PEA on Gold Project in Quebec Due Out This Quarter

The takeout potential for the company's shares is expected to increase over the next two years as derisking continues, noted a CIBC report.

O3 Mining Inc. (TSXV:OIII; OTCQX:OIIIF) updated the timeline for its flagship Marban Alliance gold project in Quebec and closed a small equity financing, reported CIBC analyst Bryce Adams in an Oct. 30 research note.

"With the updated shareholder register and continued derisking of Marban, we expect that the takeout potential for O3 shares increases within the next two years," Adams wrote.

O3 Mining is the third iteration of the successful Osisko Mining Inc. (OSK:TSX) model, focused on acquiring, exploring and developing mineral properties in Canada.

168% Return Implied

The Canadian company was trading at the time of the report at about CA$1.12 per share, and CIBC's target price on it is CA$3 per share, noted Adams. These figures reflect a potential return for investors of 168%.

O3 Mining has an Outperformer rating.

PEA Coming this Quarter

Adams presented O3's timeline for Marban Alliance and noted it aligns with CIBC's projections. The next step is completion of a preliminary economic assessment (PEA), slated for Q4/24, "which we expect will be reported on a standalone basis, with upside from potential toll milling agreements," the analyst wrote. G Mining Services now is the lead consultant on the PEA.

Next, a feasibility study will be done based on the PEA and the 2022 prefeasibility study. Targeted dates are Q1/25 to start it and Q2/25 to finish it.

Also in Q1/25, baseline environmental studies are slated for completion. Impact studies are to be started in Q2/25, and filing is slated for Q1/26.

More Strategic Investments

O3 Mining completed a non-brokered private placement of CA$1.4 million with Sidex LP and NQ Investissement Minier, two mining investment funds sponsored by the Quebec government, reported Adams. Subsequently, O3 closed a follow-up offering of US$76,800 to the company's strategic investor at the same terms.

"We view these as smaller issuances, and after model updates, our net asset value per share estimate is now one penny lower at CA$4.48 per share," Adams wrote.

O3 Mining will use the proceeds to drill at Kinebik, where it continues to consolidate land. This project shares the same formation as Hecla Mining Co.'s (HL:NYSE) Casa Berardi mine and Gold Fields Ltd.'s (GFI:NYSE; GFI:JSE) Windfall project.

Takeout Target Potential

Through its acquisition of Osisko, Gold Fields gained 100% ownership of Windfall (it previously had acquired 50% from Osisko in 2023) and 17% of O3 Mining, Adams pointed out. Gold Fields also unsuccessfully made a bid for Yamana Gold Inc.'s (YRI:TSX; AUY:NYSE; YAU:LSE) interests in the Canadian Malartic mine in Quebec earlier in 2023 and "has indicated further growth interest in Quebec."

"With Measured and Indicated resources of 2,400,000 ounces (2.4 Moz) and Inferred resources of 0.6 Moz at its flagship Marban project and near-term final permitting submission, O3 has above average takeout potential," purported Adams.

Silver Co. Releases High-Grade Results From Golden Triangle Drilling

Dolly Varden Silver Corp. (DV:TSX.V; DOLLF:OTCQX) releases new results from its 2024 drilling program at its Kitsault Valley project in British Columbia's Golden Triangle. One analyst says the company is an "attractive target" for large precious metal producers.

Dolly Varden Silver Corp. (DV:TSX.V; DOLLF:OTCQX) released results from five drill holes from its completed 2024 drilling program at its Kitsault Valley project in British Columbia's Golden Triangle.

In total, the program drilled 69 holes for 31,726 meters — 41 holes totaling more than 15,000 meters at the Dolly Varden area and 28 holes totaling more than 16,000 meters at Homestake Ridge.

Highlights of Monday's release include one hole from the Homestake Silver Deposit that found 12.23 grams per tonne gold (g/t Au) and 84 g/t silver (Ag) over 34.93 meters with high-grade breccia veins that included 166 g/t Au and 675 g/t Ag over 0.97 meters.

"The identification of a gold-rich, wide and high-grade area within the Homestake Silver Deposit is highly encouraging," said Chief Executive Officer Shawn Khunkhun. "Our geological team is encouraged by overlapping mineralizing phases of silver and gold-rich veins and breccias; the deposit remains open for expansion."

Technical Analyst Clive Maund, writing on Sunday, called Dolly Varden "one of the best pure silver companies around."

The stock's "breakout at end of last month/early this month was on big volume means it was genuine," Maund wrote.

The stock is at a "classic buy spot, although we should remain aware that it could zigzag a little lower over the short-term, but that said it looks like a strong buy here."

Highlights From Results

Highlights from the Homestake Silver Deposit include:

  • Hole HR24-432: Mineralized envelope including veins: 8.85 g/t Au and 5 g/t Ag over 48.23 meters, including an internal zone of stronger breccia vein intervals grading 29.24 g/t Au and 16 g/t Ag over 13.94 meters, including one breccia vein grading 701 g/t Au and 184 g/t Ag over 0.54 meters.
  • Hole HR24-435: Mineralized envelope including veins: 4.64 g/t Au and 38 g/t Ag over 100.80 meters, including an internal interval of stronger breccia vein mineralization grading 12.23 g/t Au and 84 g/t Ag over 34.93 meters. High-grade breccia veins include 166 g/t Au and 675 g/t Ag over 0.97 meters.
  • Hole HR24-442: Vein breccia zone: 4.58 g/t Au over 9.95 meters, including 14.96 g/t Au over 1.69 meters.

"Results from the five holes in this release suggest that the plunge of mineralization at Homestake Silver has a similar orientation as the Homestake Main Deposit, located 300 meters to the northwest," the company said in a release. "The average grades within these core areas are higher, on a precious metal silver equivalent basis, than the average grade of the silver deposits at the Dolly Varden property further south, due to the increased gold content at the Homestake Ridge Deposits."

Technical Analyst Clive Maund, writing on Sunday, called Dolly Varden "one of the best pure silver companies around."

Drill holes HR24-442 and HR24-445 are step-outs and encountered the mineralized and altered structural corridor of Homestake Silver, the company said. Drill hole HR24-442 intersected a mineralized vein breccia stockwork zone grading 4.58 g/t Au over 9.95 meters, including individual breccias with stronger pyrite mineralization grading 14.96 g/t Au over 1.69 meters.

The Homestake Ridge deposits are interpreted as structurally controlled, multi-phase epithermal vein stockwork and vein breccia system hosted in Jurassic Hazelton volcanic rocks, Dolly Varden noted. Mineralization consists of pyrite plus galena and sphalerite with visible gold in a breccia matrix within a silica breccia vein system.

"The northwest orientation of the main Homestake structural trend appears to have numerous subparallel internal structures that are interpreted to form the controls for higher grade gold and silver shoots within a broader mineralized envelope at the Homestake Silver deposit," the company said. "The main structural corridor dips steeply to the northeast at Homestake Main and rolls to vertical or steeply southwest at Homestake Silver."

Analyst's Response: 'Boom'

Jeff Valks, Senior Analyst for The Gold Advisor newsletter, reacted to the results with the word "BOOM."

"Dolly Varden Silver reports more high-grade drill results from its 2024 exploration program at the Homestake Silver Deposit in British Columbia's Golden Triangle," he wrote on Monday. "Results from five drill holes have confirmed significant gold and silver mineralization in an area targeted within the plunge of a previously undrilled high-grade zone, signaling potential expansion opportunities."

Jeff Valks, Senior Analyst for The Gold Advisor newsletter, reacted to the results with the word "BOOM."

We look forward to the remaining results." Vaks wrote. "In the meantime, the stock is flat as I write but is up over 35% year-to-date. It's not too late to buy, it's down from its recent spike, and as (editor) Jeff (Clark) has said, this is a core holding for the silver bull market. Use a stink bid if you're looking for shares. Both Jeff and I hold long positions."

Analyst Marcus Giannini of Haywood Capital Markets noted in a recent research note that Dolly Varden continues to "push the margins of known high-grade mineralization" at the project.

Gianini gave the stock a Buy rating with a CA$2.40 per share target price. "We continue to view Dolly's high-grade endowment as an increasingly attractive target for larger North American-focused precious metal producers," he noted.

The Catalyst: Analysts Point to Patience

While it has chased the record highs gold has been setting this year, silver recently broke through US$35 per ounce, reflecting a year-to-date gain of about 47%. It has since settled but held to the "crucial US$32.50 level," according to Christopher Lewis of FX Empire on Monday.

"Keep in mind that this is a market that is extraordinarily volatile and, of course, will continue to be noisy over the next couple of days as we get election results in the United States," Lewis wrote. "And of course, we also get the Federal Reserve interest rate decision on Thursday, both of which could cause chaos."

Lewis said he thinks the "least likely path is lower."

"I still favor an upside move, but I recognize that we are definitely in a little bit of a holding pattern," he wrote. "Having said that, if we do see momentum to the upside, then there's really not a whole lot here that could keep this market from trying to challenge the (US$)35 level again, obviously, a large round psychologically significant figure, but we'll just have to wait and see how that plays out."

The most conductive element in nature, silver is used to coat electrical contacts in computers, phones, cars, and appliances. It's also an important element in solar technology.

Mordor Intelligence noted that the white metal is expected to register a compound annual growth rate (CAGR) of more than 5% between 2024 and 2029.

Newsletter editor Brien Lundin encouraged investors not to get discouraged, as any price drop-off is temporary, he said. He expects the silver price to soar when the U.S. Federal Reserve doubles down on its efforts to get interest rates much lower, he wrote on Oct. 23. [OWNERSHIP_CHART-5439]

Based on silver's charts, Ron Struthers of Struthers Resource Stock Report also predicted a major run-up in the silver price.

"Back in April or early May, I highlighted the breakout from a cup and handle formation and [that] that would lead to a major upside move. This is now confirmed," he wrote on Oct. 23.

Ownership and Share Structure

According to the company's latest corporate presentation, 50% of its stock is held by institutional investors, including Fidelity Management & Research Company LLC, Sprott Asset Management LP, U.S. Global Investors Inc., and Delbrook.

About 41% is with strategic investors, including 17% with Fury Gold Mines, 14% with Hecla, and Eric Sprott owns 10% himself.

The rest, 9%, is with retail and high-net-worth investors.

The company has 301.16 million outstanding shares. Its market cap is CA$380.72 million, and its 52-week trading range is CA$0.62–1.46 per share.

Unpaid Caregivers Were Already Struggling. It's Only Gotten Worse During The Pandemic

The pandemic has taken a massive toll on people's mental health. But a new report by the U.S. Centers for Disease Control and Prevention confirms what many of us are seeing and feeling in our own lives: The impact has been particularly devastating for parents and unpaid caregivers of adults.

