The next morning we’d pull thick squares of Post-it notes out of our holiday socks and know it was from Dad, even if he’d signed it “P. A. Perclip.” Last minute gift buying is a fine tradition. Some may see the practice as thoughtless; I would argue it can be rooted in sweetness. After all, a last minute present is still a present. And now that flying has made it increasingly difficult to travel with gifts—airline luggage charges, unrelenting TSA clerks—you may as well just grab that stuff when you arrive, if you happen to be flying into Portland. Visitors may not know that our airport requires shops to maintain “street pricing,” so you won’t pay more for goods at PDX than you would in town. If you’re flying out, you’re still sitting pretty—provided they’ll let you on the plane with ten boozy advent calendars sticking precariously out of a Powell’s tote. Portland airport has seen to it that even last minute presents from PDX are—by nature of the shops onhand—thoughtful, lovely, and local. Pre-security No one is suggesting you make a trip to the airport just to shop—well, other than the Port of Portland; they would probably support the idea—but those arriving at PDX have their pick of both concourse and “the garden” shops. In keeping with our love of last minute office supplies, you’ll want to check out Paper Epiphanies, which packs an impressive variety of journals, art books, design periodicals, and a rainbow wall of markers (!) into a cute corner shop. The woman-owned business specializes in femme-empowerment messages, so you’ll find trendy books about feminist philosophy, art, and activism. On a practical level, this shop also has an entire wall of greeting cards. Pro tip: Want to hear more from someone who lives far away? A stationary set is a sweet incentive. Paper Epiphanies suzette smith Orox Leather is an epicenter of cool in this area. Not only does it sell handmade leather goods ($250 purse for when you’ve really pissed someone off; $10 leather coasters for the uncle who wants you to put one under your drink), but the people behind Orox also invited their fellow Latinx and Oaxaqueño makers from the Portland area to sell items there as well. So you can get dreamy little half-pints of Nico’s Ice Cream from a freezer in the back, and sampler packs of HAB hot sauce from shelves in the front. It’s about community, but it also appears to be about having amazing taste. Just next door, you’ll find the delicious and vegan AND gluten-free goodies of Missionary Chocolates. They have a lot on offer, but we’ll vouch for the chocolate-covered sandwich cookies. Further in, the deeply basic, but super crowd-pleasing Hello from Portland store carries THE THING YOU NEED when basically any Portland-y / Oregon-y present will do. A “Keep Portland Weird” mug it is. Concourses B and C Post-security shops have an obvious advantage because whatever you pick up after TSA’s disrobe/re-robe challenge doesn’t have to meet its dogmatic restrictions. That means booze! The GENIUSES at Straightaway Cocktails have created a craft cocktail advent calendar that has seemingly no attachment to any particular religion or any assigned set of dates. It’s just a nicely designed box with 12 mystery doors, and behind each is an alcoholic drink in a can. This is the present very good adult siblings give one another to weather the hard times of visiting family. If it helps, it’s of a similar size and shape to an oversize Bible (wink). Straightaway already has these little masterpieces available at their kiosk. Cocktail advent calendar straightaway cocktails Concourse C has a lot going for it because it’s also where you’ll find the PDX Powell’s Books pop-up. The selection has nothing on City of Books, but there’s always some interesting titles selected by staff, showcases of local authors, whatever nonfiction book everyone is mad about, a variety of card games, and the new Stephen King in stock. Now, your gate dictates what concourse you’re flying from, so some concourse options simply won’t be available, depending what side you’re on. However, if your mother sternly warned you “not to come home again without a Pendleton wool blanket”—they have booths in both C or on the way to D and E. Similarly, Westward Whiskey also put down roots on either side of security (C & E). Concourses D and E Stronghold of cute, local, and punk Tender Loving Empire manifested two locations as well, in both D and E, on the same side of security. TLE has a long history of reliably solid care packages, tailored to foodies, self-care softies, and even people who just like Jacobsen Sea Salt. They’ve got a basket for it, and if they don’t, they can make you one. While I am in no way related to Smith Tea founder Steven Smith, I do frequently receive his company’s tea as a gag gift, and I love that. Please continue this very funny joke, because the tea is a little on the spendy side. The full-leaf flavors are perfect for splurges and special occasions, though, and the company always has interesting collabs in the works. This fall, they brought back a Westward Whiskey team up, Ode to Whiskey, where black tea is aged in wet whiskey barrels to convey tasting notes/vibes—it’s perfect for your moody, lettered cousin. That’s your winter gift gauntlet handled! We hope this list helped; it’s not really just for the holidays, because there are plenty more times throughout the year when you’ll want to take along a gift to say, “I’m sorry I didn’t get a hotel,” or “so it turns out we both have the same dad.” Life can be messy, but a last minute present is still a present. dx Exploiting Friedel pairs to interpret scanning 3DXRD data from complex