A method to detect prostate cancer comprising contacting a sample of prostate cells from the patient with a set of detectably labeled probes under hybridization conditions and determining the presence of chromosomal abnormalities in prostate tumor tissue, PIN (intra-epithelial neoplasia), histologically benign tissue and benign prostatic hyperplasia (BPH); a method to combine immunofluorescence and FISH (IF-FISH) to facilitate the assessment of chromosomal abnormalities; a set of probes; and a kit comprising the set of probes and instructions for diagnosing prostate cancer in a patient.

We have discovered a protein in humans, herein referred to as collagen like gene (CLG) product (SEQ ID NOS: 12 and 13), that is expressed in human prostate cancer and breast cancer cell lines but not in normal adult, placenta, lung, liver, skeletal muscle, kidney or pancreas tissues. We have also discovered that the level of CLG mRNA expression correlates positively with the metastatic potential of the cancer cell lines tested.

In sporadic Alzheimer's disease, neurofibrillary lesion formation is preceded by extensive post-translational modification of the microtubule associated protein tau. Immunoassays have been developed recently that detect tau in biological specimens, thus providing a means for pre-mortem diagnosis of Alzheimer's disease, which has remained elusive. These assays have been improved by the analysis of relevant post-translational modifications, such as phosphorylation, however opportunity for improvement remains. The present invention addresses this issue by disclosing synthetic methylated peptides derived from the tau protein of paired helical filaments and non-diseased control brain. Alzheimer's disease specificity is provided by the presence or absence of methyl moieties on lysine residues and differences between mono-, di-, and tri-methylation. The methylated peptide is useful as an antigen and a binding partner for identifying compounds that interact with the peptide and the methylated tau protein, including antibodies that can distinguish non-diseased brain from that affected by Alzheimer's disease. The resulting antibodies are useful diagnostically and therapeutically. The compounds that specifically bind to methylated tau proteins are useful for eliminating abnormally methylated tau.

The present invention provides color-coded accessories such as wristbands, bibs, and other patient wearables, color-coded pillows and pillow cases, and a universal chart wherein the color code is associated with a specific medical diagnosis. These accessories alert healthcare providers to the medical diagnosis of the patient and enhance patient safety by reducing medical errors during administration of treatments and medicines to the patients. These accessories also provide protection to healthcare providers assisting them to identify diagnostic categories and alerting them to take proper precautions with patients, for example those with infectious diseases, thereby decreasing or preventing transmission of infectious diseases.

I have a health condition with a high mortality rate, where about one quarter of us die within the first year, and two-thirds of us don't last five years. I'm on Year 4 now and I'm doing okay, and I'm thankful to be receiving excellent care, and I'm generally optimistic that I'll get to stick around for a while.

Ramping up to my diagnosis, I thought my life was over. And that was both utterly untrue and completely true at the same time. You can't really know what it's like until you have the actual diagnosis, and even then it's been a world of surprises. You may or may not be able to do some of the things you would like to do.

For me to deal with it in a healthy way, I kind of had to create a hard line in the sand. I had to take time to grieve the person I'd been before falling ill, take stock of my accomplishments, and most importantly, I had to REALIZE MY ACCOMPLISHMENTS WERE ENOUGH for my lifetime. If I'd been hit by a bus, my life would have been over in a snap, and whatever I'd accomplished by that point would have had to be enough. Taking that pressure off myself was the kindest thing I could do for myself.

I found I had to let a lot of things go and not compare Previous Me to Sick Me. Previous Me was active, enjoyed travel, able to hold down complex and interesting work. Sick Me can't do much of that, but Sick Me does pretty okay for a sick person, and Sick Me does so much more than Dead Me could possibly do! Seriously, compared to Dead Me, Sick Me is a total winner. Sick Me can do a little modest gardening, enough to keep the weeds away. Sick Me can care for my dog and handle the occasional load of laundry. Sick Me finds a lot of joy in my friends and family and internet communities, and has transferred my social life to text, email, and the occasional dining out when the stars align and energy allows. (Metafilter is a lifesaver because I can pick it up whenever my energy level allows and people are so welcoming and understanding.)

As you're waiting for news, it's easy to fall to worry. Please be kind to yourself and don't suffer those fears and losses before you need to. Right now, you are there for your children. Don't put yourself through the punishment of losing them multiple times unnecessarily. And don't say you won't ever get to do a painting class -- I took my first painting class last fall and it was a boon to my soul and it renewed my capacity for beauty.

