Saturations from pulse oximetry (SpO2) may overestimate arterial oxygen saturations measured by CO-oximetry (SaO2). The overestimation can be affected by location of measurement, perfusion, and skin color. Previous studies are limited by small numbers of observations in a hypoxemic range.

Villavicencio, Colombia, Apr 7, 2020 / 12:00 am (CNA).- The president of the Colombian bishops’ conference, Óscar Urbina, suspended 15 priests of his archdiocese from ministry who have been accused of sexual abuse. Other jurisdictions in the country have removed four other priests.

Archbishop Óscar Urbina of Villavicencio told Colombian media that the accused priests represent 15% of the city’s priests.

The priests are accused of committing sexual abuse in Colombia, Italy and the United States, Caracol Radio reported.

Fr. Carlos Villabón, communications director and chancellor for the archdiocese of Villavicencio, told ACI Prensa, CNA’s Spanish language news partner, that the 15 priests were suspended while a canonical investigation proceeds at the Vatican.

“On March 16, 2020 these 15 priests were notified after a preliminary investigation was carried out. They are neither convicted nor acquitted by this suspension, only asked to relinquish their parish duties, cease celebrating the Eucharist and cease their ministerial service while the complete investigation is conducted,” the priest explained.

The results of the preliminary investigation “are now being sent to the Congregation for the Doctrine of the Faith at the Vatican, and there they will determine the gravity of the facts and what the Church calls a penal canonical process will be conducted,” Villabón said.

“It’s unknown how much time the canonical process will take, but the idea is that it proceed as quickly as possible, considering that in Italy and in many parts of the world there’s a quarantine because of the coronavirus,” the communications director added.
Caracol Radio published a list of the 19 priests, but Villabón told ACI Prensa that the archdiocese would neither confirm nor deny the names reported.

 “According to a witness under protection by the prosecutor’s office, the 19 priests apparently formed a network of abusers, Caracol Radio reported.

In an April 3 statement, the archdiocese of Villavicencio announced that an accusation was received Feb. 14, 2020 concerning “acts against sexual morality by some priests of this archdiocese.”

“Having as a priority the alleged victim, we expressed to him our deep pain and solidarity and have offered him psychological and spiritual accompaniment. We reaffirm our commitment to act with clarity and transparency for his good and that of the Church,” the statement said.

Following the protocols of the Archdiocesan Commission for the Protection of Minors, once the abuse was reported, the regional prosecutor’s office was notified and “we made ourselves completely available to cooperate in the investigations taking place in this case,” the statement said.

The archdiocese said that it has taken steps “to eradicate the terrible evil of abuse within and outside our institution. We ask to be informed of any situations where one of our members has finally betrayed his vocation of service and dedication to the Lord and the community.”

Accurate delay measurement between two clocks

Hi,

I am currently struggling with measuring the delay between two clocks with a sufficient accuracy. The reference one is a fixed-phase clock, and the other one is a squared clock resulting of a circuit (kind of PLL) synthesis.
As I need to run a large amount of Monte-Carlo simulations in transient noise, I need to improve the simulation speed, while keeping a satisfactory delay measurement accuracy (<0.1ps), more specifically at 0V-crossings of the differential clocks. So I cannot simply set a max timestep <0.1ps as it would be far too long to simulate.
To sum up, I would need a very relaxed timestep on clock up and down levels, and a very short timestep only at rise/fall transitions.

For this purpose, I wrote a Verilog-A script
- using a timmer function to accurately emulate the reference clock 0V-crossing times (and get the related times with $abstime)
- using @(cross to get the 0V-crossing times of the synthesized clock: but this is not accurate enough (I see simulation noise around 3ps in Conservative). Indeed, the "cross" event occures at the simulation time following the effective 0V-crossing time; this could be sometimes >3ps, far not enough accurate for my purpose.
- I have tried to replace the cross with the "above" function, but it hasn't changed anything, whatever the time_tol value I put (<0.1ps for instance), the result is the same as with the "cross" function and the points are larger than >>0.1ps, weirdly.

So I have decided to give up Verilog-A to measure the delay between my two clocks.
I am currently trying to use the "delay" function of the Cadence Calculator as I guess it will "extrapolate" the time between two simulation points and therefore give a more accurate measurement of the 0V-crossing events, but when I try to compute the delay difference between the synthesized clock and the reference clock, it returns "0".

