Unveiling the Secrets of Big Data Projects: Harnessing Machine Learning Algorithms and Maturity Domains to Predict Success

Aim/Purpose: While existing literature has extensively explored factors influencing the success of big data projects and proposed big data maturity models, no study has harnessed machine learning to predict project success and identify the critical features contributing significantly to that success. The purpose of this paper is to offer fresh insights into the realm of big data projects by leveraging machine-learning algorithms. Background: Previously, we introduced the Global Big Data Maturity Model (GBDMM), which encompassed various domains inspired by the success factors of big data projects. In this paper, we transformed these maturity domains into a survey and collected feedback from 90 big data experts across the Middle East, Gulf, Africa, and Turkey regions regarding their own projects. This approach aims to gather firsthand insights from practitioners and experts in the field. Methodology: To analyze the feedback obtained from the survey, we applied several algorithms suitable for small datasets and categorical features. Our approach included cross-validation and feature selection techniques to mitigate overfitting and enhance model performance. Notably, the best-performing algorithms in our study were the Decision Tree (achieving an F1 score of 67%) and the Cat Boost classifier (also achieving an F1 score of 67%). Contribution: This research makes a significant contribution to the field of big data projects. By utilizing machine-learning techniques, we predict the success or failure of such projects and identify the key features that significantly contribute to their success. This provides companies with a valuable model for predicting their own big data project outcomes. Findings: Our analysis revealed that the domains of strategy and data have the most influential impact on the success of big data projects. Therefore, companies should prioritize these domains when undertaking such projects. Furthermore, we now have an initial model capable of predicting project success or failure, which can be invaluable for companies. Recommendations for Practitioners: Based on our findings, we recommend that practitioners concentrate on developing robust strategies and prioritize data management to enhance the outcomes of their big data projects. Additionally, practitioners can leverage machine-learning techniques to predict the success rate of these projects. Recommendation for Researchers: For further research in this field, we suggest exploring additional algorithms and techniques and refining existing models to enhance the accuracy and reliability of predicting the success of big data projects. Researchers may also investigate further into the interplay between strategy, data, and the success of such projects. Impact on Society: By improving the success rate of big data projects, our findings enable organizations to create more efficient and impactful data-driven solutions across various sectors. This, in turn, facilitates informed decision-making, effective resource allocation, improved operational efficiency, and overall performance enhancement. Future Research: In the future, gathering additional feedback from a broader range of big data experts will be valuable and help refine the prediction algorithm. Conducting longitudinal studies to analyze the long-term success and outcomes of Big Data projects would be beneficial. Furthermore, exploring the applicability of our model across different regions and industries will provide further insights into the field.

3D imaging of magnetic domains in Nd2Fe14B using scanning hard X-ray nanotomography

Nanoscale structural and electronic heterogeneities are prevalent in condensed matter physics. Investigating these heterogeneities in 3D has become an important task for understanding material properties. To provide a tool to unravel the connection between nanoscale heterogeneity and macroscopic emergent properties in magnetic materials, scanning transmission X-ray microscopy (STXM) is combined with X-ray magnetic circular dichroism. A vector tomography algorithm has been developed to reconstruct the full 3D magnetic vector field without any prior noise assumptions or knowledge about the sample. Two tomographic scans around the vertical axis are acquired on single-crystalline Nd2Fe14B pillars tilted at two different angles, with 2D STXM projections recorded using a focused 120 nm X-ray beam with left and right circular polarization. Image alignment and iterative registration have been implemented based on the 2D STXM projections for the two tilts. Dichroic projections obtained from difference images are used for the tomographic reconstruction to obtain the 3D magnetization distribution at the nanoscale.

Structure of MltG from Mycobacterium abscessus reveals structural plasticity between composed domains

MltG, a membrane-bound lytic transglycosyl­ase, has roles in terminating glycan polymerization in peptidoglycan and incorporating glycan chains into the cell wall, making it significant in bacterial cell-wall biosynthesis and remodeling. This study provides the first reported MltG structure from Mycobacterium abscessus (maMltG), a superbug that has high antibiotic resistance. Our structural and biochemical analyses revealed that MltG has a flexible peptidoglycan-binding domain and exists as a monomer in solution. Further, the putative active site of maMltG was disclosed using structural analysis and sequence comparison. Overall, this study contributes to our understanding of the transglycosyl­ation reaction of the MltG family, aiding the design of next-generation antibiotics targeting M. abscessus.

