dba Cdc24 interacts with the septins to create a positive feedback during bud site assembly in yeast [RESEARCH ARTICLE] By jcs.biologists.org Published On :: 2020-04-29T02:35:34-07:00 Julian Chollet, Alexander Dünkler, Anne Bäuerle, Laura Vivero-Pol, Medhanie A. Mulaw, Thomas Gronemeyer, and Nils JohnssonYeast cells select the position of their new bud at the beginning of each cell cycle. The recruitment of the septins to this prospective bud site is one of the critical events in a complex assembly pathway that culminates in the outgrowth of a new daughter cell. Hereby, the septin-rods follow the high concentration of Cdc42GTP that is generated by the focused localization of its GEF Cdc24. We show that shortly before budding Cdc24 not only activates Cdc42 but also transiently interacts with Cdc11, the septin subunit that caps both ends of the septin rods. Mutations in Cdc24 reducing the affinity to Cdc11 impair septin recruitment and decrease the stability of the polarity patch. The interaction between septins and Cdc24 thus reinforces bud assembly at sites where septin structures are formed. Once the septins polymerize into the ring, Cdc24 is found at the cortex of the bud and directs its further outgrowth from this position. Full Article

They argue that, in focusing our efforts on correcting weaknesses and rounding people out, we lose the ability to get exceptional performance from them. Instead, we should focus on strengths and push everyone to shine in their own areas. To do that, companies need to rethink the way they review, pay, and promote their employees. dba Congress working remotely highlights need for better broadband connectivity, Loebsack says By feedproxy.google.com Published On :: Sat, 09 May 2020 10:44:03 PDT CEDAR RAPIDS — Like other members of the United States House, the “new normal” for Rep. Dave Loebsack involves a lot of time on the phone and in video conferences.Although the Senate returned to Washington this past week, the House may not return to the Capitol until mid-May. As representatives work from their districts, many face the same challenges as their constituents who are working from home via various phone and video meeting platforms.“They’re clunky, difficult, frustrating at times,” the 2nd District Democrat said after a 90-minute Energy and Commerce Committee teleconference meeting, “but we can get a lot of things done that way without necessarily going into Washington. It’s not the same by any means ... but I think we can get a lot of our work done that we have to get done even though were not there.”If there’s an upside to the telecommuting downside, it’s that the poor connections, dropped calls and sometimes spotty internet signals highlight an issue Loebsack has been working on — Congress and the country need better broadband connectivity. A member of the Communications and Technology subcommittee of the Energy and Commerce Committee, which has jurisdiction over telecommunications issues, Loebsack also serves as a co-chair of the Rural Broadband Caucus. In one meeting, Loebsack said, a fellow committee member was unable to ask questions because the Wi-Fi signal at his home outside of a southwestern city was so poor.“So I think we’re going to make some progress” convincing congressional colleagues of the need to connect all Americans to reliable and affordable broadband internet, Loebsack said.“In times like these, when families are at home, millions of K-12 students do not have access to the high-speed internet needed to take classes and complete their assignments online,” he said in an announcement with House Majority Whip James Clyburn and others of a House Democratic plan to connect all Americans to high-speed internet. When the transmission of COVID-19 slows, the need for high-speed internet connectivity will remain as well as the need for Congress to provide incentives for internet service providers to close the “broadband gap” by extending service to the las mile in rural areas and eliminate what he called “internet deserts” that exist in cities. The plan calls for an investment of $80 billion over five years for internet infrastructure and another $5 billion over five years for low-interest financing of projects. With more reliable connectivity, Congress could consider remote voting at times like this when members don’t want to gather in Washington. House Speaker Nancy Pelosi has been cool to the idea, but Loebsack thinks members are “more open” to voting remotely.“But we have to make sure that it’s completely secure, that the system can’t get hacked, and that votes are recorded properly,” he said.Remote voting may be a may be a generational issue, “but I think this is doable.”“I’m not making that prediction, but I would not be shocked if we move in that direction, especially the longer this (isolation) goes on,” Loebsack said.Comments: (319) 398-8375; james.lynch@thegazette.com Full Article Government

dba Output feedback stochastic MPC with packet losses. (arXiv:2004.02591v2 [math.OC] UPDATED) By arxiv.org Published On :: The paper considers constrained linear systems with stochastic additive disturbances and noisy measurements transmitted over a lossy communication channel. We propose a model predictive control (MPC) law that minimizes a discounted cost subject to a discounted expectation constraint. Sensor data is assumed to be lost with known probability, and data losses are accounted for by expressing the predicted control policy as an affine function of future observations, which results in a convex optimal control problem. An online constraint-tightening technique ensures recursive feasibility of the online optimization and satisfaction of the expectation constraint without bounds on the distributions of the noise and disturbance inputs. The cost evaluated along trajectories of the closed loop system is shown to be bounded by the optimal predicted cost. A numerical example is given to illustrate these results. Full Article
dba Exponential decay for negative feedback loop with distributed delay. (arXiv:2005.03136v1 [math.DS]) By arxiv.org Published On :: We derive sufficient conditions for exponential decay of solutions of the delay negative feedback equation with distributed delay. The conditions are written in terms of exponential moments of the distribution. Our method only uses elementary tools of calculus and is robust towards possible extensions to more complex settings, in particular, systems of delay differential equations. We illustrate the applicability of the method to particular distributions - Dirac delta, Gamma distribution, uniform and truncated normal distributions. Full Article

dba Monitoring or feedback systems and methods By www.freepatentsonline.com Published On :: Tue, 26 May 2015 08:00:00 EDT The present invention generally relates to systems and methods for monitoring and/or providing feedback for drugs or other pharmaceuticals taken by a subject. In one aspect, the present invention is directed to devices and methods for determining a species within the skin of a subject; and producing feedback to a subject based on the determination of the species. The feedback may be, for example, visual, audible, tactile, a change in temperature, etc. In some cases, information regarding the determination of the species may be transmitted to another entity, e.g., a health care provider, a computer, a relative, etc., which may then provide feedback to the subject in some fashion. In some cases, the feedback may be directly indicative of the species, e.g., whether the species is present, the concentration of the species, whether a by-product of a reaction involving the species is present, whether a compound affected by the species is present, etc. However, the feedback may also be indirect in some embodiments. For example, the subject may be presented with an external reward, e.g., based on the determination of the species within the skin. For instance, a reward such as cash, coupons, songs, discounts, personal items, etc., may be offered based on the level of compliance of the subject. Still other aspects of the invention are generally directed to kits involving such devices (with or without the drug to be monitored), methods of promoting such systems, or the like. Full Article
