Isolation and Characterization of the Novel Phage JD032 and Global Transcriptomic Response during JD032 Infection of Clostridioides difficile Ribotype 078

ABSTRACT

Insights into the interaction between phages and their bacterial hosts are crucial for the development of phage therapy. However, only one study has investigated global gene expression of Clostridioides (formerly Clostridium) difficile carrying prophage, and transcriptional reprogramming during lytic infection has not been studied. Here, we presented the isolation, propagation, and characterization of a newly discovered 35,109-bp phage, JD032, and investigated the global transcriptomes of both JD032 and C. difficile ribotype 078 (RT078) strain TW11 during JD032 infection. Transcriptome sequencing (RNA-seq) revealed the progressive replacement of bacterial host mRNA with phage transcripts. The expressed genes of JD032 were clustered into early, middle, and late temporal categories that were functionally similar. Specifically, a gene (JD032_orf016) involved in the lysis-lysogeny decision was identified as an early expression gene. Only 17.7% (668/3,781) of the host genes were differentially expressed, and more genes were downregulated than upregulated. The expression of genes involved in host macromolecular synthesis (DNA/RNA/proteins) was altered by JD032 at the level of transcription. In particular, the expression of the ropA operon was downregulated. Most noteworthy is that the gene expression of some antiphage systems, including CRISPR-Cas, restriction-modification, and toxin-antitoxin systems, was suppressed by JD032 during infection. In addition, bacterial sporulation, adhesion, and virulence factor genes were significantly downregulated. This study provides the first description of the interaction between anaerobic spore-forming bacteria and phages during lytic infection and highlights new aspects of C. difficile phage-host interactions.

IMPORTANCE C. difficile is one of the most clinically significant intestinal pathogens. Although phages have been shown to effectively control C. difficile infection, the host responses to phage predation have not been fully studied. In this study, we reported the isolation and characterization of a new phage, JD032, and analyzed the global transcriptomic changes in the hypervirulent RT078 C. difficile strain, TW11, during phage JD032 infection. We found that bacterial host mRNA was progressively replaced with phage transcripts, three temporal categories of JD032 gene expression, the extensive interplay between phage-bacterium, antiphage-like responses of the host and phage evasion, and decreased expression of sporulation- and virulence-related genes of the host after phage infection. These findings confirmed the complexity of interactions between C. difficile and phages and suggest that phages undergoing a lytic cycle may also cause different phenotypes in hosts, similar to prophages, which may inspire phage therapy for the control of C. difficile.

Histone Demethylase JMJD1A Promotes Tumor Progression via Activating Snail in Prostate Cancer

The histone demethylase JMJD1A plays a key functional role in spermatogenesis, sex determination, stem cell renewal, and cancer via removing mono- and di-methyl groups from H3K9 to epigenetically control gene expression. However, its role in prostate cancer progression remains unclear. Here, we found JMJD1A was significantly elevated in prostate cancer tissue compared with matched normal tissue. Ectopic JMJD1A expression in prostate cancer cells promoted proliferation, migration, and invasion in vitro, and tumorigenesis in vivo; JMJD1A knockdown exhibited the opposite effects. Mechanically, we revealed that JMJD1A directly interacted with the Snail gene promoter and regulated its transcriptional activity, promoting prostate cancer progression both in vitro and in vivo. Furthermore, we found that JMJD1A transcriptionally activated Snail expression via H3K9me1 and H3K9me2 demethylation at its special promoter region. In summary, our studies reveal JMJD1A plays an important role in regulating proliferation and progression of prostate cancer cells though Snail, and thus highlight JMJD1A as potential therapeutic target for advanced prostate cancer.

Implications:

Our studies identify that JMJD1A promotes the proliferation and progression of prostate cancer cells through enabling Snail transcriptional activation, and thus highlight JMJD1A as potential therapeutic target for advanced prostate cancer.

The Prognosis of Patients With Type 2 Diabetes and Nonalbuminuric Diabetic Kidney Disease Is Not Always Poor: Implication of the Effects of Coexisting Macrovascular Complications (JDDM 54)

OBJECTIVE

Nonalbuminuric diabetic kidney disease (DKD) has become the prevailing phenotype in patients with type 2 diabetes. However, it remains unclear whether its prognosis is poorer than that of other DKD phenotypes.

