T Cell Responses Induced by Attenuated Flavivirus Vaccination Are Specific and Show Limited Cross-Reactivity with Other Flavivirus Species [Vaccines and Antiviral Agents]

Members of the flavivirus genus share a high level of sequence similarity and often circulate in the same geographical regions. However, whether T cells induced by one viral species cross-react with other related flaviviruses has not been globally addressed. In this study, we tested pools of epitopes derived from dengue (DENV), Zika (ZIKV), Japanese encephalitis (JEV), West Nile (WNV), and yellow fever (YFV) viruses by intracellular cytokine staining (ICS) using peripheral blood mononuclear cells (PBMCs) of individuals naturally exposed to DENV or immunized with DENV (TV005) or YF17D vaccine. CD8 T cell responses recognized epitopes from multiple flaviviruses; however, the magnitude of cross-reactive responses was consistently severalfold lower than those to the autologous epitope pools and was associated with lower expression of activation markers such as CD40L, CD69, and CD137. Next, we characterized the antigen sensitivity of short-term T cell lines (TCL) representing 29 different individual epitope/donor combinations. TCL derived from DENV monovalent vaccinees induced CD8 and CD4 T cells that cross-reacted within the DENV serocomplex but were consistently associated with >100-fold-lower antigen sensitivity for most other flaviviruses, with no cross-recognition of YFV-derived peptides. CD8 and CD4 TCL from YF17D vaccinees were associated with very limited cross-reactivity with any other flaviviruses and in five out of eight cases >1,000-fold-lower antigen sensitivity. Overall, our data suggest limited cross-reactivity for both CD4 and CD8 T cell responses between flaviviruses and have implications for understanding immunity elicited by natural infection and strategies to develop live attenuated vaccines against flaviviral species.

IMPORTANCE The envelope (E) protein is the dominant target of neutralizing antibodies for dengue virus (DENV) and yellow fever virus (YFV). Accordingly, several DENV vaccine constructs use the E protein in a live attenuated vaccine format, utilizing a backbone derived from a heterologous flavivirus (such as YF) as a delivery vector. This backbone comprises the nonstructural (NS) and capsid (C) antigens, which are dominant targets of T cell responses. Here, we demonstrate that cross-reactivity at the level of T cell responses among different flaviviruses is very limited, despite high levels of sequence homology. Thus, the use of heterologous flavivirus species as a live attenuated vaccine vector is not likely to generate optimal T cell responses and might thus impair vaccine performance.

Decision tree as a tool for the management of coastal aquifers of limited saturated thickness

In this paper, a decision tree is presented, constructed on the basis of hydrogeological characteristics (water table depth, freshwater thickness, surface area required and distance between wells), to choose the optimal groundwater extraction method in the case of a coastal unconfined aquifer. A comparison is made of the groundwater extraction methods in a freshwater aquifer of limited thickness occurring in coastal dunes in the eastern region of the Province of Buenos Aires (Argentina). The negative effects brought about by the wrong use of the groundwater extraction methods are analysed, because, as a result of excessive extraction, such methods lead to the dramatic decrease of the freshwater reserves. The decision tree is a useful tool to assist decision-makers as it suggests the most suitable groundwater extraction method options (vertical wells or wellpoints), as well as identifying areas that are unsuitable for sustainable groundwater extraction.

The Limited Role of Glucagon for Ketogenesis During Fasting or in Response to SGLT2 Inhibition

Glucagon is classically described as a counterregulatory hormone that plays an essential role in the protection against hypoglycemia. In addition to its role in the regulation of glucose metabolism, glucagon has been described to promote ketosis in the fasted state. Sodium–glucose cotransporter 2 inhibitors (SGLT2i) are a new class of glucose-lowering drugs that act primarily in the kidney, but some reports have described direct effects of SGLT2i on α-cells to stimulate glucagon secretion. Interestingly, SGLT2 inhibition also results in increased endogenous glucose production and ketone production, features common to glucagon action. Here, we directly test the ketogenic role of glucagon in mice, demonstrating that neither fasting- nor SGLT2i-induced ketosis is altered by interruption of glucagon signaling. Moreover, any effect of glucagon to stimulate ketogenesis is severely limited by its insulinotropic actions. Collectively, our data suggest that fasting-associated ketosis and the ketogenic effects of SGLT2 inhibitors occur almost entirely independent of glucagon.

