v4 Synthesis and characterization of an organic–inorganic hybrid crystal: 2[Co(en)3](V4O13)·4H2O By journals.iucr.org Published On :: 2024-09-03 Organic–inorganic hybrid crystals have diverse functionalities, for example in energy storage and luminescence, due to their versatile structures. The synthesis and structural characterization of a new cobalt–vanadium-containing compound, 2[Co(en)3]3+(V4O13)6−·4H2O (1) is presented. The crystal structure of 1, consisting of [Co(en)3]3+ complexes and chains of corner-sharing (VO4) tetrahedra, was solved by single-crystal X-ray diffraction in the centrosymmetric space group P1. Phase purity of the bulk material was confirmed by infrared spectroscopy, scanning electron microscopy, elemental analysis and powder X-ray diffraction. The volume expansion of 1 was found to be close to 1% in the reported temperature range from 100 to 300 K, with a volume thermal expansion coefficient of 56 (2) × 10−6 K−1. The electronic band gap of 1 is 2.30 (1) eV, and magnetic susceptibility measurements showed that the compound exhibits a weak paramagnetic response down to 1.8 K, probably due to minor CoII impurities (<1%) on the CoIII site. v4 Inhibition of the SUV4-20 H1 histone methyltransferase increases frataxin expression in Friedreich's ataxia patient cells [Gene Regulation] By www.jbc.org Published On :: 2020-12-25T00:06:30-08:00 The molecular mechanisms of reduced frataxin (FXN) expression in Friedreich's ataxia (FRDA) are linked to epigenetic modification of the FXN locus caused by the disease-associated GAA expansion. Here, we identify that SUV4-20 histone methyltransferases, specifically SUV4-20 H1, play an important role in the regulation of FXN expression and represent a novel therapeutic target. Using a human FXN–GAA–Luciferase repeat expansion genomic DNA reporter model of FRDA, we screened the Structural Genomics Consortium epigenetic probe collection. We found that pharmacological inhibition of the SUV4-20 methyltransferases by the tool compound A-196 increased the expression of FXN by ∼1.5-fold in the reporter cell line. In several FRDA cell lines and patient-derived primary peripheral blood mononuclear cells, A-196 increased FXN expression by up to 2-fold, an effect not seen in WT cells. SUV4-20 inhibition was accompanied by a reduction in H4K20me2 and H4K20me3 and an increase in H4K20me1, but only modest (1.4–7.8%) perturbation in genome-wide expression was observed. Finally, based on the structural activity relationship and crystal structure of A-196, novel small molecule A-196 analogs were synthesized and shown to give a 20-fold increase in potency for increasing FXN expression. Overall, our results suggest that histone methylation is important in the regulation of FXN expression and highlight SUV4-20 H1 as a potential novel therapeutic target for FRDA. Full Article

v4 Recent Visual Experience Reshapes V4 Neuronal Activity and Improves Perceptual Performance By www.jneurosci.org Published On :: 2024-10-09T09:30:20-07:00 Recent visual experience heavily influences our visual perception, but how neuronal activity is reshaped to alter and improve perceptual discrimination remains unknown. We recorded from populations of neurons in visual cortical area V4 while two male rhesus macaque monkeys performed a natural image change detection task under different experience conditions. We found that maximizing the recent experience with a particular image led to an improvement in the ability to detect a change in that image. This improvement was associated with decreased neural responses to the image, consistent with neuronal changes previously seen in studies of adaptation and expectation. We found that the magnitude of behavioral improvement was correlated with the magnitude of response suppression. Furthermore, this suppression of activity led to an increase in signal separation, providing evidence that a reduction in activity can improve stimulus encoding. Within populations of neurons, greater recent experience was associated with decreased trial-to-trial shared variability, indicating that a reduction in variability is a key means by which experience influences perception. Taken together, the results of our study contribute to an understanding of how recent visual experience can shape our perception and behavior through modulating activity patterns in the mid-level visual cortex. I was able to ask Michiel a few interview questions about developing the posters:Cool Infographics: What’s your background and how did you get into visualizing data?Michiel Dullaert: Years ago I have been working in the boardgame and puzzle industry. Part of my work was discovering new boardgames and puzzles and introduce them to the market. I was working for two different companies, both at the time that a new product of theirs really changed the market. For the boardgame industry I worked for the company that introduced a new type of boardgame (the game of Catan) that made playing boardgames a popular hobby. After that, I worked for a company that created puzzle magazines at the time the Sudoku puzzles was introduced. And here the same happened again, a new type of puzzle that changed the way people