Pre-transplant testosterone and outcome of men after allogeneic stem cell transplantation

Testosterone is an important determinant of endothelial function and vascular health in men. As both factors play a role in mortality after allogeneic stem cell transplantation (alloSCT), we retrospectively evaluated the impact of pre-transplant testosterone levels on outcome in male patients undergoing alloSCT. In the discovery cohort (n=346), an impact on outcome was observed only in the subgroup of patients allografted for acute myeloid leukemia (AML) (n=176, hereafter termed ‘training cohort’). In the training cohort, lower pre-transplant testosterone levels were significantly associated with shorter overall survival (OS) [hazard ratio (HR) for a decrease of 100 ng/dL: 1.11, P=0.045]. This was based on a higher hazard of non-relapse mortality (NRM) (cause-specific HR: 1.25, P=0.013), but not relapse (cause-specific HR: 1.06, P=0.277) in the multivariable models. These findings were replicated in a confirmation cohort of 168 male patients allografted for AML in a different center (OS, HR: 1.15, P=0.012 and NRM, cause-specific HR: 1.23; P=0.008). Next, an optimized cut-off point for pre-transplant testosterone was derived from the training set and evaluated in the confirmation cohort. In multivariable models, low pre-transplant testosterone status (<250 ng/dL) was associated with worse OS (hazard ratio 1.95, P=0.021) and increased NRM (cause-specific HR 2.68, P=0.011) but not with relapse (cause-specific HR: 1.28, P=0.551). Our findings may provide a rationale for prospective studies on testosterone/androgen assessment and supplementation in male patients undergoing alloSCT for AML.

Impact of cytogenetic abnormalities on outcomes of adult Philadelphia-negative acute lymphoblastic leukemia after allogeneic hematopoietic stem cell transplantation: a study by the Acute Leukemia Working Committee of the Center for International Blood and

Cytogenetic risk stratification at diagnosis has long been one of the most useful tools to assess prognosis in acute lymphoblastic leukemia (ALL). To examine the prognostic impact of cytogenetic abnormalities on outcomes after allogeneic hematopoietic cell transplantation, we studied 1731 adults with Philadelphia-negative ALL in complete remission who underwent myeloablative or reduced intensity/non-myeloablative conditioning transplant from unrelated or matched sibling donors reported to the Center for International Blood and Marrow Transplant Research. A total of 632 patients had abnormal conventional metaphase cytogenetics. The leukemia-free survival and overall survival rates at 5 years after transplantation in patients with abnormal cytogenetics were 40% and 42%, respectively, which were similar to those in patients with a normal karyotype. Of the previously established cytogenetic risk classifications, modified Medical Research Council-Eastern Cooperative Oncology Group score was the only independent prognosticator of leukemia-free survival (P=0.03). In the multivariable analysis, monosomy 7 predicted post-transplant relapse [hazard ratio (HR)=2.11; 95% confidence interval (95% CI): 1.04-4.27] and treatment failure (HR=1.97; 95% CI: 1.20-3.24). Complex karyotype was prognostic for relapse (HR=1.69; 95% CI: 1.06-2.69), whereas t(8;14) predicted treatment failure (HR=2.85; 95% CI: 1.35-6.02) and overall mortality (HR=3.03; 95% CI: 1.44-6.41). This large study suggested a novel transplant-specific cytogenetic scheme with adverse [monosomy 7, complex karyotype, del(7q), t(8;14), t(11;19), del(11q), tetraploidy/near triploidy], intermediate (normal karyotype and all other abnormalities), and favorable (high hyperdiploidy) risks to prognosticate leukemia-free survival (P=0.02). Although some previously established high-risk Philadelphia-negative cytogenetic abnormalities in ALL can be overcome by transplantation, monosomy 7, complex karyotype, and t(8;14) continue to pose significant risks and yield inferior outcomes.