Two-thirds of survey respondents who identified as unpaid caregivers said they experienced mental health challenges during the pandemic, such as symptoms of anxiety or depression, or suicidal thoughts.

Only one-third of people with no caregiving responsibilities reported the same symptoms.

Of the more than 10,000 survey respondents, more than 40% identified as being unpaid caregivers.

"What is striking here is just how widespread unpaid caregiving responsibilities are in the population and how much of a burden and a toll these responsibilities" are having, says Shantha Rajaratnam, a co-author of the study and a psychologist at the Turner Institute of Brain and Mental Health at Monash University in Australia.

The study also found that people who care for both children under 18 and adults — many of them part of the sandwich generation — are faring the worst, with 85% of this group experiencing adverse mental health symptoms.

"It's an extremely important study," says psychologist Dolores Gallagher-Thompson, professor emeritus at Stanford University who has researched family caregivers and their challenges.

The study is the first to document the problems caregivers have experienced during COVID-19, she notes, and underscores "the importance of paying attention to caregiver issues, caregiver mental health" and the need for education and resources to better support them.

The contrast between caregivers and others is stark

The study, part of ongoing research by The COVID-19 Outbreak Public Evaluation (COPE) Initiative, is based on surveys conducted in December 2020 and February-March 2021.

More than half of those who identified as caregivers said they had experienced symptoms of anxiety or depression, or of disorders like PTSD related to the stress and trauma of COVID-19.

A significant number of caregivers said they had contemplated suicide. Nearly 40% reported having passive suicidal thoughts, meaning "wishing that they had gone to bed and didn't wake up," says study co-author Mark Czeisler, a graduate student at Monash University and a research trainee at Brigham and Women's Hospital in Boston.

And more than 30% had seriously considered taking their own life — about five times the number of noncaregivers, the study found.

Across the board, mental health impacts have been more severe for people who care for both children and adults. Half of this group said it had seriously considered suicide in the past month.

The pandemic worsened the challenges caregivers face

Even before the pandemic, being an unpaid caregiver was stressful and associated with a higher risk of mental health issues, says Gallagher-Thompson. The COVID-19 pandemic has made things even harder.

For instance, the pandemic has taken away many formal and informal sources of support for caregivers.

That was the case for Dr. Nicole Christian-Brathwaite. She's a Boston-based child psychiatrist and lives with her husband, her mother, her husband's father and two sons, who are 4 and 6.

Before the pandemic, her father-in-law, who has dementia, went to a day program for seniors with cognitive decline. Her mother, a survivor of breast and lung cancers, went to physical therapy twice a week, doctor appointments and met with friends.

When the pandemic hit, they lost those services and social support — at the same time Christian-Brathwaite and her husband began working from home while taking care of their sons and parents.

Life at home became much more complicated. Her sons developed behavioral problems with the transitions and stresses of the pandemic. Her mother struggled with chronic pain, and was hospitalized during the pandemic. And there were days when her father-in-law was confused, disoriented or aggressive.

"Many days I was walking around on edge waiting for something to happen because our entire setup was so very fragile and vulnerable," says Christian-Brathwaite. "It's been exhausting."

And her mental health has suffered. "I certainly was dealing with insomnia," she says. "I was short tempered. I was more irritable. I didn't have the same tolerance for things."

More support needed to help caregivers cope

The new study highlights the extent to which unpaid caregivers have struggled during the pandemic, says Gallagher-Thompson.

"There are some serious issues here that shouldn't be ignored," she says.

And yet caregivers are often ignored by the health system, which is set up to focus only on patients.

"Family members are rarely asked, 'How does this affect you? What is difficult? How can we help you? How can we support you in being able to carry out your role, your tasks, your responsibilities?'" Gallagher-Thompson says.

As the new study shows, support can make a big difference — respondents who could rely on others for help with caregiving had a lower incidence of mental health symptoms.

So it's important to educate and support caregivers. For example, physicians can start by screening their patients' caregivers for mental health symptoms and provide more resources to those who need it, says Gallagher-Thompson.

Christian-Brathwaite hopes the new study will help physicians recognize that family caregivers are just as important to consider while treating patients.

"We really need to take a step back and look at the village that's around them because our patients can't be successful without having the support from family," she says.

To Keep Your Brain Young, Take Some Tips From Our Earliest Ancestors

It's something that many of us reckon with: the sense that we're not quite as sharp as we once were.

I recently turned 42. Having lost my grandfather to Alzheimer's, and with my mom suffering from a similar neurodegenerative disease, I'm very aware of what pathologies might lurk beneath my cranium.

In the absence of a cure for Alzheimer's and other forms of dementia, the most important interventions for upholding brain function are preventivethose that help maintain our most marvelous, mysterious organ.

Based on the science, I take fish oil and broil salmon. I exercise. I try to challenge my cortex to the unfamiliar.

As I wrote my recent book, A History of the Human Brain, which recounts the evolutionary tale of how our brain got here, I began to realize that so many of the same influences that shaped our brain evolution in the first place reflect the very measures we use to preserve our cognitive function today.

Being social, and highly communicative. Exploring creative pursuits. Eating a varied, omnivorous diet low in processed foods. Being physically active.

These traits and behaviors help retrace our past, and, I believe, were instrumental in why we remain on the planet today.

And they all were, at least in part, enabled by our brain.

Social smart alecks finish first

The human saga is riddled with extinctions.

By "human," I don't just mean Homo sapiens, the species we belong to, but any member of the genus Homo. We've gotten used to being the only human species on Earth, but in our not so distant past — probably a few hundred thousand years ago – there were at least nine of us running around.

There was Homo habilis, or the "handy man." And Homo erectus, the first "pitcher." The Denisovans roamed Asia, while the more well-known Neanderthals spread throughout Europe.

But with the exception of Homo sapiens, they're all gone. And there's a good chance it was our fault.

Humans were never the fastest lot on the African plains, and far from the strongest. Cheetahs, leopards and lions held those distinctions. In our lineage, natural selection instead favored wits and wiliness.

Plenty of us became cat food, but those with a slight cognitive edge — especially Homo sapiens — lived on. In our ilk, smarts overcame strength and speed in enabling survival.

Ecology, climate, location and just sheer luck would've played important roles in who persisted or perished as well, as they do for most living beings. But the evolutionary pressure for more complex mental abilities would lead to a massive expansion in our brain's size and neurocircuitry that is surely the paramount reason we dominate the planet like no other species ever has.

Much of this "success," if you can call it that, was due to our social lives.

Primates are communal creatures. Our close monkey and ape cousins are incredibly interactive, grooming each other for hours a day to maintain bonds and relationships. Throw in a few hoots and hollers and you have a pretty complex community of communicating simians.

An active social life is now a known preserver of brain function.

Research shows that social isolation worsens cognitive decline (not to mention mental health, as many of us experienced this past year). Larger social networks and regular social activities are associated with mental preservation and slowed dementia progression.

Entwined in this new social life was an evolutionary pressure that favored innovation. Our eventual ability to generate completely novel thoughts and ideas, and to share those ideas, came to define our genus.

As we hunted and foraged together, and honed stones into hand axes, there was a collective creativity at work that gave us better weapons and tools that enabled more effective food sourcing, and, later, butchering and fire. Effectively sharing these innovations with our peers allowed information to spread faster than ever before - a seed for the larger communities and civilizations to come.

Challenging ourselves to new pursuits and mastering new skills can not only impress peers and ingratiate us to our group, but literally help preserve our brain. New hobbies. New conversations. Learning the banjo. Even playing certain video games and simply driving a new route home from work each day, as neuroscientist David Eagleman does, can keep our function high.

Whether it's honing ancient stone or taking up Sudoku, any pursuit novel and mentally challenging may help keep the neural circuits firing.

We really are what we eat

All the while, as we hunted and crafted in new and communal ways, we had to eat. And we did so with an uniquely adventurous palette.

Homo sapiens is among the most omnivorous species on the planet. Within reason we eat just about anything. Whether it's leaves, meat, fungus, or fruit, we don't discriminate. At some point, one of us even thought it might be a good idea to try the glistening, grey blobs that are oysters - and shellfish are, it turns out, among the healthiest foods for our brain.

The varied human diet is an integral part of our story. As was the near constant physicality required to source it.

On multiple occasions over the past 1 to 2 million years climate changes dried out the African landscape, forcing our ancestors out of the lush forest onto the dangerous, wide-open grasslands. As evolution pressured us to create and commune to help us survive, a diverse diet also supported our eventual global takeover.

Our arboreal past left us forever craving the dangling fruits of the forest, a supreme source of high-calorie sugars that ensured survival. Back then we didn't live long enough to suffer from Type 2 diabetes: if you encountered sweets, you ate them. And today we're stuck with a taste for cookies and candy that, given our longer lifespans, can take its toll on the body and brain.

But humans were just as amenable to dining on the bulbs, rhizomes and tubers of the savanna, especially once fire came along. We eventually became adept scavengers of meat and marrow, the spoils left behind by the big cats, who preferred more nutritive organ meat.

As our whittling improved we developed spears, and learned to trap and hunt the beasts of the plains ourselves. There is also evidence that we learned to access shellfish beds along the African coast and incorporate brain-healthy seafood into our diet.

Studying the health effects of the modern diet is tricky. Dietary studies are notoriously dubious, and often involve countless lifestyle variables that are hard to untangle.

Take blueberries. Multiple studies have linked their consumption with improved brain health. But, presumably, the berry-prone among us are also more likely to eat healthy all around, exercise, and make it to level 5 on their meditation app.

Which is why so many researchers, nutritionists, and nutritional psychiatrists now focus on dietary patterns, like those akin to Mediterranean culinary customs, rather than specific ingredients. Adhering to a Mediterranean diet is linked with preserved cognition; and multiple randomized-controlled trials suggest doing so can lower depression risk.

A similar diversity in our ancestral diet helped early humans endure an ever-shifting climate and times of scarcity. We evolved to subsist and thrive on a wide range of foods, in part because our clever brains allowed us access to them. In turn, a similarly-varied diet (minus submitting to our innate sugar craving of course) is among the best strategies to maintain brain health.

All of our hunting, and foraging, and running away from predators would have required intense physical exertion. This was certainly not unique to humans, but we can't ignore the fact that regular exercise is another effective means of preserving brain health.

Being active improves performance on mental tasks, and may help us better form memories. Long before the Peletons sold out, our brains relied on both mental and physical activity.

But overwhelmingly the evidence points to embracing a collection of lifestyle factors to keep our brain healthy, none of which existed in a Darwinian vacuum.