would puzzle and talk about puzzles. Both were very nice jobs, with a lot of enthusiastic conversations with customers about new products.In both companies I was working in the product development. I always had a large personal interest in the data behind the products. So it felt logic to do more with data, and I changed careers. I started working for a large company as an analyst. And it was not long that I found out that my colleagues were not interested in my reports. Although the reports contained interesting information it was shown in large tables and busy charts. People seemed to be not interested and if the looked into the numbers, they did not seem to understand them. My work did not seem to matter. The contrast between my previous jobs and the work as an analyst could not have been bigger. At that time I decided I would try to find a way to make people as enthusiastic about data as they were about boardgames and Sudoku puzzles. The first step was reading books on the topic of data visualization. Because I was working in the business intelligence field, the most logic choice were the books of Stephen Few. I bought them all and a whole new world opened for me. When Few was in the Netherlands to give a course I felt very lucky I could join. The workshops inspired me to learn more on this topic. So I bought more books and the next year I joined a class of Alberto Caïro. This gave me multiple perspectives on the same topic. The things I learned were applied to my work, and I was getting more and more responses on my visualizations. People actually started to read and understand them. My manager asked me to explain my choices in visualizations to my colleagues, so they could learn from me. And then I discovered how great it is to teach about data visualisation. I met a couple of UX designers and they inspired me learn more about that topic and to get an UX certification. The knowledge I gained in learning UX helped me in developing my own perspective on good data visualization. And in my workshop I try to inspire others to create such. For these workshops I wanted an overview of charts. Although there were already some great overviews, none of them had the point of view I was teaching in class. So I decided to design my own. That’s how Chart.Guide started. Cool Infographics: Who is the Chart Guide poster intended for?Michiel Dullaert: The website and the posters are intended for everyone who makes charts. For me it does not matter if you are working as a data-journalist or a data scientist, a infographic designer, project manager or a financial analyst. As long as you need help or inspiration when making charts or tables, Chart.Guide can help you. Online in the form of the website and offline in the form of the poster. Because I want to inspire as many people as possible I made the PDF of the poster free to download. I know that the poster is used in business departments, newsrooms and in school classrooms.Cool Infographics: What was your design process for the poster?Michiel Dullaert: The main source for updates to the poster or website, is the conversations I have with people I teach or work with. If they have a need for certain chart types, or make design mistakes, I try to add that topic. DPG Chart Chooser The first poster edition (picture DPG Chart Chooser) was just a collection of charts I created for my students. For each chart a few words on when to use the chart and, more important, it gave advice on what charts were not recommended. On the second edition (the first under the Chart.Guide brand) I added chart design tips. The reason, I saw people choose a good chart but then mess up the design. Last year, I got questions about maps, so I decided to add them on poster edition 4. Rearranging the layout of the Chart Guide The design process starts with insight I get from conversations I am having. Next step is cutting the old poster and rearranging everything. (see picture ChartGuide rearrange) now, will lead to more insights on the poster in the future. Although it make take some time to transform everything to the screen or paper. I still need to find some time to explain on the website why some charts are “not recommended”. Cool Infographics: You asked your followers to help choose the design of the new poster. How did that go?Michiel Dullaert: The poster is made to help people. So it seems to be logic to give them a role in the design process. As a UX designer I like to test my designs before releasing them. Most of these tests are done in class, because I like to observe the users in how they use the new design. The online voting was suggestion of a student. It did give me a lot of useful feedback. Especially when people wrote a lot of text explaining their choice. In this model, an edge between two nodes exists at discrete time $k in mathbb{N}^+$ with probability $hat{p}$ if this edge existed at $k-1$, and with probability $(1-hat{p})$ if this edge did not exist at $k-1$. Each edge is interpreted as a bidirectional communication link over which data between neighbors is shared. The initial communication graph is assumed to be an Erdos-Renyi random graph with parameters $(n,hat{p})$, hence we consider