Recent investigations underscore the involvement of a small EDRK-rich factor 1a (SERF1a) in promoting Httex1 fibrillization through interactions with its N terminus. By establishing an integrated approach with size-exclusion-column-based small- and wide-angle X-ray scattering (SEC-SWAXS), NMR, and molecular simulations using Rosetta, the analysis here reveals a tight binding of two NT17 fragments of Httex1 (comprising the initial 17 amino acids at the N terminus) to the N-terminal region of SERF1a. In contrast, examination of the complex structure of SERF1a with a coiled NT17-polyQ peptide (33 amino acids in total) indicates sparse contacts of the NT17 and polyQ segments with the N-terminal side of SERF1a. Furthermore, the integrated SEC-SWAXS and molecular-simulation analysis suggests that the coiled NT17 segment can transform into a helical conformation when associated with a polyQ segment exhibiting high helical content. Intriguingly, NT17-polyQ peptides with enhanced secondary structures display diminished interactions with SERF1a. This insight into the conformation-dependent binding of NT17 provides clues to a catalytic association mechanism underlying SERF1a's facilitation of Httext1 fibrillization. Full Article text

CheckMyMetal (CMM) is a web based tool that facilitates the interactive validation of MBS in structures determined through X-ray crystallography and cryo-electron microscopy (cryo-EM). Recent updates to CMM have significantly enhanced its capability to efficiently handle large datasets generated from cryo-EM structural analyses. In this study, we address various challenges inherent in validating MBS within both X-ray and cryo-EM structures. Specifically, we examine the difficulties associated with accurately identifying metals and modeling their coordination environments by considering the ongoing reproducibility challenges in structural biology and the critical importance of well annotated, high-quality experimental data. CMM employs a sophisticated framework of rules rooted in the valence bond theory for MBS validation. We explore how CMM validation parameters correlate with the resolution of experimentally derived structures of macromolecules and their complexes. Additionally, we showcase the practical utility of CMM by analyzing a representative cryo-EM structure. Through a comprehensive examination of experimental data, we demonstrate the capability of CMM to advance MBS characterization and identify potential instances of metal misassignment. ndi X-ray standing wave characterization of the strong metal–support interaction in Co/TiOx model catalysts By journals.iucr.org Published On :: 2024-03-31 The strong metal–support interaction (SMSI) is a phenomenon observed in supported metal catalyst systems in which reducible metal oxide supports can form overlayers over the surface of active metal nanoparticles (NPs) under a hydrogen (H2) environment at elevated temperatures. SMSI has been shown to affect catalyst performance in many reactions by changing the type and number of active sites on the catalyst surface. Laboratory methods for the analysis of SMSI at the nanoparticle-ensemble level are lacking and mostly based on indirect evidence, such as gas chemisorption. Here, we demonstrate the possibility to detect and characterize SMSIs in Co/TiOx model catalysts using the laboratory X-ray standing wave (XSW) technique for a large ensemble of NPs at the bulk scale. We designed a thermally stable MoNx/SiNx periodic multilayer to retain XSW generation after reduction with H2 gas at 600°C. The model catalyst system was synthesized here by deposition of a thin TiOx layer on top of the periodic multilayer, followed by Co NP deposition via spare ablation. A partial encapsulation of Co NPs by TiOx was identified by analyzing the change in Ti atomic distribution. This novel methodological approach can be extended to observe surface restructuring of model catalysts in situ at high temperature (up to 1000°C) and pressure (≤3 mbar), and can also be relevant for fundamental studies in the thermal stability of membranes, as well as metallurgy. ndi Bragg Spot Finder (BSF): a new machine-learning-aided approach to deal with spot finding for rapidly filtering diffraction pattern images By journals.iucr.org Published On :: 2024-04-26 Macromolecular crystallography contributes significantly to understanding diseases and, more importantly, how to treat them by providing atomic resolution 3D structures of proteins. This is achieved by collecting X-ray diffraction images of protein crystals from important biological pathways. Spotfinders are used to detect the presence of crystals with usable data, and the spots from such crystals are the primary data used to solve the relevant structures. Having fast and accurate spot finding is essential, but recent advances in synchrotron beamlines used to generate X-ray diffraction images have brought us to the limits of what the best existing spotfinders can do. This bottleneck must be removed so spotfinder software can keep pace with the X-ray beamline hardware improvements and be able to see the weak or diffuse spots required to solve the most challenging problems encountered when working with diffraction images. In this paper, we first present Bragg Spot Detection (BSD), a large benchmark Bragg spot image dataset that contains 304 images with more than 66 000 spots. We then discuss the open source extensible U-Net-based spotfinder Bragg Spot Finder (BSF), with image pre-processing, a U-Net segmentation backbone, and post-processing that includes artifact removal and watershed segmentation. Finally, we perform experiments on the BSD benchmark and obtain results that are (in terms of accuracy) comparable to or better than those obtained with two popular spotfinder software packages (Dozor and DIALS), demonstrating that this is an appropriate framework to support future extensions and improvements. It includes