Characterization of essential domains in HSD17B13 for cellular localization and enzymatic activity [Research Articles]

Human genetic studies recently identified an association of SNPs in the 17-β hydroxysteroid dehydrogenase 13 (HSD17B13) gene with alcoholic and nonalcoholic fatty liver disease development. Mutant HSD17B13 variants devoid of enzymatic function have been demonstrated to be protective from cirrhosis and liver cancer, supporting the development of HSD17B13 as a promising therapeutic target. Previous studies have demonstrated that HSD17B13 is a lipid droplet (LD)-associated protein. However, the critical domains that drive LD targeting or determine the enzymatic activity have yet to be defined. Here we used mutagenesis to generate multiple truncated and point-mutated proteins and were able to demonstrate in vitro that the N-terminal hydrophobic domain, PAT-like domain, and a putative α-helix/β-sheet/α-helix domain in HSD17B13 are all critical for LD targeting. Similarly, we characterized the predicted catalytic, substrate-binding, and homodimer interaction sites and found them to be essential for the enzymatic activity of HSD17B13, in addition to our previous identification of amino acid P260 and cofactor binding site. In conclusion, we identified critical domains and amino acid sites that are essential for the LD localization and protein function of HSD17B13, which may facilitate understanding of its function and targeting of this protein to treat chronic liver diseases.

Structural and kinetic insights into flavin-containing monooxygenase and calponin-homology domains in human MICAL3

MICAL is an oxidoreductase that participates in cytoskeleton reorganization via actin disassembly in the presence of NADPH. Although three MICALs (MICAL1, MICAL2 and MICAL3) have been identified in mammals, only the structure of mouse MICAL1 has been reported. Here, the first crystal structure of human MICAL3, which contains the flavin-containing monooxygenase (FMO) and calponin-homology (CH) domains, is reported. MICAL3 has an FAD/NADP-binding Rossmann-fold domain for mono­oxygenase activity like MICAL1. The FMO and CH domains of both MICAL3 and MICAL1 are highly similar in structure, but superimposition of the two structures shows a different relative position of the CH domain in the asymmetric unit. Based on kinetic analyses, the catalytic efficiency of MICAL3 dramatically increased on adding F-actin only when the CH domain was available. However, this did not occur when two residues, Glu213 and Arg530, were mutated in the FMO and CH domains, respectively. Overall, MICAL3 is structurally highly similar to MICAL1, which suggests that they may adopt the same catalytic mechanism, but the difference in the relative position of the CH domain produces a difference in F-actin substrate specificity.

The PRR14 heterochromatin tether encodes modular domains that mediate and regulate nuclear lamina targeting [RESEARCH ARTICLE]

A large fraction of epigenetically silent heterochromatin is anchored to the nuclear periphery via "tethering proteins" that function to bridge heterochromatin and the nuclear membrane or nuclear lamina. We identified previously a human tethering protein, PRR14, that binds heterochromatin through an N-terminal domain, but the mechanism and regulation of nuclear lamina association remained to be investigated. Here we identify an evolutionarily conserved PRR14 nuclear lamina binding domain (LBD) that is both necessary and sufficient for positioning of PRR14 at the nuclear lamina. We also show that PRR14 associates dynamically with the nuclear lamina, and provide evidence that such dynamics are regulated through phosphorylation-dephosphorylation of the LBD. Furthermore, we identified a PP2A phosphatase recognition motif within the evolutionarily conserved PRR14 C-terminal Tantalus domain. Disruption of this motif affected PRR14 localization to the nuclear lamina. The overall findings demonstrate a heterochromatin anchoring mechanism whereby the PRR14 tether simultaneously binds heterochromatin and the nuclear lamina through two separable, modular domains. The findings also describe an optimal PRR14 LBD fragment that could be used for efficient targeting of fusion proteins to the nuclear lamina.

