Novel Proteomic Profiling of Epididymal Extracellular Vesicles in the Domestic Cat Reveals Proteins Related to Sequential Sperm Maturation with Differences Observed between Normospermic and Teratospermic Individuals [Research]

Extracellular vesicles (EVs) secreted by the epididymal epithelium transfer to spermatozoa key proteins that are essential in promoting motility and subsequent fertilization success. Using the domestic cat model, the objectives were to (1) characterize and compare protein content of EVs between segments of the epididymis, and (2) compare EV protein compositions between normo- and teratospermic individuals (producing >60% of abnormal spermatozoa). Epididymal EVs from adult cats were isolated and assessed via liquid chromatography tandem MS. Both male types shared 3008 proteins in total, with 98 and 20 EV proteins unique to normospermic and teratospermic males, respectively. Expression levels of several proteins changed between epididymal segments in both male types. Several proteins in both groups were related to sperm motility (e.g. hexokinase 1, adenylate kinase isoenzyme) and zona pellucida or oolemma binding (e.g. disintegrin and metalloproteinase domain proteins, zona binding proteins 1 and 2). Interestingly, seven cauda-derived EV proteins trended downward in teratospermic compared with normospermic males, which may relate to poor sperm quality. Collective results revealed, for the first time, EV proteins related to sequential sperm maturation with differences observed between normospermic and teratospermic individuals.

Sequential Activation of Lateral Hypothalamic Neuronal Populations during Feeding and Their Assembly by Gamma Oscillations

Neural circuits supporting innate behaviors, such as feeding, exploration, and social interaction, intermingle in the lateral hypothalamus (LH). Although previous studies have shown that individual LH neurons change their firing relative to the baseline during one or more behaviors, the firing rate dynamics of LH populations within behavioral episodes and the coordination of behavior-related LH populations remain largely unknown. Here, using unsupervised graph-based clustering of LH neurons firing rate dynamics in freely behaving male mice, we identified distinct populations of cells whose activity corresponds to feeding, specific times during feeding bouts, or other innate behaviors—social interaction and novel object exploration. Feeding-related cells fired together with a higher probability during slow and fast gamma oscillations (30–60 and 60–90 Hz) than during nonrhythmic epochs. In contrast, the cofiring of neurons signaling other behaviors than feeding was overall similar between slow gamma and nonrhythmic epochs but increased during fast gamma oscillations. These results reveal a neural organization of ethological hierarchies in the LH and point to behavior-specific motivational systems, the dysfunction of which may contribute to mental disorders.

Vice President Mike Pence owes Iowans more than a photo-op when he visits Des Moines today, according to Joe Biden’s campaign.

“Iowans are seeing up close the most consequential failure of government in modern American history,” said Kate Bedingfeld, spokeswoman for the former vice president and presumptive Democratic presidential nominee.

“With nearly 300,000 Iowans filing for unemployment, rural hospitals on life support, Latino communities disproportionately suffering and workers on the job without sufficient protection, Mike Pence owes Iowans more than a photo-op — he owes them answers,” she said.

Pence, head of the White House coronavirus task force, is scheduled to meet with Iowa Gov. Kim Reynolds and U.S. Sens. Chuck Grassley and Joni Ernst, all Republicans, as well as with faith, farm and food production leaders.

Pence will talk to faith leaders about how they are using federal and state guidelines to open their houses of worship in a safe and responsible manner.

Later, he will go to Hy-Vee’s headquarters in West Des Moines for a roundtable discussion with agriculture and food supply leaders to discuss steps being taken to ensure the food supply remains secure.

Pence has called Iowa a “success story” in its response to the COVID-19, but Bedingfeld said the Trump administration failed to protect Iowa families from the virus that has claimed the lives of 231 Iowans.

“From devastating losses across the state, at meatpacking plants to rural communities, one thing is clear — it’s Iowans and the American people who are paying the price for the Trump administration’s denials and delays in response to this pandemic,” she said.

“Instead of listening to our own intelligence agencies and public health experts, Donald Trump was fed dangerous propaganda from the Chinese Communist Party — and he bought it,” she said. “Iowans deserve better — they deserve Joe Biden.”

For his part, Grassley said he welcomes the discussion with Pence.

“There’s much work to be done, and the pandemic is disrupting all of our communities,” Grassley said. “It’s important to hear directly from those who help feed the nation and the world.”

Ernst also is looking forward to the discussion of how Iowa is working to protect the health and safety of Iowa’s families and communities while reopening the state’s economy.

“We continue to take an all-hands-on-deck approach to tackling this pandemic,” she said. “Together, we will get through this.”