efficient data collection, streaming of data and metadata to a high-performance cluster (HPC), fast azimuthal data integration on the cluster, and tools for controlling the data processing steps and visualizing the data using the DIOPTAS software package. This data processing pipeline is invaluable for a great number of studies. The potential of the pipeline is illustrated with two examples of data collected on ammonia–water mixtures and multiphase mineral assemblies under high pressure. ndi The AUREX cell: a versatile operando electrochemical cell for studying catalytic materials using X-ray diffraction, total scattering and X-ray absorption spectroscopy under working conditions By journals.iucr.org Published On :: 2024-09-20 Understanding the structure–property relationship in electrocatalysts under working conditions is crucial for the rational design of novel and improved catalytic materials. This paper presents the Aarhus University reactor for electrochemical studies using X-rays (AUREX) operando electrocatalytic flow cell, designed as an easy-to-use versatile setup with a minimal background contribution and a uniform flow field to limit concentration polarization and handle gas formation. The cell has been employed to measure operando total scattering, diffraction and absorption spectroscopy as well as simultaneous combinations thereof on a commercial silver electrocatalyst for proof of concept. This combination of operando techniques allows for monitoring of the short-, medium- and long-range structure under working conditions, including an applied potential, liquid electrolyte and local reaction environment. The structural transformations of the Ag electrocatalyst are monitored with non-negative matrix factorization, linear combination analysis, the Pearson correlation coefficient matrix, and refinements in both real and reciprocal space. Upon application of an oxidative potential in an Ar-saturated aqueous 0.1 M KHCO3/K2CO3 electrolyte, the face-centered cubic (f.c.c.) Ag gradually transforms first to a trigonal Ag2CO3 phase, followed by the formation of a monoclinic Ag2CO3 phase. A reducing potential immediately reverts the structure to the Ag (f.c.c.) phase. Following the electrochemical-reaction-induced phase transitions is of fundamental interest and necessary for understanding and improving the stability of electrocatalysts, and the operando cell proves a versatile setup for probing this. In addition, it is demonstrated that, when studying electrochemical reactions, a high energy or short exposure time is needed to circumvent beam-induced effects. Full Article text

Wortel detailed Golden Cariboo's drilling campaign, which includes results such as Hole QGQ24-013, which intersected 136.51 meters at 1.77 g/t gold, including a higher-grade interval of 23.89 meters at 3.32 g/t gold. Americans are starting to believe a "sense of normal" is approaching fairly soon, however, according to a new NPR/PBS NewsHour/Marist survey. The poll also found that with the coronavirus receding in this country, mask-wearing is declining and Americans are going out more. But they remain cautious about being in large crowds. As the country continues to open up, more focus turns to the economy, which cratered during the beginning of the pandemic last year. And Americans are split by race, gender and politics on whether President Biden's ambitious policies are helping or not. Race, gender, party divides on Biden and the economy Three months ago, in a similar survey, 49% of adults said the president's policies were strengthening the economy, while 44% said they were weakening it. Now, that's declined a net of 6 points, as 44% of respondents in the new poll say Biden's policies have strengthened the economy and 45% say the opposite. The percentage who were unsure also jumped 4 points. It's all a little bit of a warning sign for Biden, as he pushes for two large — and expensive — spending packages. There are significant splits by race and gender: Just 39% of whites said Biden's policies have strengthened the economy, but 52% of people of color say they have. 54% of independent men say his policies have weakened the economy, while 56% of independent women say they've strengthened it. 45% of white male college grads say Biden has strengthened the economy, but a significantly higher 64% of white women with college degrees said so. Inflation vs. wages by party A quarter of Americans rank inflation as the U.S. economy's top concern. That's followed by wages, unemployment, housing costs, labor shortages, gas prices and interest rates. But there's a sharp political divide on the question. Republicans and independents rank inflation as their top concern, while for Democrats, it was wages. Just 4% of Republicans said wages were their top concern. Return to "normal" Americans are growing increasingly optimistic about when life will return to a "sense of normal," as the survey labels it. In April, three-quarters of Americans said they believe it will take six months or more. Now, it's just half. About a quarter (27%) say it will be less than six months, up from 15% two months ago. People are also growing more comfortable doing certain things, saying they're: dining out at restaurants (78%) and visiting unvaccinated friends and family (75%). But they are not as comfortable doing others: almost 7-in-10 are not going out to bars; about two-thirds are not attending live concerts or sporting events (65%); and a majority have also not resumed going to in-person religious services (54%). COVID-19 vaccines and going back to work While half say they are concerned about another coronavirus surge, almost 9-in-10 U.S. adults with jobs say they are at least somewhat comfortable returning to work. Notably, a majority (57%) of those with jobs do not believe employers should require COVID-19 vaccines as a condition to return to in-person work. More than a quarter of Americans say they