Branching random walks with uncountably many extinction probability vectors

Abstract:
Given a branching random walk on a set $X$, we study its extinction probability vectors $mathbf{q}(cdot,A)$. Their components are the probability that the process goes extinct in a fixed $Asubseteq X$, when starting from a vertex $xin X$. The set of extinction probability vectors (obtained letting $A$ vary among all subsets of $X$) is a subset of the set of the fixed points of the generating function of the branching random walk. In particular here we are interested in the cardinality of the set of extinction probability vectors. We prove results which allow to understand whether the probability of extinction in a set $A$ is different from the one of extinction in another set $B$. In many cases there are only two possible extinction probability vectors and so far, in more complicated examples, only a finite number of distinct extinction probability vectors had been explicitly found. Whether a branching random walk could have an infinite number of distinct extinction probability vectors was not known. We apply our results to construct examples of branching random walks with uncountably many distinct extinction probability vectors.

Abstract:
Consider a supercritical branching random walk on the real line. The consistent maximal displacement is the smallest of the distances between the trajectories followed by individuals at the $n$th generation and the boundary of the process. Fang and Zeitouni, and Faraud, Hu and Shi proved that under some integrability conditions, the consistent maximal displacement grows almost surely at rate $lambda^{*}n^{1/3}$ for some explicit constant $lambda^{*}$. We obtain here a necessary and sufficient condition for this asymptotic behaviour to hold.

Statistical inference for partially observed branching processes with application to cell lineage tracking of in vivo hematopoiesis

Abstract:
Single-cell lineage tracking strategies enabled by recent experimental technologies have produced significant insights into cell fate decisions, but lack the quantitative framework necessary for rigorous statistical analysis of mechanistic models describing cell division and differentiation. In this paper, we develop such a framework with corresponding moment-based parameter estimation techniques for continuous-time, multi-type branching processes. Such processes provide a probabilistic model of how cells divide and differentiate, and we apply our method to study hematopoiesis , the mechanism of blood cell production. We derive closed-form expressions for higher moments in a general class of such models. These analytical results allow us to efficiently estimate parameters of much richer statistical models of hematopoiesis than those used in previous statistical studies. To our knowledge, the method provides the first rate inference procedure for fitting such models to time series data generated from cellular barcoding experiments. After validating the methodology in simulation studies, we apply our estimator to hematopoietic lineage tracking data from rhesus macaques. Our analysis provides a more complete understanding of cell fate decisions during hematopoiesis in nonhuman primates, which may be more relevant to human biology and clinical strategies than previous findings from murine studies. For example, in addition to previously estimated hematopoietic stem cell self-renewal rate, we are able to estimate fate decision probabilities and to compare structurally distinct models of hematopoiesis using cross validation. These estimates of fate decision probabilities and our model selection results should help biologists compare competing hypotheses about how progenitor cells differentiate. The methodology is transferrable to a large class of stochastic compartmental and multi-type branching models, commonly used in studies of cancer progression, epidemiology and many other fields.

Emergent properties of branching morphologies modulate the sensitivity of coral calcification to high PCO2 [RESEARCH ARTICLE]

Experiments with coral fragments (i.e. nubbins) have shown that net calcification is depressed by elevated PCO2. Evaluating the implications of this finding requires scaling of results from nubbins to colonies, yet the experiments to codify this process have not been carried out. Building from our previous research demonstrating that net calcification of Pocillopora verrucosa (2–13 cm diameter) was unaffected by PCO2 (400 and 1000 µatm) and temperature (26.5 and 29.7°C), we sought generality to this outcome by testing how colony size modulates PCO2 and temperature sensitivity in a branching acroporid. Together, these taxa represent two of the dominant lineages of branching corals on Indo-Pacific coral reefs. Two trials conducted over 2 years tested the hypothesis that the seasonal range in seawater temperature (26.5 and 29.2°C) and a future PCO2 (1062 µatm versus an ambient level of 461 µatm) affect net calcification of an ecologically relevant size range (5–20 cm diameter) of colonies of Acropora hyacinthus. As for P. verrucosa, the effects of temperature and PCO2 on net calcification (mg day–1) of A. verrucosa were not statistically detectable. These results support the generality of a null outcome on net calcification of exposing intact colonies of branching corals to environmental conditions contrasting seasonal variation in temperature and predicted future variation in PCO2. While there is a need to expand beyond an experimental culture relying on coral nubbins as tractable replicates, rigorously responding to this need poses substantial ethical and logistical challenges.