In the meantime, take as much control of the situation as you need to. Write down a list of questions to review with your doctors. (I've actually typed them out and distributed copies for them to follow along.) You can google, and it's hard not to, but please never tell a doctor that your questions or concerns are coming from google. Also, do not call yourself a hypochondriac -- what you are feeling is what you are feeling, and your concerns are valid and deserving of respect.

Waiting is hard. Please be extra kind to yourself.

Insulin-like growth factors (IGFs), specifically IGF1 and IGF2, promote glucose metabolism, with their availability regulated by IGF-binding proteins (IGFBPs). We hypothesized that IGF1 and IGF2 levels, or their bioavailability, are reduced during type 1 diabetes development. Total serum IGF1, IGF2, and IGFBP1–7 levels were measured in an age-matched, cross-sectional cohort at varying stages of progression to type 1 diabetes. IGF1 and IGF2 levels were significantly lower in autoantibody (AAb)⁺ compared with AAb^– relatives of subjects with type 1 diabetes. Most high-affinity IGFBPs were unchanged in individuals with pre–type 1 diabetes, suggesting that total IGF levels may reflect bioactivity. We also measured serum IGFs from a cohort of fasted subjects with type 1 diabetes. IGF1 levels significantly decreased with disease duration, in parallel with declining β-cell function. Additionally, plasma IGF levels were assessed in an AAb⁺ cohort monthly for a year. IGF1 and IGF2 showed longitudinal stability in single AAb⁺ subjects, but IGF1 levels decreased over time in subjects with multiple AAb and those who progressed to type 1 diabetes, particularly postdiagnosis. In sum, IGFs are dysregulated both before and after the clinical diagnosis of type 1 diabetes and may serve as novel biomarkers to improve disease prediction.

Lung cancer (LC) remains the leading cause of mortality from malignant tumors worldwide. In our previous study, we surveyed both IgG and IgM-bound serological biomarkers and validated a panel of IgG-bound autoantigens for early LC diagnosis with 50% sensitivity at 90% specificity. To further improve the performance of these serological biomarkers, we surveyed HuProt arrays, comprised of 20,240 human proteins, for IgA-bound autoantigens because IgAs are a major immunoglobulin isotype in the lung. Integrating with IgG-bound autoantigens, we discovered and validated a combined biomarker panel using ELISA-format tests. Specifically, in Phase I, we obtained IgA-based autoimmune profiles of 69 early stage LC patients, 30 healthy subjects and 25 patients with lung benign lesions (LBL) on HuProt arrays and identified 28 proteins as candidate autoantigens that were significantly associated with early stage LC. In Phase II, we re-purified the autoantigens and converted them into an ELISA-format testing to profile an additional large cohort, comprised of 136 early stage LC patients, 58 healthy individuals, and 29 LBL patients. Integration of IgG autoimmune profiles allowed us to identify and validate a biomarker panel of three IgA autoantigens (i.e. BCL7A, and TRIM33 and MTERF4) and three IgG autoantigens (i.e. CTAG1A, DDX4 and MAGEC2) for diagnosis of early stage LC with 73.5% sensitivity at >85% specificity. In Phase III, the performance of this biomarker panel was confirmed with an independent cohort, comprised of 88 early stage LC patients, 18 LBL patients, and 36 healthy subjects. Finally, a blind test on 178 serum samples was conducted to confirm the performance of the biomarker panel. In summary, this study demonstrates for the first time that an integrated panel of IgA/IgG autoantigens can serve as valuable biomarkers to further improve the performance of early diagnosis of LC.

Screening for diabetes is typically done using hemoglobin A_1c (HbA_1c) or fasting plasma glucose (FPG). Polycystic ovary syndrome (PCOS) is a common female reproductive disorder that often manifests during adolescence and is associated with disruptions in health-related quality of life. Prompt evaluation and clinical support after diagnosis may prevent associated complications and optimize overall health management. This article incorporates the most recent evidence and consensus guidelines to provide an updated review of the pathogenesis, clinical presentation, diagnostic evaluation, and management strategies for adolescents with this complex condition. We will review the recent international guidelines on PCOS; because the diagnosis of PCOS remains controversial, management of this condition is inconsistent. In 2019, PCOS remains a common, yet neglected, condition, in part, because of the lack of agreement around both diagnosis and management.

Primary care providers (PCPs) identify patients with undescended testis (UDT) and refer them to surgical specialists. Referral beyond the recommended times for orchiopexy has been reported, and PCPs' accuracy in identifying and distinguishing UDTs from retractile testes has been questioned.

We describe 3 observations that are strongly correlated with UDT, that is, birth history of UDT, prematurity, and visible scrotal asymmetry. UDT diagnoses are best made by 8 months of age, to reduce confusion with testicular retraction and to facilitate timely orchiopexy. (Read the full article)
