A kainate receptor-selective RNA aptamer [Neurobiology]

Kainate and α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are two major, closely related receptor subtypes in the glutamate ion channel family. Excessive activities of these receptors have been implicated in a number of central nervous system diseases. Designing potent and selective antagonists of these receptors, especially of kainate receptors, is useful for developing potential treatment strategies for these neurological diseases. Here, we report on two RNA aptamers designed to individually inhibit kainate and AMPA receptors. To improve the biostability of these aptamers, we also chemically modified these aptamers by substituting their 2'-OH group with 2'-fluorine. These 2'-fluoro aptamers, FB9s-b and FB9s-r, were markedly resistant to RNase-catalyzed degradation, with a half-life of ∼5 days in rat cerebrospinal fluid or serum-containing medium. Furthermore, FB9s-r blocked AMPA receptor activity. Aptamer FB9s-b selectively inhibited GluK1 and GluK2 kainate receptor subunits, and also GluK1/GluK5 and GluK2/GluK5 heteromeric kainate receptors with equal potency. This inhibitory profile makes FB9s-b a powerful template for developing tool molecules and drug candidates for treatment of neurological diseases involving excessive activities of the GluK1 and GluK2 subunits.

Kai Tak archaeological expo set

About 200 archaeological Song-Yuan period finds unearthed at Kai Tak will go on display in an exhibition at the Heritage Discovery Centre from tomorrow until February 26.

      

Following the closure of Kai Tak Airport in 1998, the planning work for the Kai Tak Development Area created opportunities for archaeological studies around the former Sacred Hill in Kowloon Bay.

 

An abundance of Song-Yuan period cultural remains were subsequently unearthed, including a huge amount of ceramics produced by various kilns in Zhejiang, Fujian and Jiangxi.

 

Similar products from the same period were exported widely and could be found in shipwrecks along the maritime ceramics route.

 

Highlights of the Treasures from the Sacred Hill: Song-Yuan Period Archaeological Discoveries from Kai Tak exhibition include green glazed incense burners with an eight trigrams pattern and a green glazed dish with a moulded double fish pattern produced by Longquan Kiln.

 

Other highlights include a brown glazed dragon jar with lugs and a green glazed basin with a phoenix pattern produced by Cizao Kiln and a Daguan Tongbao bronze coin.

 

Ceramics from the same period on loan from the Art Museum of the Chinese University of Hong Kong will also be displayed.

The progressive development of microfabrics from initial deposition to slump deformation: an example from a modern sedimenary melange on the Nankai Prism

The progressive development of microfabrics from initial deposition to slump deformation and then a submarine slide was investigated in an active subduction zone using cores recovered during the Integrated Ocean Drilling Program Expedition 333. A Pleistocene–Holocene sequence was recovered at Site C0018A, which was located on a slope basin on the footwall of the megasplay fault in the Nankai Trough, SW Japan. Six mass-transport deposit units intercalated with coherent intervals were recovered from the upper 190 m of the drilled succession. The initial microfabrics in the undeformed hemipelagic sediments were characterized by random and porous fabrics composed predominantly of clay aggregations and connectors. The initial fabrics were cardhouse fabrics, which consist of clay flakes with edge-to-edge (E–E) and/or edge-to-face (E–F) contacts. These initial microfabrics developed into compacted microfabrics, which are random and consolidated fabrics (bookhouse fabrics) that consist of clay flakes with E–F and/or face-to-face (F–F) contacts and develop during burial as a pure shear deformation. During slumping, these fabrics were then deformed under simple shear to become predominantly F–F contacts and form clay chains. Thus, the microfabrics in these submarine slides are a sedimentary mélange that developed locally into a preferred clay orientation with F–F contacts.

Early in 2019, seventeen writers were recruited to participate in a box set of medical thrillers, called (with unforeseen irony) Do No Harm.

The buy-in: $750, with net income from sales going to two designated charities, and participating writers receiving a pro-rated share of any net income above those contributions. The goal: through cooperative marketing efforts, to get the set on the USA Today bestseller list.

Here's how the opportunity was presented to potential participants (this is from an email that was shared with me):


In other words, right off the bat, authors were being primed not to expect to make money.

