Differentiating through Log-Log Convex Programs. (arXiv:2004.12553v2 [math.OC] UPDATED)

We show how to efficiently compute the derivative (when it exists) of the solution map of log-log convex programs (LLCPs). These are nonconvex, nonsmooth optimization problems with positive variables that become convex when the variables, objective functions, and constraint functions are replaced with their logs. We focus specifically on LLCPs generated by disciplined geometric programming, a grammar consisting of a set of atomic functions with known log-log curvature and a composition rule for combining them. We represent a parametrized LLCP as the composition of a smooth transformation of parameters, a convex optimization problem, and an exponential transformation of the convex optimization problem's solution. The derivative of this composition can be computed efficiently, using recently developed methods for differentiating through convex optimization problems. We implement our method in CVXPY, a Python-embedded modeling language and rewriting system for convex optimization. In just a few lines of code, a user can specify a parametrized LLCP, solve it, and evaluate the derivative or its adjoint at a vector. This makes it possible to conduct sensitivity analyses of solutions, given perturbations to the parameters, and to compute the gradient of a function of the solution with respect to the parameters. We use the adjoint of the derivative to implement differentiable log-log convex optimization layers in PyTorch and TensorFlow. Finally, we present applications to designing queuing systems and fitting structured prediction models.

Methods and systems to identify and reproduce concurrency bugs in multi-threaded programs are disclosed. An example method disclosed herein includes defining a data type. The data type includes a first predicate associated with a first thread of a multi-threaded program that is associated with a first condition, a second predicate that is associated with a second thread of the multi-threaded program, the second predicate being associated with a second condition, and an expression that defines a relationship between the first predicate and the second predicate. The relationship, when satisfied, causes the concurrency bug to be detected. A concurrency bug detector conforming to the data type is used to detect the concurrency bug in the multi-threaded program.

Thumbnail images representative of recorded TV programs are generated and presented to aid a user in browsing the recorded TV programs. In one implementation, a temporary thumbnail image is generated when a TV program first starts recording. The temporary thumbnail is used to populate any user interface (UI) screens that reference the recoded TV program. Once the TV program has reached a threshold amount of recording (e.g., a prescribed duration of recording, or completion of the recording), a permanent thumbnail image is generated and associated with the TV program. The permanent thumbnail is then presented in any subsequent UI screens, replacing the temporary thumbnail. In another implementation, display of the thumbnail images in the UI screens may be further controlled by setting preferences, such as parental controls.

The neonatal microenvironment programs innate γδ T cells through the transcription factor STAT5

IL-17–producing RORγt+ γδ T cells (γδT17 cells) are innate lymphocytes that participate in type 3 immune responses during infection and inflammation. Herein, we show that γδT17 cells rapidly proliferate within neonatal lymph nodes and gut, where, upon entry, they upregulate T-bet and coexpress IL-17, IL-22, and IFN-γ in a STAT3- and retinoic acid–dependent manner. Neonatal expansion was halted in mice conditionally deficient in STAT5, and its loss resulted in γδT17 cell depletion from all adult organs. Hyperactive STAT5 mutant mice showed that the STAT5A homolog had a dominant role over STAT5B in promoting γδT17 cell expansion and downregulating gut-associated T-bet. In contrast, STAT5B preferentially expanded IFN-γ–producing γδ populations, implying a previously unknown differential role of STAT5 gene products in lymphocyte lineage regulation. Importantly, mice lacking γδT17 cells as a result of STAT5 deficiency displayed a profound resistance to experimental autoimmune encephalomyelitis. Our data identify that the neonatal microenvironment in combination with STAT5 is critical for post-thymic γδT17 development and tissue-specific imprinting, which is essential for infection and autoimmunity.

Parental metabolic syndrome epigenetically reprograms offspring hepatic lipid metabolism in mice

The prevalence of nonalcoholic fatty liver disease (NAFLD) is increasing worldwide. Although gene-environment interactions have been implicated in the etiology of several disorders, the impact of paternal and/or maternal metabolic syndrome on the clinical phenotypes of offspring and the underlying genetic and epigenetic contributors of NAFLD have not been fully explored. To this end, we used the liver-specific insulin receptor knockout (LIRKO) mouse, a unique nondietary model manifesting 3 hallmarks that confer high risk for the development of NAFLD: hyperglycemia, insulin resistance, and dyslipidemia. We report that parental metabolic syndrome epigenetically reprograms members of the TGF-β family, including neuronal regeneration–related protein (NREP) and growth differentiation factor 15 (GDF15). NREP and GDF15 modulate the expression of several genes involved in the regulation of hepatic lipid metabolism. In particular, NREP downregulation increases the protein abundance of 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR) and ATP-citrate lyase (ACLY) in a TGF-β receptor/PI3K/protein kinase B–dependent manner, to regulate hepatic acetyl-CoA and cholesterol synthesis. Reduced hepatic expression of NREP in patients with NAFLD and substantial correlations between low serum NREP levels and the presence of steatosis and nonalcoholic steatohepatitis highlight the clinical translational relevance of our findings in the context of recent preclinical trials implicating ACLY in NAFLD progression.