Extensive multilineage analysis in patients with mixed chimerism after allogeneic transplantation for sickle cell disease: insight into hematopoiesis and engraftment thresholds for gene therapy

Although studies of mixed chimerism following hematopoietic stem cell transplantation in patients with sickle cell disease (SCD) may provide insights into the engraftment needed to correct the disease and into immunological reconstitution, an extensive multilineage analysis is lacking. We analyzed chimerism simultaneously in peripheral erythroid and granulomonocytic precursors/progenitors, highly purified B and T lymphocytes, monocytes, granulocytes and red blood cells (RBC). Thirty-four patients with mixed chimerism and ≥12 months of follow-up were included. A selective advantage of donor RBC and their progenitors/precursors led to full chimerism in mature RBC (despite partial engraftment of other lineages), and resulted in the clinical control of the disease. Six patients with donor chimerism <50% had hemolysis (reticulocytosis) and higher HbS than their donor. Four of them had donor chimerism <30%, including a patient with AA donor (hemoglobin >10 g/dL) and three with AS donors (hemoglobin <10 g/dL). However, only one vaso-occlusive crisis occurred with 68.7% HbS. Except in the patients with the lowest chimerism, the donor engraftment was lower for T cells than for the other lineages. In a context of mixed chimerism after hematopoietic stem cell transplantation for SCD, myeloid (rather than T cell) engraftment was the key efficacy criterion. Results show that myeloid chimerism as low as 30% was sufficient to prevent a vaso-occlusive crisis in transplants from an AA donor but not constantly from an AS donor. However, the correction of hemolysis requires higher donor chimerism levels (i.e. ≥50%) in both AA and AS recipients. In the future, this group of patients may need a different therapeutic approach.

Metabolic Disorders with Kidney Transplant

Metabolic disorders are highly prevalent in kidney transplant candidates and recipients and can adversely affect post-transplant graft outcomes. Management of diabetes, hyperparathyroidism, and obesity presents distinct opportunities to optimize patients both before and after transplant as well as the ability to track objective data over time to assess a patient’s ability to partner effectively with the health care team and adhere to complex treatment regimens. Optimization of these particular disorders can most dramatically decrease the risk of surgical and cardiovascular complications post-transplant. Approximately 60% of nondiabetic patients experience hyperglycemia in the immediate post-transplant phase. Multiple risk factors have been identified related to development of new onset diabetes after transplant, and it is estimated that upward of 7%–30% of patients will develop new onset diabetes within the first year post-transplant. There are a number of medications studied in the kidney transplant population for diabetes management, and recent data and the risks and benefits of each regimen should be optimized. Secondary hyperparathyroidism occurs in most patients with CKD and can persist after kidney transplant in up to 66% of patients, despite an initial decrease in parathyroid hormone levels. Parathyroidectomy and medical management are the options for treatment of secondary hyperparathyroidism, but there is no randomized, controlled trial providing clear recommendations for optimal management, and patient-specific factors should be considered. Obesity is the most common metabolic disorder affecting the transplant population in both the pre- and post-transplant phases of care. Not only does obesity have associations and interactions with comorbid illnesses, such as diabetes, dyslipidemia, and cardiovascular disease, all of which increase morbidity and mortality post-transplant, but it also is intimately inter-related with access to transplantation for patients with kidney failure. We review these metabolic disorders and their management, including data in patients with kidney transplants.

In Situ Immune Profiling of Heart Transplant Biopsies Improves Diagnostic Accuracy and Rejection Risk Stratification

Recognizing that guideline-directed histologic grading of endomyocardial biopsy tissue samples for rejection surveillance has limited diagnostic accuracy, quantitative, in situ characterization was performed of several important immune cell types in a retrospective cohort of clinical endomyocardial tissue samples. Differences between cases were identified and were grouped by histologic grade versus clinical rejection trajectory, with significantly increased programmed death ligand 1+, forkhead box P3+, and cluster of differentiation 68+ cells suppressed in clinically evident rejections, especially cases with marked clinical-histologic discordance. Programmed death ligand 1+, forkhead box P3+, and cluster of differentiation 68+ cell proportions are also significantly higher in "never-rejection" when compared with "future-rejection." These findings suggest that in situ immune modulators regulate the severity of cardiac allograft rejection.