Finding food was as social an endeavor as it was mental and physical. Our creative brains harnessed information; gossiping, innovating, and cooking our spoils around the campfire.

Researchers are beginning to piece together the complex pathology behind the inevitable decline of the human brain, and despite a parade of failed clinical trials in dementia, there should be promising treatments ahead.

Until then, in thinking about preserving the conscious experience of our world and relationships — and living our longest, happiest lives — look to our past.

He Inherited A Devastating Disease. A CRISPR Gene-Editing Breakthrough Stopped It

Patrick Doherty had always been very active. He trekked the Himalayas and hiked trails in Spain.

But about a year and a half ago, he noticed pins and needles in his fingers and toes. His feet got cold. And then he started getting out of breath any time he walked his dog up the hills of County Donegal in Ireland where he lives.

"I noticed on some of the larger hill climbs I was getting a bit breathless," says Doherty, 65. "So I realized something was wrong."

Doherty found out he had a rare, but devastating inherited disease — known as transthyretin amyloidosis — that had killed his father. A misshapen protein was building up in his body, destroying important tissues, such as nerves in his hands and feet and his heart.

Doherty had watched others get crippled and die difficult deaths from amyloidosis.

"It's terrible prognosis," Doherty says. "This is a condition that deteriorates very rapidly. It's just dreadful."

So Doherty was thrilled when he found out that doctors were testing a new way to try to treat amyloidosis. The approach used a revolutionary gene-editing technique called CRISPR, which allows scientists to make very precise changes in DNA.

"I thought: Fantastic. I jumped at the opportunity," Doherty says.

On Saturday, researchers reported the first data indicating that the experimental treatment worked, causing levels of the destructive protein to plummet in Doherty's body and the bodies of five other patients treated with the approach.

"I feel fantastic," Doherty says. "It's just phenomenal."

The advance is being hailed not just for amyloidosis patients but also as a proof-of-concept that CRISPR could be used to treat many other, much more common diseases. It's a new way of using the innovative technology.

"This is a major milestone for patients," says Jennifer Doudna of the University of California, Berkeley, who shared a Nobel Prize for her work helping develop CRISPR.

"While these are early data, they show us that we can overcome one of the biggest challenges with applying CRISPR clinically so far, which is being able to deliver it systemically and get it to the right place," Doudna says.

CRISPR has already been shown to help patients suffering from the devastating blood disorders sickle cell disease and beta thalassemia. And doctors are trying to use it to treat cancer and to restore vision to people blinded by a rare genetic disorder.

But those experiments involve taking cells out of the body, editing them in the lab, and infusing them back in or injecting CRISPR directly into cells that need fixing.

The study Doherty volunteered for is the first in which doctors are simply infusing the gene-editor directly into patients and letting it find its own way to the right gene in the right cells. In this case, it's cells in the liver making the destructive protein.

"This is the first example in which CRISPR-Cas9 is injected directly into the bloodstream — in other words systemic administration — where we use it as a way to reach a tissue that's far away from the site of injection and very specifically use it to edit disease-causing genes," says John Leonard, the CEO of Intellia Therapeutics, which is sponsoring the study.

Doctors infused billions of microscopic structures known as nanoparticles carrying genetic instructions for the CRISPR gene-editor into four patients in London and two in New Zealand. The nanoparticles were absorbed by their livers, where they unleashed armies of CRISPR gene-editors. The CRISPR editor honed in on the target gene in the liver and sliced it, disabling production of the destructive protein.

Within weeks, the levels of protein causing the disease plummeted. Researchers reported at the Peripheral Nerve Society Annual Meeting and in a paper published in The New England Journal of Medicine.

"It really is exciting," says Dr. Julian Gillmore, who is leading the study at the University College London, Royal Free Hospital.

"This has the potential to completely revolutionize the outcome for these patients who have lived with this disease in their family for many generations. It's decimated some families that I've been looking after. So this is amazing," Gillmore says.

The patients will have to be followed longer, and more patients will have to be treated, to make sure the treatment's safe, and determine how much it's helping, Gillmore stresses. But the approach could help those struck by amyloidosis that isn't inherited, which is a far more common version of the disease, he says.

Moreover, the promising results potentially open the door for using the same approach to treatment of many other, more common diseases for which taking cells out of the body or directly injecting CRISPR isn't realistic, including heart disease, muscular dystrophy and brain diseases such as Alzheimer's.

"This is really opening a new era as we think about gene-editing where we can begin to think about accessing all kinds of different tissue in the body via systemic administration," Leonard says.

Other scientists who are not involved in the research agree.

"This is a wonderful day for the future of gene-editing as a medicine,"
agree Fyodor Urnov, a professor of genetics at the University of California, Berkeley. "We as a species are watching this remarkable new show called: our gene-edited future."

Doherty says he started feeling better within weeks of the treatment and has continued to improve in the weeks since then.

"I definitely feel better," he told NPR. "I'm speaking to you from upstairs in our house. I climbed stairs to get up here. I would have been feeling breathless. I'm thrilled."

A Hospital Charged More Than $700 For Each Push Of Medicine Through Her IV

Claire Lang-Ree was in a lab coat taking a college chemistry class remotely in the kitchen of her Colorado Springs, Colo., home, when a profound pain twisted into her lower abdomen. She called her mom, Jen Lang-Ree, a nurse practitioner who worried it was appendicitis and found a nearby hospital in the family's health insurance network.

After a long wait in the emergency room of Penrose Hospital, Claire received morphine and an anti-nausea medication delivered through an IV. She also underwent a CT scan of her abdomen and a series of tests.

Hospital staffers ruled out appendicitis and surmised Claire was suffering from a ruptured ovarian cyst, which can be a harmless part of the menstrual cycle but can also be problematic and painful. After a few days — and a chemistry exam taken through gritted teeth — the pain went away.

Then the bill came.

Patient: Claire Lang-Ree, a 21-year-old Stanford University student who was living in Colorado for a few months while taking classes remotely. She's insured by Anthem Blue Cross through her mom's work as a pediatric nurse practitioner in Northern California.

Total Bill: $18,735.93, including two $722.50 fees for a nurse to "push" drugs into her IV, a process that takes seconds. Anthem's negotiated charges were $6,999 for the total treatment. Anthem paid $5,578.30, and the Lang-Rees owed $1,270.45 to the hospital, plus additional bills for radiologists and other care. (Claire also anted up a $150 copay at the ER.)

What Gives: As hospitals disaggregate charges for services once included in an ER visit, a hospitalization or a surgical procedure, there has been a proliferation of newfangled fees to increase billing. In the health field, this is called "unbundling." It's analogous to the airlines now charging extra for each checked bag or for an exit row seat. Over time, in the medical industry, this has led to separate fees for ever-smaller components of care. A charge to put medicine into a patient's IV line — a "push fee" — is one of them.

Though the biggest charge on Claire's bill, $9,885.73, was for a CT scan, in many ways Claire and her mom found the push fees most galling. (Note to readers: Scans are frequently many times more expensive when ordered in an ER than in other settings.)

"That was so ridiculous," says Claire, who adds she had previously taken the anti-nausea drug they gave her; it's available in tablet form for the price of a cup of coffee, no IV necessary. "It works really well. Why wasn't that an option?"

In Colorado, the average charge for the code corresponding to Claire's first IV push has nearly tripled since 2014, and the dollars hospitals actually get for the procedure has doubled. In Colorado Springs specifically, the cost for IV pushes rose even more sharply than it did statewide.

A typical nurse in Colorado Springs makes about $35 an hour. At that rate, it would take nearly 21 hours to earn the amount of money Penrose charged for a push of plunger that likely took seconds or at most minutes.

The hospital's charge for just one "IV push" was more than Claire's portion of the monthly rent in the home she shared with roommates. In the end, Anthem did not pay the push fees in its negotiated payment. But claims data shows that in 2020 Penrose typically received upward of $1,000 for the first IV push. And patients who didn't have an insurer to dismiss such charges would be stuck with them. Colorado hospitals on average received $723 for the same code, according to the claims database.

"It's insane the variation that we see in prices, and there's no rhyme or reason," says Cari Frank with the Center for Improving Value in Health Care, a Colorado nonprofit that runs a statewide health care claims database. "It's just that they've been able to negotiate those prices with the insurance company and the insurance company has decided to pay it."

To put the total cost in context, Penrose initially charged more money for Claire's visit than the typical Colorado hospital would have charged for helping someone give birth, according to data published by the Colorado Division of Insurance.

Even with the negotiated rate, "it was only $1,000 less than an average payment for having a baby," Frank says.

In an email statement, Centura said it "conducted a thorough review and determined all charges were accurate" and went on to explain that "an Emergency Room (ER) must be prepared for anything and everything that comes through the doors," requiring highly trained staff, plus equipment and supplies. "All of this adds up to large operating costs and can translate into patient responsibility."

As researchers have found, little stands in the way of hospitals charging through the roof, especially in a place like an emergency room, where a patient has few choices. A report from National Nurses United found that hospital markups have more than doubled since 1999, according to data from the United States Bureau of Labor Statistics. In an email, Anthem called the trend of increasing hospital prices "alarming" and "unsustainable."

But Ge Bai, an associate professor of accounting and health policy at Johns Hopkins University, says when patients see big bills it isn't only the hospital's doing — a lot depends on the insurer, too. For one, the negotiated price depends on the negotiating power of the payer, in this case, Anthem.

"Most insurance companies don't have comparable negotiating or bargaining power with the hospital," said Bai. Prices in a state like Michigan, where Bai said the UAW union covers a big proportion of Michigan patients, will look very different from those in Colorado.

Also, insurers are not the wallet defenders patients might assume them to be.

"In many cases, insurance companies don't negotiate as aggressively as they can, because they earn profit from the percentage of the claims," she says. The more expensive the actual payment is, the more money they get to extract.

Though Anthem negotiated away the push fees, it paid the hospital 30% more than the average Level IV emergency department visit in Colorado that year, and it paid quadruple what Medicare would allow for her CT scan.

Resolution: Claire and her mom decided to fight the bill, writing letters to the hospital and searching for information on what the procedures should have cost. The cost of the IV pushes and CT scan infuriated them — the hospital wanted more than double for a CT than what top-rated hospitals typically charged in 2019.

But the threat of collections wore them out and ultimately they paid their assigned share of the bill — $1,420.45, which was mostly coinsurance.

"Eventually it got to the point where I was like, 'I don't really want to go to collections, because this might ruin my credit score,'" says Claire, who didn't want to graduate from college with dinged credit.