Lipid composition of cell membranes is linked to metabolic rate and lifespan in mammals and birds but very little information is available for fishes. In this study, three fish species of the short-lived annual genus Nothobranchius with different maximum lifespan potentials (MLSP) and the longer-lived outgroup species Aphyosemion australe were studied to test whether they conform to the predictions of the longevity-homeoviscous adaptation (LHA) theory of aging. Lipid analyses were performed in whole fish samples and peroxidation indexes (PIn) for every PL class and for the whole membrane, were calculated. Total PL content was significantly lower in A. australe and N. korthausae, the two species with the highest MLSP, and a negative correlation between membrane total PIn and fish MLSP was found, this meaning that the longer-lived fish species have more saturated membranes and therefore, a lower susceptibility to oxidative damage, as the LHA theory posits.

The mechanisms that control intrinsic axon growth potential, and thus axon regeneration following injury, are not well understood. Developmental axon regrowth of Drosophila mushroom body -neurons during neuronal remodeling offers a unique opportunity to study the molecular mechanisms controlling intrinsic growth potential. Motivated by the recently uncovered developmental expression atlas of -neurons, we here focus on the role of the actin-severing protein cofilin during axon regrowth. We show that Twinstar (Tsr), the fly cofilin, is a crucial regulator of both axon growth and branching during developmental remodeling of -neurons. tsr mutant axons demonstrate growth defects both in vivo and in vitro, and also exhibit actin-rich filopodial-like structures at failed branch points in vivo. Our data is inconsistent with Tsr being important for increasing G-actin availability. Furthermore, analysis of microtubule localization suggests that Tsr is required for microtubule infiltration into the axon tips and branch points. Taken together, we show that Tsr promotes axon growth and branching, likely by clearing F-actin to facilitate protrusion of microtubules.

'IPL 2020 postponed indefinitely': BCCI to franchises

The BCCI on Tuesday told IPL franchises that the event stands "postponed indefinitely" after the lockdown to combat the COVID-19 pandemic was extended till May 3, leaving no scope for the league to take place during its designated window. While the development hasn't been officially announced as yet but it is learnt that the BCCI has reached out to all the stakeholders, including the eight franchises and the broadcasters, to convey that the event stands "postponed and not cancelled".

"Yes, the BCCI has intimated us that IPL has been postponed for the time being but they are hopeful of finding a window later this year as and when things normalise," a veteran franchise official told PTI on conditions of anonymity. It is understood that BCCI president Sourav Ganguly and secretary Jay Shah were waiting for the government directive and once the lockdown was extended till May 3 on Tuesday, they saw no possibility of holding the IPL during its traditional April-May window. Ganguly had repeatedly made it clear that the game cannot be a priority given the crisis that the country is facing. India currently has over 10,000 positive cases with over 350 recorded deaths, making it virtually impossible for the government to lift any of the restrictions put in place last month.

The IPL was supposed to be held from March 29 to May 24. However, on March 13, the BCCI announced that the league would be postponed till April 15 due to the restrictions placed on the entry of foreigners, which ruled out any international participation. The only window for an IPL in the current year would be between September and November, provided Cricket Australia and the ICC agree to reschedule the T20 World Cup. The six-month travel restrictions in Australia end on September 30 if the situation becomes normal and the World Cup is scheduled in October-November. The other option is having a curtailed IPL in six weeks between September and early October as it's highly unlikely that the Asia Cup will happen. England are also unlikely to travel for their white-ball series in India.

