Lipid-rich myelin forms electrically insulating, axon-wrapping multilayers that are essential for neural function, and mature myelin is traditionally considered metabolically inert. Surprisingly, we discovered that mature myelin lipids undergo rapid turnover, and quaking (Qki) is a major regulator of myelin lipid homeostasis. Oligodendrocyte-specific Qki depletion, without affecting oligodendrocyte survival, resulted in rapid demyelination, within 1 week, and gradually neurological deficits in adult mice. Myelin lipids, especially the monounsaturated fatty acids and very-long-chain fatty acids, were dramatically reduced by Qki depletion, whereas the major myelin proteins remained intact, and the demyelinating phenotypes of Qki-depleted mice were alleviated by a high-fat diet. Mechanistically, Qki serves as a coactivator of the PPARβ-RXRα complex, which controls the transcription of lipid-metabolism genes, particularly those involved in fatty acid desaturation and elongation. Treatment of Qki-depleted mice with PPARβ/RXR agonists significantly alleviated neurological disability and extended survival durations. Furthermore, a subset of lesions from patients with primary progressive multiple sclerosis were characterized by preferential reductions in myelin lipid contents, activities of various lipid metabolism pathways, and expression level of QKI-5 in human oligodendrocytes. Together, our results demonstrate that continuous lipid synthesis is indispensable for mature myelin maintenance and highlight an underappreciated role of lipid metabolism in demyelinating diseases.

IBM announced today a new experimental laboratory for nanotechnology research in Brazil. The NanoLab is part of a $4M investment within the recently upgraded IBM Research-Brazil lab in Rio de Janeiro and will focus on projects related to research in Oil & Gas, Agriculture and Health across Latin America.

Part of how I got out of debt last time was to be frank with myself and others about my finances. For the first 10 years we were together, James and I had zero debt, and it took some pushing past shame to admit we'd accumulated $10K+ in consumer debt, but admitting it was the first step to eliminate it. This time, the debt is even higher (but it was important, she says with some shame, to fix our teeth and our car, and and and...) but it's not any different: the key to getting rid of it for me is facing it. Well, facing it and having a plan.

Facing it, having a plan, and an almost fanatical devotion to the Pope.

Another thing I have to face is that I can't afford to go part-time yet. Not until either James gets the job he applied for or I get more editing work. I'm a little sad about that, but it's okay -- I can do this for a year while we knock out debt, and then I can decide what to do after that happens. When we don't have debt, we have SO MUCH FREEDOM. I just need to keep that in mind.

In completely other news, I don't get a "Least Complicated" earworm often, in day-to-day life, but something about getting to the "Music" box on the DW posting page brings it to mind; it's my most frequent "Music" entry, and it seems to just be a conditioned response to seeing the input box. Weird.

Ještě minulý týden byly úřední hodiny kontaktních pracovišť Úřadu práce ČR velmi omezené. Pobočky sice v pondělí otevřely, ani dnes ale na úřad nemusíte osobně, většinu záležitostí vyřídíte online. Poradíme jak na to a shrneme, kdy a v jaké výši máte nárok na podporu v nezaměstnanosti.

Finanční znalosti Čechů se začínají zlepšovat. V indexu finanční gramotnosti, který každoročně měří Česká bankovní asociace, dosáhli Češi 61 procentních bodů ze 100. Oproti loňskému roku to je o čtyři body více, což je doposud nejvyšší hodnota za posledních několik let. Problém ale je, že pětina lidí přiznává, že má finanční polštář vytvořený jen na pouhý měsíc.

• 1 head romaine, chopped ($1.70)
• 1 orange, peeled and sectioned ($.50)
• 1 banana, sliced ($.30)
• 2 tablespoons sliced onion

• 2 oranges, peeled and sectioned ($1.00)
• 2 tablespoons balsamic vinegar ($.20)
• 2 tablespoons maple syrup (or two droppers stevia) ($.40)
• 3 tablespoons tahini ($.40)
• 1 teaspoon onion powder
• 1 teaspoon garlic powder
• 1 teaspoon salt
• 1/2 teaspoon pepper
• crushed red pepper flakes for the top

directions

1. Prepare the salad ingredients.
2. Place all the dressing ingredients except the crushed red pepper flakes into a blender and puree until very smooth. 
3. Assemble the salad ingredients on plates or in bowls. Add the orange sections, banana slices, and onion to the top.
4. Pour salad dressing over and add a sprinkle of black pepper or crushed red pepper flakes. I added both.

Either way this one is really aimed at trolling computer scientists, and so may be impenetrable if you don't have the background; sorry about that!

Allergies and various things have got me down recently but it's also getting nice out, which should provide a burst of energy!! This weekend is the Marathon in Pittsburgh, which I intend to run. No costume plans but sometimes I get last minute inspiration / compulsion. Feel free to taunt me with your ideas.