Bai and Frank say the state of Maryland can provide a useful benchmark for medical bills, since it sets the prices that hospitals can charge for each procedure. Data provided by the Maryland Health Care Commission shows that Anthem and Claire paid seven times what she likely would have paid for the CT scan there, and nearly 10 times what they likely would have paid for the emergency department Level IV visit. In Maryland, intravenous pushes typically cost about $200 apiece in 2019. A typical Maryland hospital would have received only about $1,350 from a visit like Claire's, and the Lang-Rees would have been on the hook for about $270.

Claire's pain has come back a few times, but never as bad as that night in Colorado. She has avoided reentering an emergency room since then. After visiting multiple specialists back home in California, she learned she might have had a condition called ovarian torsion.

The Takeaway: Even at an in-network facility and with good insurance, patients can get hurt financially by visiting the ER. A few helpful documents can help guide the way to fighting such charges. The first is an itemized bill.

"I just think it's wrong in the U.S. to charge so much," says Jen Lang-Ree. "It's just a little side passion of mine to look at those and make sure I'm not being scammed."

Bai, of Johns Hopkins, suggests asking for an itemized explanation of benefits from the insurance company, too. That will show what the hospital actually received for each procedure.

Find out if the hospital massively overcharged. The Medicare price lookup tool can be useful for getting a benchmark. And publicly available data on health claims in Colorado and at least 17 other states can help, too.

Vincent Plymell with the Colorado Division of Insurance encourages patients to reach out if something on a bill looks sketchy. "Even if it's not a plan we regulate," he wrote in an email, departments such as his "can always arm the consumer with info."

Finally, make scrutinizing such charges fun. Claire and Jen made bill-fighting their mother-daughter hobby for the winter. They recommend pretzel chips and cocktails to boost the mood.

There are more than 2 million people across the United States who have no option when it comes to health insurance. They're in what's known as the "coverage gap" — they don't qualify for Medicaid in their state, and make too little money to be eligible for subsidized health plans on the Affordable Care Act insurance exchanges.

Briana Wright is one of those people. She's 27, lives near Jackson, Miss., works at McDonalds, and doesn't have health insurance. So to figure out her options when she recently learned she needed to have surgery to remove her gallbladder, she called Health Help Mississippi, a nonprofit that helps people enroll in health insurances.

Because she lives in Mississippi, "I wasn't going to be eligible for Medicaid — because I don't have children [and] I'm not pregnant," she tells NPR. When she had her income checked for Healthcare.gov, it was just shy of the federal poverty line — the minimum to qualify for subsidies. "It was $74 [short]. I was like, oh wow," she says.

Wright's inability to get a subsidized policy on Healthcare.gov is related to how the Affordable Care Act was originally designed. People needing insurance who were above the poverty line were supposed to be funneled via the federal and state insurance exchanges to private policies — with federal subsidies to help make those policies affordable. People who were under the poverty line were to be funneled to a newly-expanded version of Medicaid — the public health insurance program that is jointly funded by states and the federal government. But the Supreme Court made Medicaid expansion essentially optional in 2012, and many Republican-led states declined to expand. Today, there are 12 holdout states that have not expanded Medicaid, and Mississippi is one of them.

So, Wright is still uninsured. Her gallbladder is causing her pain, but she can't afford the surgery without shuffling household bills, and risking leaving something else unpaid. "I'm stressed out about it. I don't know what I'm going to do," she says. "I'm going to just have to pay it out of pocket or get on some payment plan until it all gets paid for."

Hoping to finally find a fix for Wright and the millions like her who are in Medicaid limbo, several teams of Democratic lawmakers have recently been hashing out several options — hoping to build on the momentum of the latest Supreme Court confirmation that the ACA is here to stay.

OPTION 1: Sweet-talk the 12 holdout states

The COVID-19 relief bill passed in March included financial enticements for these 12 states to expand Medicaid. Essentially, the federal government will cover 90% of the costs of the newly eligible population, and an additional 5% of the costs of those already enrolled.

It's a good financial deal. An analysis by the nonprofit Kaiser Family Foundation estimates that the net benefit for these states would be $9.6 billion. But, so far — publicly, at least — no states have indicated they intend to take the federal government up on its offer.

"If that is not getting states to move, then that suggests that the deep root of their hesitation is not about financial constraint," says Jamila Michener, a professor of government at Cornell University and author of the book Fragmented Democracy: Medicaid Federalism And Unequal Politics.

Instead, Michener says, the reluctance among some Republican-led legislatures and governors to expand Medicaid may be a combination of partisan resistance to President Obama's signature health law, and not believing "this kind of government intervention for these groups of people is appropriate."

What's Next: When asked about progress on this front in an April press briefing, Biden's press secretary Jen Psaki said "the President is certainly supportive of — and an advocate for — states expanding Medicaid," but did not answer a follow up about whether the White House was directly reaching out to governors regarding this option.

OPTION 2: Create a federal public option to fill the gap

Some have advocated for circumventing these holdout states and creating a new, standalone federal Medicaid program that people who fall into this coverage gap could join. It would be kind of like a tailored public option just for this group.

This idea was included in Biden's 2022 budget, which says, in part: "In States that have not expanded Medicaid, the President has proposed extending coverage to millions of people by providing premium-free, Medicaid-like coverage through a Federal public option, paired with financial incentives to ensure States maintain their existing expansions."

But it wouldn't be simple. "That can be quite complex — to implement a federal program that's targeted to just these 2.2 million people across a handful of states," says Robin Rudowitz, co-director of the Medicaid program at the Kaiser Family Foundation, who wrote a recent analysis of the policy options.

It also may be a heavy lift, politically, says Michener. "Anything that expanded the footprint of the federal government and its role in subsidizing health care would be especially challenging," she says.

What's next: This idea was raised as a possible solution in a letter last month from Georgia's Democratic senators to Senate leaders, and Sen. Raphael Warnock said this week he plans to introduce legislation soon.

OPTION 3: Get around stubborn states by letting cities expand Medicaid

Instead of centralizing the approach, this next idea goes even more local. The COVER Now Act, introduced by Rep. Lloyd Doggett, D-Texas, would empower local jurisdictions to expand Medicaid. So, if you live in Austin, Texas, maybe you could get Medicaid, even if someone in Lubbock still couldn't.

The political and logistical challenges would be tough, policy analysts say. Logistically, such a plan would require counties and cities to create new infrastructure to run a Medicaid program, Rudowitz notes, and the federal government would have to oversee how well these new local programs complied with all of Medicaid's rules.

"It does not seem feasible politically," Michener says. "The legislators who would have to vote to make this possible would be ceding quite a bit of power to localities." It also might amplify geographic equity concerns, she says. People's access to health insurance would not just "be arbitrarily based on what state you live in — which is the current state of affairs — It's also going to be arbitrary based on what county you live in, based on what city you live in."

What's next: Doggett introduced the bill earlier this month. There's no guarantee it would get a vote on the House floor and — even if it did — it wouldn't survive a likely filibuster in the evenly divided Senate.

OPTION 4: Change the ACA to open up the exchanges

A fourth idea, Rudowitz says, is to change the law to remove the minimum cutoff for the private health insurance exchanges, since "right now, individuals who are below poverty are not eligible for subsidies in the marketplace." With this option, states wouldn't be paying any of the costs, since the federal government pays premium subsidies, Rudowitz says, but "there are issues around beneficiary protections, benefits, out-of-pocket costs."

What's next: This idea hasn't yet been included in any current congressional bills.

Will any of these ideas come to fruition?

Even with a variety of ideas on the table, "there's no slam dunk option, it's a tough policy issue," Rudowitz says. All of these would be complicated to pull off.

It's possible Democrats will include one of these ideas in a reconciliation bill that could pass without the threat of a Republican filibuster. But that bill has yet to be written, and what will be included is anyone's guess.

Even so, Michener says she's glad the discussion of the Medicaid coverage gap is happening, because it's sensitizing the public, as well as people in power, to the problem and potentially changing the political dynamic down the line. "Even in policy areas where you don't have any kind of guaranteed victory, it is often worth fighting the fight," she says. "Politics is a long game."

Hospitals Have Started Posting Their Prices Online. Here's What They Reveal

A colonoscopy might cost you or your insurer a few hundred dollars — or several thousand, depending on which hospital or insurer you use.

Long hidden, such price variations are supposed to be available in stark black and white under a Trump administration price transparency rule that took effect at the start of this year. It requires hospitals to post a range of actual prices — everything from the rates they offer cash-paying customers to costs negotiated with insurers.

Many have complied.

But some hospitals bury the data deep on their websites or have not included all the categories of prices required, according to industry analysts. A sizable minority of hospitals have not disclosed the information at all.

While imperfect and potentially of limited use right now to the average consumer, the disclosures that are available illustrate the huge differences in prices — nationally, regionally and within the same hospital. But they're challenging for consumers and employers to use, giving a boost to a cottage industry that analyzes the data.

While it's still an unanswered question whether price transparency will lead to overall lower prices, KHN took a dive into the initial trove of data to see what it reveals. Here are five takeaways from the newly public data and tips for how you might be able to use it to your benefit

1) As expected, prices are all over the map

The idea behind the requirement to release prices is that the transparency may prompt consumers to shop around, weighing cost and quality. Perhaps they could save a few hundred dollars by getting their surgery or imaging test across town instead of at the nearby clinic or hospital.

Under the Trump-era rule, hospitals must post what they accept from all insurers for thousands of line items, including each drug, procedure or treatment they provide. In addition, hospitals must present this in a format easily readable by computers and include a consumer-friendly separate listing of 300 "shoppable" services, bundling the full price a hospital accepts for a given treatment, such as having a baby or getting a hip replacement.

The negotiated rates now being posted publicly often show an individual hospital accepting a wide range of prices for the same service, depending on the insurer, often based on how much negotiating power each has in a market.

In some cases, the cash-only price is less than what insurers pay. And prices may vary widely within the same city or region.

In Virginia, for example, the average price of a diagnostic colonoscopy is $2,763, but the range across the state is from $208 to $10,563, according to a database aggregated by San Diego-based Turquoise Health, one of the new firms looking to market the data to businesses, while offering some information free of charge to patients.

2) Patients can look up the information, but it's incomplete

Patients can try to find the price information themselves by searching hospital websites, but even locating the correct tab on a hospital's website is tricky.

Typically, consumers don't comparison-shop, preferring to choose convenience or the provider their doctor recommends. A recent Peterson-KFF Health System Tracker brief, for instance, found that 85% of adults said they had not researched online the price of a hospital treatment.

And hospitals say the transparency push alone won't help consumers much, because each patient's situation is different and may vary from the average— and individual deductibles and insurance plans complicate matters.

