India and China want to join Russia's lunar nuclear station project

India and China evince interest in the Russian nuclear power plant project on the Moon, TASS reports with reference to Rosatom chief Alexey Likhachev. "The new solution that we are asked to implement is a version of a lunar nuclear power plant with an energy capacity of up to half a megawatt,” Likhachev said at the Eastern Economic Forum. In May, Roscosmos Director Yuri Borisov said that a Russian-made nuclear installation for a joint Russian-Chinese station would be sent to the Moon after 2036. Preliminary X-ray diffraction and ligand-binding analyses of the N-terminal domain of hypothetical protein Rv1421 from Mycobacterium tuberculosis H37Rv

Mycobacterium tuberculosis can reside and persist in deep tissues; latent tuberculosis can evade immune detection and has a unique mechanism to convert it into active disease through reactivation. M. tuberculosis Rv1421 (MtRv1421) is a hypothetical protein that has been proposed to be involved in nucleotide binding-related metabolism in cell-growth and cell-division processes. However, due to a lack of structural information, the detailed function of MtRv1421 remains unclear. In this study, a truncated N-terminal domain (NTD) of MtRv1421, which contains a Walker A/B-like motif, was purified and crystallized using PEG 400 as a precipitant. The crystal of MtRv1421-NTD diffracted to a resolution of 1.7 Å and was considered to belong to either the C-centered monoclinic space group C2 or the I-centered orthorhombic space group I222, with unit-cell parameters a = 124.01, b = 58.55, c = 84.87 Å, β = 133.12° or a = 58.53, b = 84.86, c = 90.52 Å, respectively. The asymmetric units of the C2 or I222 crystals contained two or one monomers, respectively. In terms of the binding ability of MtRv1421-NTD to various ligands, uridine diphosphate (UDP) and UDP-N-acetylglucosamine significantly increased the melting temperature of MtRv1421-NTD, which indicates structural stabilization through the binding of these ligands. Altogether, the results reveal that a UDP moiety may be required for the interaction of MtRv1421-NTD as a nucleotide-binding protein with its ligand.

Ternary structure of Plasmodium vivax N-myristoyltransferase with myristoyl-CoA and inhibitor IMP-0001173

Plasmodium vivax is a major cause of malaria, which poses an increased health burden on approximately one third of the world's population due to climate change. Primaquine, the preferred treatment for P. vivax malaria, is contraindicated in individuals with glucose-6-phosphate dehydrogenase (G6PD) deficiency, a common genetic cause of hemolytic anemia, that affects ∼2.5% of the world's population and ∼8% of the population in areas of the world where P. vivax malaria is endemic. The Seattle Structural Genomics Center for Infectious Disease (SSGCID) conducted a structure–function analysis of P. vivax N-myristoyltransferase (PvNMT) as part of efforts to develop alternative malaria drugs. PvNMT catalyzes the attachment of myristate to the N-terminal glycine of many proteins, and this critical post-translational modification is required for the survival of P. vivax. The first step is the formation of a PvNMT–myristoyl–CoA binary complex that can bind to peptides. Understanding how inhibitors prevent protein binding will facilitate the development of PvNMT as a viable drug target. NMTs are secreted in all life stages of malarial parasites, making them attractive targets, unlike current antimalarials that are only effective during the plasmodial erythrocytic stages. The 2.3 Å resolution crystal structure of the ternary complex of PvNMT with myristoyl-CoA and a novel inhibitor is reported. One asymmetric unit contains two monomers. The structure reveals notable differences between the PvNMT and human enzymes and similarities to other plasmodial NMTs that can be exploited to develop new antimalarials.

Optimization of synchrotron radiation parameters using swarm intelligence and evolutionary algorithms

Alignment of each optical element at a synchrotron beamline takes days, even weeks, for each experiment costing valuable beam time. Evolutionary algorithms (EAs), efficient heuristic search methods based on Darwinian evolution, can be utilized for multi-objective optimization problems in different application areas. In this study, the flux and spot size of a synchrotron beam are optimized for two different experimental setups including optical elements such as lenses and mirrors. Calculations were carried out with the X-ray Tracer beamline simulator using swarm intelligence (SI) algorithms and for comparison the same setups were optimized with EAs. The EAs and SI algorithms used in this study for two different experimental setups are the Genetic Algorithm (GA), Non-dominated Sorting Genetic Algorithm II (NSGA-II), Particle Swarm Optimization (PSO) and Artificial Bee Colony (ABC). While one of the algorithms optimizes the lens position, the other focuses on optimizing the focal distances of Kirkpatrick–Baez mirrors. First, mono-objective evolutionary algorithms were used and the spot size or flux values checked separately. After comparison of mono-objective algorithms, the multi-objective evolutionary algorithm NSGA-II was run for both objectives – minimum spot size and maximum flux. Every algorithm configuration was run several times for Monte Carlo simulations since these processes generate random solutions and the simulator also produces solutions that are stochastic. The results show that the PSO algorithm gives the best values over all setups.