Sequential state elements in triple-mode redundant (TMR) state machines

The disclosure relates generally to triple-redundant sequential state (TRSS) machines formed as integrated circuits on a semiconductor substrate, such as CMOS, and computerized methods and systems of designing the triple-redundant sequential state machines. Of particular focus in this disclosure are sequential state elements (SSEs) used to sample and hold bit states. The sampling and holding of bits states are synchronized by a clock signal thereby allowing for pipelining in the TRSS machines. In particular, the clock signal may oscillate between a first clock state and a second clock state to synchronize the operation of the SSE according to the timing provided by the clock states. The SSEs has a self-correcting mechanism to protect against radiation induced soft errors. The SSE may be provided in a pipeline circuit of a TRSS machine to receive and store a bit state of bit signal generated by combinational circuits within the pipeline circuit.

Method and sequential monitoring overlay system for track circuits

A sequential monitoring system is for an interlocking logic system and a track circuit system including a plurality of track circuits. The sequential monitoring system includes an interface between the interlocking logic system and the track circuit system; and a processor structured to monitor a state of each of the track circuits, validate a sequence of state changes of the track circuits, and interrupt and correct invalid track circuit state indications between the track circuit system and the interlocking logic system. The methods include the sequential application of vacuum and mechanical force through a two-stroke process performed by a lamination apparatus having one or more force-producing stroke cylinders. The sealed chamber may be evacuated of air by application of a vacuum. Abstract:
We propose a method to generate random distributions with known quantile distribution, or, more generally, with known distribution for some form of generalized quantile. The method takes inspiration from the random Sequential Barycenter Array distributions (SBA) proposed by Hill and Monticino (1998) which generates a Random Probability Measure (RPM) with known expected value. We define the Sequential Quantile Array (SQA) and show how to generate a random SQA from which we can derive RPMs. The distribution of the generated SQA-RPM can have full support and the RPMs can be both discrete, continuous and differentiable. We face also the problem of the efficient implementation of the procedure that ensures that the approximation of the SQA-RPM by a finite number of steps stays close to the SQA-RPM obtained theoretically by the procedure. Finally, we compare SQA-RPMs with similar approaches as Polya Tree.

Sequential Aggregation of Probabilistic Forecasts -- Applicaton to Wind Speed Ensemble Forecasts. (arXiv:2005.03540v1 [stat.AP])

In the field of numerical weather prediction (NWP), the probabilistic distribution of the future state of the atmosphere is sampled with Monte-Carlo-like simulations, called ensembles. These ensembles have deficiencies (such as conditional biases) that can be corrected thanks to statistical post-processing methods. Several ensembles exist and may be corrected with different statistiscal methods. A further step is to combine these raw or post-processed ensembles. The theory of prediction with expert advice allows us to build combination algorithms with theoretical guarantees on the forecast performance. This article adapts this theory to the case of probabilistic forecasts issued as step-wise cumulative distribution functions (CDF). The theory is applied to wind speed forecasting, by combining several raw or post-processed ensembles, considered as CDFs. The second goal of this study is to explore the use of two forecast performance criteria: the Continous ranked probability score (CRPS) and the Jolliffe-Primo test. Comparing the results obtained with both criteria leads to reconsidering the usual way to build skillful probabilistic forecasts, based on the minimization of the CRPS. Minimizing the CRPS does not necessarily produce reliable forecasts according to the Jolliffe-Primo test. The Jolliffe-Primo test generally selects reliable forecasts, but could lead to issuing suboptimal forecasts in terms of CRPS. It is proposed to use both criterion to achieve reliable and skillful probabilistic forecasts.

Asymptotic genealogies of interacting particle systems with an application to sequential Monte Carlo

Abstract:
We study weighted particle systems in which new generations are resampled from current particles with probabilities proportional to their weights. This covers a broad class of sequential Monte Carlo (SMC) methods, widely-used in applied statistics and cognate disciplines. We consider the genealogical tree embedded into such particle systems, and identify conditions, as well as an appropriate time-scaling, under which they converge to the Kingman $n$-coalescent in the infinite system size limit in the sense of finite-dimensional distributions. Thus, the tractable $n$-coalescent can be used to predict the shape and size of SMC genealogies, as we illustrate by characterising the limiting mean and variance of the tree height. SMC genealogies are known to be connected to algorithm performance, so that our results are likely to have applications in the design of new methods as well. Our conditions for convergence are strong, but we show by simulation that they do not appear to be necessary.