geological materials By journals.iucr.org Published On :: 2024-11-08 The present study introduces a processing strategy for synchrotron scanning 3D X-ray diffraction (s3DXRD) data, aimed at addressing the challenges posed by large, highly deformed, polyphase materials such as crystalline rocks. Leveraging symmetric Bragg reflections known as Friedel pairs, our method enables diffraction events to be precisely located within the sample volume. This method allows for fitting the phase, crystal structure and unit-cell parameters at the intra-grain scale on a voxel grid. The processing workflow incorporates several new modules, designed to (i) efficiently match Friedel pairs in large s3DXRD datasets containing up to 108 diffraction peaks; (ii) assign phases to each pixel or voxel, resolving potential ambiguities arising from overlap in scattering angles between different crystallographic phases; and (iii) fit the crystal orientation and unit cell locally on a point-by-point basis. We demonstrate the effectiveness of our technique on fractured granite samples, highlighting the ability of the method to characterize complex geological materials and show their internal structure and mineral composition. Additionally, we include the characterization of a metal gasket made of a commercial aluminium alloy, which surrounded the granite sample during experiments. The results show the effectiveness of the technique in recovering information about the internal texture and residual strain of materials that have undergone high levels of plastic deformation. dx Scaling and merging macromolecular diffuse scattering with mdx2 By journals.iucr.org Published On :: 2024-04-12 Diffuse scattering is a promising method to gain additional insight into protein dynamics from macromolecular crystallography experiments. Bragg intensities yield the average electron density, while the diffuse scattering can be processed to obtain a three-dimensional reciprocal-space map that is further analyzed to determine correlated motion. To make diffuse scattering techniques more accessible, software for data processing called mdx2 has been created that is both convenient to use and simple to extend and modify. mdx2 is written in Python, and it interfaces with DIALS to implement self-contained data-reduction workflows. Data are stored in NeXus format for software interchange and convenient visualization. mdx2 can be run on the command line or imported as a package, for instance to encapsulate a complete workflow in a Jupyter notebook for reproducible computing and education. Here, mdx2 version 1.0 is described, a new release incorporating state-of-the-art techniques for data reduction. The implementation of a complete multi-crystal scaling and merging workflow is described, and the methods are tested using a high-redundancy data set from cubic insulin. It is shown that redundancy can be leveraged during scaling to correct systematic errors and obtain accurate and reproducible measurements of weak diffuse signals. Full Article text

dx Test of Nikon D800e, a DX lens and a Teleconverter By niels-henriksen.blogspot.com Published On :: Thu, 02 Aug 2012 15:59:00 +0000 I was fortunate to finally get a Nikon D800e (36 Mpx) after a brief 3 month wait. I didn't, and I also think its the same with Nikon, expect that there would be such a huge demand for this camera. I don't know why Nikon seemed to leap so far ahead with its next generation but I'm grateful. I think there are millions (well a whole bunch anyway) more who also think the same.This is a full frame camera and a DX lens will work but is cropped due to limited image area. The camera even has mode where it will auto detect a DX lens and crop accordingly. Knowing that there is still a little more image available with the DX crop I decided not to crop and use whatever was available. Nikon D800e with 70-200mm f2.8 VR. The image on right is actual size of window pane My favorite DX lens is the 18-200mm f3.5 VR and as I travel around I notice others who like this lens. While it may have some pincushion and barrel distortion, thanks to Photoshop lens correction, this is almost all eliminated. Nikon D800e with 70-200mm f2.8 VR set to 1.2 crop. The image on right is actual size of window pane. I didn't realize at first that the crop factor also applied to FX lens. It was while I was performing a resolution test with my D300 and the new D800e by using both the 18-200mm f3.5 VR and the 70-200mm f2.8 VR that I noticed that when full zoomed on the DX lens that vignetting seemed to disappear, but looked like a dime at 18mm.This excited me more than the current test and therefore I decided to pursue this further.The 18-200mm is really like a 24-300mm on a DX camera due to the 1.5 crop factor and this range will meet about 90% of my needs. With a 1.4 teleconverter on the FX camera it's now almost the same range as on a DX camera.The test I performed was hand-held as I didn't need with this test to check for perfect resolution. I stood in front of a large apartment building in the parking lot across the street and remained at the same position while I changed the lens focal length.In the 1st test, see film strip below, there is significant vignetting in the 18mm and slowly reduces as the focal length is increased. Even at 200mm there is still slight vignetting at the corners. When the teleconverter was added then magic seemed to appear. At 18mm there is only slight vignetting at the corners and disappears by 24mm. The film strip below shows the changes with teleconverter added. Now a favorite lens becomes useful on my full frame camera.Most lens that have such a wide range of focal