will not get vaccinated. The most resistant to getting vaccinated continue to be supporters of former President Donald Trump. Half of them say they won't get the shot, the highest of any group surveyed. Trump has touted the vaccine and got it himself. Since Centers for Disease Control and Prevention guidelines came out, noting that Americans who have been vaccinated can largely set masks aside, there's been a double-digit decline in those saying they wear a mask even when it's not required. There's also been a double-digit increase in those saying they generally do not wear a mask. In May, 49% said they wore masks even when it was not required. Now, that's just 36%. One-in-five said they generally do not wear masks. Two months ago, it was less than one-in-10. Affordability, not coronavirus, limiting vacations Speaking of getting back to normal, a majority of Americans say they plan to take a vacation this summer. But of the significant minority (45%) who say they aren't taking one, almost three times as many cited affordability (35%) as the main reason for not going, as opposed to concerns about COVID-19 (12%). Methodology: The poll of 1,115 U.S. adults was conducted using live telephone interviewers from June 22 through June 29. Survey questions were available in English or Spanish. The push is being led by Senators Elizabeth Warren, Democrat of Massachusetts and Bernie Sanders, a Vermont Independent, In the December relief package, Congress increased the number of low-income college students eligible for Supplemental Nutrition Assistance Program (or SNAP) benefits for the duration of the pandemic. That included students who are eligible for work study, have an Expected Family Contribution of zero dollars, or qualify for a maximum Pell Grant on their federal financial aid form. According to The Century Foundation, this expansion affects about 3 million college students. The legislation proposed Tuesday would make these changes permanent, including requiring the U.S. Education Department to notify students they may be eligible for SNAP when they fill out their student aid applications. The bill would also require the Department to collect data on hunger and food insecurity, and would create a $1 billion-a-year grant program for institutions to address hunger on campus. "Far too many college students struggle to meet their basic needs while they get their education and the pandemic has made this problem even worse," Warren said in a statement to NPR. "As students take on a mountain of student loan debt, they shouldn't have to choose between paying tuition and eating." The push comes amid new research that shows 39% of two-year college students are facing food insecurity; for students at 4-year schools the number affected is 29%, according to Temple University's Hope Center for College, Community and Justice. Before the pandemic, in 2019, the U.S. Government Accountability Office issued a report on humger among college students, concluding that over a third of students don't always have enough to eat, and that federal systems already in place, including SNAP, could do a better job of helping them. Eric Holcomb in an attempt to block the state's push to end pandemic unemployment benefits on June 19. Indiana Legal Services, an organization providing free legal assistance, and the Concerned Clergy of Indianapolis filed the lawsuit on behalf of five unnamed plaintiffs who are set to lose their jobless benefits. The complaint was filed Monday in Marion County Superior Court. This lawsuit may be the first of its kind that aims to stop states from ending these benefits earlier than Congress mandated. The unemployment insurance program "has served as a vital lifeline for thousands of Hoosiers," the complaint, reviewed by NPR, says. "By prematurely deciding t0 stop administering these federal benefits, Indiana has violated the clear mandates 0f Indiana's unemployment statute—to secure all rights and benefits available for unemployed individuals." Indiana is one of 25 Republican-led states that decided to end jobless aid in an effort to get people to return to work. Indiana and seven other states are set to end expanded unemployment benefits as soon as this weekend. This is despite Congress's authorization for extra payments until early September. Those benefits include the extra $300 a week in federal aid and the special pandemic program for gig workers that allows them to receive jobless benefits. Ordinarily, independent contractors wouldn't be eligible. Plaintiffs, as well as many other Indiana residents, rely entirely on the unemployment benefits to pay for food and rent and to care for their families, the complaint alleges. Attorneys in this case are requesting the judge approve a preliminary injunction that would allow people to receive their benefits while the case continues. Holcomb says it's time to get back to work Holcomb told The Indianapolis Star that people no longer need unemployment benefits as the state has a plethora of jobs open. "Eliminating these pandemic programs will not be a silver bullet for employers to find employees, but we currently have about 116,000 available jobs in the state that need filled now," he said. According to the governor's office, Indiana's unemployment rate has recovered to 3.9% after climbing to 17% at the height of the pandemic. The lawsuit challenges Holcomb's assertion. Each of the five plaintiffs say they are unable to return to work due to lingering injuries or disability, health conditions that put them at risk for COVID-19 exposure, dependent children at home and no childcare available, or no positions that are available in their career field. Workers of color feel the loss of unemployment the most The National Employment Law Project says ending these jobless benefits early threatens the livelihoods of workers of color the most. Millions of Americans still heavily rely on jobless aid as the country slowly