RESEARCH DESIGN AND METHODS

A total of 2,953 Japanese patients with type 2 diabetes and estimated glomerular filtration rate (eGFR) ≥30 mL/min/1.73 m2, enrolled in an observational cohort study in 2004, were followed until 2015. On the basis of albuminuria (>30 mg/g creatinine) and reduced eGFR (<60 mL/min/1.73 m2) at baseline, participants were classified into the four DKD phenotypes—no-DKD, albuminuric DKD without reduced eGFR, nonalbuminuric DKD with reduced eGFR, and albuminuric DKD with reduced eGFR—to assess the risks of mortality, cardiovascular disease (CVD), and renal function decline.

RESULTS

During the mean follow-up of 9.7 years, 113 patients died and 263 developed CVD. In nonalbuminuric DKD, the risks of death or CVD were not higher than those in no-DKD (adjusted hazard ratio 1.02 [95% CI 0.66, 1.60]) and the annual decline in eGFR was slower than in other DKD phenotypes. The risks of death or CVD in nonalbuminuric DKD without prior CVD were similar to those in no-DKD without prior CVD, whereas the risks in nonalbuminuric DKD with prior CVD as well as other DKD phenotypes were higher.

CONCLUSIONS

Nonalbuminuric DKD did not have a higher risk of mortality, CVD events, or renal function decline than the other DKD phenotypes. In nonalbuminuric DKD, the presence of macrovascular complications may be a main determinant of prognosis rather than the renal phenotype.

FWICE's president BN Tiwari: JD Majethia has sent a wrong message to the daily wagers

On Monday, JD Majethia, chairman of the TV wing of the Indian Film and TV Producers' Council (IFTPC), stated that he intends to approach the government to seek permission to resume shoots of television shows with small crews from May 4. The move has sent the daily-wage workers of the television industry into a tizzy as they have been relentlessly calling the Federation of Western India Cine Employees (FWICE) to enquire whether they can report to work post-May 3.

BN Tiwari, president, FWICE, says, "[With his statement], JD Majethia has sent a wrong message to the daily wagers who now feel that producers are willing to work, but the FWICE is imposing unnecessary restrictions. That is not the case. Maharashtra is at high risk, and we are only concerned about our workers' safety. We have issued a letter to Majethia, asking him not to make such statements."

On March 15, various cine bodies had unanimously decided to suspend all shoots in the wake of the pandemic. "Similarly, the decision to resume work will have to be collectively taken by IFTDA, FWICE, Producers' Guild, and other associations," insists Tiwari.

Majethia was keen to propose the resumption of shoots as it would bring relief to the daily wagers. However, Tiwari points out that the associations are standing by those in need. "The industry — including Salman Khan, Ajay Devgn, Rohit Shetty and Yash Raj Films — has come together to help the workers. Even if the lockdown is lifted on May 3, we may not begin work until May 25. In case the stay-at-home orders are extended to June, we will continue to take care of our workers."

When mid-day reached out to Majethia, he agreed that the recommencement of shoots will be "a collective decision of the industry" with the approval of the government. "It is not as if we have already taken a call. We will start shooting only when it is safe for everyone, but we must start planning for it in advance."

