The Andlinger Center is hosting a virtual event featuring Bernard Bigot of The ITER Organization. Bigot is the director-general of the 35-country scientific collaboration known as ITER (International Thermonuclear Experimental Reactor) tasked to build a 500-megawatt fusion plant in southern France. This project is slated to be the first fusion experiment to produce more energy than it consumes. PPPL is an active collaborator on the project, and physicist Egemen Kolemen is the seminar’s host.   Bigot is based in France and will Zoom to an international audience 12pm EST/4pm GMT. This seminar is an example of Princeton convening leadership at the highest levels, specifically around energy and environmental issues and solutions. Fusion is seen as a potential source of limitless energy, and Bigot will discuss how ITER puts fusion on a pathway to commercialization, and the advances and challenges with the project. Spatial trajectories are ubiquitous and complex signals. Their analysis is crucial in many research fields, from urban planning to neuroscience. Several approaches have been proposed to cluster trajectories. They rely on hand-crafted features, which struggle to capture the spatio-temporal complexity of the signal, or on Artificial Neural Networks (ANNs) which can be more efficient but less interpretable. In this paper we present a novel ANN architecture designed to capture the spatio-temporal patterns characteristic of a set of trajectories, while taking into account the demographics of the navigators. Hence, our model extracts markers linked to both behaviour and demographics. We propose a composite signal analyser (CompSNN) combining three simple ANN modules. Each of these modules uses different signal representations of the trajectory while remaining interpretable. Our CompSNN performs significantly better than its modules taken in isolation and allows to visualise which parts of the signal were most useful to discriminate the trajectories.

The only way to solubilize many antigens for immunoprecipitation is by denaturation. This cell lysis protocol is ideally suited for this purpose to release proteins from complex structures or reveal antibody epitopes hidden within native proteins. Short linear epitopes may not be accessible to antibodies within the native tertiary and quaternary protein structures, but they become exposed upon the unraveling of proteins, exposing their secondary structure. Antibodies otherwise not suitable for the immunoprecipitation of proteins prepared under nondenaturing conditions are now able to bind these antigens of interest in cell lysates prepared under denaturing conditions. These antibodies may also work well for immunoblotting purposes when the protein target is completely denatured. Harvested cells in this protocol are washed in tris-buffered saline (TBS) before lysis in 2% sodium dodecyl sulfate (SDS)-containing Lysis buffer for 10 min at 100°C. The resulting sample is diluted 20-fold in TBS to reduce the SDS concentration to ≤0.1% before the addition of an antibody for immunoprecipitation. Addition of 2% bovine serum albumin (BSA) or 0.1% Nonidet P-40 to the TBS before an immunoprecipitation, respectively, ensures either removal of SDS from the target protein or retaining denatured proteins in solution.
