulic pressure to the disc brake.

A method of monitoring a well for unwanted formation fluid influx is disclosed. Measurements of well outflow are acquired during a period in which drilling operations are performed for the well. Occurrences of stagnant flow events during the period are determined. An outflow signature is generated from the well outflow measurements for each stagnant flow event. The outflow signatures are displayed sequentially in time of occurrence. Each outflow signature is analyzed for an anomaly.

A pump for pumping molten metal and delivering flux includes a refractory base that can be submerged in molten metal including an impeller chamber, an inlet and an outlet. A refractory shaft sleeve has upper and lower end portions and is fastened to the base at the lower end portion. A motor is disposed near the upper end portion of the shaft sleeve. A refractory shaft extends in the shaft sleeve and is connected to the motor near the upper end portion of the shaft sleeve. A refractory impeller is connected to the shaft and is rotatable in the impeller chamber. A flux feeding device feeds flux into the shaft sleeve. Also featured is a method for delivering flux in the shaft sleeve of the pump and a method for cleaning flux accretions in the shaft sleeve.

Various processes and apparatus are discussed for an ultra-high heat flux chemical reactor. A thermal receiver and the reactor tubes are aligned to 1) absorb and re-emit radiant energy, 2) highly reflect radiant energy, and 3) any combination of these, to maintain an operational temperature of the enclosed ultra-high heat flux chemical reactor. Particles of biomass are gasified in the presence of a steam carrier gas and methane in a simultaneous steam reformation and steam biomass gasification reaction to produce reaction products that include hydrogen and carbon monoxide gas using the ultra-high heat flux thermal energy radiated from the inner wall and then into the multiple reactor tubes. The multiple reactor tubes and cavity walls of the receiver transfer energy primarily by radiation absorption and re-radiation, rather than by convection or conduction, to the reactants in the chemical reaction to drive the endothermic chemical reaction flowing in the reactor tubes.

A probabilistic digitizer for extracting information from a Josephson comparator is disclosed. The digitizer uses statistical methods to aggregate over a set of comparator readouts, effectively increasing the sensitivity of the comparator even when an input signal falls within the comparator's gray zone. Among other uses, such a digitizer may be used to discriminate between states of a qubit.

A system of systems to monitor data for carbon flux, for example, at scales capable of managing regional net carbon flux and pricing carbon financial instruments is disclosed. The system of systems can monitor carbon flux in forests, soils, agricultural areas, body of waters, flue gases, and the like. The system includes a means to identify and quantify sources of carbon based on simultaneous measurement of isotopologues of carbon dioxide, for example, industrial, agricultural or natural sources, offering integration of same in time and space. Carbon standards are employed at multiple scales to ensure harmonization of data and carbon financial instruments.

Provided herein is a resin fluxed solder paste that exhibits a desirable solder bump reinforcement effect without requiring an underfill process. The disclosure also provides a mount structure. The resin fluxed solder paste includes a non-resinic powder containing a solder powder and an inorganic powder; and a flux containing a first epoxy resin, a curing agent, and an organic acid. The non-resinic powder accounts for 30 to 90 wt % of the total, and the surface of the inorganic powder is covered with an organic resin.