Plasma DNA End-Motif Profiling as a Fragmentomic Marker in Cancer, Pregnancy, and Transplantation [Research Briefs]

Plasma DNA fragmentomics is an emerging area of research covering plasma DNA sizes, end points, and nucleosome footprints. In the present study, we found a significant increase in the diversity of plasma DNA end motifs in patients with hepatocellular carcinoma (HCC). Compared with patients without HCC, patients with HCC showed a preferential pattern of 4-mer end motifs. In particular, the abundance of plasma DNA motif CCCA was much lower in patients with HCC than in subjects without HCC. The aberrant end motifs were also observed in patients with other cancer types, including colorectal cancer, lung cancer, nasopharyngeal carcinoma, and head and neck squamous cell carcinoma. We further observed that the profile of plasma DNA end motifs originating from the same organ, such as the liver, placenta, and hematopoietic cells, generally clustered together. The profile of end motifs may therefore serve as a class of biomarkers for liquid biopsy in oncology, noninvasive prenatal testing, and transplantation monitoring.

Significance:

Plasma DNA molecules originating from the liver, HCC and other cancers, placenta, and hematopoietic cells each harbor a set of characteristic plasma DNA end motifs. Such markers carry tissue-of-origin information and represent a new class of biomarkers in the nascent field of fragmentomics.

[PERSPECTIVES] Regulating Preimplantation Genetic Testing across the World: A Comparison of International Policy and Ethical Perspectives

Preimplantation genetic testing (PGT) is a reproductive technology that, in the course of in vitro fertilization (IVF), allows prospective parents to select their future offspring based on genetic characteristics. PGT could be seen as an exercise of reproductive liberty, thus potentially raising significant socioethical and legal controversy. In this review, we examine—from a comparative perspective—variations in policy approaches to the regulation of PGT. We draw on a sample of 19 countries (Australia, Austria, Belgium, Brazil, Canada, China, France, Germany, India, Israel, Italy, Japan, Mexico, Netherlands, Singapore, South Korea, Switzerland, United Kingdom, and the United States) to provide a global landscape of the spectrum of policy and legislative approaches (e.g., restrictive to permissive, public vs. private models). We also explore central socioethical and policy issues and contentious applications, including permissibility criteria (e.g., medical necessity), nonmedical sex selection, and reproductive tourism. Finally, we further outline genetic counseling requirements across policy approaches.

More than 900 COVID-19 cases at Cargill plant, but governments allow it to reopen

Cargill Incorporated is the largest privately held company in the United States, and that means it is essentially a family business. 

You cannot buy Cargill shares on the Toronto, New York or any other stock exchange. The descendants of William Cargill, who founded the company in 1865 as a grain storage operation, own 90 per cent of the company.

But if it is a family business, Cargill is no mom-and-pop operation. 

The company has grown over the past century and a half into a multi-tentacled corporate behemoth, involved in everything from grain to livestock to potash to steel to transport to financial services. In 2018, Cargill and its various subsidiaries reported revenues of over $110 billion

Cargill has operations on five continents, in more than 70 countries, including Canada, and the company's meat-packing plant in High River, Alberta is a tiny piece of that worldwide empire. 

In this country, however, the High River plant has an extremely high profile. It is one of the epicentres of COVID-19 in Canada -- in all of North America, in fact -- with over 900 reported cases out of 2,000 employees. That's almost half the workforce.  

Two people have died in connection with the Cargill outbreak -- one, a plant worker originally from Vietnam; the other, an infected plant worker's father, who had been visiting from the Philippines.

Cargill initially resisted pleas from workers and their union to close the plant, but finally relented, in late April. After only two weeks, it hastily reopened, on Monday, May 4, giving the largely immigrant workforce the Hobson's choice of either going back to a potentially fatal workplace or losing their jobs. 

Neither the workers, nor their union think the plant has become safe. 

The union, the United Food and Commercial Workers (UFCW), has gone to court to force a shutdown, until Cargill can absolutely guarantee safe and healthy conditions for all employees. 