But if you do want to try, here's one tip: "You can Google the hospital name and the words 'price transparency' and see where that takes you," says Caitlin Sheetz, director and head of analytics at the consulting firm ADVI Health in the Washington, D.C., metro area.

Typing in "MedStar Health hospital transparency," for example, likely points to the MedStar Washington Hospital Center's "price transparency disclosure" page, with a link to its full list of prices, as well as its separate list of 300 shoppable services.

By clicking on the list of shoppable services, consumers can download an Excel file. Searching it for "colonoscopy" pulls up several variations of the procedure, along with prices for different insurers, such as Aetna and Cigna, but a "not available" designation for the cash-only price. The file explains that MedStar does not have a standard cash price but makes determinations case by case.

Performing the same Google search for the nearby Inova health system results in less useful information.

Inova's website links to a long list of thousands of charges, which are not the discounts negotiated by insurers, and the list is not easily searchable. The website advises those who are not Inova patients or who would like to create their own estimate to log into the hospitals' "My Chart" system, but a search on that for "colonoscopy" failed to produce any data.

3) Third-party firms are trying to make searching prices simpler – and cash in

Because of the difficulty of navigating these websites — or locating the negotiated prices once there — some consumers may turn to sites like Turquoise. Another such firm is Health Cost Labs, which will have pricing information for 2,300 hospitals in its database when it goes live July 1.

Doing a similar search for "colonoscopy" on Turquoise shows the prices at MedStar by insurer, but the process is still complicated. First, a consumer must select the "health system" button from the website's menu of options, click on "surgical procedures," then click again on "digestive" to get to it.

There is no similar information for Inova because the hospital has not yet made its data accessible in a computer-friendly format, said Chris Severn, CEO of Turquoise.

Inova spokesperson Tracy Connell said in a written statement that the health system will create personalized estimates for patients and is "currently working to post information on negotiated prices and discounts on services."

Firms like Turquoise and Health Cost Labs aim to sell the data gathered from hospitals nationally to insurers, employers and others. In turn, those groups may use it in negotiations with hospitals over future prices. While that may drive down prices in areas with a lot of competition, it might do the opposite where there are few hospitals to choose from, or in situations where a hospital raises its prices to match competitors.

4) Consumers could use this data to negotiate, especially if they're paying cash

For consumers who go the distance and can find price data from their hospitals, it may prove helpful in certain situations:

  • Patients who are paying cash or who have unmet deductibles may want to compare prices among hospitals to see if driving farther could save them money.
  • Uninsured patients could ask the hospital for the cash price or attempt to negotiate for the lowest amount the facility accepts from insurers.
  • Insured patients who get a bill for out-of-network care may find the information helpful because it could empower them to negotiate a discount off the hospitals' gross charges for that care.

While there's no guarantee of success, "if you are uninsured or out of network, you could point to some of those prices and say, 'That's what I want,'" says Barak Richman, a contract law expert and professor of law at Duke University School of Law.

But the data may not help insured patients who notice their prices are higher than those negotiated by other insurers.

In those cases, legal experts say the insured patients are unlikely to get a bill changed because they have a contract with that insurer, which has negotiated the price with their contracted hospitals.

"Legally, a contract is a contract," says Mark Hall, a health law professor at Wake Forest University.

Richman agrees.

"You can't say, 'Well, you charged that person less,'" he notes, but neither can they say they'll charge you more.

Getting the data, however, relies on the hospital having posted it.

5) Hospitals still aren't really on board

When it comes to compliance, "we're seeing the range of the spectrum," says Jeffrey Leibach, a partner at the consulting firm Guidehouse, which found earlier this year that about 60% of 1,000 hospitals surveyed had posted at least some data, but 30% had reported nothing at all.

Many in the hospital industry have long fought transparency efforts, even filing a lawsuit seeking to block the new rule. The suit was dismissed by a federal judge last year.

They argue the rule is unclear and overly burdensome. Additionally, hospitals haven't wanted their prices exposed, knowing that competitors might then adjust theirs, or health plans could demand lower rates. Conversely, lower-cost hospitals might decide to raise prices to match competitors.

The rule stems from requirements in the Affordable Care Act. The Obama administration required hospitals to post their chargemaster rates, which are less useful because they are generally inflated, hospital-set amounts that are almost never what is actually paid.

Insurers and hospitals are also bracing for next year when even more data is set to come online. Insurers will be required to post negotiated prices for medical care across a broader range of facilities, including clinics and doctors' offices.

In May, the Centers for Medicare & Medicaid Services sent letters to some of the hospitals that have not complied, giving them 90 days to do so or potentially face penalties, including a $300-a-day fine.

"A lot of members say until hospitals are fully compliant, our ability to use the data is limited," says Shawn Gremminger, director of health policy at the Purchaser Business Group on Health, a coalition of large employers.

His group and others have called for increasing the penalty for noncomplying hospitals from $300 a day to $300 a bed per day, so "the fine would be bigger as the hospital gets bigger," Gremminger says. "That's the kind of thing they take seriously."

Already, though, employers or insurers are eyeing the hospital data as leverage in negotiations, says Severn, Turquoise's CEO. Conversely, some employers may use it to fire their insurers if the rates they're paying are substantially more than those agreed to by other carriers.

"It will piss off anyone who is overpaying for health care, which happens for various reasons," he says.

New Report Finds Major US Metro Areas, Greater Los Angeles Among Them, Are More Segregated Now Than 30 Years Ago

Despite the racial reckoning going on in America right now, and despite the fact that attitudes towards race, inclusion and representation are different now than they were 30 years ago, new research from UC Berkeley shows that a large majority of American metro areas are more segregated now than they were in 1990. The new report from Berkeley’s Institute covers a number of topic areas, but among the key findings were from the national segregation report component of the project, which found Los Angeles to be the sixth-most segregated metro area with more than 200,000 people.

Today on AirTalk, we’ll talk with the lead researcher on the new report and a local historian to talk about how we see the findings of the report play out in Southern California.

LA City Council to Vote on New Measure to Restrict Homeless Encampments

The Los Angeles City Council votes Thursday on a proposal to ban sleeping or camping in certain parts of the city, including near schools, parks, libraries, and other “sensitive” facilities like daycares. It would also ban tents and encampments from blocking sidewalks if wheelchair users cannot access them. The motion is a departure from the city’s previous approach to the homelessness crisis.

Council members voted 12 to 3 on Tuesday to pull the draft ordinance out of Homelessness and Poverty Committee, where it had been stuck since November, and directed City Attorney Mike Feuer’s office to draft the new rules. Lawmakers Reach A Bipartisan Agreement On Police Reform

Lawmakers in Washington, D.C., have reached a preliminary, bipartisan agreement on police reform after months of closely watched debate on the topic.

Sens. Tim Scott, R-S.C., and Cory Booker, D-N.J., and Rep. Karen Bass, D-Calif., announced the agreement on Thursday evening.

"After months of working in good faith, we have reached an agreement on a framework addressing the major issues for bipartisan police reform," the lawmakers said in a joint statement.

"There is still more work to be done on the final bill, and nothing is agreed to until everything is agreed to. Over the next few weeks we look forward to continuing our work toward getting a finalized proposal across the finish line."

The exact details of the plan were not immediately clear.

The issue of reforming qualified immunity, to make it easier to sue police officers over allegations of brutality, had been a sticking point in negotiations. The police use of chokeholds was another debated provision.

The effort to reform U.S. policing comes after several years of increasing pressure to better understand and regulate the way officers interact with the communities they patrol.

The high-profile deaths of several Black people — many unarmed — at the hands of police — who have in some notable instances been white — have been the catalyst for the police reform movement.

The Democratic-led House had approved the George Floyd Justice in Policing Act — named after one of those Black people killed by police — in early March, and President Biden had hoped Congress would pass the reform effort by the first anniversary of Floyd's death in late May.

But Bass had said then that getting "a substantive piece of legislation" is "far more important than a specific date."

Floyd's murderer, former Minneapolis police officer Derek Chauvin, is set to be sentenced to prison on Friday.

White House press secretary Jen Psaki said in a statement Thursday that Biden "is grateful to Rep. Bass, Sen. Booker, and Sen. Scott for all of their hard work on police reform, and he looks forward to collaborating with them on the path ahead."

The topic of police reform has divided the nation across party lines, with progressives accusing the right of seeking to maintain an antiquated and all-too-powerful law enforcement apparatus. Conservatives say the left has blamed the actions of some officers on the institution itself, turning the topic of police support and "blue lives" into more ammunition for the ongoing culture war.

We Just Got Our Clearest Picture Yet Of How Biden Won In 2020

We know that President Biden won the 2020 election (regardless of what former President Donald Trump and his allies say). We just haven't had a great picture of how Biden won.

That is until Wednesday, when we got the clearest data yet on how different groups voted, and crucially, how those votes shifted from 2016. The Pew Research Center just released its validated voters' report, considered a more accurate measure of the electorate than exit polls, which have the potential for significant inaccuracies.

The new Pew data shows that shifts among suburban voters, white men and independents helped Biden win in November, even while white women and Hispanics swung toward Trump from 2016 to 2020.

To compile the data, Pew matches up survey respondents with state voter records. Those voter files do not say how a person voted, but they do allow researchers to be sure that a person voted, period. That helps with accuracy, eliminating the possibility of survey respondents overreporting their voting activity. In addition, the Pew study uses large samples of Americans — more than 11,000 people in 2020.

It's a numbers-packed report, but there are some big takeaways about what happened in 2020 (and what it might tell us about 2022 and beyond):

Suburban voters (especially white suburban voters) swung toward Biden

Suburban voters appear to have been a major factor helping Biden win. While Pew found Trump winning the suburbs by 2 points in 2016, Biden won them by 11 points in 2020, a 13-point overall swing. Considering that the suburbs accounted for just over half of all voters, it was a big demographic win for Biden.

That said, Trump gained in both rural and urban areas. He won 65% of rural voters, a 6-point jump from 2016. And while cities were still majority-Democratic, his support there jumped by 9 points, to 33%.

Men (especially white men) swung toward Biden

In 2020, men were nearly evenly split, with 48% choosing Biden to Trump's 50%. That gap shrank considerably from 2016, when Trump won men by 11 points. In addition, this group that swung away from Trump grew as a share of the electorate from 2016 — signaling that in a year with high turnout, men's turnout grew more.

White men were a big part of the swing toward Biden. In 2016, Trump won white men by 30 points. In 2020, he won them again, but by a substantially slimmer 17 points.

In addition, Biden made significant gains among married men and college-educated men. All of these groups overlap, but they help paint a more detailed portrait of the type of men who might have shifted or newly participated in 2020.

However, we can't know from this data what exactly was behind these shifts among men — for example, exactly what share of men might have sat on the sidelines in 2016, as opposed to 2020.