reopens from pandemic-induced lockdowns, according to the organization. As of May 22, more than 15.3 million people still needed some form of unemployment benefit—nearly twice the number who received payments when the aid programs began in late March 2020, NELP said. According to its analysis, over 46% of unemployment insurance recipients in the states ending the programs early are people of color. ndi The Supreme Court Will Hear A Case On The Funding Of Religious Schools By www.scpr.org Published On :: Fri, 02 Jul 2021 15:20:08 -0700 Eric Singerman | NPRAfter issuing its final decisions of the term Thursday, the Supreme Court on Friday granted a religious liberty case for next term and turned away challenges to longstanding decisions on qualified immunity and defamation, prompting dissents from the court's conservatives. Court agrees to hear one religious liberty case, but rejects another The justices agreed to consider a constitutional challenge to a school funding program in Maine that excludes private schools that teach religion. Only half the school districts in Maine run their own high schools. The rest pay for students to attend public schools in other districts or to attend private schools. The state, however, will not fund students who attend any school that offers religious teaching. Parents who wanted to send their children to a private Christian school challenged the law, alleging it violated their right to exercise their religion freely. The First Circuit disagreed, but now the high court will hear their case. The justices, however, declined to hear another case about religious liberty – this one brought by a Washington state florist who refused to provide flowers for a same-sex wedding. She alleged that the state's antidiscrimination law violated her First Amendment rights, and in 2017, Washington's supreme court ruled against her. Though the justices on Friday declined to hear her appeal, three of the court's conservatives—Samuel Alito, Clarence Thomas, and Neil Gorsuch—would have taken it for next term. Thomas calls to do away with qualified immunity Also on Friday, Justice Thomas once again called for the court to do away with qualified immunity, the legal shield for police officers that has come under intense scrutiny in the last year of racial justice protests. Thomas was dissenting from the court's refusal to hear the case of a college student promoting Turning Point USA, a right-wing organization known for publishing lists of university professors it deems hostile to conservatives. The student alleged campus police at Arkansas State University violated her First Amendment rights when they stopped her from advertising the organization near the student union. But the campus officers escaped liability in the lower court because of qualified immunity, a doctrine created by the Supreme Court in 1967 that has evolved into a near-impenetrable bulwark for the police. "Why should university officers," wrote Thomas, "receive the same protection as a police officer who makes a split-second decision to use force in a dangerous setting?" Going further, Thomas questioned whether the judicially-created doctrine should exist at all, an opinion that has garnered more and more bipartisan consensus in the wake of George Floyd's murder. Thomas and Gorsuch call to overturn landmark Free Speech precedent The court declined to hear a defamation case brought by a Miami-born international arms dealer—portrayed in the 2016 movie War Dogs—against the author of a book about his life. The lower court dismissed the suit. It pointed to a landmark 1964 First Amendment decision, in which the high court said that publishers are immune from libel suits brought by public figures, so long as the publishers either didn't know, or had no reason to know, that the information they published was false. Both Thomas and Gorsuch dissented, arguing the court should overturn the nearly 50-year-old precedent. In the era of disinformation, "lies impose real harm," wrote Thomas. "Instead of continuing to insulate those who perpetrate lies," said Thomas, the court should narrow First Amendment protections. In a separate dissent, Gorsuch agreed. In 1964, publishers needed protection against libel for unpopular opinions to survive. Indeed, the court's 1964 decision was first used to protect civil rights leaders who had published a New York Times ad criticizing the Montgomery, Alabama police for repeatedly arresting Martin Luther King Jr. But, said Gorsuch, in 2021, "it's less obvious what force [libel protections have] in a world in which everyone carries a soapbox in their hands," referring to smartphones. Now, Gorsuch wrote, "the deck seems stacked against those with traditional (and expensive) journalistic standards—and in favor of those who can disseminate the most sensational information as efficiently as possible without any particular concern for truth." Another execution On top of its decisions about cases next term, the justices gave Alabama the green light to execute Matthew Reeves, whose death sentence was recently overturned by the 11th Circuit Court of Appeals. This is the second time the justices have ruled against Reeves, who in 1998 was convicted for murder in Alabama. In 2002, Reeves first challenged his sentence in state court. He argued that because of his low IQ, his lawyer should have hired an expert to evaluate him for an intellectual disability. After 15 years of appeals, the Supreme Court denied his claim in 2017. So Reeves appealed his claim through the federal system. But on Friday, the high court again rejected his challenge, thus allowing Alabama to move forward with his execution. Justice Sotomayor, joined by Justice Kagan, dissented, criticizing the state court for its brusque dismissal of Reeves's claim. Sotomayor drew attention to "a troubling trend in which this court strains to reverse summarily any grants of relief to those facing execution."