The UFCW does not think the notoriously low-paid plant workers should have to risk their lives to fatten the balance sheet of a U.S.-based transnational corporation that ranks number 15 on the Fortune 500. 

Kenney and Trump on the same wavelength

Alberta Premier Jason Kenney has a different view from that of the union and the workers it represents. 

The premier, and former Harper Conservative government cabinet minister, appropriates a concept meant to describe access to necessary basic foodstuffs we all need for sustenance – food security – and applies it to the much different situation of the High River plant. The Cargill workers have to do their part, the Alberta premier argues, to ensure food security for Canadians. 

The truth is that Canada's food security does not depend on meat from Cargill or any other commercial operation. 

If our local butcher runs out of hamburger for the barbecue, we all have other nutritious options. There are, for instance, the protein-packed pulses -- chickpeas, lentils and the like -- that farmers in Saskatchewan grow in great quantity. 

In the U.S., as in Canada, COVID-19 has been particularly hard on the meat-packing industry, forcing more than 20 plant closures, and causing meat shortages on grocery shelves. Some fast food chains have even had to take hamburgers off the menu. 

Corporate executives in the meat industry told U.S. President Trump that they were reluctant to reopen their U.S.-based plants for fear of lawsuits. The U.S. is a far more litigious country than Canada. 

The president's response was to give the corporations cover, by invoking the U.S. Defense Protection Act (DPA). In effect, the president is forcing the corporations to reopen their plants. 

The purpose of the DPA is to allow a president to harness the resources of private industry to serve public needs in time of war or national emergency. Many have urged Trump to invoke the act to assure production of personal protective equipment for front-line workers during the pandemic, but he has refused. 

Now, Trump is using the extraordinary powers of the DPA to force workers back to dangerous plants, while shielding their bosses from responsibility.

As for the High River Cargill plant workers, they fall under provincial labour jurisdiction. And the Alberta premier has already indicated he will not lift a finger to protect them. But there might be a way that federal authorities could step in.

Jagmeet Singh urges Trudeau government to act

In Canada, it is the federal government that has authority over food safety, and NDP Leader Jagmeet Singh believes the Trudeau team should assertively use that power to protect the Cargill workers.

Singh put the question to Deputy Prime Minister Chrystia Freeland during the House of Commons' weekly face-to-face session on Wednesday, May 6.

"Food safety and worker safety cannot be divorced," Singh told the House. "Will the government ensure that the Cargill workers are in safe work conditions?"

Freeland, in a manner all-too-typical of Liberal politicians, dissimulated, offering sympathy but no action.

"The member opposite is quite right that where the federal government has particular authority in food processing is to guarantee the safety of the foods processed there for Canadians to eat," she said, and then expressed some vague sentiments of concern. "When it comes to Cargill and food processing, I agree with the member opposite that it's something we all need to be particularly concerned about, and we have been."

The NDP leader was not satisfied. 

"Will the government commit to using the authority that it has under food safety to ensure that workers are also safe, because there's no way that food can truly be safe if workers are in dangerous conditions and if workers are contracting COVID-19?" Singh asked, adding: "If workers are dying, the food can't be safe."

Freeland would not budge. The Trudeau government wants to get credit for caring, without pushing the envelope in dealing with the most prickly and confrontational provincial government in the country, Alberta's. 

"I think we all understand there is a very clear difference between the duty to inspect food which is produced and to ensure that that food is safe for Canadians, and even more sacred duty to ensure that workers are working in safe conditions," Freeland answered. "We take both of those extremely seriously and we are aware what falls specifically in our jurisdictions. Having said that, we care very much about all Canadian workers." 

Freeland's assertion that responsibility for the safety of a product that consumers eat does not include making sure a processing plant is not an active breeder of a deadly virus reflects a narrow and limited understanding of the federal role. 

There is no evidence of food borne transmission of COVID-19, or of food packaging carrying the virus, according to authorities in both the U.S. and Canada.

But experts have not always got it right about COVID-19 since the outbreak at the beginning of this year. At this stage, all we know for sure is that there remain many unanswered questions about it.