Women (especially white women) swung toward Trump

The idea that a majority of white women voted for Trump quickly became one of the 2016 election's most-cited statistics, as many Hillary Clinton supporters — particularly women — were outraged to see other women support Trump.

While that statistic was repeated over and over, Pew's data ultimately said this wasn't true — they found that in 2016, white women were split 47% to 45%, slightly in Trump's favor but not a majority.

This year, however, it appears that Trump did win a majority of white women. Pew found that 53% of white women chose Trump this year, up by 6 points from 2016.

This support contributed to an overall shift in women's numbers — while Clinton won women of all races by 15 points in 2016, Biden won them by 11 points in 2020. Combined with men's shifts described above, it shrank 2016's historic gender gap.

Notably, the swing in white women's margin (5 points altogether) was significantly smaller than white men's swing toward Biden (13 points altogether).

Hispanic voters swung toward Trump

Trump won 38% of Hispanic voters in 2020, according to Pew, up from 28% in 2016.

That 38% would put Trump near George W. Bush's 40% from 2004 — a recent high-water mark for Republicans with Hispanic voters. That share fell off substantially after 2004, leading some Republican pollsters and strategists to wonder how the party could regain that ground. Trump in 2016 intensified those fears, with his nativist rhetoric and hard-line immigration policies.

There are some important nuances to these Hispanic numbers. Perhaps most notably, there is a sizable education gap. Biden won college-educated Hispanic voters by 39 points, but the Democrat won those with some college education or less by 14 points.

That gap mirrors the education gap regularly seen in the broader voting population.

Unfortunately, Pew's sample sizes from 2016 weren't big enough to break down Hispanic voters by gender that year, so it's impossible to see if this group's gender gap widened.

Nonwhite voters leaned heavily toward Biden

Unlike white and Hispanic voters, Black voters didn't shift significantly from 2016. They remained Democratic stalwarts, with 92% choosing Biden — barely changed from four years earlier.

Nearly three-quarters of Asian voters also voted for Biden, along with 6 in 10 Hispanic voters and 56% of voters who chose "other" as their race. (Those groups' sample sizes also weren't big enough in 2016 to draw a comparison over time.)

2018 trends stuck around ... but diminished

In many of these cases where there were substantial shifts in how different groups voted, they weren't surprising, given how voters in the last midterms voted. For example, white men voted more for Democrats in 2018 than they did in 2016, as did suburban voters.

What it means for 2022

The data signals that Democrats' strength with Hispanic voters has eroded, but that the party succeeded in making further inroads in the suburbs, including among suburban whites.

It suggests that these groups, already major focuses for both parties, will continue to be so in 2022, with Republicans trying to cement their gains among Hispanics (and regain suburban voters), while Democrats do Hispanic outreach and try to hold onto the suburbs.

However, it's hard to project much into the future about what voters will do based on the past two elections because of their unique turnout numbers.

"It's hard to interpret here, because 2018 was such a high turnout midterm election, and then our last data point, 2014, was a historically low turnout midterm election," said Ruth Igielnik, senior researcher at Pew Research Center.

In the past couple of weeks, we’ve lost several industry icons, including Paul Mooney, Charles Grodin and Norman Lloyd. 

Actor and comedian Paul Mooney was a boundary-pushing comedian who was Richard Pryor’s longtime writing partner and whose bold, incisive musings on racism and American life made him a revered figure in stand-up. He was 79. Charles Grodin was an offbeat actor and writer who scored as a caddish newlywed in “The Heartbreak Kid” and later had roles ranging from Robert De Niro’s counterpart in the comic thriller “Midnight Run” to the bedeviled father in the “Beethoven” comedies. He was 86. Norman Lloyd’s role as kindly Dr. Daniel Auschlander on TV’s “St. Elsewhere” was a single chapter in a distinguished stage and screen career that put him in the company of Orson Welles, Alfred Hitchcock, Charlie Chaplin and other greats. He was 106. Lloyd’s son, Michael Lloyd, said his father died at his home in the Brentwood neighborhood of Los Angeles. Today on FilmWeek, our critics reflect on their work. Plus they share a couple of their favorite films of the 2021 so far.

Alamo Drafthouse Founder On The Return Of Cinema, Movie Going In A Streaming Era And More

Movie theaters are starting to reopen, and moviegoers are starting to return. All eight of the Laemmle’s theaters are now reopened, its Glendale location the last to do so a couple weeks ago. Tickets are now on sale for the first time in a year at American Cinematheque's Aero theater. 

Last weekend, “A Quiet Place: Part II” opened with very strong box office grosses. And one of the locations that sold a lot of tickets for the sequel was the Alamo Drafthouse in downtown Los Angeles. The Texas-based boutique chain filed for bankruptcy reorganization in early March. Unlike the Arclight and Pacfic theaters, Alamo was able to come back quickly with many of its theaters reopening in May.

KPCC’s John Horn called up Tim League, Alamo’s founder and executive chairman, to talk about his circuit’s return, the future of moviegoing in a streaming era, and whether or not Alamo might be a buyer of the closed Cinerama Dome in Hollywood.

Wearable sensors detect what's in your sweat

Needle pricks not your thing? A team of National Science Foundation-funded scientists is developing wearable skin sensors that can detect what's in your sweat. They hope that one day, monitoring perspiration could bypass the need for more invasive procedures like blood draws, and provide real-time updates on health problems such as dehydration or fatigue. In a new paper, the team describes a new sensor design that can be rapidly manufactured using a "roll-to-roll" processing technique that essentially prints the sensors onto a sheet of plastic like words on a newspaper. They used the sensors to monitor the sweat rate, and the electrolytes and metabolites in sweat, from volunteers who were exercising, and others who were experiencing chemically induced perspiration. The new sensors contain a spiraling microscopic tube, or microfluidic, that wicks sweat from the skin. By tracking how fast the sweat moves through the microfluidic, the sensors can report how much a person is sweating, or their sweat rate. The microfluidics are also outfitted with chemical sensors that can detect concentrations of electrolytes like potassium and sodium, and metabolites like glucose.

Earth's last magnetic field reversal took far longer than once thought

Earth's magnetic field seems steady and true -- reliable enough to navigate by. Yet, largely hidden from daily life, the field drifts, waxes and wanes. The magnetic North Pole is currently shifting toward Siberia, forcing the Global Positioning System that underlies modern navigation to update its software sooner than expected. Every several hundred thousand years, the magnetic field dramatically shifts and reverses its polarity. Magnetic north flips to the geographic South Pole and, eventually, back again. This reversal has happened countless times over Earth's history, but scientists' understanding of why and how the field reverses is limited. The researchers find that the most recent field reversal 770,000 years ago took at least 22,000 years to complete, several times longer than previously thought. The results call into question controversial findings that some reversals could occur within a human lifetime.

Study finds big increase in ocean carbon dioxide absorption along West Antarctic Peninsula

A new study shows that the West Antarctic Peninsula is experiencing some of the most rapid climate change on Earth, featuring dramatic increases in temperatures, retreats in glaciers and declines in sea ice. The Southern Ocean absorbs nearly half of the carbon dioxide -- the key greenhouse gas linked to climate change -- that is absorbed by all the world's oceans. The study tapped an unprecedented 25 years of oceanographic measurements in the Southern Ocean and highlights the need for more monitoring in the region. The research revealed that carbon dioxide absorption by surface waters off the West Antarctic Peninsula is linked to the stability of the upper ocean, along with the amount and type of algae present. A stable upper ocean provides algae with ideal growing conditions. During photosynthesis, algae remove carbon dioxide from the surface ocean, which in turn draws carbon dioxide out of the atmosphere. From 1993 to 2017, changes in sea ice dynamics off the West Antarctic Peninsula stabilized the upper ocean, resulting in greater algal concentrations and a shift in the mix of algal species. That's led to a nearly five-fold increase in carbon dioxide absorption during the summertime. The research also found a strong north-south difference in the trend of carbon dioxide absorption. The southern portion of the peninsula, which to date has been less impacted by climate change, experienced the most dramatic increase in carbon dioxide absorption, demonstrating the poleward progression of climate change in the region.

New way for bridges to withstand earthquakes: Support column design

Bridges make travel faster and more convenient, but, in an earthquake, these structures are subject to forces that can cause extensive damage and make them unsafe. Now civil and environmental engineer Petros Sideris of Texas A&M University is leading a National Science Foundation (NSF)-funded research project to investigate the performance of hybrid sliding-rocking (HSR) columns. HSR columns provide the same support as conventional bridge infrastructure columns but are more earthquake-resistant. HSR columns are a series of individual concrete segments held together by steel cables that allow for controlled sliding and rocking. This allows the columns to shift without damage, while post-tensioning strands ensure that at the end of an earthquake the columns are pushed back to their original position. Conventional bridges are cast-in-place monolithic concrete elements that are strong but inflexible. Structural damage in these bridge columns, typically caused by a natural disaster, often forces a bridge to close until repairs are completed. But bridges with HSR columns can withstand large earthquakes with minimal damage and require minor repairs, likely without bridge closures. Such infrastructure helps with post-disaster response and recovery and can save thousands in taxpayer dollars. In an earthquake, HSR columns provide "multiple advantages to the public," Sideris said. "By preventing bridge damage, we can maintain access to affected areas immediately after an event for response teams to be easily deployed, and help affected communities recover faster. In mitigating losses related to post-event bridge repairs and bridge closures, more funds can be potentially directed to supporting the recovery of the affected communities." According to Joy Pauschke, NSF program director for natural hazards engineering, "NSF invests in fundamental engineering research so that, in the future, the nation's infrastructure can be more resilient to earthquakes, hurricanes, and other forces of nature."

Silver Break Out Confirmed

The silver break out is confirmed, and Ron Struthers of Struthers Resource Stock Report expects a move to $50. He explains why he believes Coeur Mining Inc. (CDE:NYSE) offers good value and shares one biotech stock he believes is currently a sell.

Silver is up again today, currently $34.72 up about $0.64.

This confirms yesterdays breakout and if you remember back in April or early May, I highlighted the breakout from a cup and handle formation and that would lead to a major upside move. This is not confirmed and I see $50 as the near term target.

Similar to gold, investor participation is still quite low. Volumes into the silver etf SLF are up some but no where near 2020 volumes. There are all kinds of silver bullion available at the coin dealer I use.

Our silver stocks are not dragging down the average performance of our gold stocks as much now, and I would like to add another one to the list.

