ehr Ehrlichia chaffeensis Uses an Invasin To Suppress Reactive Oxygen Species Generation by Macrophages via CD147-Dependent Inhibition of Vav1 To Block Rac1 Activation By mbio.asm.org Published On :: 2020-04-21T01:31:26-07:00 ABSTRACT The obligatory intracellular pathogen Ehrlichia chaffeensis lacks most factors that could respond to oxidative stress (a host cell defense mechanism). We previously found that the C terminus of Ehrlichia surface invasin, entry-triggering protein of Ehrlichia (EtpE; EtpE-C) directly binds mammalian DNase X, a glycosylphosphatidylinositol-anchored cell surface receptor and that binding is required to induce bacterial entry and simultaneously to block the generation of reactive oxygen species (ROS) by host monocytes and macrophages. However, how the EtpE-C–DNase X complex mediates the ROS blockade was unknown. A mammalian transmembrane glycoprotein CD147 (basigin) binds to the EtpE-DNase X complex and is required for Ehrlichia entry and infection of host cells. Here, we found that bone marrow-derived macrophages (BMDM) from myeloid cell lineage-selective CD147-null mice had significantly reduced Ehrlichia-induced or EtpE-C-induced blockade of ROS generation in response to phorbol myristate acetate. In BMDM from CD147-null mice, nucleofection with CD147 partially restored the Ehrlichia-mediated inhibition of ROS generation. Indeed, CD147-null mice as well as their BMDM were resistant to Ehrlichia infection. Moreover, in human monocytes, anti-CD147 partially abrogated EtpE-C-induced blockade of ROS generation. Both Ehrlichia and EtpE-C could block activation of the small GTPase Rac1 (which in turn activates phagocyte NADPH oxidase) and suppress activation of Vav1, a hematopoietic-specific Rho/Rac guanine nucleotide exchange factor by phorbol myristate acetate. Vav1 suppression by Ehrlichia was CD147 dependent. E. chaffeensis is the first example of pathogens that block Rac1 activation to colonize macrophages. Furthermore, Ehrlichia uses EtpE to hijack the unique host DNase X-CD147-Vav1 signaling to block Rac1 activation. IMPORTANCE Ehrlichia chaffeensis is an obligatory intracellular bacterium with the capability of causing an emerging infectious disease called human monocytic ehrlichiosis. E. chaffeensis preferentially infects monocytes and macrophages, professional phagocytes, equipped with an arsenal of antimicrobial mechanisms, including rapid reactive oxygen species (ROS) generation upon encountering bacteria. As Ehrlichia isolated from host cells are readily killed upon exposure to ROS, Ehrlichia must have evolved a unique mechanism to safely enter phagocytes. We discovered that binding of the Ehrlichia surface invasin to the host cell surface receptor not only triggers Ehrlichia entry but also blocks ROS generation by the host cells by mobilizing a novel intracellular signaling pathway. Knowledge of the mechanisms by which ROS production is inhibited may lead to the development of therapeutics for ehrlichiosis as well as other ROS-related pathologies. Full Article

ehr Coronavirus outbreak: BMC takes over Nehru Centre for COVID-19 fight By www.mid-day.com Published On :: 7 May 2020 02:00:35 GMT A slice of the verdant Nehru Science Centre in Worli has been taken over by the civic authorities for what Centre director Dr S Khened thinks must be for "an isolation centre, or for quarantine." Khened said, "I do not know exactly for which. We got official communication a couple of days earlier that a portion of the centre's space will be acquired. The front portion of the Centre, just as you enter the main gate, has been taken. We usually use that space to hold Science exhibitions. We have also held a vintage car exhibition two years ago in that venue, so it is a massive space." Centre officials said preparations are in full swing, with beds and mattresses and even mobile toilets being brought in. The Science Centre officials have tried to talk to those in charge at the site for details but without too much success. They reasoned, "it is evident that the officials are extremely stressed and we are in fact, prepared for the worst. Authorities are working extremely hard to get this ready," they stated. Khened said that the environs of the Centre are very green and will need to be maintained. "We need to have some skeletal staff inside to maintain the green patches, the trees, the many plants that we have. They need water and regular upkeep," he said adding that it was important that the Coronavirus facility is completely sealed off from the other parts of the sprawling complex. The Centre