Helming the endeavor was a five-year-old PR company called Genius Media Inc., owned by a man called Wid Bastian (full name: Widtsoe T. Bastian). Per this long 2017 discussion on KBoards, one of Genius Media's not-so-genius MOs was to cold-contact writers by form email and offer glowingly-described Kindle Unlimited promotions. "For you, my estimate on an eBook promo is 10,000 plus downloads and 700 plus sales, positive ROI right out of the gate and huge page read income."

The cost: $2,000.

KBoards members urged caution, especially after expert analysis of Genius's claims indicated that its promotions weren't as successful as it advertised, and information offered by a writer who'd paid for a promo suggested that Genius was violating the TOS of the advertising platforms it used. Also noteworthy: this post from a writer who bought two promos from Genius, and lost money on each one. "That is what Genius Media told me to expect, that the first promo would not show a profit, but that by the second or third promo, they would show a profit." (The writer bought a third promo.)

As it turns out, these concerns were a sign of things to come.

Do No Harm was published, as promised, and made the USA Today bestseller list, also as promised. On October 3 (or possibly October 4), it was unpublished--three (or possibly four) days after the contractually-stipulated end date of September 30.

Per the contract (which you can see here), final reporting and payment was due to authors "no later than December 1". December 1 came...and went. Bastian promised anxious authors they'd get everything by December 15.

One day past that deadline, they did receive a report...but not from Genius Media. Between early December and December 16, with no notice or warning, a company called Kairos Phoenix had purportedly acquired Genius. Other than its business registration--in Wyoming, just like Genius's--and hometown--Logan, Utah, again just like Genius's--Kairos was a black box, with zero web presence. It had incorporated less than a month earlier, on November 22. (You can see the report here.)

That wasn't all that was suspicious. Here's Kairos's financial breakdown:


In other words, USA Today bestselling box set Do No Harm hadn't just failed to make a profit, it had lost money. But...where was the revenue from the $750 buy-in fees--which, with 17 authors, totaled $12,750? (Kairos's explanation: it wasn't included because it was "ordinary income" for Genius Media. "As stated in the contract, the fee was paid to 'participate' in DNH Collab by the author and for no other purpose".) Where were revenues for the days the set had been on sale past the unpublish deadline? (Kairos: "The contractual period for DNH Collab was strictly defined in the contract" as September 30, so any revenue past that date was "irrelevant".)

Equally troubling, why were there more than $15,000 in expenses--three-quarters of which were for "labor"--when the contract stipulated that expenses were not to exceed the total of the buy-in fees? According to Kairos, this wasn't really an expense cap: "This provision was specifically and intentionally included in the contract language to avoid the possibility of a 'cash call' – Genius Media asking authors to contribute more to DNH Collab to achieve the goal of making USA Today Bestseller status. No 'cash call' was ever made in the DNH Collab."

Here's the actual contract wording, though (my bolding):
For the purposes of the USA Today Bestseller Medical Thriller Author Publishing Collaborative Boxed Set program, Genius Media shall not incur any publication and promotion expenses of any nature in excess of the fees paid under the terms of its author agreements and shall have no power to obligate [redacted] or any other author for any publication and promotion expense above author fees paid whatsoever.
There's a "cash call" prohibition and an expense cap. But Kairos wanted writers to believe otherwise, so it could inflate expenses and ensure a loss.

It was obvious to Do No Harm participants that Kairos was taking the money and running--avoiding the substantial payouts it would otherwise have to make by retroactively interpreting contract language, and also enabling Bastian himself to claim he was blameless because of Genius's supposed takeover by an unrelated company. (No one was under any illusion that Kairos Phoenix was anything other than Bastian in a different guise.)

Authors were furious. On December 22, two of them, Christoph Fischer and Dan Alatorre, went public with their experience. Others posted warnings on Kairos's corporate business listing.

Do No Harm isn't the first time Bastian has run this scheme, either.


The box set in question appears to be Tales From Big Country, which was published around the same time as Do No Harm. A third set, Galaxia, was pubbed in September, with profits supposedly going to the Well Aware clean water charity. I've been told that Bastian is recruiting for other sets, including a collection of thrillers.

So who is Wid Bastian, a.k.a. Widtsoe T. Bastian?

His LinkedIn profile ("EXPOSE your new book to develop your author brand and sell more books!") identifies him as the owner of Genius Media, and also of an ebook promotion program called Book Dynamite. In an earlier profile on a freelancers' job site, he describes himself as a "published novelist and screenwriter" (more on that below) who "makes most of my daily bread as a ghostwriter." He also has an IMDB profile, presumably because of his efforts to make a film of the life of Greek Orthodox priest Fr. Themi Adamopoulos.

