Monocyte Invasion into the Retina Restricts the Regeneration of Neurons from Mu&#x0308;ller Glia

Endogenous reprogramming of glia into neurogenic progenitors holds great promise for neuron restoration therapies. Using lessons from regenerative species, we have developed strategies to stimulate mammalian Müller glia to regenerate neurons in vivo in the adult retina. We have demonstrated that the transcription factor Ascl1 can stimulate Müller glia neurogenesis. However, Ascl1 is only able to reprogram a subset of Müller glia into neurons. We have reported that neuroinflammation from microglia inhibits neurogenesis from Müller glia. Here we found that the peripheral immune response is a barrier to CNS regeneration. We show that monocytes from the peripheral immune system infiltrate the injured retina and negatively influence neurogenesis from Müller glia. Using CCR2 knock-out mice of both sexes, we found that preventing monocyte infiltration improves the neurogenic and proliferative capacity of Müller glia stimulated by Ascl1. Using scRNA-seq analysis, we identified a signaling axis wherein Osteopontin, a cytokine highly expressed by infiltrating immune cells is sufficient to suppress mammalian neurogenesis. This work implicates the response of the peripheral immune system as a barrier to regenerative strategies of the retina.

15 arrested in multi-agency raid on Joo Chiat public entertainment outlets

12 men and three women, aged between 19 and 63, were arrested for various offences during a multi-agency enforcement operation against errant public entertainment outlets on Friday (Nov 8) night. The offences include working without a valid work permit, suspected drink driving, driving without a valid license and insurance, suspected consumption and possession of controlled drugs, and the possession of scheduled weapons.  Members of the media, including AsiaOne, were invited to observe the enforcement operations, led by Bedok Police Division, in Joo Chiat on Friday night.  The enforcement operation is part of their ongoing efforts to clamp down on illicit activities and regulatory offences in the area, said the police in a statement on Wednesday. Upon arriving at the scene just after midnight on Saturday, members of the media were led to the second floor of the first KTV outlet.

"Love Wins" is Bell's first book since his break from Zondervan, the Christian publisher based in Grand Rapids, Michigan, that published Bell's first four books and also publishes the New International Version [NIV] of the Bible, one of the most popular translations of the Bible among evangelicals. Bell's split from Zondervan came in part over this new book. "The break with Zondervan was amicable," Tauber said. "In the end the president of Zondervan made the decision. The proposal came in and they said, 'This proposal doesn't fit in with our mission.'" Zondervan would not discuss its relationship with Bell but released a statement: Zondervan has published four books by Rob Bell, as well as numerous Nooma videos in which Rob was featured. We published these titles because we believed they were consistent with Zondervan's [fooling of Christians - occult] mission statement and publishing philosophy. We still believe these titles are impactful with their message and positive contribution and intend to continue to publish them. Tauber said when he got the call that Bell's new book was up for bid, HarperOne jumped at the chance. "There were at least four or five major publishers that were involved in bidding for this book," he said. *When pressed for financial figures of the deal, he said, "We're talking a six-figure deal for the advance, but I can't say more than that." Athanasius is counted as one of the Great Doctors of the Church in Eastern Orthodoxy where he is also labeled the "Father of Orthodoxy". He is also one of the four Great Doctors of the Church from the East in the Roman Catholic Church. He is renowned in the Protestant churches, who label him "Father of The Canon". Athanasius is venerated as a Christian saint, whose feast day is 2 May in Western Christianity, 15 May in the Coptic Orthodox Church, and 18 January in the other Eastern Orthodox churches. Purpose of Writing: The Book of Malachi is an oracle: The word of the Lord to Israel through Malachi (1:1). This was God's warning through Malachi to tell the people to turn back to God. As the final book of the Old Testament closes, the pronouncement of God's just and the promise of His restoration through the coming Messiah is ringing in the ears of the Israelites. Four hundred years of silence ensues, ending with a similar message from God's next prophet, John the Baptist, proclaiming, "Repent, for the kingdom of heaven is near" (Matthew 3:2). Key Verses: Malachi 1:6, "A son honors his father, and a servant his master. If I am a father, where is the honor due me? If I am a master, where is the respect due me?" says the Lord Almighty. "It is you, O priests, who show contempt for my name." Malachi 3:6-7, "I the Lord do not change. So you, O descendants of Jacob, are not destroyed. Ever since the time of your forefathers you have turned away from my decrees and have not kept them. Return to me, and I will return to you, says the Lord Almighty." Brief Summary: Malachi wrote the words of the Lord to God's chosen people who had gone astray, especially the priests who had turned from the Lord. Priests were not treating the sacrifices they were to make to God seriously. Animals with blemishes were being sacrificed even though the law demanded animals without defect (Deuteronomy 15:21). The men of Judah were dealing with the wives of their youth treacherously and wondering why God would not accept their sacrifices. Also, people were not tithing as they should have been (Leviticus 27:30, 32). But in spite of the people's sin and turning away from God, Malachi reiterates God's love for His people (Malachi 1:1-5) and His promises of a coming Messenger (Malachi 2:17-3:5). Foreshadowings: Malachi 3:1-6 is a prophecy concerning John the Baptist. He was the Messenger of the Lord sent to prepare the way (Matthew 11:10) for the Messiah, Jesus Christ. John preached repentance and baptized in the name of the Lord, thus preparing the way for Jesus' first advent. But the Messenger who comes "suddenly to the Temple" is Christ Himself in His second advent when He comes in power and might (Matthew 24). At that time, He will "purify the sons of Levi" (v. 3), meaning that those who exemplified the Mosaic Law would themselves need purification from sin through the blood of the Savior. Only then will they be able to offer "an offering in righteousness" because it will be the righteousness of Christ imputed to them through faith (2 Corinthians 5:21).

Regenerative Gel Restores Spinal Cord in Mice

This is fantastic news that will hopefully turn into a treatment for people with spinal cord injuries and other nerve injuries.

A self-assembling gel injected at the site of spinal cord injuries in paralysed mice has enabled them to walk again after four weeks.

The gel mimics the matrix that is normally found around cells, providing a scaffold that helps cells to grow. It also provides signals that stimulate nerve regeneration.

Samuel Stupp at Northwestern University in Chicago and his colleagues created a material made of protein units, called monomers, that self-assemble into long chains, called supramolecular fibrils, in water.

When they were injected into the spinal cords of mice that were paralysed in the hind legs, these fibrils formed a gel at the injury site.

The researchers injected 76 paralysed mice with either the fibrils or a sham treatment made of salt solution, a day after the initial injury. They found that the gel enabled paralysed mice to walk by four weeks after the injection, whereas mice given the placebo didn't regain the ability to walk.

The team found that the gel helped regenerate the severed ends of neurons and reduced the amount of scar tissue at the injury site, which usually forms a barrier to regeneration. The gel also enhanced blood vessel growth, which provided more nutrients to the spinal cord cells.