also has skeletal staff quarters on the premises where currently eight senior officers are living with their families, "so it is vital that those who are going to be shifted into the facilities for Coronavirus, be confined to that space. I am sure this has been thought out and will be done," said Khened. Physical spaceThe Science centre has moved its activities online, like most other institutions, with two virtual lectures scheduled on their YouTube channel on May 9 and May 11 respectively. The former will be a talk by Dr Vasant Shinde on Rakhigarhi Excavation and the latter is Dr Anil Kakodkar, on 'dealing with energy needs in the context of climate crisis'. Yet, said Dr Khened, "We will open again as a physical space when the government gives the green light. Children, the main visitors at the Centre, may have some apprehension about visiting since this will be used as a Coronavirus facility. There is a great deal of wariness as the Centre is just next to the Jijamata Nagar slum area, which is a COVID hotspot. "However, with all the trepidation, we too understand that this is for the greater good, and everybody has to do their part in this fight," the director said. Line upThe Worli area, with an unenviable record number of Coronavirus cases in Mumbai, is also being taken over as quarantine/isolation areas rapidly. Just around the corner from the Science Centre, the Nehru Centre, which is next to the Nehru Planetarium has also been acquired for Corona purposes. S P Verma, Jt Director of Nehru Centre, said, "The two exhibition halls on the ground floor have been acquired, not the Nehru Planetarium. I think beds and other amenities will be moved in shortly, I do not know when exactly. We had been told to hand over possession of the spaces and have done so," he finished. The Nehru Centre joins neighbour, NSCI club's The Dome and Mahalaxmi racecourse as an entire line of Haji Ali institutions, whose space has been acquired for Corona combat. Former India pacer Ashish Nehra and spinner Harbhajan Singh feel that saliva's use in shining the ball is a "must". While discussions are at nascent stage, questions are already being asked about what external substances could be used if ball tampering becomes legal? Is it going to be bottle cap in pocket to scuff up one side of the ball, vaseline to shine (made famous by English cricketer John Lever) or chain zipper? "Get one thing clear at the onset. The ball will not swing if you don't apply sweat or saliva on the ball. That's basic necessity of swing bowling. The moment ball gets scuffed up from one side, sweat and saliva must be applied on the other side," said Nehra, who completely shot down the idea of using external substances. "Now let's understand why do you need saliva? Sweat is heavier than saliva but both are heavy enough to make one side of the ball heavier for reverse swing. Vaseline comes into the picture only after sweat and saliva, not before that. "It is lighter and doesn't even ensure conventional swing. It can keep the shine, but doesn't make the ball heavy," he added. Harbhajan also agreed that saliva if one has already chewed mint, which has sugar in it, makes it heavier. But when it comes to using external substance, he wants to know what can be the options. "It's not that murray mint can be used without putting it in your mouth. The coat of sugar on the saliva makes it heavier after one side gets scuffed. A scuffed up ball is also good for spinners as it ensures a better grip than a shiny new ball. Dhoni was always open to a chat with his players after a match and his hotel room would always be open for them after the games. "People think that Dhoni doesn't talk much. That is not the case. His room would remain open in the night after matches. Anyone could walk in, order food and discuss cricket," Nehra said while speaking to former India opener Aakash Chopra on his show 'Aakash Vani'. "Be it in CSK or in the Indian team, Dhoni would pass on the message on what he thinks a player needs to do during a conversation and that would be enough for the players to get the drift," added Nehra who played a lot of cricket under Dhoni's captaincy both for India and at Chennai Super Kings in the Indian Premier League (IPL). Nehra also spoke on former India captain and current Board of Control for Cricket in India (BCCI) President Sourav Ganguly, saying he was a leader who used to back his players to the hilt. "Dhoni had senior players around him when he took over and the challenge was how to manage them. Dada had a team of juniors and he backed them to the hilt.
