A measure of review consistency is introduced to determine the degree to which the review sentiment of textual content conforms with the review rating score. A theoretical model grounded in signaling theory is adopted to explore how different variables (review sentiment, review rating, review length, and review rating variance) affect review consistency and the relationship between review consistency and review helpfulness. Background: Online reviews vary in their characteristics and hence their different quality features and degrees of helpfulness. High-quality online reviews offer consumers the ability to make informed purchase decisions and improve trust in e-commerce websites. The helpfulness of online reviews continues to be a focal research issue regardless of the independent or joint effects of different factors. This research posits that the consistency between review content and review rating is an important quality indicator affecting the helpfulness of online reviews. The review consistency of online reviews is another important requirement for maintaining the significance and perceived value of online reviews. Incidentally, this parameter is inadequately discussed in the literature. A possible reason is that review consistency is not a review feature that can be readily monitored on e-commerce websites. Methodology: More than 100,000 product reviews were collected from Amazon.com and preprocessed using natural language processing tools. Then, the quality reviews were identified, and relevant features were extracted for model training. Machine learning and sentiment analysis techniques were implemented, and each review was assigned a consistency score between 0 (not consistent) and 1 (fully consistent). Finally, signaling theory was employed, and the derived data were analyzed to determine the effect of review consistency on review helpfulness, the effect of several factors on review consistency, and their relationship with review helpfulness. Contribution: This research contributes to the literature by introducing a mathematical measure to determine the consistency between the textual content of online reviews and their associated ratings. Furthermore, a theoretical model grounded in signaling theory was developed to investigate the effect on review helpfulness. This work can considerably extend the body of knowledge on the helpfulness of online reviews, with notable implications for research and practice. Findings: Empirical results have shown that review consistency significantly affects the perceived helpfulness of online reviews. The study similarly finds that review rating is an important factor affecting review consistency; it also confirms a moderating effect of review sentiment, review rating, review length, and review rating variance on the relationship between review consistency and review helpfulness. Overall, the findings reveal the following: (1) online reviews with textual content that correctly explains the associated rating tend to be more helpful; (2) reviews with extreme ratings are more likely to be consistent with their textual content; and (3) comparatively, review consistency more strongly affects the helpfulness of reviews with short textual content, positive polarity textual content, and lower rating scores and variance. Recommendations for Practitioners: E-commerce systems should incorporate a review consistency measure to rank consumer reviews and provide customers with quick and accurate access to the most helpful reviews. Impact on Society: Incorporating a score of review consistency for online reviews can help consumers access the best reviews and make better purchase decisions, and e-commerce systems improve their business, ultimately leading to more effective e-commerce. Future Research: Additional research should be conducted to test the impact of review consistency on helpfulness in different datasets, product types, and different moderating variables. Full Article

We aim to develop a noninvasive prediction model for meningioma consistency utilizing MR elastography and DTI. MATERIALS AND METHODS: Ninety-four patients (52 ± 22 years old, 69 women, 25 men) diagnosed with meningioma were recruited in the study. Each patient underwent preoperative T1WI, T2WI, DTI, and MR elastography. Combined MR elastography–DTI model was developed based on multiple logistic regression. Intraoperative tumor descriptions served as clinical criteria for evaluating meningioma consistency. The diagnostic efficacy in determining meningioma consistency was evaluated by using a receiver operating characteristic curve. Further validation was conducted in 27 stereotactic biopsies by using indentation tests and underlying mechanism was investigated by histologic analysis. RESULTS: Among all the imaging modalities, MR elastography demonstrated the highest efficacy with the shear modulus magnitude (|G*|) achieving an area under the curve (AUC) of 0.81 (95% CI: 0.699–0.929). When combined with DTI, the diagnostic accuracy further increased (AUC: 0.88, 95% CI: 0.784–0.971), surpassing any technique alone. Indentation measurement based on stereotactic biopsies further demonstrated that the MR elastography–DTI model was suitable for predicting intratumor consistency. Histologic analysis suggested that meningioma consistency may be correlated with tumor cell density and fibrous content. CONCLUSIONS: The MR elastography–DTI combined model is effective in noninvasive prediction of meningioma consistency. Full Article