a emph{stationary} Markov model $overline{mathcal{G}}_{n,hat{p}}$. The asymptotic "$u_0$-expected flooding time" of $overline{mathcal{G}}_{n,hat{p}}$ is defined as the expected number of iterations required to transmit the data $d_0$ from $u_0$ nodes to $n$ nodes, in the limit as $n$ approaches infinity. Although most previous results on the asymptotic flooding time in graphical Markov models are either emph{almost sure} or emph{with high probability}, the bounds obtained here are emph{in expectation}. We successfully detect the product of two species masses as a feature to determine boundedness, gradient estimates, blow-up and $W^{j,infty}(1leq jleq 3)$-exponential convergence of classical solutions for the corresponding IBVP. More specifically, we first show generally a smallness on the product of both species masses, thus allowing one species mass to be suitably large, is sufficient to guarantee global boundedness, higher order gradient estimates and $W^{j,infty}$-convergence with rates of convergence to constant equilibria; and then, in a special case, we detect a straight line of masses on which blow-up occurs for large product of masses. Our findings provide new understandings about the underlying model, and thus, improve and extend greatly the existing knowledge relevant to this model. Full Article

Let $G$ be a finitely generated group admitting a $C^{1,alpha}$ action $ ho$ with a free orbit on the circle, and such that the logarithms of derivatives of group elements are uniformly bounded at some point of the circle. We prove that if $G$ has spherical growth bounded by $c n^{d-1}$ and if the function $1/alpha^d$ is integrable near zero, then under some mild technical assumptions on $alpha$, there is a sequence of exceptional $C^{1,alpha}$ actions of $G$ which converge to $ ho$ in the $C^1$ topology. As a consequence for a single diffeomorphism, we obtain that if the function $1/alpha$ is integrable near zero, then there exists a $C^{1,alpha}$ exceptional diffeomorphism of the circle. This corollary accounts for all previously known moduli of continuity for derivatives of exceptional diffeomorphisms. We also obtain a partial converse to our main result. For finitely generated free abelian groups, the existence of an exceptional action, together with some natural hypotheses on the derivatives of group elements, puts integrability restrictions on the modulus $alpha$. These results are related to a long-standing question of D. McDuff concerning the length spectrum of exceptional $C^1$ diffeomorphisms of the circle. Full Article

v4 PACT: Privacy Sensitive Protocols and Mechanisms for Mobile Contact Tracing. We created this document to suggest three functionalities on how we might best harness computing technologies to supporting the goals of public health organizations in minimizing morbidity and mortality associated with the spread of COVID-19, while protecting the civil liberties of individuals. In particular, this work advocates for a third-party free approach to assisted mobile contact tracing, because such an approach mitigates the security and privacy risks of requiring a trusted third party. We also explicitly consider the inferential risks involved in any contract tracing system, where any alert to a user could itself give rise to de-anonymizing information. More generally, we hope to participate in bringing together colleagues in industry, academia, and civil society to discuss and converge on ideas around a critical issue rising with attempts to mitigate the COVID-19 pandemic. v4 Unsupervised Domain Adaptation on Reading Comprehension. (arXiv:1911.06137v4 [cs.CL] UPDATED) By arxiv.org Published On :: Reading comprehension (RC) has been studied in a variety of datasets with the boosted performance brought by deep neural networks. However, the generalization capability of these models across different domains remains unclear. To alleviate this issue, we are going to investigate unsupervised domain adaptation on RC, wherein a model is trained on labeled source domain and to be applied to the target domain with only unlabeled samples. We first show that even with the powerful BERT contextual representation, the performance is still unsatisfactory when the model trained on one dataset is directly applied to another target dataset. To solve this, we provide a novel conditional adversarial self-training method (CASe). Specifically, our approach leverages a BERT model fine-tuned on the source dataset along with the confidence filtering to generate reliable pseudo-labeled samples in the target domain for self-training. On the other hand, it further reduces domain distribution discrepancy through conditional adversarial learning across domains. Extensive experiments show our approach achieves comparable accuracy to supervised models on multiple large-scale benchmark datasets. Full Article
