The noncanonical biosynthesis of curacozole by Streptomyces curacoi is shown to be dependent on the bldA-encoded Leu-tRNA^UUA. The minimal biosynthetic gene cluster is defined, and the functions of key biosynthetic enzymes are established in vitro.