To better understand the formation of cocrystals on a molecular level, the effects of disposition of functional groups on the formation of cocrystals were systematically and comprehensively investigated using cresol isomers (o-, m-, p-cresol) as model compounds. Consistency and variability in these cocrystals containing positional isomers were found and analyzed. The structures, molecular recognition and self-assembly mechanism of supramolecular synthons in solution and in their corresponding cocrystals were verified by a combined experimental and theoretical calculation approach. It was found that the heterosynthons (heterotrimer or heterodimer) combined with O—H⋯N hydrogen bonding played a significant role. Hirshfeld surface analysis and computed interaction energy values were used to determine the hierarchical ordering of the weak interactions. The quantitative analyses of charge transfers and molecular electrostatic potential were also applied to reveal and verify the reasons for consistency and variability. Finally, the molecular recognition, self-assembly and evolution process of the supramolecular synthons in solution were investigated. The results confirm that the supramolecular synthon structures formed initially in solution would be carried over to the final cocrystals, and the supramolecular synthon structures are the precursors of cocrystals and the information memory of the cocrystallization process, which is evidence for classical nucleation theory. We also derive the posterior convergence rates for precision matrices and Cholesky factors with respect to various matrix norms. The obtained posterior convergence rates are the fastest among those of the existing Bayesian approaches. In particular, we prove that our posterior convergence rates for Cholesky factors are the minimax or at least nearly minimax depending on the relative size of true sparseness for the entire dimension. Through simulation studies, we demonstrate that our model selection procedure can outperform other Bayesian methods which treat the scale parameter as given, and commonly used penalized likelihood methods, in a range of simulation settings. New RNA families are often detected and analyzed using comparative methods based on multiple sequence alignments. Accordingly, a number of programs have been developed for aligning and deriving secondary structures from sets of RNA sequences. Yet, the best tools for these tasks remain unclear because existing benchmarks contain too few sequences belonging to only a small number of RNA families. RNAconTest (RNA consistency test) is a new benchmarking approach relying on the observation that secondary structure is often conserved across highly divergent RNA sequences from the same family. RNAconTest scores multiple sequence alignments based on the level of consistency among known secondary structures belonging to reference sequences in their output alignment. Similarly, consensus secondary structure predictions are scored according to their agreement with one or more known structures in a family. Comparing the performance of 10 popular alignment programs using RNAconTest revealed that DAFS, DECIPHER, LocARNA, and MAFFT created the most structurally consistent alignments. The best consensus secondary structure predictions were generated by DAFS and LocARNA (via RNAalifold). Many of the methods specific to noncoding RNAs exhibited poor scalability as the number or length of input sequences increased, and several programs displayed substantial declines in score as more sequences were aligned. Overall, RNAconTest provides a means of testing and improving tools for comparative RNA analysis, as well as highlighting the best available approaches. Regulatory agencies and the biotechnology industry consider cell line "clonal origin" an important aspect of maintaining process control. Demonstration of clonal origin of the cell substrate, or production cell line, has received considerable attention in the past few years, and the industry has improved methods and devised standards to increase the probability and/or assurance of clonal derivation. However, older production cell lines developed before the implementation of these methods, herein referred to as "legacy cell lines," may not meet current regulatory expectations for demonstration of clonal derivation. In this article, the members of the IQ Consortium Working Group on Clonality present our position that the demonstration of process consistency and product comparability of critical quality attributes throughout the development life cycle should be sufficient to approve a license application without additional genetic analysis to support clonal origin, even for legacy cell lines that may not meet current day clonal derivation standards. With this commentary, we discuss advantages and limitations of genetic testing methods to support clonal derivation of legacy cell lines and wish to promote a mutual understanding with the regulatory authorities regarding their optional use during early drug development, subsequent to Investigational New Drug (IND) application and before demonstration of product and process consistency at Biologics License Applications (BLA) submission.