Could you please give me hints to dramatically improve my 0V-crossing time measurements while relaxing the simulation time?
- either by helping me in writing a more suitable Verilog-A script
- or by helping me in using the "delay" function of the calculator
- or maybe by providing me a "magic" Skill function?

WASHINGTON: The United States on Friday accused China and Russia of stepping up cooperation to spread false narratives over the coronavirus pandemic, saying Beijing was increasingly adopting techniques honed by Moscow.

“Even before the Covid-19 crisis we assessed a certain level of coordination between Russia and the PRC (Peoples Republic of China) in the realm of propaganda,” said Lea Gabrielle, coordinator of the State Department’s Global Engagement Center, which tracks foreign propaganda.

“But with this pandemic the cooperation has accelerated rapidly,” she told reporters.

“We see this convergence as a result of what we consider to be pragmatism between the two actors who want to shape public understanding of the Covid pandemic for their own purposes,” she said.

The Global Engagement Center earlier said that thousands of Russian-linked social media accounts were spreading conspiracies about the pandemic, including charging that the virus first detected last year in the Chinese metropolis of Wuhan was created by the United States.

China outraged the United States when a foreign ministry spokesman tweeted a conspiracy that the US military brought the virus to Wuhan, but the two countries reached an informal rhetorical truce in late March after telephone talks between President Donald Trump and his counterpart Xi Jinping.

Tensions have again soared as Secretary of State Mike Pompeo pushes the theory that the virus originated in a Chinese laboratory, even though both the World Health Organisation and the US government’s top epidemiologist say there is no evidence of this.

According to the Global Engagement Centre, China has again intensified its online campaign to defend its handling of the pandemic, which has killed some 270,000 people worldwide, and criticise the United States.

“Beijing is adapting in real time and increasingly using techniques that have long been employed by Moscow,” Gabrielle said.

China has increasingly used bot networks to amplify its message, Gabrielle said.

She said that official Chinese diplomatic accounts suddenly witnessed a surge in late March, going from adding around 30 new followers daily to more than 720, often from freshly created accounts.

She said that China was first observed using such online methods to “sow political discord” in its autonomous territory of Hong Kong, which has witnessed major pro-democracy demonstrations.

YejM Modulates Activity of the YciM/FtsH Protease Complex To Prevent Lethal Accumulation of Lipopolysaccharide

ABSTRACT

Lipopolysaccharide (LPS) is an essential glycolipid present in the outer membrane (OM) of many Gram-negative bacteria. Balanced biosynthesis of LPS is critical for cell viability; too little LPS weakens the OM, while too much LPS is lethal. In Escherichia coli, this balance is maintained by the YciM/FtsH protease complex, which adjusts LPS levels by degrading the LPS biosynthesis enzyme LpxC. Here, we provide evidence that activity of the YciM/FtsH protease complex is inhibited by the essential protein YejM. Using strains in which LpxC activity is reduced, we show that yciM is epistatic to yejM, demonstrating that YejM acts upstream of YciM to prevent toxic overproduction of LPS. Previous studies have shown that this toxicity can be suppressed by deleting lpp, which codes for a highly abundant OM lipoprotein. It was assumed that deletion of lpp restores lipid balance by increasing the number of acyl chains available for glycerophospholipid biosynthesis. We show that this is not the case. Rather, our data suggest that preventing attachment of lpp to the peptidoglycan sacculus allows excess LPS to be shed in vesicles. We propose that this loss of OM material allows continued transport of LPS to the OM, thus preventing lethal accumulation of LPS within the inner membrane. Overall, our data justify the commitment of three essential inner membrane proteins to avoid toxic over- or underproduction of LPS.

IMPORTANCE Gram-negative bacteria are encapsulated by an outer membrane (OM) that is impermeable to large and hydrophobic molecules. As such, these bacteria are intrinsically resistant to several clinically relevant antibiotics. To better understand how the OM is established or maintained, we sought to clarify the function of the essential protein YejM in Escherichia coli. Here, we show that YejM inhibits activity of the YciM/FtsH protease complex, which regulates synthesis of the essential OM glycolipid lipopolysaccharide (LPS). Our data suggest that disrupting proper communication between LPS synthesis and transport to the OM leads to accumulation of LPS within the inner membrane (IM). The lethality associated with this event can be suppressed by increasing OM vesiculation. Our research has identified a completely novel signaling pathway that we propose coordinates LPS synthesis and transport.