Signal-to-noise and spatial resolution in in-line imaging. 1. Basic theory, numerical simulations and planar experimental images

Signal-to-noise ratio and spatial resolution are quantitatively analysed in the context of in-line (propagation based) X-ray phase-contrast imaging. It is known that free-space propagation of a coherent X-ray beam from the imaged object to the detector plane, followed by phase retrieval in accordance with Paganin's method, can increase the signal-to-noise in the resultant images without deteriorating the spatial resolution. This results in violation of the noise-resolution uncertainty principle and demonstrates `unreasonable' effectiveness of the method. On the other hand, when the process of free-space propagation is performed in software, using the detected intensity distribution in the object plane, it cannot reproduce the same effectiveness, due to the amplification of photon shot noise. Here, it is shown that the performance of Paganin's method is determined by just two dimensionless parameters: the Fresnel number and the ratio of the real decrement to the imaginary part of the refractive index of the imaged object. The relevant theoretical analysis is performed first, followed by computer simulations and then by a brief test using experimental images collected at a synchrotron beamline. More extensive experimental tests will be presented in the second part of this paper.

Chem. 23, 3160–3166] showing the first crystal structures of μ-F-bridged penta­fluoro­germanates. Herein, we report the second crystal structure of trans-penta­fluoro­germanate anions present in the crystal structure of sodium trans-penta­fluoro­germanate(IV) bis­(hy­dro­gen fluoride), Na[GeF5]·2HF. The crystal structure [ortho­rhom­bic Pca21, a = 12.3786 (3), b = 7.2189 (2), c = 11.4969 (3) Å and Z = 8] is built up from infinite chains of trans-linked [GeF6]2− octa­hedra, extending along the b axis and spanning a network of penta­gonal bipyramidal distorted Na-centred polyhedra. These [NaF7] polyhedra are linked in a trans-edge fashion via hy­dro­gen fluoride mol­ecules, in analogy to already known sodium hy­dro­gen fluorides and potassium hy­dro­gen fluorides.

The 'Human Error' That's Snarling The New York City Mayor's Race

The closely-watched New York City mayoral primary election tumbled into chaos this week as the NYC Board of Elections announced it had released incorrect preliminary results on Tuesday.

City officials admitted they failed to remove 135,000 test ballots from the election management system before starting to count the real votes from Election Day and early voting, skewing the results.

"The Board apologizes for the error and has taken immediate measures to ensure the most accurate up to date results are reported," the agency tweeted.

The error is complicated by the fact that New York City is using ranked-choice voting, in which each round of vote counting hinges on the results from the previous round.

Some of the top candidates vying to lead the country's largest city blasted the board's mistake as they — and about 8.5 million other New Yorkers — awaited the results of a revised tally expected to be released on Wednesday.

Ranked-choice voting, explained

Instead of choosing just one candidate to vote for, New York City voters in last week's election were able to rank their top five candidates in order of preference.

It was the first time in decades New York used ranked-choice voting, which city voters overwhelmingly approved in a 2019 ballot measure.

NPR's Domenico Montanaro explained how the process works:

  1. "If someone gets 50% plus one after all the first-choice votes are counted, then the election is over and that candidate wins. 
  2. "But if no one gets 50% plus one, it's on to Round 2.
  3. "The person with the lowest number of first-place votes is eliminated, and that candidate's voters' second choices get redistributed as votes for other candidates.
  4. "This reallocation of votes goes on until someone reaches 50% plus one."

If just two candidates remain at the end, the candidate with the most votes wins.

What happened this week

On Tuesday, the city Board of Elections released the first ranked-choice voting reports from the election.

With only first-preference votes counted as of election night, Brooklyn Borough President Eric Adams boasted a nine-point lead over attorney Maya Wiley. Those first reported ranked-choice results shrank Adam's lead to just two points ahead of former Sanitation Commissioner Kathryn Garcia, Gothamist reported.

But just hours later, the board tweeted that it had become aware of a "discrepancy" in the ranked-choice voting results and pulled them from its website.

In a follow-up apology, the board acknowledged that it had erroneously left 135,000 test votes in its election system, producing "additional records" that likely impacted an accurate tally.

"At this point it really seems like an issue of human error," WNYC reporter Brigid Bergin told NPR's Morning Edition.

"The board does conduct a lot of pre-election testing to make sure their systems are working and, obviously, that was even more important this time, because it was the first time they were using this new ranked-choice voting system," she added.