It is well-known that a celebrated J"{o}rgens-Calabi-Pogorelov theorem for Monge-Amp`ere equations states that any classical (viscosity) convex solution of $det(D^2u)=1$ in $mathbb{R}^n$ must be a quadratic polynomial. Therefore, it is an interesting topic to study the existence and uniqueness theorem of such fully nonlinear partial differential equations' Dirichlet problems on exterior domains with suitable asymptotic conditions at infinity. As a continuation of the works of Caffarelli-Li for Monge-Amp`ere equation and of Bao-Li-Li for $k$-Hessian equations, this paper is devoted to the solvability of the exterior Dirichlet problem of Hessian quotient equations $sigma_k(lambda(D^2u))/sigma_l(lambda(D^2u))=1$ for any $1leq l<kleq n$ in all dimensions $ngeq 2$. By introducing the concept of generalized symmetric subsolutions and then using the Perron's method, we establish the existence theorem for viscosity solutions, with prescribed asymptotic behavior which is close to some quadratic polynomial at infinity.

We first give a sufficient condition, issued from pluripotential theory, for an unbounded domain in the complex Euclidean space $mathbb C^n$ to be Kobayashi hyperbolic. A LiDAR-based real-time capable 3D Perception System for Automated Driving in Urban Domains. (arXiv:2005.03404v1 [cs.RO])

We present a LiDAR-based and real-time capable 3D perception system for automated driving in urban domains. The hierarchical system design is able to model stationary and movable parts of the environment simultaneously and under real-time conditions. Our approach extends the state of the art by innovative in-detail enhancements for perceiving road users and drivable corridors even in case of non-flat ground surfaces and overhanging or protruding elements. We describe a runtime-efficient pointcloud processing pipeline, consisting of adaptive ground surface estimation, 3D clustering and motion classification stages. Based on the pipeline's output, the stationary environment is represented in a multi-feature mapping and fusion approach. Movable elements are represented in an object tracking system capable of using multiple reference points to account for viewpoint changes. We further enhance the tracking system by explicit consideration of occlusion and ambiguity cases. Our system is evaluated using a subset of the TUBS Road User Dataset. We enhance common performance metrics by considering application-driven aspects of real-world traffic scenarios. The perception system shows impressive results and is able to cope with the addressed scenarios while still preserving real-time capability.

The ribosome biogenesis factor Las1 is an essential endoribonuclease that is well-conserved across eukaryotes and a newly established member of the higher eukaryotes and prokaryotes nucleotide-binding (HEPN) domain-containing nuclease family. HEPN nucleases participate in diverse RNA cleavage pathways and share a short HEPN nuclease motif (RφXXXH) important for RNA cleavage. Most HEPN nucleases participate in stress-activated RNA cleavage pathways; Las1 plays a fundamental role in processing pre-rRNA. Underscoring the significance of Las1 function in the cell, mutations in the human LAS1L (LAS1-like) gene have been associated with neurological dysfunction. Two juxtaposed HEPN nuclease motifs create Las1's composite nuclease active site, but the roles of the individual HEPN motif residues are poorly defined. Here using a combination of in vivo experiments in Saccharomyces cerevisiae and in vitro assays, we show that both HEPN nuclease motifs are required for Las1 nuclease activity and fidelity. Through in-depth sequence analysis and systematic mutagenesis, we determined the consensus HEPN motif in the Las1 subfamily and uncovered its canonical and specialized elements. Using reconstituted Las1 HEPN-HEPN' chimeras, we defined the molecular requirements for RNA cleavage. Intriguingly, both copies of the Las1 HEPN motif were important for nuclease function, revealing that both HEPN motifs participate in coordinating the RNA within the Las1 active site. We also established that conformational flexibility of the two HEPN domains is important for proper nuclease function. The results of our work reveal critical information about how dual HEPN domains come together to drive Las1-mediated RNA cleavage.