    Navajo Nation Suffers Third-Highest COVID-19 Infection Rate in U.S. with Limited Healthcare & Water

    We get an update from two doctors treating patients with the Navajo Nation, the largest Indigenous reservation in the country, which has been hard hit by the coronavirus pandemic. Dr. Michelle Tom is a member of the Navajo Nation and a family physician treating COVID-19 patients at the Winslow Indian Health Care Center and Little Colorado Medical Center in northern Arizona near the Navajo reservation. In Gallup, New Mexico, Dr. Sriram Shamasunder is leading a medical volunteer group of 21 nurses and doctors from the University of California, San Francisco as part of the HEAL Initiative. He says the coronavirus hit harder on the Navajo Nation due to a "trajectory of an underfunded health system," and notes the Indian Health Service is funded at one-third the rate per capita as Medicare. "The level of inequity that you're seeing … it's part of this pattern."

    Shiv Sena may have discovered a way to bend the BJP its way. The political precision with which the party snapped up the disgruntled son of a late BJP parliamentarian for fielding in the Palghar Lok Sabha bypoll has taken Uddhav's resolve of outsmarting the saffron frenemy to a decisive level. Uddhav wants to fly solo, escape BJP's trap for emerging as the single largest party in the state next year.

    Palghar, a testing lab, may decide the fate of the strained relation between the two partners, which the Sena is using to try and carve a niche for itself by playing a detractor within and outside the government.

    Uddhav has a spring in his step, as Sena prepares to run Palghar, and the guts to ask the BJP to withdraw from the race. It would be interesting to see who blinks first in a barter proposed by Uddhav. The Sena chief says if BJP opts out in Palghar, he will support the ally in Gondia-Bhandara, which too will go to Lok Sabha bypoll on the same day (May 28).

    But will they reach a compromise? The possibility of the two parties backing out seem remote. There are others in the fray who too want the BJP defeated. The BJP has fielded a former Congress junior minister, Rajendra Gavit, who had started his political career with Janata Dal. Gavit came in amid high drama after Sena's nomination of BJP's late MP Chintaman Wanga's son Shrinivas surprised all. Gavit had lost two Assembly elections in a row for the Congress, and before that, he was unceremoniously asked to pull out for a BVS candidate in 2014 Lok Sabha polls.

    'Well played, Sena'
    The BJP camp does not admit it openly, but insiders say the Sena was tactically much superior this time. Sena explored the emotional turmoil in the Wanga family, which felt ignored by the BJP. The family members accused the BJP high command, including the CM, of not giving them audience. In response, Fadnavis blamed the poaching on Sena. He said it was unexpected of the ally, and that the BJP would win, with or without Sena.

    The Palghar bypoll is important for the BJP to reaffirm its influence in the tribal belt. In the 2014 Lok Sabha elections, four out of the 48 seats were reserved for Scheduled Tribes, of which all went to the BJP, an unprecedented result. BJP had been contesting Dahanu (delimited and named Palghar) for many decades but with limited success.

    In the Assembly, 25 of the 288 seats are reserved for STs. Of these, BJP won 11, followed by Congress's five, NCP's four, Sena's three and one each by CPI(M) and Bahujan Vikas Aghadi (BVA).

    The results of Lok Sabha bypolls in Maharashtra should measure BJP's influence in the four years of government. Sena hasn't fielded in Gondia-Bhandara because its poor strength will never secure it a prominent place. Knowing his weakness in the region well, Uddhav paid an impromptu visit to Nagpur last week. Again, his was an attempt to create some nuisance for the BJP, where they expect BJP and NCP to fight straight.

    Other factors in Palghar
    For Sena, Palghar has turned out to be a sharper tool to cut the BJP to size. Uddhav has nothing lose in a skirmish that will be four-way in case some candidates don't pull out, while a victory would be a shot in the arm for Sena.

    Former allies Congress and influential local outfit, led by Hitendra Thakur, BVA are also in the fray. BVA's role will be significant in splitting votes because it had won the LS seat in 2009, and lost the next to BJP, despite having its vote share of 30 per cent intact. In 2014, BJP increased its vote share by a whopping 25 per cent. BJP and Sena were together then, while Congress had withdrawn Gavit (now BJP's candidate) for supporting Thakur's candidate. BVA continues to be a big player in two urban Assembly segments of Vasai and Nalasopara, which Thakur and son represent.