Movie Review: Ujda Chaman

Over the years, there have been several films that have tackled various conditions that lower a person’s self-esteem like skin colour, obesity, stunted growth etc. Premature balding, however, has rarely been the central plot point of a film. Two very lesser known films like I M 24 [2012], HAIR IS FALLING [2011] etc have dealt with the issue but they sank without a trace. Producer Abhishek Pathak for his directorial debut, UJDA CHAMAN, tackles this idea in the romcom space. So does UJDA CHAMAN manage to entertain and move audiences? Or does it fail to entertain? Chaman Kohli (Sunny Singh) is a 30-year-old lecturer in Delhi’s Hansraj College. He has faced acute hair loss in the last five years, affecting his self-esteem and also his marriage prospects. His parents, Shashi Kohli (Atul Kumar) and Sushma Kohli (Grusha Kapoor) put his five-year-old picture in his marriage bio-data and when he goes to see a prospective bride, he wears a cap. Sadly, the cap comes off and the prospective bride and her family kick the Kohli’s from their house. Their family astrologer (Saurabh Shukla) adds to their tension as he claims that if Chaman doesn’t get married before his 31<sup>st</sup> birthday, he’ll remain celibate for life. Chaman tries to find love, first at the marriage ceremony of a friend but it proves futile. Later, he tries to woo a fellow lecturer, Ekta (Aishwarya Sakhuja) but she rejects him as she is already dating someone. All of a sudden, a first-year student, Aaina Ali Khan (Karishma Sharma) shows interest in Chaman. Chaman falls for her but in no time, he realizes that she hung out with him for vested interests. With no other option, Chaman accepts his father’s suggestion and downloads Tinder. He uploads his picture, but by cropping his bald patch. In no time, he matches with Apsara (Maanvi Gagroo), who is overweight and just like Chaman, she too puts a close up picture to hide her so-called imperfection. Both decide to meet and obviously get shocked looking at each other. What happens next forms the rest of the film. UJDA CHAMAN is an official remake of the 2017 Kannada film, ONDU MOTTEYA KATHE (Writer: Raj B Shetty). Danish J Singh's adapted story has immense potential as a lot of people can relate to the goings on shown in the film. But Danish J Singh's screenplay plays spoilsport. The narrative is engaging but only in parts. Danish J Singh's dialogues are funny and matter-of-fact. Abhishek Pathak's direction is not worthy enough. The execution seems a bit amateurish and over-the-top. The manner in which Chaman is ridiculed almost on a daily basis by the students because of his receding hairline is too much. Moreover, the direction style is not consistent. The second half looks very different from the first half. Also, with nothing unique happening in the film, the narrative becomes extremely predictable. On the positive side, a few scenes work like the track of Aaina and the hospital scene. UJDA CHAMAN begins on a fun but very loud note with the initial portions being mildly engaging. The humour looks forced in the scene where Chaman attempts to assault his brother Goldy (Gagan Arora). The track of Aaina is thankfully better but still, the film doesn’t reach a high. The intermission point is interesting and the film seems promising from here on. And the initial portions of the second half are very entertaining as Chaman and Apsara get close. But after a point, nothing novel or interesting happens. The film gets dragging and predictable. The final scene is cute but comes too late in the film. <span style="text-decoration: underline;"><strong>Ujda Chaman: Public Review | First Day First Show | Sunny Singh Nijjar | Maanvi Gagroo</strong></span> <iframe id="jwiframe" class="playerFrame" src="https://www.bollywoodhungama.com/videos/first-day-first-show/ujda-chaman-public-review-first-day-first-show-sunny-singh-nijjar-maanvi-gagroo/?jwembed=1" width="800" height="340" frameborder="0" allowfullscreen="allowfullscreen"></iframe> Sunny Singh gives his best but the desired impact is not made. The manner in which he seems so serious and lifeless throughout the film is difficult to digest. Even his narration in the beginning is needlessly done in a sombre tone. Maanvi Gagroo does quite better and is a natural. Her issues and predicaments seem very relatable and she plays the part nicely. Atul Kumar also does a genuinely good job. Grusha Kapoor is very loud. Gagan Arora is passable. Karishma Sharma plays her part as per the requirement. Aishwarya Sakhuja has a great screen presence. Sharib Hashmi (Raj) is good but sadly his track is predictable. Saurabh Shukla is strictly okay. Music is disappointing. Only <em>'Chand Nikla'</em> gets registered. The rest of the songs like <em>'Twinkle Twinkle', 'Outfit'</em> and <em>'Oh Bandeya'</em> are forgettable. Hitesh Sonik's background score is too loud and dramatic. Sudhir K Chaudhary's cinematography is neat. Tarpan Srivastav's production design is in sync with the film’s setting. Preetisheel Singh's make up and prosthetics are flawless. Rahil Raja and Himanshi Nijhawan's costumes are realistic. Mitesh Soni's editing is sans complaint and the film’s length is appropriate at 120 minutes. On the whole, UJDA CHAMAN has a promising and relatable story but the inconsistent execution and predictable narrative spoils the show. At the box office, thanks to limited buzz and lack of popular names, the film will have a difficult time.
