A graph $G=(V,E)$ is $delta$-hyperbolic if for any four vertices $u,v,w,x$, the two larger of the three distance sums $d(u,v)+d(w,x)$, $d(u,w)+d(v,x)$, and $d(u,x)+d(v,w)$ differ by at most $2delta geq 0$. Recent work shows that many real-world graphs have small hyperbolicity $delta$. This paper describes the eccentricity terrain of a $delta$-hyperbolic graph. The eccentricity function $e_G(v)=max{d(v,u) : u in V}$ partitions the vertex set of $G$ into eccentricity layers $C_{k}(G) = {v in V : e(v)=rad(G)+k}$, $k in mathbb{N}$, where $rad(G)=min{e_G(v): vin V}$ is the radius of $G$. The paper studies the eccentricity layers of vertices along shortest paths, identifying such terrain features as hills, plains, valleys, terraces, and plateaus. It introduces the notion of $eta$-pseudoconvexity, which implies Gromov's $epsilon$-quasiconvexity, and illustrates the abundance of pseudoconvex sets in $delta$-hyperbolic graphs. In particular, it shows that all sets $C_{leq k}(G)={vin V : e_G(v) leq rad(G) + k}$, $kin mathbb{N}$, are $(2delta-1)$-pseudoconvex. Additionally, several bounds on the eccentricity of a vertex are obtained which yield a few approaches to efficiently approximating all eccentricities. An $O(delta |E|)$ time eccentricity approximation $hat{e}(v)$, for all $vin V$, is presented that uses distances to two mutually distant vertices and satisfies $e_G(v)-2delta leq hat{e}(v) leq {e_G}(v)$. It also shows existence of two eccentricity approximating spanning trees $T$, one constructible in $O(delta |E|)$ time and the other in $O(|E|)$ time, which satisfy ${e}_G(v) leq e_T(v) leq {e}_G(v)+4delta+1$ and ${e}_G(v) leq e_T(v) leq {e}_G(v)+6delta$, respectively. Thus, the eccentricity terrain of a tree gives a good approximation (up-to an additive error $O(delta))$ of the eccentricity terrain of a $delta$-hyperbolic graph.

A modulator generates a baseband digital signal from an information-bearing digital signal. The baseband signal has time-varying phase and amplitude defined by a sequence of complex data words, each having an in-phase (I) component and a quadrature (Q) component. A noise-shaping modulator generates a noise-shaped digital signal from the baseband digital signal such that quantization noise in the noise-shaping modulator is attenuated by a spectral null of its noise transfer function. The spectral null is selected by a noise-shaping parameter corresponding to a selected one of a plurality of output frequencies. A signal converter generates an analog signal conveying the information of the information-bearing digital signal on an analog carrier signal having the selected output frequency.

Typically, complex systems require a separate and expensive equalizer at the output of an analog-to-digital converter (ADC). Rather than providing a separate equalizer, the effective Signal Transfer Function (STF) of a Multi-stAge noise SHaping (MASH) ADC can be modified by leveraging available digital filtering hardware necessary for quantization noise cancellation. The modification can involves adding calculations in the software previously provided for computing digital quantization noise cancellation filter coefficients, where the calculations are added to take into account equalization as well. As a result, the signal transfer function can be modified to meet ADC or system-level signal-chain specifications without additional equalization hardware. The method is especially attractive for high-speed applications where magnitude and phase responses are more challenging to meet.

Reduction of protein phosphatase 2A (PP2A) complexity reveals cellular functions and dephosphorylation motifs of the PP2A/B'{delta} holoenzyme [Enzymology]

Protein phosphatase 2A (PP2A) is a large enzyme family responsible for most cellular Ser/Thr dephosphorylation events. PP2A substrate specificity, localization, and regulation by second messengers rely on more than a dozen regulatory subunits (including B/R2, B'/R5, and B″/R3), which form the PP2A heterotrimeric holoenzyme by associating with a dimer comprising scaffolding (A) and catalytic (C) subunits. Because of partial redundancy and high endogenous expression of PP2A holoenzymes, traditional approaches of overexpressing, knocking down, or knocking out PP2A regulatory subunits have yielded only limited insights into their biological roles and substrates. To this end, here we sought to reduce the complexity of cellular PP2A holoenzymes. We used tetracycline-inducible expression of pairs of scaffolding and regulatory subunits with complementary charge-reversal substitutions in their interaction interfaces. For each of the three regulatory subunit families, we engineered A/B charge–swap variants that could bind to one another, but not to endogenous A and B subunits. Because endogenous Aα was targeted by a co-induced shRNA, endogenous B subunits were rapidly degraded, resulting in expression of predominantly a single PP2A heterotrimer composed of the A/B charge–swap pair and the endogenous catalytic subunit. Using B'δ/PPP2R5D, we show that PP2A complexity reduction, but not PP2A overexpression, reveals a role of this holoenzyme in suppression of extracellular signal–regulated kinase signaling and protein kinase A substrate dephosphorylation. When combined with global phosphoproteomics, the PP2A/B'δ reduction approach identified consensus dephosphorylation motifs in its substrates and suggested that residues surrounding the phosphorylation site play roles in PP2A substrate specificity.