Genius Media's website has been taken offline, but traces remain in the form of cached pages, and here's how it looked in January 2016, courtesy of the Internet Archive (more recent versions haven't been archived). The company has a D+ rating from the BBB. Bastian also owned or was an officer with at least three other companies during the early to mid-1990s: Off & Flying, Prospex Interntaional [sic] (yes, it really was registered with a typo in the name), and Nevada Pension Investment Fund. Both Off & Flying and Prospex had their statuses "permanently revoked" a few years after incorporating. All three are long dead.

There is also a Widtsoe T. Bastian who pleaded guilty to 13 felony counts including embezzlement, money laundering, and bankruptcy fraud in US District Court in North Carolina, and in 2005 was sentenced to one hundred and forty-four months in prison and restitution of more than $3,000,000. Nothing I can find online directly connects Widtsoe T. Bastian of Genius Media to Widtsoe T. Bastian of North Carolina, so it may be a different person. But Widtsoe T. Bastian is quite an unusual name, as a websearch will make clear.

Finally...I can't say "what goes around comes around", since this pre-dates the ripoff that's the subject of this post, but it certainly seems like a case of advance karma: Bastian's own 2010 novel, Solomon's Porch, was published by none other than Tate Publishing & Enterprises, a notorious vanity publisher that scammed thousands of authors and a multitude of staff, and whose owners pleaded guilty in 2017 to an array of felony charges very similar to the ones described in the previous paragraph.

Tate authors suffered terribly at the hands of their unscrupulous publisher, but Wid Bastian is one Tate author I can't feel all that sorry for.

UPDATE 12/31/19: PACER was down yesterday, so I wasn't able to do a case search. I did so this morning, and here's what's there for Widtsoe T. Bastian:


Some of the listings are redundant, and several cite court documents without any links to those documents. But there's enough available to paint a tangled picture of a 1995 Chapter 11 bankruptcy in Nevada involving several companies in addition to the three mentioned above (the case was finally closed in 2002); a 1999 indictment in Nevada "on charges related to the operation of [Bastian's] venture capital firm"; failure to appear in a Nevada court in 2001; and a 2002 arrest in North Carolina, leading to the plea of guilty on 13 felony counts referenced above.

In 2012, Bastian was placed on probation or supervised release, and jurisdiction over his case was transferred to US District Court for Utah. His probation ended on May 4, 2015.

UPDATE 2/21/19: I'm featuring this comment from yesterday, as it's more indication of a pattern (the commenter has shared documentation with me that confirms what they say below):
In 2013-15, I hired Mr. Bastian as a ghost writer and later co-author of a novel we wrote together called Henry and Tom, sold through Amazon/Kindle Direct Publishing (KDP).

In mid 2015, Mr. Bastian embezzled money from a joint bank account we opened to pay for marketing campaigns for Henry and Tom. Rather than pursue embezzlement charges, I hired an expensive lawyer in Washington DC to transfer all rights for Henry and Tom to me in exchange for a $4K payment to Wid and a non-disparagement clause for both of us. This seemed at the time like the most expedient and cost effective way to deal with all the problems Mr. Bastian had caused.

I thought this matter was concluded until recently when Amazon/KDP informed me that a debt collection firm had placed a lien on Henry and Tom due to a bankruptcy filed by Wid Bastian/Genius Media Inc. KDP is currently sending my royalties to the collector per the lien. I have filed formal complaints with Attorneys General in Connecticut, New Jersey, and New York in an attempt to have the lien removed in states where the debt collectors are registered. I have also filed formal complaints against Wid and Genius Media Inc. with the Attorney General of Utah and the County Attorneys Office in Providence, Utah.

I think blogs like this are creating awareness of the depth and extent of Wid’s continuing criminal activities so we can all work together to stop him from doing this to others and get him to face justice.
You'll note a reference to a bankruptcy. Wid Bastian filed a Chapter 7 bankruptcy petition on January 13, 2020.


A number of victims of his box set schemes are listed as unsecured creditors; if you're one of them, you should have received a snail mail notice, but if you haven't, and are wondering if you're included, here's the Certificate of Notice, with all Bastian's claimed creditors.

The bankruptcy filings also confirm what everybody already knew: Kairos Phoenix was Wid Bastian. The reason he set up this company probably won't surprise you, either.