If you liked Ajay Devgn's 2018 mystery thriller Raid, then you will love this bit of news! It's being reported that Devgn's film will soon get a sequel, as the actor and producer Bhushan Kumar are already said to be working on it. Plus, according to a report, Raid will be turned into a multi-film franchise. Excited yet?

A source close to the development informed Mumbai Mirror, "The franchise seeks to bring into the public eye heroes who work closely with intelligence agencies to track white-collar crimes. To this day, nobody knows the identity of the officer on whom Ajay's Amay was modelled. Raid 2 will also be a tribute to men who don’t wear the uniform."

Bhushan Kumar, who produced the first film, told the publication, "It will be a huge multi-film franchise and Ajay Ji and I, along with Kumar Mangat ji, are looking to take Raid forward. The script of Part 2 is being developed. Since the original was a much-loved film, the sequel is a huge responsibility. After the super success of Tanhaji: The Unsung Warrior, there's a lot of pressure on this collaboration to live up to the expectation. We have always aimed at creating high concept films."

Raid, directed by Raj Kumar Gupta, starred Ajay Devgn an IRS officer, Amay Patnaik. While Devgn and Bhushan Kumar are already collaborating on Bhuj: The Pride Of India, news of them reuniting for the Raid series is much welcome for fans of the film!

'Jurassic Park' child star Joe Mazzello not back in franchise yet

"Jurassic Park" child star Joe Mazzello says he is not coming back to the franchise yet. Mazzello as a child essayed the role of Tim Murphy, grandson of the park founder John Hammond, in 1993's "Jurassic Park" and the 1997 sequel "The Lost World: Jurassic Park". Recently, actor Chris Pratt said that the original cast is slated to return for "Jurassic World 3".

According to a source, Mazzello hasn't yet been contacted by the studio to reprise his role in the film franchise, reports pagesix.com.

"I hope we get to find out someday (what happens to Tim)," Mazzello told fans during a YouTube Q&A recently after watching the movie on a live-stream together.

The 36-year-old then hinted that he wasn't reprising his character, saying: "I do think about it all the time because there's so much I think that you could like do with this character."

He said: "Like, he was like sort of the obvious heir to, like, John Hammond's fortune. He was the heir to Jurassic Park and, like, would this experience have turned him away from dinosaurs and make him hate dinosaurs?

"I do think it would be amazing to see what happened to Tim Murphy, you know as an adult, and I don't know … there's so much you can do with that character and yeah, you never know, but maybe one day we'll be able to find out," he added. "They got to make a lot more movies, that's what I'm saying, OK? A lot more films," he added.

According to the source, "millions of 'Jurassic Park' fans have been extremely upset" since Mazzello made the remarks.

"This character is iconic and part of so many of the fans' lives. Joe's comments alluded that there needed to be more movies in order for his character to possibly come back. It's not up to Joe, it's up to the studio and director if they want the entire original cast or not," added the source.

The film has "no set timetable to start production" amid the ongoing coronavirus shutdown. It has a tentative release date of June 11, 2021.

MS Dhoni's wife Sakshi Dhoni apparently shared ain Instagram Live video in which Mahi can be seen giving his daughter Ziva a ride on a motorbike in their farmhouse. 

The video was also shared by MS Dhoni's Indian Premier League (IPL) team Chennai Super Kings on Twitter immediately after it went viral. CSK went on to caption the photo with some fun play of words: "Thala Suthifying, literally. #WhistlePodu VC: @SakshiSRawat."

Sakshi Dhoni had also shared a cute photo of herself and MS Dhoni during lockdown as she playfully goes to lick his toes while he was immersed in his mobile phone.

In normal circumstances, MS Dhoni would have been currently leading three-time champions CSK in the 13th edition of the IPL. However, due to the coronavirus pandemic, the cash-rich league has been postponed indefinitely by the BCCI.

The former captain's participation at this year's T20 World Cup remains in doubt as he hasn't played international cricket since India's defeat at the hands of New Zealand in the World Cup semi-final last year.