v4 Biologic and Prognostic Feature Scores from Whole-Slide Histology Images Using Deep Learning. In this study, we introduced feature scores generated from hematoxylin and eosin histology images based on deep learning (DL) models developed for prostate pathology. We demonstrated that these feature scores were significantly prognostic for time to event endpoints (biochemical recurrence and cancer-specific survival) and had simultaneously molecular biologic associations to relevant genomic alterations and molecular subtypes using already trained DL models that were not previously exposed to the datasets of the current study. Further, we discussed the potential of such feature scores to improve the current tumor grading system and the challenges that are associated with tumor heterogeneity and the development of prognostic models from histology images. Our findings uncover the potential of feature scores from histology images as digital biomarkers in precision medicine and as an expanding utility for digital pathology. Full Article
v4 A Fast and Accurate Algorithm for Spherical Harmonic Analysis on HEALPix Grids with Applications to the Cosmic Microwave Background Radiation. (arXiv:1904.10514v4 [math.NA] UPDATED) By arxiv.org Published On :: The Hierarchical Equal Area isoLatitude Pixelation (HEALPix) scheme is used extensively in astrophysics for data collection and analysis on the sphere. The scheme was originally designed for studying the Cosmic Microwave Background (CMB) radiation, which represents the first light to travel during the early stages of the universe's development and gives the strongest evidence for the Big Bang theory to date. Refined analysis of the CMB angular power spectrum can lead to revolutionary developments in understanding the nature of dark matter and dark energy. In this paper, we present a new method for performing spherical harmonic analysis for HEALPix data, which is a central component to computing and analyzing the angular power spectrum of the massive CMB data sets. The method uses a novel combination of a non-uniform fast Fourier transform, the double Fourier sphere method, and Slevinsky's fast spherical harmonic transform (Slevinsky, 2019). For a HEALPix grid with $N$ pixels (points), the computational complexity of the method is $mathcal{O}(Nlog^2 N)$, with an initial set-up cost of $mathcal{O}(N^{3/2}log N)$. This compares favorably with $mathcal{O}(N^{3/2})$ runtime complexity of the current methods available in the HEALPix software when multiple maps need to be analyzed at the same time. Using numerical experiments, we demonstrate that the new method also appears to provide better accuracy over the entire angular power spectrum of synthetic data when compared to the current methods, with a convergence rate at least two times higher. v4 Keeping out the Masses: Understanding the Popularity and Implications of Internet Paywalls. The most common funding method, online advertising, is rife with well-known performance and privacy harms, and an intractable subject-agent conflict: many users do not want to see advertisements, depriving the site of needed funding. Because of these negative aspects of advertisement-based funding, paywalls are an increasingly popular alternative for websites. This shift to a "pay-for-access" web is one that has potentially huge implications for the web and society. Instead of a system where information (nominally) flows freely, paywalls create a web where high quality information is available to fewer and fewer people, leaving the rest of the web users with less information, that might be also less accurate and of lower quality. Despite the potential significance of a move from an "advertising-but-open" web to a "paywalled" web, we find this issue understudied. This work addresses this gap in our understanding by measuring how widely paywalls have been adopted, what kinds of sites use paywalls, and the distribution of policies enforced by paywalls. A partial list of our findings include that (i) paywall use is accelerating (2x more paywalls every 6 months), (ii) paywall adoption differs by country (e.g. 18.75% in US, 12.69% in Australia), (iii) paywalls change how users interact with sites (e.g. higher bounce rates, less incoming links), (iv) the median cost of an annual paywall access is $108 per site, and (v) paywalls are in general trivial to circumvent. Finally, we present the design of a novel, automated system for detecting whether a site uses a paywall, through the combination of runtime browser instrumentation and repeated programmatic interactions with the site. We intend this classifier to augment future, longitudinal measurements of paywall use and behavior. Full Article
v4 Machine learning topological phases in real space. (arXiv:1901.01963v4 [cond-mat.mes-hall] UPDATED) By arxiv.org Published On :: We develop a supervised machine learning algorithm that is able to learn topological phases for finite condensed matter systems from bulk data in real lattice space. The algorithm employs diagonalization in real space together with any supervised learning algorithm to learn topological phases through an eigenvector ensembling procedure. We combine our algorithm with decision trees and random forests to successfully recover topological phase diagrams of Su-Schrieffer-Heeger (SSH) models from bulk lattice data in real space and show how the Shannon information entropy of ensembles of lattice eigenvectors can be used to retrieve a signal detailing how topological