We have addressed the question whether 5' and 3' ends meet by diffusion-controlled encounter "through solution" or by a mechanism involving the RNA backbone. For this purpose, we used a translation system derived from Drosophila embryos that displays two types of 5'–3' interactions: Cap-dependent translation initiation is stimulated by the poly(A) tail and inhibited by Smaug recognition elements (SREs) in the 3' UTR. Chimeric RNAs were made consisting of one RNA molecule carrying a luciferase coding sequence and a second molecule containing SREs and a poly(A) tail; the two were connected via a protein linker. The poly(A) tail stimulated translation of such chimeras even when disruption of the RNA backbone was combined with an inversion of the 5'–3' polarity between the open reading frame and poly(A) segment. Stimulation by the poly(A) tail also decreased with increasing RNA length. Both observations suggest that contacts between the poly(A) tail and the 5' end are established through solution, independently of the RNA backbone. In the same chimeric constructs, SRE-dependent inhibition of translation was also insensitive to disruption of the RNA backbone. Thus, tracking of the backbone is not involved in the repression of cap-dependent initiation. However, SRE-dependent repression was insensitive to mRNA length, suggesting that the contact between the SREs in the 3' UTR and the 5' end of the RNA might be established in a manner that differs from the contact between the poly(A) tail and the cap. The actor believes the video—titled #LockdownOnDomesticViolence and featuring a string of celebrities, including Karan Johar, Sachin Tendulkar, Madhuri Dixit Nene, Virat Kohli and Anushka Sharma—was the need of the hour as cases of domestic violence have witnessed a spike amid the lockdown. "The video had to be in three languages—Marathi, because the majority of our state understands the language, and Hindi, for the migrant population in the state. The English campaign has been noticed by people the world over," says Bose. The state government has launched several numbers, including the uniform state helpline number 100, where women can register their complaints. One can probably determine the endeavour's success by studying if there has been a drop in the cases reported since the video released. However, he disagrees, "We have to consider the other possibility that domestic violence is continuing, and yet, women can't report it." Ever since the release of Kabir Singh in 2019, and consequently, Thappad earlier this year, there has been a larger discussion about representation of casual violence on screen. The actor, however, believes movies have minimal effect on social behaviour. "There's little co-relation between the two factors. Films don't need to carry a positive message, but they shouldn't carry a negative one, that does disservice. A ground-up campaign, in which the same message is passed on consistently, film on film, year on year, can change behaviour more than a movie can." To make his case, he cites how the state of California launched a multi-media campaign against tobacco consumption, leading to a 12 per cent drop in sales after 20 years. "So, looking at the misogyny and status of women in India, we need a ground campaign over decades to attain gender justice. For him, it is important that the story should have the essence of humanism and not the glorification of hatred. "Honestly speaking, my rules of saying yes to a project do not change. For me, the story matters and not the length of my character. I think by now people know that. I am not the person running after fame or concern about how many people are watching my work. For me, the story has to be important enough to engage, as an actor and as an audience," Rahul told IANS. The actor has worked in the Netflix original "Bulbul", besides the web series "Bombay Begums" directed by Alankrita Srivastava. You see, the story has to have some amount of humanism. You can show a character as the biggest evil (person) of the world and still can come up with the right message -- not one that is destructive or negative. A message cannot glorify hatred, and if it does, I do not want to be part of that. Of course, my character can be the villain but not the overall message of the film," Rahul said. He explained: "In the recent past I am offered a film that shows a very, very narrow view of nationalism and it was quite a jingoistic story. I can never be part of that. I am the same actor who was part of a film like "Shaurya" that sent out an absolutely right message -- that is, no matter which religion, caste or region you are from, when you are a man in a uniform, all you are upholding is the security of your country." Amid the nationwide lockdown, a long queue could be seen every morning at Subhasgram comprising of local rickshaw pullers, daily wage earners or small time hawkers who come to collect their daily ration. Bose, a member of the 'Blue Tigers' would be at the other end, happily distributing packets containing rice, pulses, potatoes, onions and other staple food items. This is his way of paying back to the society. "The rickshaw pullers who have so many times offered me free rides to-and-fro to local matches, that local vendor and shopkeeper who had gifted me so many free packets of food after heart-warming performances -- I felt I needed to pay them back," the India left-back told PTI of his daily regime since Friday. "It was such a satisfaction to hand over some food items to lot many known faces whom I have known in my locality over my upbringing." India had imposed a nationwide lockdown since March 24 to curb the spread of COVID-19 pandemic and the worst-hit were people from the lower strata of the society whose life revolve around their daily wages. One fine morning, the 24-year-old felt restless and was awakened by the Swami Vivekananda's soul-stirring words 'Arise, Awake, And Stop Not Till the Goal is Reached!' "It kept on reverberating between my ears. My conscience did not allow me to shy away -- rather it kept on pushing me to give it back to the society where I have grown up." "Somewhere down the line, however, I felt restless. It's understandable that even as we need to Stay Home Stay Safe, there have been so many brave hearts who have cared less for their lives, and are out to serve the country in this troubled time -- the doctors, nurses, cleaners, police, journalists, etc. It's not only the frontline workers, who are put their lives on line but citizens need to do their bit, reckons Bose. "But are they enough? Can we all just sit back on our cosy rooms, and expect the privilege of others keeping us safe? Don't we need more volunteers? Till then stay home, and stay safe," he added. The Mumbai City FC defender was looking forward to the Indian camp when the pandemic halted all sporting activities. "A longer camp for the national team was planned and we were forward to it. But under current circumstances all sporting action around the world has been postponed." Asked whether he's missing the game, he replied: "Human lives are of utmost importance, and like everyone around us, I look forward for normalcy to return soon." The defender however regrets that he's not able to go out for runs so he's making the best use of the time at his private gym. "I cannot go out for my runs. So I am trying to keep myself as much fit as possible indoors. I try to sweat out at least two hours a day in my personal gym so that I stay in proper shape when the season resumes.
