The crystal and solution SAXS structures of a fragment of human leucocyte common antigen-related protein show that it is less flexible than the homologous proteins tyrosine phosphatase receptors δ and σ.

The title compound, C12H21NO7, (I), is conformationally unstable; the predominant form present in its solution is the β-pyran­ose form (74.3%), followed by the β- and α-furan­oses (12.1 and 10.2%, respectively), α-pyran­ose (3.4%), and traces of the acyclic carbohydrate tautomer. In the crystalline state, the carbohydrate part of (I) adopts the 2C5 β-pyran­ose conformation, and the amino acid portion exists as a zwitterion, with the side chain cyclo­pentane ring assuming the E9 envelope conformation. All heteroatoms are involved in hydrogen bonding that forms a system of anti­parallel infinite chains of fused R33(6) and R33(8) rings. The mol­ecule features extensive intra­molecular hydrogen bonding, which is uniquely multicentered and involves the carboxyl­ate, ammonium and carbohydrate hy­droxy groups. In contrast, the contribution of inter­molecular O⋯H/H⋯O contacts to the Hirshfeld surface is relatively low (38.4%), as compared to structures of other d-fructose-amino acids. The 1H NMR data suggest a slow rotation around the C1—C2 bond in (I), indicating that the intra­molecular heteroatom contacts survive in aqueous solution of the mol­ecule as well.

Leucocyte common antigen-related protein (LAR) is a post-synaptic type I transmembrane receptor protein that is important for neuronal functionality and is genetically coupled to neuronal disorders such as attention deficit hyperactivity disorder (ADHD). To understand the molecular function of LAR, structural and biochemical studies of protein fragments derived from the ectodomain of human LAR have been performed. The crystal structure of a fragment encompassing the first four FNIII domains (LARFN1–4) showed a characteristic L shape. SAXS data suggested limited flexibility within LARFN1–4, while rigid-body refinement of the SAXS data using the X-ray-derived atomic model showed a smaller angle between the domains defining the L shape compared with the crystal structure. The capabilities of the individual LAR fragments to interact with heparin was examined using microscale thermophoresis and heparin-affinity chromatography. The results showed that the three N-terminal immunoglobulin domains (LARIg1–3) and the four C-terminal FNIII domains (LARFN5–8) both bound heparin, while LARFN1–4 did not. The low-molecular-weight heparin drug Innohep induced a shift in hydrodynamic volume as assessed by size-exclusion chromatography of LARIg1–3 and LARFN5–8, while the chemically defined pentameric heparin drug Arixtra did not. Together, the presented results suggest the presence of an additional heparin-binding site in human LAR.

The impact that national energy sectors have on international freshwater resources has been demonstrated in the first global study of its kind. The analysis of 129 countries showed differences between countries and sectors in their reliance on international freshwater resources. For example, although the petroleum industries of North America and China are similar in magnitude, the North American industry consumes three times as much international freshwater. Demands from economically developed countries on less economically developed countries, which may have pre-existing water-scarcity issues, compound these problems and complicate the creation of policies that ensure both water and energy security.

In a new study, scientists have analysed the outcome of millions of barrels of oil and gas discharged into the Gulf of Mexico during the Deepwater Horizon disaster. By measuring changes in the composition of the fluid, scientists now have a better understanding of how pollution behaves in the deep ocean, to improve estimates of environmental impact.

A study of the pollution caused by ship scrapping in Alang, India, shows significantly higher levels of heavy metal and petroleum hydrocarbons in sediment and seawater, compared to a control site. The researchers also found reduced populations of zooplankton — a critical food source for marine biota — and increased numbers of pathogenic bacteria.

Living near busy roads is associated with an increased risk of childhood leukaemia, according to French researchers. The study found that children living within 500 metres of major roads were more likely to develop leukaemia than those who lived far from such roads. Bacteria taken from underground petroleum reserves could be used to effectively break down crude oil from spills at sea, new research has found. The study measured the breakdown of crude oil components in simulated seawater by four bacterial strains that had been isolated from petroleum reservoirs, as well as by four genetically modified stains. The findings raise the possibility of tailor-making organisms to clean up specific types of contamination.
