Engineering a surrogate human heteromeric α/β glycine receptor orthosteric site exploiting the structural homology and stability of acetylcholine-binding protein

Protein-engineering methods have been exploited to produce a surrogate system for the extracellular neurotransmitter-binding site of a heteromeric human ligand-gated ion channel, the glycine receptor. This approach circumvents two major issues: the inherent experimental difficulties in working with a membrane-bound ion channel and the complication that a heteromeric assembly is necessary to create a key, physiologically relevant binding site. Residues that form the orthosteric site in a highly stable ortholog, acetylcholine-binding protein, were selected for substitution. Recombinant proteins were prepared and characterized in stepwise fashion exploiting a range of biophysical techniques, including X-ray crystallography, married to the use of selected chemical probes. The decision making and development of the surrogate, which is termed a glycine-binding protein, are described, and comparisons are provided with wild-type and homomeric systems that establish features of molecular recognition in the binding site and the confidence that the system is suited for use in early-stage drug discovery targeting a heteromeric α/β glycine receptor.

CLIC4 is a cytokinetic cleavage furrow protein that regulates cortical cytoskeleton stability during cell division [RESEARCH ARTICLE]

During mitotic cell division, the actomyosin cytoskeleton undergoes several dynamic changes that play key roles in progression through mitosis. While the regulators of cytokinetic ring formation and contraction are well-established, proteins that regulate cortical stability during anaphase and telophase have been understudied. Here, we describe a role for CLIC4 in regulating actin and actin-regulators at the cortex and cytokinetic cleavage furrow during cytokinesis. We first describe CLIC4 as a new component of the cytokinetic cleavage furrow that is required for successful completion of mitotic cell division. We also demonstrate that CLIC4 regulates the remodeling of sub-plasma membrane actomyosin network within the furrow by recruiting MST4 kinase and regulating ezrin phosphorylation. This work identifies and characterizes new molecular players involved in regulating cortex stiffness and blebbing during late stages of cytokinetic furrowing.

DE-cadherin and Myosin II balance regulates furrow length for onset of polygon shape in syncytial Drosophila embryos [RESEARCH ARTICLE]

Cell shape morphogenesis from spherical to polygonal occurs in epithelial cell formation in metazoan embryogenesis. In syncytial Drosophila embryos, the plasma membrane incompletely surrounds each nucleus and is organized as a polygonal epithelial-like array. Each cortical syncytial division cycle shows circular to polygonal plasma membrane transition along with furrow extension between adjacent nuclei from interphase to metaphase. In this study, we assess the relative contribution of DE-cadherin and Myosin II at the furrow for polygonal shape transition. We show that polygonality initiates during each cortical syncytial division cycle when the furrow extends from 4.75 to 5.75 µm. Polygon plasma membrane organization correlates with increased junctional tension, increased DE-cadherin and decreased Myosin II mobility. DE-cadherin regulates furrow length and polygonality. Decreased Myosin II activity allows for polygonality to occur at a lower length than controls. Increased Myosin II activity leads to loss of lateral furrow formation and complete disruption of polygonal shape transition. Our studies show that DE-cadherin-Myosin II balance regulates an optimal lateral membrane length during each syncytial cycle for polygonal shape transition.

California Drought News: My burger, my burrito, my poor wallet

Tuesday's drought news makes you question whether waiting so long in the drive-thru line will continue to be worth it.

Food:

  • Have you noticed the increased food prices at the grocery store? Well, now you're going to see it at your fast food joints too. In-N-Out and Chipotle are having to raise prices on their food. Starbucks has also.
In-N-Out raised the cost of its hamburgers and cheeseburgers by a dime and their famous Double-Double jumped 15 cents to $3.45. French fries were unchanged but soft drinks went up a nickel. (San Gabriel Valley Tribune)

Oil and Water:

  • They don't mix, but they separate pretty well. The New York Times looks at how an oil field in the Central Valley also pumps 760,000 gallons of water each day that it sells to a local water district. Article goes on to look at the fight over water use in fracking. (NY Times)

Looting:

  • The lowering water levels at Lake Oroville have revealed more prehistoric artifacts. Volunteers are helping rangers by keeping tabs on looters who are digging up the relics. My favorite factoid is about how meth heads are among the many culprits. Does smoking meth cause an uncontrollable digging impulse?
Though many who disturb artifacts may not know any better, others can be troublesome. Among them are insomniac "tweakers" high on methamphetamine. "They just dig and dig like little squirrels," Dobis said. (LA Times)

  • Dowsers have been the media darlings of the current drought year. Benjamin Radford pokes at the practice and points out why we should trust our suspicions about the water-finding trick.