Katrina Kaif condoles her 'New York' co-star Irrfan Khan's demise

Bollywood superstar Katrina Kaif on Wednesday condoled the demise of her 'New York' co-star and critically acclaimed actor Irrfan Khan. Kaif took to Instagram to post a picture of the departed actor and recalled the time she spent with him during the shoot of the film.

"RIP... an irreplaceable loss ... a true genius ..... will be greatly missed .... can never forget our laughs during New York," she wrote in the caption.

The two actors worked together in 2009's thriller flick 'New York.' The blockbuster hit was based on the terrorist attack on World Trade Center which took place in September 2011.

The film also featured actors John Abraham and Niel Nitin Mukesh.

Irrfan Khan's last rites were performed at the Versova cemetery earlier today.

The internationally renowned actor passed away at Mumbai's Kokilaben Hospital on Wednesday while battling rare cancer. He was 53. Irrfan was last seen on screen in 'Angrezi Medium'. He was not a part of the film's promotions owing to his ill health.

With critically acclaimed films like 'The Lunchbox' and 'Paan Singh Tomar' to his credit, Irrfan was regarded as one of the finest actors in Hindi cinema.

The film stars Akshay Kumar in a pivotal role. Due to the Coronavirus scare, her film Sooryavanshi has been postponed indefinitely, keeping in mind the sensitive atmosphere. KL Rahul should only be used as backup wicket-keeper: Mohammad Kaif

Former cricketer Mohammad Kaif on Wednesday said KL Rahul should only be used as a backup wicket-keeper so that workload on the player can be managed.

"People think that Rahul may become our main keeper in the future. But I feel that Rahul should be used as a backup wicketkeeper, if the main keeper is injured or he is not with the team then Rahul should be there because if you rely on him as the main keeper then chances of injuries will grow as the workload will increase on him," Kaif told ANI.

Kaif also feels that MS Dhoni should feature in the ICC Men's T20 World Cup and ruling the former captain out from the tournament will be 'unfair'.

"See people may have eyes on Dhoni that how he is going to play in the IPL (Indian Premier League) and then there will be talks of the T20 World Cup but my view is different from others. I do not judge Dhoni based on his IPL form. He is a great batsman and he is fit now, he wants to play IPL, do captaincy and is showing his availability. He has a winning mentality and knows how to win games under pressure," Kaif said.

"So, I think ruling him out will be unfair. See a lot of cricket is still left in Dhoni and when a player plays for this long, there are ups and downs in his career. It happens with every cricketer not just Dhoni," he added.

The ICC T20 World Cup 2020 is scheduled to be played between October 18 and November 15 at seven venues across Australia.

As the country continues to fight against the coronavirus crisis, Kaif urged people to adhere to the guidelines laid by the government and stay home.

"My message for the society is to be at home and follow what the government is saying then only we can beat this virus. At this stage, this is something very important for one and all to be at home," he said.

A lot of cricket left in MS Dhoni, should play T20 World Cup: Mohammad Kaif

Former cricketer Mohammad Kaif on Wednesday backed MS Dhoni to play in the ICC Men's T20 World Cup, saying that the wicket-keeper batsman has 'a lot of cricket' left in him and ruling him out will be 'unfair'.

"See, people may have eyes on Dhoni that how he is going to play in the IPL (Indian Premier League) and then there will be talks of the T20 World Cup but my view is different from others. I do not judge Dhoni based on his IPL form. He is a great batsman and he is fit now. He wants to play IPL, do captaincy and is showing his availability. He has a winning mentality and knows how to win games under pressure," Kaif told ANI.

"So, I think ruling him out will be unfair. See a lot of cricket is still left in Dhoni and when a player plays for this long, there are ups and downs in his career. It happens with every cricketer, not just Dhoni," he added.

Dhoni last played competitive cricket during the 2019 World Cup. India was knocked out of the tournament after suffering a semi-final defeat at the hands of New Zealand. Dhoni was run out in the 49th over after scoring 50 runs. After his dismissal, India failed to chase the 240-run target.

Despite Dhoni failing to take India over the line, Kaif termed him a 'champion player'.

"The semifinal of World Cup which we lost, everyone was expecting Dhoni to win it but it did not happen. So, from there on, people thought that Dhoni should not be there. But for me, Dhoni is a champion player. People should look at Dhoni's record in the last 10 to 15 years. See, the current form is important but at the same time his form was not that bad that you rule him out," Kaif said.