Coeur Mining

Shares Outstanding - 399 million

Coeur Mining Inc. (CDE:NYSE) has been a laggard in this bull rally thus far because it is not well understood. Investors seem to remember more of their legacy than who they are today. Many investors know Coeur as a silver company, but for many years now, most of their revenue and profits have come from gold. Around $7.20, the stock is well below its 2021 highs of around $11.50 and 2016 highs of $16

In Q2 2024, gold sales were $154.1 million, and silver sales were $67.9 million. This makes gold sales almost 70% of revenues. The stock should have responded more to the rising gold price, but as I said, I think investors were still viewing Coeur as mostly a silver company.

That said, they do have large leverage to silver because their resource base they are almost 60% silver. The company is maintaining its full-year production guidance ranges of 310,000 - 355,000 gold ounces and 10.7 - 13.3 million silver ounces. Full-year CAS guidance at Palmarejo and Wharf has been reduced to reflect strong cost management efforts, while Rochester's second-half CAS guidance ranges have been increased to reflect the timing of ounces placed under leach.

Other significant news on the silver front was just a couple of weeks ago, on October 4. Coeeur announced that they entered into a definitive agreement to acquire all of the issued and outstanding shares of SilverCrest pursuant to a court-approved plan of arrangement.

Under the terms of the Agreement, SilverCrest shareholders will receive 1.6022 Coeur common shares for each SilverCrest common share. The Exchange Ratio implies a consideration of $11.34 per SilverCrest common share, based on the closing price of Coeur common shares on the New York Stock Exchange ("NYSE") on October 3, 2024. This represents an 18% premium based on 20-day volume-weighted average prices of Coeur and SilverCrest each as of October 3, 2024.

It turns out, this was a very well timed acquisition ahead of the silver price rise and it will make Coeur the world's largest pure silver producer at about 21 million ounces per year. Their silver production should be neck and neck or just a little behind Pan American Silver not shown on this graphic.

Another key positive fundamental is the expansion of their Rochester Mine this year. In mid-September, they announced that the new three-stage crushing circuit continues to deliver greatly enhanced levels of flexibility to accommodate the full range of mined ore in Rochester.

For the month of August, approximately 2.7 million tons were placed on the new Stage VI leach pad, representing a 39% increase over July placement levels. Rochester remains on track to place 7.0 – 8.0 million tons per quarter during the second half of 2024 and to achieve its full-year 2024 production guidance of 4.8 – 6.6 million ounces of silver and 37,000 – 50,000 ounces of gold.

Rochester is the largest open pit heap leach operation in North America and the largest silver reserve asset in the U.S.

At the end of 2023, Coeur had 3.2 million ounces of proven and probable gold reserves and 243.9 million ounces of silver. At the long-term reference of 60 to 1 ratio, their reserves are 58% silver and 42% gold.

With the significant expansion at Rochester and the acquisition of SilverCrest it will make a significant positive effect to increased profits and cash flow. According to Coeur's presentation, using Factset Street Research data, they will be the leader among peers. The Pro Forma adding SilverCrest is significant. The higher silver price is more gravy on top.

The chart looks good, too. Volume is picking up, and it looks like the stock will break through the resistance area and head to $11.

You can get some more leverage with Call options. Because this is a low-priced stock, I would take advantage of low premiums on long dates. The December 2025 $5.50 Call option is about $2.75 and is $1.87 in the money, so a premium of less than $1.00 for almost 14 months. Nektar Therapeutics (NKTR:NASDAQ)

Opinion - Sell

There is nothing wrong with Nektar Therapeutics (NKTR:NASDAQ), but I am concerned we could get a significant market correction, and I don't like the fact the stock has not done better in a bullish market.

That said, I think this reflects how concentrated this bull market is and does not have good breadth.

The stock is just below cash value but the stock is near resistance on the chart and besides that we have over 100% profits in about 8 months, lets take them.

Regenerative Medicine Co. May Have Solution to Delivering Cell Treatments

Sernova Corp. (SVA:TSX.V; SEOVF:OTCQB; PSH:XERTA) and its Cell Pouch technology could be the solution to existing challenges involving the delivery of medical treatments to patients, such as the ones described here.

Diabetic patients in resource-limited settings are having to revert back to one of the less favored, alternative ways to take insulin, via syringes or glass vials, because Danish pharmaceutical company, Novo Nordisk A/S (NVO:NYSE), will stop making its insulin pens, The Guardian reported. Patients generally prefer this method for dosing themselves with insulin, as shown in a 2024 survey, because it is more convenient and more accurate.

Type 1 diabetic patients already are being impacted as Novo stopped supplying its insulin pens to certain regions, South Africa for instance. Patients there have switched back to using glass vials.

In a second situation, Novo Nordisk is working to bring stem cell-based therapies to patients more efficiently and, in seeking a solution, formed a partnership with Evotec SE (EVO:NASDAQ) to develop technologies that will achieve this, noted Evotec is a Germany-based global biotech firm with its own cell therapy and partnered cell types all in preclinical development for various indications, including diabetes, oncology, cardiology, and ophthalmology.

Per the agreement, Novo Nordisk is to provide research and development funding and potentially monetary incentives to Evotec, and Evotec is to develop the desired new technologies. Novo has the option to obtain exclusive rights to use, in a predefined medical indication, the product(s) born out of this collaboration agreement. Novo's areas of focus, along with diabetes, are cardiovascular diseases, rare diseases, growth hormone-related diseases, hemophilia, nonalcoholic steatohepatitis, and weight management.

Safe, Effective Therapeutic Cell Delivery

Sernova Corp.'s Cell Pouch is a vehicle for delivering various types of therapeutic cells to patients, such as donor islet cells to insulin-dependent diabetics.

When used, the Cell Pouch's containment channels are filled with the appropriate therapeutic cells, and then the device is implanted in the patient. In situ, the cells release therapeutic proteins or hormones the patient's body completely or partially lacks. The device creates a vascularized, organ-like environment that protects the therapeutic cells from immune system attacks, keeping them alive and functioning.

"The Cell Pouch is the most advanced encapsulation device in development," Ventum Capital Markets Analyst Stefan Quenneville wrote in a Sept. 12 research report.

Sernova is testing its Cell Pouch in the clinic, specifically in Type 1 diabetes. In its ongoing Phase 1/2 study, the Canadian company is evaluating the treatment of insulin-dependent diabetes with donor islets implanted via the Cell Pouch, with added immunosuppression therapy. Study data so far have shown the Cell Pouch to be safe and well tolerated and the treatment, effective, reported Dr. Joseph Pantginis, analyst at H.C. Wainwright & Co., in a Sept. 12 research report.

Seven patients, all six of Cohort A and one in Cohort B, achieved sustained insulin independence, between 5.5 and 50 months in duration, free of hypoglycemic episodes. Their blood sugar levels were controlled in the nondiabetic range (i.e.,) HbA1c less than 6.5%.

"The Cell Pouch is the most advanced encapsulation device in development," Ventum Capital Markets Analyst Stefan Quenneville wrote.

A Cell Pouch removed from one of the study patients showed it still contained functioning insulin, glucagon, and somatostatin-producing cells. No evidence was seen of detrimental fibrotic tissue, too many T-cells, material degradation, or changes in the device architecture.

"We believe the impressive response rates and observed durability support Sernova's strategy and justify further investigation while positioning the technology for potential commercial success," noted Pantginis.

The results add to an expanding collection of evidence that the Cell Pouch is functioning as it should. The data also support the "impressive" results already reported from this study and help derisk future related trials.

"If Sernova is successful in bringing its functional cure for insulin-dependent diabetes to the stage where it can go into commercial production, the global market for it will be massive," wrote Technical Analyst Clive Maund in a Sept. 16 note.

In another of its programs, Sernova, in collaboration with Evotec, is developing an implantable off-the-shelf, induced pluripotent stem cell (iPSC)-based islet replacement therapy, Maund reported.

"This partnership provides Sernova a potentially unlimited supply of insulin-producing cells to treat millions of patients with insulin-dependent diabetes (Type 1 and Type 2)," he added.

Market Growth Predicted to 2030

The global live cell encapsulation market, encompassing drug delivery, regenerative medicine and cell transplantation, is expected to continue growing through at least 2030, according to Grand View Research. The market's value, US$210.7 million in 2022, is forecasted to increase at a 3.97% compound annual growth rate between that year and 2030.

"If Sernova is successful in bringing its functional cure for insulin-dependent diabetes to the stage where it can go into commercial production, the global market for it will be massive," wrote Technical Analyst Clive Maund.

Along with diabetes, live cell encapsulation is being used to treat neurological disorders like Parkinson's disease, The market research firm noted. Further, it has been proven to be a suitable way to deliver treatment for other types of diseases, including cancer, anemia, heart failure and more.

Several factors are expected to keep driving market growth during the forecast period, Grand View noted. A significant one is the increasing use of live cell encapsulation in regenerative medicine to replace disease or damaged tissues. A related contributor is rising public and private funding and investments in cell and gene therapies.

The advantages of live cell encapsulation in controlled drug delivery are boosting the market, too. They include enhanced therapeutic effects, lowered drug dose, reduced cytotoxicity, improved patient convenience and better patient compliance.

Novel new products and technological advancements are expected to add value to the market as well.

The Catalysts: Progress With Programs

Various potential stock-moving events are slated for Sernova, according to its September 2024 Corporate Presentation.

Two catalysts are expected by Sernova in 2025, related to the company's ongoing Phase 1/2 clinical trial in Type 1 diabetes. One is results for the remaining Cohort B patients. The other is commencement of Cohort C, who will receive, along with the islet cells, an optimized immune suppression regimen.

Several analysts are bullish on Sernova. One of them is Loe, who rates it as a Speculative Buy. His price target on the life sciences firm implies a 455% return from its current share price.

Next year, Sernova plans to start a Phase 1/2 trial of the regeneratively produced islet cells to result from its partnership with Evotec, delivered via the Cell Pouch to Type 1 diabetes patients.

Other catalysts are expected to come as a result of Sernova advancing its preclinical programs. One is a personalized treatment with patient corrected cells via Cell Pouch for hypothyroidism. Another is a Cell Pouch-delivered, ex vivo lentiviral factor VIII gene therapy for hemophilia, being developed in partnership with the European Haemacure Consortium.

Also, through partnerships, Sernova is developing technologies that would eliminate the need for concurrent immunosuppression during Cell Pouch-delivered cell treatment, a "blue sky objective," Douglas Loe, a Leede Financial Inc. analyst, noted in a Sept. 12 research report.

"Any advances in this regard could be incorporated into future Cell Pouch studies," he wrote. "We do not consider the need for such therapy to be relevant to Cell Pouch function itself."

Analyst: Company is "Very Undervalued"

Several analysts are bullish on Sernova. One of them is Loe, who rates it as a Speculative Buy. His price target on the life sciences firm implies a 455% return from its current share price.

