Mature myelin maintenance requires Qki to coactivate PPARβ-RXRα–mediated lipid metabolism

Lipid-rich myelin forms electrically insulating, axon-wrapping multilayers that are essential for neural function, and mature myelin is traditionally considered metabolically inert. Surprisingly, we discovered that mature myelin lipids undergo rapid turnover, and quaking (Qki) is a major regulator of myelin lipid homeostasis. Oligodendrocyte-specific Qki depletion, without affecting oligodendrocyte survival, resulted in rapid demyelination, within 1 week, and gradually neurological deficits in adult mice. Myelin lipids, especially the monounsaturated fatty acids and very-long-chain fatty acids, were dramatically reduced by Qki depletion, whereas the major myelin proteins remained intact, and the demyelinating phenotypes of Qki-depleted mice were alleviated by a high-fat diet. Mechanistically, Qki serves as a coactivator of the PPARβ-RXRα complex, which controls the transcription of lipid-metabolism genes, particularly those involved in fatty acid desaturation and elongation. Treatment of Qki-depleted mice with PPARβ/RXR agonists significantly alleviated neurological disability and extended survival durations. Furthermore, a subset of lesions from patients with primary progressive multiple sclerosis were characterized by preferential reductions in myelin lipid contents, activities of various lipid metabolism pathways, and expression level of QKI-5 in human oligodendrocytes. Together, our results demonstrate that continuous lipid synthesis is indispensable for mature myelin maintenance and highlight an underappreciated role of lipid metabolism in demyelinating diseases.

If you were to Google the definition of a comedian you would see it defined by Dictionary.com as an entertainer whose act is designed to make an audience laugh. Likewise the New York Times has a comedy critic, Jason Zinoman, who defines comedy in a moment of reflecting on his own career of analyzing them.

This often dictates the form of my column, since while the goal of comedy is to make you laugh, what’s fascinating about the art form — especially these days, when it’s so fragmented and aesthetically diverse — is that there are many ways artists accomplish that goal.

However, if you were to ask one furry who considers himself one, 2 Gryphon, you’d find an entirely different etymology of the word, and what the job of a comic is.

It is this quote that we are going to be over-analysing today. I have broken this down into three main points as to why this definition of the job of a comedian is not only a fundamental misunderstanding of the role, but also a resignation of the foundational principles of comedy.

Dissecting Mechanisms of Financial Crises: Intermediation and Sentiment -- by Arvind Krishnamurthy, Wenhao Li

We develop a model of financial crises with both a financial amplification mechanism, via frictional intermediation, and a role for sentiment, via time-varying beliefs about an illiquidity state. We confront the model with data on credit spreads, equity prices, credit, and output across the financial crisis cycle. In particular, we ask the model to match data on the frothy pre-crisis behavior of asset markets and credit, the sharp transition to a crisis where asset values fall, disintermediation occurs and output falls, and the post-crisis period characterized by a slow recovery in output. We find that a pure amplification mechanism quantitatively matches the crisis and aftermath period but fails to match the pre-crisis evidence. Mixing sentiment and amplification allows the model to additionally match the pre-crisis evidence. We consider two versions of sentiment, a Bayesian belief updating process and one that overweighs recent observations. We find that both models match the crisis patterns qualitatively, generating froth pre-crisis, non-linear behavior in the crisis, and slow recovery. The non-Bayesian model improves quantitatively on the Bayesian model in matching the extent of the pre-crisis froth.

Media in the US: lax campaign financing bad for democracy but good for media companies

US campaign financing rules have been relaxed even further in recent years, making a worrying problem about the state of American democracy worse (because those with money have even more ability to try and buy votes or influence policy).

Media and advertising: manipulating photos of people; going too far?

It has long been known that advertisers will “photoshop” (slang for editing photos to touch up or airbrush out imperfections) photos to make the subject more attractive. But many have pointed out that this subtle manipulation often goes too far. Young people, girls in particular, are bombarded with images of how they are supposed to look, contributing to many social and health problems as a result.

The American National Advertising Division (the US advertising industry’s self-regulating watchdog) for example, recently moved to ban the misleading use of photoshopping and enhanced post-production in cosmetics adverts as it could be too misleading. In the UK some adverts have been banned because the effects of airbrushing has resulted in very misleading cosmetic adverts. France has even considered legislation to explicitly identify retouched images.

Some will claim it is up to parents to teach their children how to recognize these things, but parents cannot often win against an army of professional psychologists, marketers and others all focused on pushing their own images onto children, when studies also say that children are not old enough tell the difference between commercial and other content.

Elon Musk says Tesla will 'immediately' leave California after coronavirus shutdowns forced the company to close its main car factory (TSLA)

After a week of decrying coronavirus shelter-in-place orders that have left Tesla's main factory shuttered and unable to produce vehicles, Elon Musk says the company may move its factory out of the state.

"Tesla is filing a lawsuit against Alameda County immediately," the chief executive said on Twitter Saturday morning. "The unelected & ignorant 'Interim Health Officer' of Alameda is acting contrary to the Governor, the President, our Constitutional freedoms & just plain common sense!"

That was followed up with a threat to move Tesla's headquarters outside the state.

"Frankly, this is the final straw," he replied. "Tesla will now move its HQ and future programs to Texas/Nevada immediately. If we even retain Fremont manufacturing activity at all, it will be dependent on how Tesla is treated in the future. Tesla is the last carmaker left in CA."

