AFC U23 Asian Cup 2023

Asia’s rising stars discovered their opponents following the conclusion of the AFC U23 Asian Cup Uzbekistan 2022 Qualifiers Draw in Tashkent. The Qualifiers are scheduled for October 23 to 31, 2021. Out of 47 AFC Member Associations, a total of 42 teams entered the competition and were drawn into nine groups of four teams and two groups of three team. The 11 group winners and four best second-placed teams will seal their qualification to the Finals, along with Uzbekistan who have already confirmed their place as hosts. The Finals will be held between 1-19 June 2022. Players born on or after 1 January 1999 are eligible to compete in the tournament.



India is hoping to qualify to the Finals of the tournament for the first time since its inception and was drawn in Group E alongside Kyrgyz Republic, Oman and UAE, who will host the group.

Representative cancer-associated U2AF2 mutations alter RNA interactions and splicing [Molecular Bases of Disease]

High-throughput sequencing of hematologic malignancies and other cancers has revealed recurrent mis-sense mutations of genes encoding pre-mRNA splicing factors. The essential splicing factor U2AF2 recognizes a polypyrimidine-tract splice-site signal and initiates spliceosome assembly. Here, we investigate representative, acquired U2AF2 mutations, namely N196K or G301D amino acid substitutions associated with leukemia or solid tumors, respectively. We determined crystal structures of the wild-type (WT) compared with N196K- or G301D-substituted U2AF2 proteins, each bound to a prototypical AdML polypyrimidine tract, at 1.5, 1.4, or 1.7 Å resolutions. The N196K residue appears to stabilize the open conformation of U2AF2 with an inter-RNA recognition motif hydrogen bond, in agreement with an increased apparent RNA-binding affinity of the N196K-substituted protein. The G301D residue remains in a similar position as the WT residue, where unfavorable proximity to the RNA phosphodiester could explain the decreased RNA-binding affinity of the G301D-substituted protein. We found that expression of the G301D-substituted U2AF2 protein reduces splicing of a minigene transcript carrying prototypical splice sites. We further show that expression of either N196K- or G301D-substituted U2AF2 can subtly alter splicing of representative endogenous transcripts, despite the presence of endogenous, WT U2AF2 such as would be present in cancer cells. Altogether, our results demonstrate that acquired U2AF2 mutations such as N196K and G301D are capable of dysregulating gene expression for neoplastic transformation.

Location of Cu2+ in CHA zeolite investigated by X-ray diffraction using the Rietveld/maximum entropy method

Rietveld/MEM analysis applied to synchrotron powder X-ray diffraction data of dehydrated CHA zeolites with catalytically active Cu2+ reveals Cu2+ in both the six- and eight-membered rings in the CHA framework, providing the first complete structural model that accounts for all Cu2+. Density functional theory calculations are used to corroborate the experimental structure and to discuss the Cu2+ coordination in terms of the Al distribution in the framework.

α-SrZn5-Type solid solution, BaZn2.6Cu2.4

Single crystals of the title compound barium zinc copper, BaCu2.6Zn2.4, were obtained from a sample prepared by heating metal chips of Ba, Cu, and Zn in an Ar atmosphere up to 973 K, followed by slow cooling. Single-crystal X-ray structure analysis revealed that BaCu2.6Zn2.4 crystallizes in an ortho­rhom­bic cell [a = 12.9858 (3), b = 5.2162 (1), and c = 6.6804 (2) Å] with an α-SrZn5-type structure (space group Pnma). The three-dimensional framework consists of Cu and Zn atoms, with Ba atoms in the tunnels extending in the b-axis direction. Although the Ba atom is larger than the Sr atom, the cell volume of BaCu2.6Zn2.4 [452.507 (19) Å3] is smaller than that of α-SrZn5 [466.08 Å3]. This decrease in volume can be attributed to the partial substitution of Cu atoms by Zn atoms in the framework because the Cu—Zn and Cu—Cu bonds are shorter than the Zn—Zn bond. The increase in Ba—Zn inter­atomic distances from the Sr—Zn distances is cancelled out by the partial replacement of Zn with Cu atoms, which leads to shorter average Ba—Zn/Cu distances.

Structure of the MICU1–MICU2 heterodimer provides insights into the gatekeeping threshold shift

Mitochondrial calcium uptake proteins 1 and 2 (MICU1 and MICU2) mediate mitochondrial Ca2+ influx via the mitochondrial calcium uniporter (MCU). Its molecular action for Ca2+ uptake is tightly controlled by the MICU1–MICU2 heterodimer, which comprises Ca2+ sensing proteins which act as gatekeepers at low [Ca2+] or facilitators at high [Ca2+]. However, the mechanism underlying the regulation of the Ca2+ gatekeeping threshold for mitochondrial Ca2+ uptake through the MCU by the MICU1–MICU2 heterodimer remains unclear. In this study, we determined the crystal structure of the apo form of the human MICU1–MICU2 heterodimer that functions as the MCU gatekeeper. MICU1 and MICU2 assemble in the face-to-face heterodimer with salt bridges and me­thio­nine knobs stabilizing the heterodimer in an apo state. Structural analysis suggests how the heterodimer sets a higher Ca2+ threshold than the MICU1 homodimer. The structure of the heterodimer in the apo state provides a framework for understanding the gatekeeping role of the MICU1–MICU2 heterodimer.