The ICC T20 World Cup 2020 is scheduled to be played between October 18 and November 15 at seven venues across Australia.

As the nationwide lockdown as been extended to May 3 due to the coronavirus, the IPL has been postponed indefinitely. "IPL 2020 season has been postponed indefinitely," sources within BCCI told ANI on Wednesday.

Kaif feels that postponing the league is the right decision as life is more important than sports at the moment.

"In the present scenario, life is more important than sports. I think the decision of BCCI is right. When the situation improves, we will think of sports. Sporting events all over the world have either been postponed or cancelled. In our country, cases are rising. So, as of now, the situation is not under control. So I think it's the right decision because at this stage first priority is to beat coronavirus and save the people," he said.

When asked about his views on the window for IPL, Kaif said: "I feel when things are sorted out and there is the possibility of games to be played, then they (BCCI) will look for options."

Australia opener David Warner is making most of the forced break by spending quality time with his family at home as the cricket world remains on a standstill due to coronavirus outbreak. On Saturday, Warner shared a TikTok video in which he is dancing with his daughter on a popular Bollywood track.

Revealed! Here's how Mohammad Kaif got the nickname 'bhai sahab'

Former India cricketer Sachin Tendulkar recalled a hilarious incident which ended up with his former teammate Mohammed Kaif getting a new nickname. The Master Blaster was referring to an incident that took place recently in the first match of Road Safety Series against West Indies XI.

Tendulkar stated that Kaif was giving his all in the field despite being 39 and that prompted others to caution him that it is the first match of the tournament and there is still a long way to go.

"We had to tell Kaif to be cautious. We nicknamed him 'bhai sahab, bhai sahab, thoda sambhalke.' It's the first match of the tournament and there are a lot of matches to be played," Tendulkar said in a video uploaded on 100 MB YouTube channel.

"What if you pull a hamstring or something. It (diving and fielding quickly) comes naturally to him. He thinks if you are fit, you continue doing those things. He was a really fit guy. He was an outstanding fielder in team," he added.

Kaif was regarded as one of the finest fielders ever to don the iconic blue jersey. He, along with Yuvraj Singh, marshalled the cover-point area with great aplomb in an era where fielding wasn't Team India's strongest suit.

Despite his incredible fielding skills, Kaif is fondly remembered by almost every Indian cricket fan for his Man of the Match performance in the Natwest Trophy final against England at Lord's in 2002.

I thought we would lose when Yuvraj Singh got out: Mohammed Kaif on NatWest final

Former India batsman Mohammad Kaif had thought the famous NatWest Series final, which India won defying odds against England, was lost when Yuvraj Singh got out in their daunting chase of 326 at Lord's in 2002.

India, led by Sourav Ganguly, won the final by two wickets with three balls to spare riding Yuvraj and Kaif's sixth-wicket stand of 121 runs. The Men in Blue were 146/5 at one stage when the pair joined hands and from there the rest was history.

Kaif remained unbeaten on 87 to take India home with Zaheer Khan at the other end but Yuvraj got out in the 42nd over on 69, with India still needing 59 for victory and four wickets in hand.

"When you (Yuvraj) got out, I thought the match is gone. I did not think we will win," Kaif said in an Instagram Live chat with Yuvraj.

"I was set, you were there. So I believed if we played till the end India win will. But you got out and India lost hope and my heart broke," Kaif added.

The victory, also remembered for Ganguly celebrating by opening his shirt at the Lord's balcony, changed Indian cricket forever in ways more than one.

Kaif also picked out Yuvraj's 25-ball 58 against Australia in the 2000 U-19 World Cup where the former was the captain as one of his best knocks despite the decorated southpaw smashing six sixes and having many other memorable knocks in his illustrious career.

"That innings, the way you dominated the likes of Mitchell Johnson, Shane Watson was amazing. You have many superb knocks, you hit six sixes...but to me at the U-19 level to see someone bat like that...was special. We all knew you will go far and play for a long time," Kaif said.

Rated as one of the best fielders in India, Kaif said he worked on his fielding as he wanted to stand out in one of the departments.

"I always wanted to be different and I wanted to focus on fielding. I worked really hard on my fielding."

Yuvraj added that he and Kaif changed the momentum for India on the field during those days.

"At point and cover we changed the momentum in a lot of ways. Now the team has so many good fielders but I feel we changed the momentum," said Yuvraj, himself a livewire.