information is distributed in the bulk. The discovery of Shannon information entropy signals associated with topological phase transitions from the analysis of data from several thousand SSH systems illustrates how model explainability in machine learning can advance the research of exotic quantum materials with properties that may power future technological applications such as qubit engineering for quantum computing. v4 Performance of the smallest-variance-first rule in appointment sequencing. (arXiv:1812.01467v4 [math.PR] UPDATED) By arxiv.org Published On :: A classical problem in appointment scheduling, with applications in health care, concerns the determination of the patients' arrival times that minimize a cost function that is a weighted sum of mean waiting times and mean idle times. One aspect of this problem is the sequencing problem, which focuses on ordering the patients. We assess the performance of the smallest-variance-first (SVF) rule, which sequences patients in order of increasing variance of their service durations. While it was known that SVF is not always optimal, it has been widely observed that it performs well in practice and simulation. We provide a theoretical justification for this observation by proving, in various settings, quantitative worst-case bounds on the ratio between the cost incurred by the SVF rule and the minimum attainable cost. We also show that, in great generality, SVF is asymptotically optimal, i.e., the ratio approaches 1 as the number of patients grows large. While evaluating policies by considering an approximation ratio is a standard approach in many algorithmic settings, our results appear to be the first of this type in the appointment scheduling literature. v4 SilhoNet: An RGB Method for 6D Object Pose Estimation. Methods using RGB-D data have shown great success in solving this problem. However, there are situations where cost constraints or the working environment may limit the use of RGB-D sensors. When limited to monocular camera data only, the problem of object pose estimation is very challenging. In this work, we introduce a novel method called SilhoNet that predicts 6D object pose from monocular images. We use a Convolutional Neural Network (CNN) pipeline that takes in Region of Interest (ROI) proposals to simultaneously predict an intermediate silhouette representation for objects with an associated occlusion mask and a 3D translation vector. The 3D orientation is then regressed from the predicted silhouettes. We show that our method achieves better overall performance on the YCB-Video dataset than two state-of-the art networks for 6D pose estimation from monocular image input. Full Article

For covariance functions that are Green's functions of elliptic boundary value problems and homogeneously-distributed sampling points, we show how to identify a subset $S subset { 1 , dots , N }^2$, with $# S = O ( N log (N) log^{d} ( N /epsilon ) )$, such that the zero fill-in incomplete Cholesky factorisation of the sparse matrix $Theta_{ij} 1_{( i, j ) in S}$ is an $epsilon$-approximation of $Theta$. This factorisation can provably be obtained in complexity $O ( N log( N ) log^{d}( N /epsilon) )$ in space and $O ( N log^{2}( N ) log^{2d}( N /epsilon) )$ in time, improving upon the state of the art for general elliptic operators; we further present numerical evidence that $d$ can be taken to be the intrinsic dimension of the data set rather than that of the ambient space. The algorithm only needs to know the spatial configuration of the $x_{i}$ and does not require an analytic representation of $G$. Furthermore, this factorization straightforwardly provides an approximate sparse PCA with optimal rate of convergence in the operator norm. Hence, by using only subsampling and the incomplete Cholesky factorization, we obtain, at nearly linear complexity, the compression, inversion and approximate PCA of a large class of covariance matrices. By inverting the order of the Cholesky factorization we also obtain a solver for elliptic PDE with complexity $O ( N log^{d}( N /epsilon) )$ in space and $O ( N log^{2d}( N /epsilon) )$ in time, improving upon the state of the art for general elliptic operators. v4 TRPV4 helps Piezo1 put the squeeze on pancreatic acinar cells By www.jci.org Published On :: Alterations in calcium signaling in pancreatic acinar cells can result in pancreatitis. Although pressure changes in the pancreas can elevate cytosolic calcium (Ca2+) levels, it is not known how transient pressure-activated elevations in calcium can cause prolonged calcium changes and consequent pancreatitis. In this issue of the JCI, Swain et al. describe roles for the mechanically activated plasma membrane calcium channels Piezo1 and transient receptor potential vanilloid subfamily 4 (TRPV4) in acinar cells. The authors used genetic deletion models and cell culture systems to investigate calcium signaling. Notably, activation of the Piezo1-dependent TRPV4 pathway was independent of the cholecystokinin (CCK) stimulation pathway. These results elegantly resolve an apparent discrepancy in calcium signaling and the pathogenesis of pancreatitis in pancreatic acinar cells. Full Article