Interim PET evaluation in diffuse large B-cell lymphoma employing published recommendations: Comparison of the Deauville 5-point scale and the {Delta}SUVmax method

The value of interim 18F-fluorodeoxyglucose positron emission tomography (iPET) guided treatment decisions in patients with diffuse large B-cell lymphoma (DLBCL) has been the subject of much debate. This investigation focuses on a comparison of the Deauville score and the deltaSUVmax (SUVmax) approach – two methods to assess early metabolic response to standard chemotherapy in DLBCL. Methods: Of 609 DLBCL patients participating in the Positron Emission Tomography-guided Therapy of Aggressive non-Hodgkin Lymphomas (PETAL) trial, iPET scans of 596 patients originally evaluated using the SUVmax method were available for post-hoc assessment of the Deauville score. A commonly used definition of an unfavorable iPET result according to the Deauville score is an uptake greater than that of the liver, whereas an unfavorable iPET scan with regard to the SUVmax approach is characterized as a relative reduction of the maximum standardized uptake value between baseline and iPET staging of less than or equal to 66%. We investigated the two methods’ correlation and concordance by Spearman’s rank correlation coefficient and the agreement in classification, respectively. We further used Kaplan-Meier curves and Cox regression to assess differences in survival between patient subgroups defined by the pre-specified cut-offs. Time-dependent receiver operating curve analysis provided information on the methods’ respective discrimination performance. Results: Deauville score and SUVmax approach differed in their iPET-based prognosis. The SUVmax approach outperformed the Deauville score in terms of discrimination performance – most likely due to a high number of false-positive decisions by the Deauville score. Cut-off-independent discrimination performance remained low for both methods, but cut-off-related analyses showed promising results. Both favored the SUVmax approach, e.g. for the segregation by iPET response, where the event-free survival hazard ratio was 3.14 (95% confidence interval (CI): 2.22 – 4.46) for SUVmax and 1.70 (95% CI: 1.29 – 2.24) for the Deauville score. Conclusion: When considering treatment intensification, the currently used Deauville score cut-off of an uptake above that of the liver seems to be inappropriate and associated with potential harm for DLBCL patients. The SUVmax criterion of a relative reduction of the maximum standardized uptake value of less than or equal to 66% should be considered as an alternative.

PI3K{delta} as a Novel Therapeutic Target in Pathological Angiogenesis

Diabetic retinopathy is the most common microvascular complication of diabetes, and in the advanced diabetic retinopathy appear vitreal fibrovascular membranes that consist of a variety of cells, including vascular endothelial cells (ECs). New therapeutic approaches for this diabetic complication are urgently needed. Here, we report that in cultured human retinal microvascular ECs, high glucose induced expression of p110, which was also expressed in ECs of fibrovascular membranes from patients with diabetes. This catalytic subunit of a receptor-regulated PI3K isoform is known to be highly enriched in leukocytes. Using genetic and pharmacological approaches, we show that p110 activity in cultured ECs controls Akt activation, cell proliferation, migration, and tube formation induced by vascular endothelial growth factor, basic fibroblast growth factor, and epidermal growth factor. Using a mouse model of oxygen-induced retinopathy, p110 inactivation was found to attenuate pathological retinal angiogenesis. p110 inhibitors have been approved for use in human B-cell malignancies. Our data suggest that antagonizing p110 constitutes a previously unappreciated therapeutic opportunity for diabetic retinopathy.

Two additional Delta Dental member companies announced details April 27-28 of programs that will provide economic assistance and post-COVID-19 pandemic relief for members of its independent provider networks across 16 states and the District of Columbia.