lens is all about compromises when being designed and built. The same with the 18-200mm f3.5 VR. Photoshop easily handles the barrel distortion. But this lens at the glass edge, maybe more is being used now, also has a fair amount of chromatic appellation. This is also handled well with Photoshop. Since most artistic images don't need to be sharp or well defined at the corners of a photograph then any additional distortion caused by using more lens area is not really a problem with digital-editing.So when and if you do decide to go full frame one day then your DX glass can be useful with a teleconverter.The reason this works is because the lens is now further from the sensor and therefore increases in size on sensor chip. Take a magnifying glass and hold it to show an image appearing upside down on a piece of paper. Now move the magnifying glass further away and watch how the image on the paper grows larger. dx Novel pathogenic PDX1 gene variant in a Korean family with maturity-onset diabetes of the young [VARIANT DISCREPANCY RESOLUTION] By molecularcasestudies.cshlp.org Published On :: 2024-01-10T08:13:38-08:00 The diagnosis of maturity-onset diabetes of the young (MODY), a monogenic form of diabetes mellitus caused by a mutation in a single gene, is often uncertain until genetic testing is performed. We report a 13-yr-old Korean boy who was initially diagnosed with type 2 diabetes (T2DM). MODY was suspected because of his nonobese body habitus and family history of multiple affected members. Targeted panel sequencing of all MODY-related genes was performed using the NextSeq 550Dx platform (Illumina). Sanger sequencing was performed using blood samples from the parents, siblings, and other relatives. A frameshift variant in the 3' region of the last exon of PDX1 was detected in the patient and his family members with diabetes. PP1_Moderate criterion was applied and this variant was confirmed to be the genetic cause of diabetes in the family and classified as likely pathogenic. The study highlights the importance of genetic testing for nonobese, early-onset diabetic patients with multiple affected family members. Increased awareness and aggressive genetic testing for MODY are needed. Full Article
dx Novel inherited CDX2 variant segregating in a family with diverse congenital malformations of the genitourinary system [RAPID COMMUNICATION] By molecularcasestudies.cshlp.org Published On :: 2024-01-10T08:13:38-08:00 Anorectal malformations (ARMs) constitute a group of congenital defects of the gastrointestinal and urogenital systems. They affect males and females, with an estimated worldwide prevalence of 1 in 5000 live births. These malformations are clinically heterogeneous and can be part of a syndromic presentation (syndromic ARM) or as a nonsyndromic entity (nonsyndromic ARM). Despite the well-recognized heritability of nonsyndromic ARM, the genetic etiology in most patients is unknown. In this study, we describe three siblings with diverse congenital anomalies of the genitourinary system, anemia, delayed milestones, and skeletal anomalies. Genome sequencing identified a novel, paternally inherited heterozygous Caudal type Homeobox 2 (CDX2) variant (c.722A > G (p.Glu241Gly)), that was present in all three affected siblings. The variant identified in this family is absent from population databases and predicted to be damaging by most in silico pathogenicity tools. So far, only two other reports implicate variants in CDX2 with ARMs. Remarkably, the individuals described in these studies had similar clinical phenotypes and genetic alterations in CDX2. CDX2 encodes a transcription factor and is considered the master regulator of gastrointestinal development. This variant maps to the homeobox domain of the encoded protein, which is critical for interaction with DNA targets. Our finding provides a potential molecular diagnosis for this family's condition and supports the role of CDX2 in anorectal anomalies. It also highlights the clinical heterogeneity and variable penetrance of ARM predisposition variants, another well-documented phenomenon. Finally, it underscores the diagnostic utility of genomic profiling of ARMs to identify the genetic etiology of these defects. Full Article

However, as a long time fan of Wicked, that could change as Xfinity has released a new 4DX ad ahead of Wicked the movie's November 22 release date. The ad is a short film titled Stay Connected to Your Dreams and was directed by Alice Brooks, Wicked's director of photography. It centers on Wicked's leads Cynthia Erivo and Ariana Grande, who play Elphaba and Glinda, respectively. The short film shows viewers Erivo and Grande's childhoods and their love of Wicked, Grande's decade and a half old tweet wishing she'd one day play Glinda and the calls when each actor learned they'd been cast in the movie. According to Xfinity, the 4DX film will allow the audience to "feel the magic of Oz" and includes synchronized motion seats, wind, lightning and, of course, bubbles (IYKYK). The company also claims it's the first 4DX spot to be played nationwide. While we've not always been fans of 4DX, Wicked lovers might enjoy the slightly more immersive experience. You can experience the short film in select 4DX Regal theaters before the movie premieres or sans-4DX here. The second part of Wicked should arrive next year, on November 21, 2025. The 3D strain field of a tin grain, located within a sample of approximately 70 grains, is analysed and compared across reconstruction methods. It is verified that the two proposed methods remedy these problems by taking the spatial properties of the inverse problem into account.