'The worst company in the world'

What is not in doubt is the kind of company we're dealing with. 

Not too long ago the U.S. environmental organization Mighty Earth undertook a study of the social and environmental impact of Cargill's operations and issued a report they called "The Worst Company in the World."

The report opens by stating "when it comes to addressing the most important problems facing our world, including the destruction of the natural environment, the pollution of our air and water, the warming of the globe, the displacement of Indigenous peoples, child labor, and global poverty, Cargill is not only consistently in last place, but is driving these problems at a scale that dwarfs their closest competitors."

The report details how Cargill has become more powerful than governments and has betrayed repeated promises to adhere to high environmental standards. 

"Nowhere is Cargill's pattern of deception and destruction more apparent than in its participation in the destruction of the lungs of the planet, the world's forests. Despite repeated and highly publicized promises to the contrary, Cargill has continued to bulldoze ancient ecosystems, sometimes within the bounds of lax laws -- and, too often, outside those bounds as well."

With the advent to power of virulently anti-environmental Trump in the U.S. and Jair Bolsonaro in Brazil, there is now virtually no limit, Mighty Earth says, to Cargill's capacity to ravage rainforests, savannahs and other vital habitats. 

Mighty Earth cites many examples. 

One of those is that of "the Gran Chaco, a 110-million-hectare ecosystem spanning Argentina, Bolivia, and Paraguay." 

This ecosystem "is one of the largest remaining continuous tracts of native vegetation in South America, second in size only to the Amazon rainforest. These forests are home to vibrant communities of Indigenous Peoples … who have depended on and coexisted with the Chaco forest for millennia."

Cargill, the report tells us, is now actively endangering both the people and other inhabitants of the Gran Chaco to produce a cash crop -- soy -- that feeds the animals which become Big Macs and Whoppers.

"Once the impenetrable stronghold of creatures like the screaming hairy armadillo, the jaguar, and the giant anteater, Cargill has infiltrated the Gran Chaco, bulldozing and burning to make way for vast fields of genetically modified soy."

Mighty Earth also documents Cargill's use of violence to subdue Indigenous peoples, its exploitative labour practices, including child labour, and its predatory practices that have driven competitors out of certain businesses. 

This is the company that Jason Kenney says must be allowed to operate, uninhibited by health concerns, to assure our food security. 

If you believe that, you might also believe that injecting bleach into your veins can cure COVID-19, or that, as many opinion leaders in the U.S. say, it is necessary to accept that thousands must die in the interests of what they call the economy. 

The owners of Cargill are not personally offering to sacrifice their lives. They are offering their employees' lives instead.

Along with forest scientists like Suzanne Simard and Peter Wohlleben, Gagliano raises profound scientific and philosophical questions about the nature of intelligence and the possibility of “vegetal consciousness.” But what’s unusual about Gagliano is her willingness to talk about her experiences with shamans and traditional healers, along with her use of psychedelics. For someone who’d already received fierce pushback from other scientists, it was hardly a safe career move to reveal her personal experiences in otherworldly realms.

Gagliano considers her explorations in non-Western ways of seeing the world to be part of her scientific work. “Those are important doors that you need to open and you either walk through or you don’t,” she told me. “I simply decided to walk through.” Sometimes, she said, certain plants have given her precise directions on how to conduct her experiments, even telling her which plant to study. But it hasn’t been easy. “Like Alice, [I] found myself tumbling down a rather strange rabbit hole,” she wrote in a 2018 memoir, Thus Spoke the Plant. “I did doubt my own sanity many times, especially when all these odd occurrences started—and yet I know I do not suffer from psychoses.”

Shortly before the COVID-19 lockdown, I talked with Gagliano at Dartmouth College, where she was a visiting scholar. We spoke about her experiments, the new field of plant intelligence, and her own experiences of talking with plants.

You are best known for an experiment with Mimosa pudica, commonly known as the “sensitive plant,” which instantly closes its leaves when it’s touched. Can you describe your experiment?