According to H.C. Wainwright's Pantginis, the deepening responses of Type 1 diabetes patients in its Phase 1/2 trial continue to "crystallize Sernova stock's possible upside." The upside reflected in Pantginis' price target is 2,122%. The analyst recommends the company as a Buy.

Ventum's Quenneville also has a Buy on Sernova, and his target price reflects an 826% return on investment. In his report, the analyst highlighted the impressive efficacy and tolerability of the Cell Pouch up to five years post-implantation, as shown in the Phase 1/2 clinical trial data.

"This represents the longest-lasting implanted encapsulation device containing functioning islets without fibrosis," Quenneville wrote.

According to Technical Analyst Maund, Sernova is "very undervalued here given its huge potential" in the Type 1 diabetes market, as indicated on the stock charts. A total of 2,138,766 special warrants of the company were offered at CA$0.75 per special warrant with proceeds of CA$1.6 million. The company also said 466,666 units of the company were offered at CA$0.75 per unit for proceeds of CA$349,999.50.

The company said it intends to use the proceeds for working capital purposes.

"We are very encouraged by the support from our investors with their overwhelming response to the financing and the subsequent over-subscription," Chief Executive Officer Dr. Essam Hamza said. "This money will help expedite our aggressive growth plans over the next year."

New Blood Cancer Treatment Shows Continued Response

The biotech behind this cell therapy has numerous near-term catalysts related to its pipeline, noted a Wedbush report.

TScan Therapeutics Inc. (TCRX:NASDAQ) Phase 1 ALLOHA study, evaluating its lead therapeutic candidates TSC-100 and TSC-101 in hematologic malignancies, showed patients continuing to have a positive response after one year, reported Wedbush analyst Dr. David Nierengarten in a Nov. 5 research note. TSC-100 and TSC-101 are T-cell receptor-engineered T-cell therapies (TCR-Ts).

"We see a catalyst-rich next few months ahead with data building in prominence on stock impact," Nierengarten wrote.

87% Return Potential

Wedbush has a US$10 per share target price on the Massachusetts-based biotech, trading at the time of the report at about US$5.36 per share, noted the analyst. The difference between these figures implies an 87% return potential for investors.

TScan Therapeutics remains rated Outperform.

Durability of Response Data

Nierengarten presented the clinical trial's latest results. As of the July 8, 2024 data cutoff date, in Phase 1 of ALLOHA, 16 patients with hematologic tumors had been administered TSC-100 or TSC-101, and 11 patients had been given a placebo. Median follow-ups had occurred at 5.8 months and 5.3 months, respectively.

At the time, none of the patients in the treatment arm had had a relapse. In the control arm, however, three, or 27% of, the 11 patients had, and the median time to relapse was 159 days. The analyst explained that this is typical for patients receiving a hematopoietic stem cell transplant after reduced-intensity conditioning.

One year out from treatment, five patients were evaluable, and all remained relapse free and minimal residual disease negative at the time. These data underscore the durability of response to this TCR-T treatment, Nierengarten commented. Its safety profile was shown to be favorable still, with no patients experiencing dose-limiting toxicities or adverse events associated with allogeneic hematopoietic cell transplantation.

"Enrollment continues in dose expansion cohorts, and results could support a registrational trial as early as 2025, pending regulatory feedback," Nierengarten wrote.

On the Horizon

TScan Therapeutics has several catalysts related to its clinical programs on the horizon, which Nierengarten listed.

On Nov. 8 and 9, the company will present preclinical data in the poster sessions at the annual Society for Immunotherapy of Cancer meeting. One poster will show in vitro combinatorial data for T-Plex, TScan's cellular therapy for treating solid tumors. It is comprised of two to three different TCR-Ts that target different tumor antigens on different human leukocyte antigen (HLA) types.

A second poster will detail the expansion of ImmunoBank, the biotech's diverse bank of therapeutic T-cell receptors (TCRs) that recognize diverse targets and are associated with multiple HLA types. The third will depict development of a target agnostic platform to evaluate how TCR-Ts affect primary human tissues.

On Dec. 9, TScan Therapeutics will present updated one-year data from ALLOHA, at the American Society for Hematology Annual Meeting in December.

By year-end, the biotech will announce initial data from administering singleplex therapy, cell therapy engineered using a single TCR, to patients with solid tumors. This treatment is being given to establish safety before administering multiplex therapy, cell therapy engineered from multiple TCRs.

In 2025, TScan Therapeutics will provide long-term duration of response data for multiplex therapy in solid tumors and will potentially commence a registrational trial for TSC-100 and TSC-101.

'I'm Not A Cover Girl': Halima Aden On Why She Decided To Leave A Modeling Career

For Halima Aden, the decision to walk away from a career as the world's first hijab-wearing supermodel was fairly clear cut. She's felt used for so long, she says — by the modeling industry and by UNICEF, the organization she was photographed by as a child in a refugee camp in Kenya and later served as an ambassador for.

Aden has been featured on the covers of Vogue, Elle and Allure magazines. And she walked the runway for Rihanna's Fenty Beauty and Kanye West's Yeezy.

She tells Morning Edition host Rachel Martin she wanted to be a role model for young girls while being true to herself, but she wasn't accomplishing either. Modeling, she realized, was in "direct conflict" with who she is.

"I'm not a cover girl, I'm Halima from Kakuma," she says. "I want to be the reason why girls have confidence within themselves, not the reason for their insecurity."

Aden was raised in the Kakuma refugee camp in northwestern Kenya. She and her family moved to Minnesota in 2004 when she was 7.

It was there her journey as a model began, competing for Miss Minnesota USA in 2016, seeking a scholarship. She finished in the semifinals, and says from there, modeling "fell from the sky" into her lap.


Interview Highlights

You saw [modeling] not just as a chance to wear gorgeous clothes and to have your photo in magazines but also as a way to help people.

Growing up in America, not seeing representation, not seeing anybody who dressed like me look like me, it did make me feel like, wow, what's wrong with me, you know? And I'm sure if I had if I would have had representation growing up, I would have been so much more confident to wear my hijab, to be myself, to be authentic. But to be that person, to grow up and be on the cover of magazines, I've covered everything from Vogue to Allure, some of the biggest publications in fashion. And yet I still couldn't relate personally to my own image because that's not who I really am. That's not how I really dress. That's not how my hijab really looks. And, you know, fashion, it can be a very creative field, and I completely appreciate that. But my hijab was just getting spread so thin that I knew I had to give it all away, give it up. I'm not a cover girl. I am Halima from Kakuma. I want to be the reason why girls have confidence within themselves, not the reason for their insecurity.

When you say your hijab was being kind of styled out of existence, what passed for a hijab as you were walking down those runways?

Everything. Oh, my goodness. I had jeans at one point on my head as a hijab. I had Gucci pants styled as a turban. It just didn't even make sense, and I felt so far removed from the image itself.

During the pandemic you decided to walk away from fashion and UNICEF. Was it a complicated decision?

I'll be honest with you, the feelings that I've had towards the fashion industry and UNICEF, it was just multiplying as the years went on, so it was just festering. You know, because the fashion industry is very known to use these young girls and boys while their young, age 14 to like 24, I think is the average career of a model. And then they just replace them and move on to a newer model. And same with UNICEF. They've been photographing me and using me since the time I was a baby in a refugee camp. I remember getting those headshots taken and it made me feel, it's very dehumanizing. And so I wanted to show UNICEF, too. How does it feel to be used? It's not a good feeling. And so let's stop using people.

What are you going to do [next]?

For me right now, I don't know what's next. And that's OK. That's OK, because I'm young and I have time to figure it out. And I'm grateful. I'm grateful to the people that I've met. I'm grateful to the agents that I worked with. I'm grateful for the experiences I was able to have these last four years. But at the same time, I just am also grateful that I don't have to do that anymore because it was in direct conflict with who I am as an individual, as a human being.

George R.R. Martin Signs New Five Year Deal With HBO And HBO Max

George R.R. Martin has entered into a five year deal with HBO to develop content for both HBO and HBO Max, the network said in a statement on Monday.

The best-selling fantasy author and four-time Emmy award winner, best known for his book series A Song of Ice and Fire and its television adaptation Game of Thrones, is attached to multiple projects in the pipeline for the media company and streaming service.

The Game of Thrones television series followed powerful families aiming for the iron throne in the continent of Westeros. It ran for eight years on HBO, with the final season wrapping in 2019.

Martin has a new drama series in the works, House of the Dragon, which is based on his Fire & Blood book. The Game of Thrones prequel follows House Targaryen and is set 300 years before the events of Game of Thrones.

The show has been given a ten episode order. Paddy Considine, Olivia Cooke, Matt Smith, and Emma D'Arcy have joined the series, with additional cast members to be announced.

The show is co-created by Martin and Ryan Condal, whose prior work includes Colony, Rampage, and Hercules. Condal and Miguel Sapochnik (Game of Thrones, Altered Carbon) serve as showrunners and producers with Martin and Vince Gerardis (Game of Thrones).

Martin is also set to executive produce HBO's Who Fears Death, a fantasy post-apocalyptic series with Tessa Thompson attached to star and adapted from the novel by Nnedi Okorafor; and Roadmarks, an adaptation of the sci-fi novel from Roger Zelazny.

Poetry Challenge: Create A List Poem That Grapples With Rise Of Anti-Asian Racism

Over the years, NPR's poetry community has turned both painful and joyful experiences into magnificent work.

As the world still endures the coronavirus pandemic, the U.S. also grieves over increased violence against Asian Americans and a mass shooting in Georgia that left six women of Asian descent dead.

"Let's be clear: Anti-Asian violence and discrimination are not new. But, this racism seems to be heightened," says Kwame Alexander, NPR's resident poet. "And the onus is not on Asian Americans to figure this out. Frankly, it's on white people, it's on the rest of us — individually, systemically, to talk about it, to pay attention to, advocate against it."

"Between Autumn Equinox and Winter Solstice, Today," by Emily Jungmin Yoon, is a list poem that reflects the coldness of the world and how it wears on us. Yoon is a South Korean-born poet pursuing her Ph.D. in Korean literature at the University of Chicago.

Alexander and Morning Edition's Rachel Martin ask listeners: How do you cope with recent anti-Asian violence and discrimination? Tell us in a list poem.

Your poem doesn't have to rhyme. It just needs to have an ordered list with details that show your state of mind — and must begin with the word "today."

Share your poem through the form below. Then Alexander will take lines from some of your pieces and create a community crowdsourced poem. Alexander and Martin will read it on air, and NPR will publish it online, where contributors will be credited.