New $G$-formula for the sequential causal effect and blip effect of treatment in sequential causal inference

Abstract:
In sequential causal inference, two types of causal effects are of practical interest, namely, the causal effect of the treatment regime (called the sequential causal effect) and the blip effect of treatment on the potential outcome after the last treatment. The well-known $G$-formula expresses these causal effects in terms of the standard parameters. In this article, we obtain a new $G$-formula that expresses these causal effects in terms of the point observable effects of treatments similar to treatment in the framework of single-point causal inference. Based on the new $G$-formula, we estimate these causal effects by maximum likelihood via point observable effects with methods extended from single-point causal inference. We are able to increase precision of the estimation without introducing biases by an unsaturated model imposing constraints on the point observable effects. We are also able to reduce the number of point observable effects in the estimation by treatment assignment conditions.

Abstract:
In this paper, we consider the knot-matching problem arising in computational forestry. The knot-matching problem is an important problem that needs to be solved to advance the state of the art in automatic strength prediction of lumber. We show that this problem can be formulated as a quadripartite matching problem and develop a sequential decision model that admits efficient parameter estimation along with a sequential Monte Carlo sampler on graph matching that can be utilized for rapid sampling of graph matching. We demonstrate the effectiveness of our methods on 30 manually annotated boards and present findings from various simulation studies to provide further evidence supporting the efficacy of our methods.

Sequential Monte Carlo Samplers with Independent Markov Chain Monte Carlo Proposals

Abstract:
Sequential Monte Carlo (SMC) methods for sampling from the posterior of static Bayesian models are flexible, parallelisable and capable of handling complex targets. However, it is common practice to adopt a Markov chain Monte Carlo (MCMC) kernel with a multivariate normal random walk (RW) proposal in the move step, which can be both inefficient and detrimental for exploring challenging posterior distributions. We develop new SMC methods with independent proposals which allow recycling of all candidates generated in the SMC process and are embarrassingly parallelisable. A novel evidence estimator that is easily computed from the output of our independent SMC is proposed. Our independent proposals are constructed via flexible copula-type models calibrated with the population of SMC particles. We demonstrate through several examples that more precise estimates of posterior expectations and the marginal likelihood can be obtained using fewer likelihood evaluations than the more standard RW approach.

Control of Type I Error Rates in Bayesian Sequential Designs

Abstract:
Bayesian approaches to phase II clinical trial designs are usually based on the posterior distribution of the parameter of interest and calibration of certain threshold for decision making. If the posterior probability is computed and assessed in a sequential manner, the design may involve the problem of multiplicity, which, however, is often a neglected aspect in Bayesian trial designs. To effectively maintain the overall type I error rate, we propose solutions to the problem of multiplicity for Bayesian sequential designs and, in particular, the determination of the cutoff boundaries for the posterior probabilities. We present both theoretical and numerical methods for finding the optimal posterior probability boundaries with $alpha$ -spending functions that mimic those of the frequentist group sequential designs. The theoretical approach is based on the asymptotic properties of the posterior probability, which establishes a connection between the Bayesian trial design and the frequentist group sequential method. The numerical approach uses a sandwich-type searching algorithm, which immensely reduces the computational burden. We apply least-square fitting to find the $alpha$ -spending function closest to the target. We discuss the application of our method to single-arm and double-arm cases with binary and normal endpoints, respectively, and provide a real trial example for each case.

Randomized Trial of Oral Versus Sequential IV/Oral Antibiotic for Acute Pyelonephritis in Children

The standard initial management for infants and children with acute pyelonephritis is intravenous antibiotic treatment.

Our results support the use of an oral cefixime treatment of initial episodes of acute pyelonephritis involving a gram-negative bacteria strain in children aged 1 month to 3 years who are without urological abnormalities and without clinical hemodynamic impairment. (Read the full article)

Wrong Constraint Values in Sequential Cell Characterization

Hi,

I am trying to characterize a D flip-flop for low voltage operation (0.6V) using Cadence Liberate (V16). This is a positive edge triggered D flip flop based on true-single-phase clocking scheme. After the characterization, the measurements reported for hold constraint arcs seem to deviate significantly from its (spectre) spice simulation.

I specifically used set_var constraint_combinational 2 in the settings, in case the Bisection pass/fail mode fails to capture the constraints. In my spice simulation, the hold_rise (D=1, CLK=R, Q=R) arc at-least requires ~250 ps for minimum CLK/D slew combination (for the  by default smallest capacitive load as per Liberate)  while Liberate reports only ~30 ps. The define_cell template to this flip flop is pretty generic, which does not have any user specified arcs. So which settings most likely affecting the constraint measurements in Liberate and how can I debug this issue ?