Bergin said the board is expected to release a corrected ranked-choice voting report Wednesday, but it will still be preliminary and it won't include 124,000 absentee ballots.

How the candidates are reacting

All of the mayoral contenders expressed frustration with the board's blunder.

"Today's mistake by the Board of Elections was unfortunate," Adams tweeted Tuesday. "It is critical that New Yorkers are confident in their electoral system, especially as we rank votes in a citywide election for the first time."

Garcia, who was fleetingly thrust into second place by the incorrect ranked-choice voting report, called for a more thorough accounting of what went wrong.

"The Board of Elections' release of incorrect ranked choice votes is deeply troubling and requires a much more transparent and complete explanation. Every ranked choice and absentee vote must be counted accurately so that all New Yorkers have faith in our democracy and our government," she tweeted.

Progressive candidate Maya Wiley said this week's misstep was just the latest in a string of mistakes by the board.

"This error by the Board of Elections is not just failure to count votes properly today, it is the result of generations of failures that have gone unaddressed," Wiley said. "Today, we have once again seen the mismanagement that has resulted in a lack of confidence in results, not because there is a flaw in our election laws, but because those who implement it have failed too many times."

WNYC's Bergin said she thought the misstep would not cause voters to question the election results but that it may diminish the board's reputation in the eyes of the public.

"This agency is really the last bastion of true patronage politics in New York," she said. "There's been a push to overhaul the agency, to give the staff more authority over political appointees. But ultimately that's all up to state lawmakers to do."

Aashima Gupta from Google Cloud shared insights on these tools' transformative capabilities, while Tony Farah from Highmark Health cited an 85% reduction in provider administrative costs after automating prior authorizations. Helen Waters from Meditech added, "We believe that gen AI and AI overall is transforming how healthcare professionals access and use information to make powerful decisions confidently," reflecting the positive impact of AI tools on healthcare workflows and decision-making.

In A Narrow Ruling, Supreme Court Hands Farmworkers Union A Loss

The Supreme Court on Wednesday tightened the leash on union representatives and their ability to organize farmworkers in California and elsewhere. At issue in the case was a California law that allows union organizers to enter farms to speak to workers during nonworking hours — before and after work, as well as during lunch — for a set a number of days each year.

By a 6-3 vote along ideological lines, the court ruled that the law — enacted nearly 50 years ago after a campaign by famed organizer Cesar Chavez — unconstitutionally appropriates private land by allowing organizers to go on farm property to drum up union support.

"The regulation appropriates a right to physically invade the growers' property," Chief Justice John Roberts wrote for the court's conservative majority. "The access regulation amounts to simple appropriation of private property."

The decision is a potentially mortal blow that threatens the very existence of the farmworkers union. However, the ruling stopped short of upending other laws that allow government officials to enter private property to inspect and enforce health and safety rules that cover everything restaurants to toxic chemical sites.

Indeed, as Roberts wrote: "Under this framework, government health and safety inspection regimes will generally not constitute takings."

The court's decision on Wednesday was only the latest in a series of decisions that have aimed directly at the heart of organized labor in the United States. In 2018, the court hamstrung public-sector unions' efforts to raise money for collective bargaining. In that decision, the court by a 5-4 vote overturned a 40-year precedent that had allowed unions to collect limited "fair share" fees from workers not in the union but who benefited from the terms of the contract that the union negotiated.

The case decided by the court on Wednesday began in 2015 at Cedar Point Nursery, near the Oregon border. The nursery's owner, Mike Fahner, said union organizers entered the farm at 5 a.m. one morning, without the required notice, and began harassing his workers with bullhorns. The general counsel for the United Farm Workers, Mario Martinez, countered that the people with bullhorns were striking workers, not union organizers.

When Cedar Point filed a complaint with the California Agricultural Labor Relations Board, the board found no illegal behavior and dismissed the complaint. Cedar Point, joined by another California grower, appealed all the way to the Supreme Court, arguing they should be able to exclude organizers from their farms.

Writing for the court's three liberals, Justice Stephen Breyer said the access in the case was "temporary" and so did not constitute a "taking" under the law.

The rule, he wrote , is "not functionally equivalent to the classic taking in which government directly appropriates private property or ousts the owner from his domain."

"In my view, the majority's conclusion threatens to make many ordinary forms of regulation unusually complex or impractical," he wrote.

The court's decision could be disastrous for unions in general, but especially those that represent low-income workers. The growers asserted that unions should have no problem organizing workers in the era of the internet. But many of the workers at Cedar Point don't own smartphones and don't have internet access. What's more, many speak Spanish or indigenous languages and live scattered throughout the area, in motels, in labor camps or with friends and family, often moving after just a few weeks when the seasonal harvest is over.