Dehydroascorbate Reductases and Glutathione Set a Threshold for High-Light-Induced Ascorbate Accumulation

Plants require a high concentration of ascorbate as a redox buffer for survival under stress conditions, such as high light. Dehydroascorbate reductases (DHARs) are enzymes that catalyze the reduction of DHA to ascorbate using reduced glutathione (GSH) as an electron donor, allowing rapid ascorbate recycling. However, a recent study using an Arabidopsis (Arabidopsis thaliana) triple mutant lacking all three DHAR genes (herein called dhar) did not find evidence for their role in ascorbate recycling under oxidative stress. To further study the function of DHARs, we generated dhar Arabidopsis plants as well as a quadruple mutant line combining dhar with an additional vtc2 mutation that causes ascorbate deficiency. Measurements of ascorbate in these mutants under low- or high-light conditions indicated that DHARs have a nonnegligible impact on full ascorbate accumulation under high light, but that they are dispensable when ascorbate concentrations are low to moderate. Because GSH itself can reduce DHA nonenzymatically, we used the pad2 mutant that contains ~30% of the wild-type GSH level. The pad2 mutant accumulated ascorbate at a wild-type level under high light; however, when the pad2 mutation was combined with dhar, there was near-complete inhibition of high-light–dependent ascorbate accumulation. The lack of ascorbate accumulation was consistent with a marked increase in the ascorbate degradation product threonate. These findings indicate that ascorbate recycling capacity is limited in dhar pad2 plants, and that both DHAR activity and GSH content set a threshold for high-light–induced ascorbate accumulation.

Atorvastatin Reduces In Vivo Fibrin Deposition and Macrophage Accumulation, and Improves Primary Patency Duration and Maturation of Murine Arteriovenous Fistula

Background

Arteriovenous fistulas placed surgically for dialysis vascular access have a high primary failure rate resulting from excessive inward remodeling, medial fibrosis, and thrombosis. No clinically established pharmacologic or perisurgical therapies currently address this unmet need. Statins’ induction of multiple anti-inflammatory and antithrombotic effects suggests that these drugs might reduce arteriovenous fistula failure. Yet, the in vivo physiologic and molecular effects of statins on fistula patency and maturation remain poorly understood.

Methods

We randomized 108 C57Bl/6J mice to receive daily atorvastatin 1.14 mg/kg or PBS (control) starting 7 days before end-to-side carotid artery–jugular vein fistula creation and for up to 42 days after fistula creation. We then assessed longitudinally the effects of statin therapy on primary murine fistula patency and maturation. We concomitantly analyzed the in vivo arteriovenous fistula thrombogenic and inflammatory macrophage response to statin therapy, using the fibrin-targeted, near-infrared fluorescence molecular imaging agent FTP11-CyAm7 and dextranated, macrophage-avid nanoparticles CLIO-VT680.

Results

In vivo molecular-structural imaging demonstrated that atorvastatin significantly reduced fibrin deposition at day 7 and macrophage accumulation at days 7 and 14, findings supported by histopathologic and gene-expression analyses. Structurally, atorvastatin promoted favorable venous limb outward remodeling, preserved arteriovenous fistula blood flow, and prolonged primary arteriovenous fistula patency through day 42 (P<0.05 versus control for all measures).

Conclusions

These findings provide new in vivo evidence that statins improve experimental arteriovenous fistula patency and maturation, indicating that additional clinical evaluation of statin therapy in patients on dialysis undergoing arteriovenous fistula placement is warranted.

Assessing the accuracy of direct-coupling analysis for RNA contact prediction [ARTICLE]

Many noncoding RNAs are known to play a role in the cell directly linked to their structure. Structure prediction based on the sole sequence is, however, a challenging task. On the other hand, thanks to the low cost of sequencing technologies, a very large number of homologous sequences are becoming available for many RNA families. In the protein community, the idea of exploiting the covariance of mutations within a family to predict the protein structure using the direct-coupling-analysis (DCA) method has emerged in the last decade. The application of DCA to RNA systems has been limited so far. We here perform an assessment of the DCA method on 17 riboswitch families, comparing it with the commonly used mutual information analysis and with state-of-the-art R-scape covariance method. We also compare different flavors of DCA, including mean-field, pseudolikelihood, and a proposed stochastic procedure (Boltzmann learning) for solving exactly the DCA inverse problem. Boltzmann learning outperforms the other methods in predicting contacts observed in high-resolution crystal structures.
