Crystal structures of the Bacillus subtilis prophage lytic cassette proteins XepA and YomS

As part of the Virus-X Consortium that aims to identify and characterize novel proteins and enzymes from bacteriophages and archaeal viruses, the genes of the putative lytic proteins XepA from Bacillus subtilis prophage PBSX and YomS from prophage SPβ were cloned and the proteins were subsequently produced and functionally characterized. In order to elucidate the role and the molecular mechanism of XepA and YomS, the crystal structures of these proteins were solved at resolutions of 1.9 and 1.3 Å, respectively. XepA consists of two antiparallel β-sandwich domains connected by a 30-amino-acid linker region. A pentamer of this protein adopts a unique dumbbell-shaped architecture consisting of two discs and a central tunnel. YomS (12.9 kDa per monomer), which is less than half the size of XepA (30.3 kDa), shows homology to the C-terminal part of XepA and exhibits a similar pentameric disc arrangement. Each β-sandwich entity resembles the fold of typical cytoplasmic membrane-binding C2 domains. Only XepA exhibits distinct cytotoxic activity in vivo, suggesting that the N-terminal pentameric domain is essential for this biological activity. The biological and structural data presented here suggest that XepA disrupts the proton motive force of the cytoplasmatic membrane, thus supporting cell lysis.

A charger includes a loading chamber into which a battery pack is insertably/removably loaded. An insertion opening into which the battery pack is inserted is formed on an upper surface of the main body. The loading chamber is forwardly inclined to a front surface of the main body, and a bottom surface of the insertion opening is inclined with respect to a horizontal direction so that one end of the front surface side is located at a lower end and the other end of the back surface side is located at an upper end. A connector for supplying power is disposed at the upper end side of the bottom surface. Even when the dust, rubbish or fluid entered from the insertion opening drops to the bottom surface, it flows down to the lower end side, so that less dirt adheres to the connector.

A surgical cassette clamping system includes a mounting plate having a first side and a second side. A bracket system may be disposed adjacent the first side of the mounting plate. A clamp motor may be disposed adjacent the mounting plate and fixed relative to the mounting plate. The clamp motor may be operably connected to the bracket system to displace the bracket system relative to the mounting plate. The system may also include a plurality of pivot arms pivotably connected to the bracket system and extending adjacent the second side of the mounting plate. An engagement portion may be attached to each of the plurality of pivot arms that cooperatively engages the second side of the mounting plate, the engagement portion being operable to pivot the pivot arms when the bracket system moves relative to the mounting plate.

X-linked adrenoleukodystrophy (X-ALD) is an inherited disorder caused by deleterious mutations in the ABCD1 gene. The ABCD1 protein transports very long-chain FAs (VLCFAs) from the cytosol into the peroxisome where the VLCFAs are degraded through β-oxidation. ABCD1 dysfunction leads to VLCFA accumulation in individuals with X-ALD. FAs are activated by esterification to CoA before metabolic utilization. However, the intracellular pools and metabolic profiles of individual acyl-CoA esters have not been fully analyzed. In this study, we profiled the acyl-CoA species in fibroblasts from X-ALD patients and in ABCD1-deficient HeLa cells. We found that hexacosenoyl (26:1)-CoA, but not hexacosanoyl (26:0)-CoA, was the most abundantly concentrated among the VLCFA-CoA species in these cells. We also show that 26:1-CoA is mainly synthesized from oleoyl-CoA, and the metabolic turnover rate of 26:1-CoA was almost identical to that of oleoyl-CoA in both WT and ABCD1-deficient HeLa cells. The findings of our study provide precise quantitative and metabolic information of each acyl-CoA species in living cells. Our results suggest that VLCFA is endogenously synthesized as VLCFA-CoA through a FA elongation pathway and is then efficiently converted to other metabolites, such as phospholipids, in the absence of ABCD1.