Nanodomains can persist at physiologic temperature in plasma membrane vesicles and be modulated by altering cell lipids [Research Articles]

The formation and properties of liquid-ordered (Lo) lipid domains (rafts) in the plasma membrane are still poorly understood. This limits our ability to manipulate ordered lipid domain-dependent biological functions. Giant plasma membrane vesicles (GPMVs) undergo large-scale phase separations into coexisting Lo and liquid-disordered lipid domains. However, large-scale phase separation in GPMVs detected by light microscopy is observed only at low temperatures. Comparing Förster resonance energy transfer-detected versus light microscopy-detected domain formation, we found that nanodomains, domains of nanometer size, persist at temperatures up to 20°C higher than large-scale phases, up to physiologic temperature. The persistence of nanodomains at higher temperatures is consistent with previously reported theoretical calculations. To investigate the sensitivity of nanodomains to lipid composition, GPMVs were prepared from mammalian cells in which sterol, phospholipid, or sphingolipid composition in the plasma membrane outer leaflet had been altered by cyclodextrin-catalyzed lipid exchange. Lipid substitutions that stabilize or destabilize ordered domain formation in artificial lipid vesicles had a similar effect on the thermal stability of nanodomains and large-scale phase separation in GPMVs, with nanodomains persisting at higher temperatures than large-scale phases for a wide range of lipid compositions. This indicates that it is likely that plasma membrane nanodomains can form under physiologic conditions more readily than large-scale phase separation. We also conclude that membrane lipid substitutions carried out in intact cells are able to modulate the propensity of plasma membranes to form ordered domains. This implies lipid substitutions can be used to alter biological processes dependent upon ordered domains.

Here using a combination of in vivo experiments in Saccharomyces cerevisiae and in vitro assays, we show that both HEPN nuclease motifs are required for Las1 nuclease activity and fidelity. Through in-depth sequence analysis and systematic mutagenesis, we determined the consensus HEPN motif in the Las1 subfamily and uncovered its canonical and specialized elements. Using reconstituted Las1 HEPN-HEPN' chimeras, we defined the molecular requirements for RNA cleavage. Intriguingly, both copies of the Las1 HEPN motif were important for nuclease function, revealing that both HEPN motifs participate in coordinating the RNA within the Las1 active site. We also established that conformational flexibility of the two HEPN domains is important for proper nuclease function. The results of our work reveal critical information about how dual HEPN domains come together to drive Las1-mediated RNA cleavage.

The ribosome biogenesis factor Las1 is an essential endoribonuclease that is well-conserved across eukaryotes and a newly established member of the higher eukaryotes and prokaryotes nucleotide-binding (HEPN) domain-containing nuclease family. HEPN nucleases participate in diverse RNA cleavage pathways and share a short HEPN nuclease motif (RφXXXH) important for RNA cleavage. Most HEPN nucleases participate in stress-activated RNA cleavage pathways; Las1 plays a fundamental role in processing pre-rRNA. Underscoring the significance of Las1 function in the cell, mutations in the human LAS1L (LAS1-like) gene have been associated with neurological dysfunction. Two juxtaposed HEPN nuclease motifs create Las1's composite nuclease active site, but the roles of the individual HEPN motif residues are poorly defined. Nanodomains can persist at physiologic temperature in plasma membrane vesicles and be modulated by altering cell lipids [Research Articles]

The formation and properties of liquid-ordered (Lo) lipid domains (rafts) in the plasma membrane are still poorly understood. This limits our ability to manipulate ordered lipid domain-dependent biological functions. Giant plasma membrane vesicles (GPMVs) undergo large-scale phase separations into coexisting Lo and liquid-disordered lipid domains. However, large-scale phase separation in GPMVs detected by light microscopy is observed only at low temperatures. Comparing Förster resonance energy transfer-detected versus light microscopy-detected domain formation, we found that nanodomains, domains of nanometer size, persist at temperatures up to 20°C higher than large-scale phases, up to physiologic temperature. The persistence of nanodomains at higher temperatures is consistent with previously reported theoretical calculations. To investigate the sensitivity of nanodomains to lipid composition, GPMVs were prepared from mammalian cells in which sterol, phospholipid, or sphingolipid composition in the plasma membrane outer leaflet had been altered by cyclodextrin-catalyzed lipid exchange. Lipid substitutions that stabilize or destabilize ordered domain formation in artificial lipid vesicles had a similar effect on the thermal stability of nanodomains and large-scale phase separation in GPMVs, with nanodomains persisting at higher temperatures than large-scale phases for a wide range of lipid compositions. This indicates that it is likely that plasma membrane nanodomains can form under physiologic conditions more readily than large-scale phase separation. We also conclude that membrane lipid substitutions carried out in intact cells are able to modulate the propensity of plasma membranes to form ordered domains. This implies lipid substitutions can be used to alter biological processes dependent upon ordered domains.