I built a little contraption that allowed me to drop the plants from a height of maybe 15 centimeters. So it’s not too high. When they fall, they land in a softly padded base. This plant closes its leaves when disturbed, especially if the disturbance is a potential predator. When the leaves are closed, big, spiny, pointy things stick out, so they might deter a predator. In fact, they not only close the leaf, but literally droop, like, “Look, I’m dead. No juice for you here.”

You did this over and over, dropping the plants repeatedly.

Exactly. It makes no sense for a plant or animal to repeat a behavior that is actually useless, so we learn pretty quick that whatever is useless, you don’t do anymore. You’re wasting a lot of energy trying to do something that doesn’t actually help. So, can the plant—in this case, Mimosa—learn not to close the leaves when the potential predator is not real and there are no bad consequences afterward?

After how many drops did they stop closing their leaves?

The test is for a specific type of learning that is called habituation. I decided they would be dropped continuously for 60 times. Then there was a big pause to let them rest and I did it again. But the plants were already re-opening their leaves after the first three to six drops. So within a few minutes, they knew exactly what was going on—like, “Oh my god, this is really annoying but it doesn’t mean anything, so I’m just not going to bother closing. Because when my leaves are open, I can eat light.” So there is a tradeoff between protecting yourself when the threat is real and continuing to feed and grow. I left the plants undisturbed for a month and then came back and repeated the same experiment on those individuals. And they showed they knew exactly what was going on. They were trained.

This is who I am. And nobody has the right to tell me that it’s not real.

You say these plants “understand” and “learn” that there’s no longer a threat. And you’re suggesting they “remember.” You’re not using these words metaphorically. You mean this literally?

Yes, that’s what they’re doing. This is definitely memory. It’s the same kind of experiment we do with a bee or a mouse. So using the words “memory” and “learning” feels totally appropriate. I know that some of my colleagues accuse me of anthropomorphizing, but there is nothing anthropomorphic about this. These are terms that refer to certain processes. Memory and learning are not two separate processes. You can’t learn unless you remember. So if a plant is ticking all the boxes and doing what you would expect a rat or a mouse or a bee to do, then the test is being passed.

Do you think these plants are actually making decisions about whether or not to close their leaves?

This experiment with Mimosa wasn’t designed to test that specific question. But later, I did experiments with other plants, with peas in particular, and yes, there is no doubt the plants make choices in real decision-making. This was tested in the context of a maze, where the test is actually to make a choice between left and right. The choice is based on what you might gain if you choose one side or the other. I did one study with peas that showed the plants can choose the right arm in a maze based on where the sound of water is coming from. Of course, they want water. So they will use the signal to follow that arm of the maze as they try to find the source of water.

So plants can hear water?

Oh, yeah, of course. And I’m not talking about electrical signals. We have also discovered that plants emit their own sounds. The acoustic signal comes out of the plant.

What kind of sounds do they make?

We call them clicks, but this is where language might fail because we are trying to describe something we’re not familiar enough with to create the language that really describes the picture. We worked out that, yes, plants not only produce their own sound, which is amazing, but they are listening to sounds. We are surrounded by sound, so there are studies, like my own study, of plants moving toward certain frequencies and then responding to sounds of potential predators chewing on leaves, which other plants that are not yet threatened can hear. “Oh, that’s a predator chewing on my neighbor’s leaves. I better put my defenses up.” And more recently, there was some work done in Israel on the sound of bees and how flowers prepared themselves and become very nice and sweet, literally, to be more attractive to the bee. So the level of sugars gets increased as a bee passes by.

You are describing a surprising level of sophistication in these plants. Do you have a working definition of “intelligence?”

That’s one of those touchy subjects. I use the Latin etymology of the word and “intelligere” literally means something like “choosing between.” So intelligence really underscores decision-making, learning, memory, choice. As you can imagine, all those words are also loaded. They belong in the cognitive realm. That’s why I define all of this work as “cognitive ecology.”

Do you see parallels between this kind of intelligence in plants and the collective intelligence that we associate with social insects in ant colonies or beehives?