    If it were to ensure BJP's defeat, will Congress withdraw the candidature of former MP Damu Shingda this time? Congress insiders say supporting BVA may not be a good idea, and instead, the party should have a tacit understanding with the Sena for beating the BJP at the hands of the ally. But will that be acceptable to Congress, which is working to emerge as a strong alternative to BJP?

    All eyes will be on developments taking place on Monday, the last day of withdrawal of candidatures.

    Coronavirus outbreak: Limited trains move out of red zone MMR

    Trains ferrying migrant workers have been on the move with about 14 of them having left from the Central Railway section and 62 from the Western Railway section across different divisions. However, there continues to be limitations in the number of trains leaving from the Mumbai Metropolitan Region (MMR) as it is a red zone.

    So far, eight trains have left from the MMR region with a couple of more planned in the coming days. A Bhivandi Road-Gorakhpur one was the first train to leave the MMR region on May 2 with 1,104 migrants.

    The same day another train left from Vasai Road for Gorakhpur with 1,200 migrants. After this, two trains left for Jaipur — one from Bhivandi and another from Dahanu — on May 4, two from Kalyan to Darbhanga and Kalyan to Guntkul on May 5 and another two — Panvel to Rewa and Bhivandi to Patna in the wee hours of May 6.

    Meanwhile, the first train to arrive in Maharashtra was from Rayanapadu in Vijaywada. It reached Chandrapur with 1,212 migrants around 9.10 am on May 6. The passengers were provided with food and water by the state government and later taken for health check-ups by the district collector before being sent to their homes.

    One of the group leaders, who was onboard the train, said the migrants from Maharashtra had gone to Rayanapadu for red chilli plucking and transport was organised by the Krishna district administration. "They were taken to the station in 48 buses with 25 passengers each. The passengers included senior citizens, women and children too," he told mid-day over phone. Western Railway chief PRO, Ravinder Bhakar said the railways would run these trains as long as there was demand from other state governments.

    Ravi Shastri is convinced that the class of India's 1985 team, of which he was a vital cog, was such it could trouble even the current team led by Virat Kohli. Shastri was the key man back in 1985 when the team under Sunil Gavaskar won the World Championship of Cricket. He was hero of India's triumph in Australia and had won the famous Audi car for being 'player of the tournament'. He is still influential in India's cricket set up, sitting in head coach chair, having played a significant role in team's ascendancy in world cricket across formats.

    "No question about that. They (team of 85) will give any team that India puts up in white ball cricket, a run for their money. That team of 85 will give this team a run for money," Shastri said during 'Sony Ten Pit Stop' show on channel's FB page. Shastri also has a firm belief that that the team of 1985 was qualitatively better than the side that won the 1983 World Cup because it was a perfect blend of youth and experience. "I go one step ahead and say the team of 1985 was a stronger team compared to 1983.

    "You know, I was part of both teams, I played in the 1983 World Cup and 1985, when you look man to man, 80 per cent of that 83 team was still there, but then some of the youngsters that you got in there like a Sivaramakrishnan, Sadanand Vishwanath, Azharuddin, those kind of guys came in to add to the experience you already had of 83 (and it) was fantastic." Shastri said that winning Test series in Australia in 2018-19 for the first time in 71 years was special but when it came to white ball cricket, 85 was special.

    "It is great to be part of those two teams, as a coach that series in Australia, is going to be very very special and hard to beat, because that team wasn't beaten in 71 years by any Asian team. "There have been plenty who have gone there, tried their best and everyone knows how tough it is to beat Australia in Australia in Test match cricket, but in white ball cricket you can't beat 1985 as a player. To win both is fantastic."

    Shastri then shared a hilarious anecdote about a team meeting before the final against Pakistan. Shastri imitated what Kapil Dev said in his typical English. "Kapil said, 'if I win car, I keep 25 percent (by selling) and rest share. Jimmy came and (said) "Yaar jisko mila mila' (whoever gets it, gets it). When my turn came, I said, if I win it, I am keeping the car and I can only share the stepney," Shastri said in typical tongue-in-cheek manner.