It wasn't immediately clear if a suit had yet been filed, or in which court Tesla will file the lawsuit. Most state and federal courts are closed on weekends and do not allow filing. In a subsequent Tweet, Musk alsourged shareholders to file a class action suit for damages caused by shutdown. 

Tesla's press relations department did not immediately respond to a request for comment. Alameda County did not immediately respond to a request for comment. 

Alameda County — the East Bay locale which includes Fremont, California, and Tesla's gigafactory about 30 miles southeast of San Francisco — extended its shelter-in-place order on April 29 "until further notice." Local authorities have not allowed Tesla to reopen the factory, and all manufacturing remains prohibited under the order.

The San Francisco Chronicle reported that Tesla was planning to resume some manufacturing operations at the factory as soon as last Wednesday, May 6. Local officials said it did not have permission to do so.

"Right now, the same health order is in place so nothing has changed," Fremont Police Department spokeswoman Geneva Bosques told Business Insider at the time. "Operating the assembly line was determined early on to be a violation."

Last week, following Tesla's first-quarter earnings announcement, Musk decried the shutdowns as a substantial risk to the company's financials.

"Frankly, I would call it forcible imprisoning of people in their homes against all of, their constitutional rights, in my opinion," he said on a conference call. "It's breaking people's freedoms in ways that are horrible and wrong and not why they came to America or built this country. What the f---. Excuse me. Outrage. Outrage."

"If somebody wants to stay in their house, that's great and they should be able to," he continued. "But to say they cannot leave their house and that they will be arrested if they do, that's fascist. That is not democratic — this is not freedom. Give people back their goddamn freedom."

Some states, including Texas, Georgia, and others, have begun to slowly allow certain businesses to re-open in recent weeks.

Musk praised counties neighboring Alameda, like San Joaquin for what he said were more "reasonable" responses. In a podcast released May 7, he told Joe Rogan that the company had learned from the coronavirus in China, where it briefly forced Tesla to close its Shanghai factory — a claim he repeated on Twitter Saturday. 

"Our castings foundry and other faculties in San Joaquin have been working 24/7 this entire time with no ill effects. Same with Giga Nevada," Musk said. "Tesla knows far more about what needs to be done to be safe through our Tesla China factory experience than an (unelected) interim junior official in Alameda County." 

As Musk began to complain about factory shutdowns in April, workers at Tesla's Fremont factory told Business Insider that the comments made them anxious.

"I'm for going back to work, but only if it is safe for me, my family, coworkers," said one production employee. "I don't feel like I'm being forced to stay home or that my freedom has been taken away. It's for the good of California."

AllVideos v5.0.0 released - now with web-native media playback

We have just released a major update to AllVideos, version 5.0.0, for Joomla versions 1.5 to 3.x.

It introduces web-native media playback with no 3rd party dependencies.

Here's what's been added or changed in this new release of AllVideos:

  • AllVideos now supports web-native media only (for self-hosted files) and uses browser-native playback controls, without a single 3rd party dependency. AllVideos will now load a single CSS file for its template and a single JS file to assist in self-hosted media playback - a total of less than 7KBs. As such, both Clappr and JW Player have been removed from AllVideos, as well as any helper JS files that were previously utilized for media playback (e.g. for QuickTime).
  • Modern media playback includes video and audio formats like H.264/MPEG-4 (.mp4/.m4v/.m4a), WebM (.webm), Ogg Theora Vorbis (.ogg/.ogv/.oga), MP3 (.mp3), WAVE PCM (.wav) & FLAC (.flac). AllVideos will also playback .avi and .mkv files as they are used as container formats for H.264/MPEG-4 most of the times. Every other format that was previously supported by AllVideos (e.g. Flash Video, QuickTime or Windows Media), will now appear as a link to download the file as browsers no longer support playback directly or require plugin activation to do so. By utilizing modern media playback only, we ensure that AllVideos can be used as your go-to solution for media embedding across any device.
  • By using web-native media formats in AllVideos, we can now do cool stuff like auto-generate poster/preview images for videos, without the need for server-side software (like FFMPEG). If you already uploaded your own poster/preview images, well, don't worry as they will still take precedence over the auto-generated ones.
  • Added support for custom poster/preview images for both video and audio utilizing the .webp format. This means you can now use all 4 web-native image formats for previews (jpg/png/gif/webp). For added fun, try using animated GIFs as your media preview :)
  • Support for small 3rd party video providers has been dropped, as most are now either defunct or using a service like YouTube for serving content (e.g. Funny or Die or GodTube). This means that AllVideos will now support only the 6 major media providers: YouTube, Vimeo, DailyMotion, Facebook (videos), Flickr (videos) and SoundCloud.
  • All 3 templates in AllVideos have been re-configured and tested across all device types. The "Responsive" template is of course still the default template when you install AllVideos for the first time.
  • A new option has been added that allows you to limit the maximum width responsive media can occupy. This will come handy with sites that extend beyond 1200px in width as media can be limited to a percentage of the page's width or to a fixed pixel size (e.g. no more than 1000px width for media playback).
  • You can now allow your visitors to directly download a self-hosted video file (previously possible for audio files only).
  • The options have been significantly simplified with the removal of Clappr & JW Player as well as the removal of QuickTime support.
  • AllVideos continues to be PHP 5 to 7 compatible, for Joomla 1.5 to 3.x. There is really no reason to ditch support for PHP 5 and/or Joomla 1.5/2.5 when it's easy to maintain support for them. Neither security nor performance are sacrificed either way.