That kind of intelligence might be referred to as “distributed intelligence” or “collective intelligence.” We are testing those questions right now. Plants don’t have neurons. They don’t have a brain, which is often what we assume is the base for all of these behaviors. But like slime molds and other basal animals that don’t have neural systems, they seem to be doing the same things. So the short answer is yes.

What you’re saying is very controversial among scientists. The common criticism of your views is that an organism needs a brain or at least a nervous system to be able to learn or remember. Are you saying neurons are not required for intelligence?

Science is full of assumptions and presuppositions that we don’t question. But who said the brain and the neurons are essential for any form of intelligence or learning or cognition? Who decided that? And when I say neurons and brains are not required, it’s not to say they’re not important. For those organisms like ourselves and many animals who do have neurons and brains, it’s amazing. But if we look at the base of the animal kingdom, sponges don’t have neurons. They look like plants because when they’re adults, they settle on the bottom of the ocean and pretty much just sit there forever. Yet if you look at the sponge’s genome, they have the genetic code for the neural system. It’s almost like from an evolutionary perspective, they simply decided that developing a neural system was not useful. So they went a different way. Why would you invest that energy if you don’t need it? You can achieve the same task in different ways.

Your food is psychedelic. It changes your brain chemistry all the time.

Your critics say these are just automatic adaptive responses. This is not really learning.

You know, they just say plants do not learn and do not remember. Then you do this study and stumble on something that actually shows you otherwise. It’s the job of science to be humble enough to realize that we actually make mistakes in our thinking, but we can correct that. Science grows by correcting and modifying and adjusting what we once thought was the fact. I went and asked, can plants do Pavlovian learning? This is a higher kind of learning, which Pavlov did with his dogs salivating, expecting dinner. Well, it turns out plants actually can do it, but in a plant way. So plants do not salivate and dinner is a different kind of dinner. Can you as a scientist create the space for these other organisms to express their own, in this case, “plantness,” instead of expecting them to become more like you?

There’s an emerging field of what’s called “vegetal consciousness.” Do you think plants have minds?

What is the mind? [Laughs] You see, language is very inadequate at the moment in describing this field. I could ask you the same question in referring to humans. Do you think humans have a mind? And I could answer again, what is the mind? Of course, I have written a paper with the title “The Mind of Plants” and there is a book coming called The Mind of Plants. In this context, language is used to capture aspects of how plants can change their mind, and also whether they have agency. Is there a “person” there? These questions are relevant beyond science because they have ethical repercussions. They demand a change in our social attitude toward the environment. But I already have a problem with the language we are using because the question formulated in that way demands a yes or no answer. And what if the answer cannot be yes or no?

Let me ask the question a different way. Do you think plants have emotional lives? Can they feel pain or joy?

It’s the same question. Where do feelings arise from, and what are feelings? These are yes or no questions, usually. But to me, they are yes and no. It depends on what you mean by “feeling” and “joy.” It also depends on where you are expecting the plant to feel those things, if they do, and how you recognize them in a human way. I mean, plants might have more joy than we do. It’s just that we don’t know because we’re not plants.

We have only talked about this from the scientific perspective, which is the Western view of the world. But I’ve also had a close relationship with plants from a very different perspective, the indigenous world view. Why is that less valuable? And when you actually do explore those perspectives, they require your experience. You can’t just understand them by thinking about them. My own personal experience tells me that plants definitely feel many things. I don’t know if they would use those words to describe joy or sadness, but they are feeling bodies. We are feeling bodies.

You’ve studied with shamans in indigenous cultures and you’ve taken ayahuasca and other psychoactive plants. Why did you seek out those experiences?

I didn’t. They sought me. So I just followed. They just arrived in my life. You know, those are important doors that you need to open and you either walk through or you don’t. I simply decided to walk through. I had this weird series of three dreams while I was in Australia doing my normal life. By the time the third dream came, it was very clear that the people that I was dreaming of were real people. They were waiting somewhere in this reality, in this world. And the next thing, I’m buying a ticket and going to Peru and my partner at the time is looking at me like, “What are you doing?” [laughs] I have no idea, but I need to go. As a scientist, I find this is the most scientific approach that I’ve ever had. It’s like there is something asking a question and is calling you to meet the answer. The answer is already there and is waiting for you, if you are prepared to open the door and cross through. And I did.

What did you do in Peru?

The first time I went, I found this place that was in my dream. It was just exactly the same as what I saw in my dream. It was the same man I saw in my dream, grinning in the same way as he was in my dream. So I just worked with him, trying to learn as much as I could about myself with his support.

This was a local shaman whom you identify as Don M. And there was a particular plant substance, a hallucinogen, that you took.

I did what they call a “dieta,” which is basically a quiet, intense time in isolation that you do on your own in a little hut. You are just relating with the plant that the elder is deciding on. So for me, the plant that I worked with wasn’t by itself a psychedelic in the normal way of thinking about it. But of course, all plants are psychedelic. Even your food is psychedelic because it changes your brain chemistry and your neurobiology all the time you eat. Sugars, almonds, all sorts of neurotransmitters are flying everywhere. So, again, even the idea of what a psychedelic experience is needs to be revised, because a lot of people might think that it’s only about certain plants that they have a very strong, powerful transformation. And I find that all plants are psychedelic. I can sit in my garden. I don’t have to ingest anything and I can feel very altered by that experience.

You’ve said the plant talked to you. Did you actually hear words?

When you’re trying to describe this to people haven’t had the experience, it probably doesn’t make much sense because this kind of knowledge requires your participation. I don’t hear someone talking to me as if from the outside, talking to me in words and sound. But even that is not correct because inside my head it does sound exactly like a conversation. Not only that, but I know it’s not me. There is no way that I would know about some of the information that’s been shared with me.

Are you saying these plants had specific information to tell you about your life and your work?

Yeah, I mean, some of the plants tell me exactly how wrong I was in thinking about my experiments and how I should be doing them to get them to work. And I’m like, “Really?” I’m scribbling down without really understanding. Then I go in the lab and try what they say. And even then, there is a part of me that doesn’t really believe it. For one experiment, the one on the Pavlovian pea, I was trying to address that question the year before with a different plant. I was using sunflowers. And while I was doing my dieta with a different tree back in Peru, the plant just turned up and said, “By the way, not sunflowers, peas.” And I’m like, “what?” People always think that when you have these experiences, you’re supposed to understand the secrets of the universe. No, my plants are usually quite practical. [laughs] And they were right.

Do you think you are really encountering the consciousness of that plant? Maybe your imagination has opened up to see the world in new ways, but it’s all just a projection of your own mind. How do you know you are actually encountering another intelligence?

If you had this experience of connecting with plants the way I have described—and there are plenty of people who have—the experience is so clear that you know that it’s not you; it’s someone else talking. If you haven’t had that experience, then I can totally see it’s like, “No way, it must be your mind that makes it up.” But all I can say is that I have had exchanges with plants who have shared things about topics and asked me to do things that I have really no idea about.

What have plants asked you to do?

I’m not a medical scientist, but I’ve been given information by plants about their medical properties. And these are very specific bits of information. I wrote them in my diary. I would later check and I did find them in the medical literature: “This plant is for this and we know this.” I just didn’t know. So maybe I’m tapping into the collective consciousness.

What do you do with these kinds of personal experiences? You are a scientist who’s been trained to observe and study and measure the physical world. But this is an entirely different kind of reality. Can you reconcile these two different realities?

I think there are some presuppositions that a scientist should just explore the consensus reality that most of us experience in more or less the same way. But I don’t really have a conflict because I find this is just part of experimenting and exploring. If anything, I found that it has enriched and expanded the science I do. This is a work in progress, obviously, but I think I’m getting better at it. And in the writing of my book, which for a scientist was a very scary process because it was laying bare some parts of me that I knew would likely compromise my career forever, it also became liberating because once it was written, now the world knows. And it’s my truth. This is how I operate. This is who I am. And nobody has the right or the authority to tell me that it’s not real.
